The objective of this work was the transformation of tobacco and 'Valencia' sweet orange with the GUS gene driven by the citrus phenylalanine ammonia-lyase (PAL) gene promoter (CsPP). Transformation was accomplished by co-cultivation of tobacco and 'Valência' sweet orange explants with Agrobacterium tumefaciens containing the binary vector CsPP-GUS/2201. After plant transformation and regeneration, histochemical analyses using GUS staining revealed that CsPP promoter preferentially, but not exclusively, conferred gene expression in xylem tissues of tobacco. Weaker GUS staining was also detected throughout the petiole region in tobacco and citrus CsPP transgenic plants.

ABSTRACT This study aimed to evaluate the effect of factors that may affect the genetic transformation of cotiledonary explants of Eucalyptus saligna mediated by EHA105 strain of Agrobacterium tumefaciens. The vector pBI121 carrying gus gene under control of 35S CaMV promoter was used. The effect of the following factors was evaluated: explant pre-culture, use of different antibiotics and presence of acetosyringone (AS) in co-culture media. An antioxidant solution was also used during excision, containing ascorbic acid (250mg.L-1), citric acid (25mg.L-1) and PVP-40 (1g.L-1). Pre-culture of the explants before the co-culture with bacteria was done over a 4-day period in MS culture medium supplemented with 4.4µM BAP and 2.7ìM NAA. After theco-culture period, three concentrations of kanamycin (12.5;25 and 50mg.L-1) combined with 300mg.L-1 Augmentin® in the culture medium were tested The influence of the antibiotic was also evaluated by keeping the explants in a medium containing 50mg.L-1 Km and 300mg.L-1 Augmentin® or 500mg.L-1 cefotaxime. It was concluded that Augmentin® stimulates organogenesis, that a Km concentration of 12.5mg.L-1 allows selection of explants transformed with gus gene and, finally, the addition of AS (50ìM) to the liquid and solid co-culture media has a positive effect on gus gene expression. Moreover, the use of an antioxidant solution during cotyledon excision is dispensable and the pre-culture of the explants has no effect on bud regeneration or gus gene expression. A transformation efficiency of 1.5% was reached.

The expression of GUS and ipt genes under control of a chalcone synthase (chs) promoter (PCHS) has been determined in tobacco (Nicotiana tabacum L.) plants and related to the development of plants expressing the chimaeric PCHS -ipt gene. GUS gene expression, which served as a model for the expression of the ipt gene, was highest in the internal phloem tissue of stems, in mature leaf laminae and in the upper part of corollas when fully open. Expression of the PCHS -ipt gene was assessed by quantifying the cytokinins produced, by determining incorporation of [3H]adenine into cytokinins and by quantifying ipt mRNA. Results from these studies were in general agreement with those based on expression of the PCHS -GUS gene. The chs promoter controlled expression of the ipt gene with some degree of tissue and temporal specificity. Expression of the ipt gene markedly elevated the cytokinin level in mature leaf laminae and the upper stems of flowering plants. The former was associated with retardation of leaf senescence and increased rates of transpiration due to changes in number, size and aperture of stomata, while the latter was associated with development of lateral shoots. In shoot tip cultures, 2-fold elevations in endogenous cytokinin level caused clear changes in development and this is discussed in relation to current concepts concerning the hormonal control of plant development. Using the transgenic tobacco tissues, it was shown that cis-zeatin is a substrate for cytokinin oxidase, that cis-zeatin is not converted to trans-zeatin in these tissues and that the endogenous cytokinin level influences the level of cytokinin oxidase activity in tissue and the rate of degradation of exogenous zeatin riboside to adenosine.
