Is inquiry possible in light of accountability?: A quantitative comparison of the relative effectiveness of guided inquiry and verification laboratory instruction

ABSTRAK Tidak adanya buku petunjuk praktikum yang jelas dapat menyulitkan siswa dalam pelaksanaan praktikum. Penelitian ini bertujuan untuk mengembangkan petunjuk praktikum biologi berbasis inkuiri terbimbing pada materi sistem pencernaan kelas XI SMA Negeri 5 Pontianak yang dapat digunakan sebagai bahan ajar praktikum. Model pengembangan yang digunakan dalam penelitian ini adalah model pengembangan 4-D yang dimodifikasi meliputi tahap pendefiinisian (Define), perancangan (Design), dan pengembangan (Develop). Berdasarkan penilaian para ahli, nilai kevalidan hasil validasi ahli materi sebesar 3,87 artinya memenuhi kriteria kevalidan, ahli tampilan sebesar 4 memenuhi ktiteria kevalidan, dan ahli bahasa sebesar 3,6 kriteria valid sehingga petunjuk praktikum layak untuk digunakan. Nilai kepraktisan berdasarkan penilaian angket respon yang diberikan kepada siswa sebesar 82,291% pada uji coba awal dan pada uji coba utama respon yang diberikan oleh siswa sebesar 85,18% sehingga memberikan respon positif terhadap penggunaan petunjuk praktikum yang telah direvisi pada kegiatan praktikum biologi materi sistem pencernaan. Nilai keefektifan berdasarkan perbedaan nilai pretest dan posttest pada saat uji coba utama, 0,61 dengan kategori sedang.Kata Kunci: Inkuiri Terbimbing, Petunjuk Praktikum, Sistem Pencernaan ABSTRACTThe unavalability of the standardized lab instruction leads to the students’ problem in conducting lab activities. This study aimed at developing the guided- inquiry laboratory instruction in ‘Digestive System’ class at XI grade class of SMA Negeri 5 Pontianak. The development model used in this study was 4D model (Define, Design, Develop). The results of the validation experts (content, language, appereance) showed that the material /the content score was 3,87 which means that the product was valid, the material appereance was 4 (valid), and the language use was 3,6 (valid). Therefore, the lab instruction was considered proper to be used. The result of the questionnaire revealed that the respondents gave positive response to the validated product. It can be seen from the practicality score at the first (82,291%) and the second try out (85,18%). Similarly,the effectiveness score based on the main try out of pre and post tests was 0,61 (average category).Keywords: guided inquiry, lab instruction/manual, digestive system

An understanding of the specific interactions of actin with regulatory proteins has been limited by the lack of information about the structure of the actin filament. Molecular actin has been studied in actin-DNase I complexes by single crystal X-ray analysis, to a resolution of about 0.6nm, and in the electron microscope where two dimensional actin sheets have been reconstructed to a maximum resolution of 1.5nm. While these studies have shown something of the structure of individual actin molecules, essential information about the orientation of actin in the filament is still unavailable.The work of Egelman & DeRosier has, however, suggested a method which could be used to provide an initial quantitative estimate of the orientation of actin within the filament. This method involves the quantitative comparison of computed diffraction data from single actin filaments with diffraction data derived from synthetic filaments constructed using the molecular model of actin as a building block. Their preliminary work was conducted using a model consisting of two juxtaposed spheres of equal size.

For many years the concept of quantitative diffraction contrast experiments might have consisted of the determination of dislocation Burgers vectors using a g.b = 0 criterion from several different 2-beam images. Since the advent of the personal computer revolution, the available computing power for performing image-processing and image-simulation calculations is enormous and ubiquitous. Several programs now exist to perform simulations of diffraction contrast images using various approximations. The most common approximations are the use of only 2-beams or a single systematic row to calculate the image contrast, or calculating the image using a column approximation. The increasing amount of literature showing comparisons of experimental and simulated images shows that it is possible to obtain very close agreement between the two images; although the choice of parameters used, and the assumptions made, in performing the calculation must be properly dealt with. The simulation of the images of defects in materials has, in many cases, therefore become a tractable problem.
