Our previous study found saikosaponin b2 (SSb2) increased high-density lipoprotein (HDL) uptake in HepG2 cells. SSb2 is only found in aqueous Bupleuri Radix extract, and it is one of the secondary saponins derived from saikosaponin d (SSd), which exists in the methanolic extract. This study aimed to compare the effect of aqueous extract of Bupleuri Radix on hepatic uptake of HDL with methanolic extract and to reveal the underlying mechanism of enhancing HDL uptake in mice fed with high-fat diet (HFD). Cellular HDL uptake in each group was quantified by flow cytometry. Bioactive components bound to the HepG2 cytomembrane were detected with HPLC-DAD. RNA sequencing was performed to screen the underlying target on hepatic HDL-uptake, and western blotting was conducted to verify differential protein expression. Significant increases of HDL uptake by HepG2 cells were observed in all groups of aqueous extract of Bupleuri Radix, while no effect or negative effect was observed in the methanolic extract. Saikosaponin b1 (SSb1) and SSb2 were detected in the desorption elute of the aqueous extract from the HepG2 cytomembrane, while saikosaponin a (SSa) and SSd were not found. Remarkable upregulation of FGF21 in HFD-fed mice liver was affirmed after treatment with aqueous extract. This study suggested that aqueous Bupleuri Radix extract could promote hepatic HDL uptake in vitro but methanolic extract could not, and FGF21 might be the potential target.

Recent studies have reported that dietary fiber improved metabolic syndrome (MetS). However, the effects of fucoidans on MetS were still not clear. In this study, we evaluated the activity of fucoidan from Fucus vesiculosus (FvF) on attenuating MetS and first elucidated the underlying mechanism. In vitro, FvF treatment remarkably lowered the level of reactive oxygen species (ROS) compared with the sodium palmitate (PA)-induced insulin resistance (IR) group. The phosphorylation level of c-Jun N-terminal kinase (JNK) was significantly decreased, while phosphorylation of protein kinase B (pAkt) level increased, compared with that of the HepG2 cells treated with PA. Thus, FvF increased glucose consumption and relieved IR via ROS-mediated JNK and Akt signaling pathways. In addition, these changes were accompanied by the activation of adenosine 5′-monophosphate-ativated protein kinase (AMPK) and its downstream targets (e.g., HMG-CoA reductase (HMGCR), acetyl-CoA carboxylase (ACC), and sterol-regulatory element-binding protein-1c (SREBP-1C)), which improved lipid metabolism in IR HepG2 cells. In vivo, FvF improved hyperglycemia and decreased serum insulin level in mice with MetS. Furthermore, we evaluated the inhibition of glucose transport by in vitro (Caco-2 monolayer model), semi-in vivo (everted gut sac model) and oral glucose tolerance test (OGTT), which indicated that FvF could significantly reduce the absorption of glucose into the blood stream, thus it could improve blood-glucose levels and IR in mice with MetS. Moreover, FvF decreased serum triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) levels and liver lipid accumulation, while increased the serum high density lipoprotein cholesterol (HDL-C) level in mice with MetS. Therefore, FvF could be considered as a potential candidate for the treatment of MetS by alleviating IR, inhibiting glucose transportation, and regulating lipid metabolism.

Introdution: Hepatocellular carcinoma (HCC) is one of the most common cancers. It is the third leading cause of cancer related death. This study aimed to evaluate the phytochemical composition of ethanolic and aqueous Glycyrrhiza glabra root extracts its apoptotic effects against human hepatocarcinoma HepG2 cell line. Methods: In the current experimental study, the phyochemical composition of aqueous and ethanolic Glycyrrhiza glabra extract was evaluated by GC/MS method. The HepG2 cells were grown in RPMI1640 supplemented with 10 % (v/v) FBS at 37°C with 5 %carbon dioxide (CO2) atmosphere. The cytotoxicity activities of extracts were measured using MTT after 24 hours. To evaluate the ability of extracts to trigger apoptosis in HepG2 cells, the Hosest8778 staining method was carried out. Results: The phytochemical data of aqueous extract revealed 18 compounds, including Dodecane (13.34%) and 2',6'-Dihydroxyacetophenone bis (trimethylsilyl) ether (12.48%) as the major constitutes. The chemical composition of ethanolic extract showed 17 compounds, including Pentadecane (7.78%), 2,6,11,15-Tetramethylhexadecane (4.67%) as the prominent compounds. The MTT results revealed that the extract-treated cells had a dose-related toxicity effects. The results of Hoechst 33258 staining showed that the nuclear DNA fragmentation was increased in the ethanolic extract treatment compared to aqueous extract. Conclusion: The ethanolic extract of Glycyrrhiza glabra possesses more apoptotic effect on cancer cells and further researches are needed for investigating its potential mechanism.
