AbstractAnalysis of growth and death kinetics at single-cell resolution is a key step in understanding the complexity of the nonreplicating growth phenotype of the bacterial pathogen Mycobacterium tuberculosis. Here, we developed a single-cell-resolution microfluidic mycobacterial culture device that allows time-lapse microscopy-based long-term phenotypic visualization of the live replication dynamics of mycobacteria. This technology was successfully applied to monitor the real-time growth dynamics of the fast-growing model strain Mycobacterium smegmatis (M. smegmatis) while subjected to drug treatment regimens during continuous culture for 48 h inside the microfluidic device. A clear morphological change leading to significant swelling at the poles of the bacterial membrane was observed during drug treatment. In addition, a small subpopulation of cells surviving treatment by frontline antibiotics was observed to recover and achieve robust replicative growth once regular culture media was provided, suggesting the possibility of identifying and isolating nonreplicative mycobacteria. This device is a simple, easy-to-use, and low-cost solution for studying the single-cell phenotype and growth dynamics of mycobacteria, especially during drug treatment.

Amebiasis remains a major health problem in Mexico. Therefore, the search for better culture media and low-cost diagnostic and therapeutic tools is fundamental. We present a new culture medium forEntamoeba histolyticawhich allows the microbe to preserve its virulence factors and ability to induce hepatic abscesses in animal models. The novel CLUPS medium is an improved version of the PEHPS medium, previously designed in our laboratory. The main difference is the substitution of raw beef liver in PEHPS by raw beef lung in the CLUPS medium. To compare the performance of three-culture media (traditional TYI-S-33, PEHPS, and CLUPS),E. histolyticatrophozoites were cultured in quintuplicate, followed by the evaluation of phospholipase activity and the induction of liver abscesses in golden hamsters.E. histolyticatrophozoites grew significantly better in CLUPS medium than in TYI-S-33. Likewise, CLUPS-cultured trophozoites produced significantly more phospholipases than TYI-S-33-cultured trophozoites. Finally, trophozoites grown in any of the three tested media had similar potential to induce liver abscesses.

Genetic engineering was used to produce an elastin-like polymer (ELP) with precise amino acid composition, sequence and length, resulting in the absolute control of MW and stereochemistry. A synthetic monomer DNA sequence encoding for (VPAVG)20, was used to build a library of concatemer genes with precise control on sequence and size. The higher molecular weight polymer with 220 repeats of VPAVG was biologically produced in Escherichia coli and purified by hot and cold centrifugation cycles, based on the reversible inverse temperature transition property of ELPs. The use of low cost carbon sources like lactose and glycerol for bacteria cells culture media was explored using Central Composite Design approach allowing optimization of fermentation conditions. Due to its self-assembling behaviour near 33 °C stable spherical microparticles with a size ~ 1µm were obtained, redissolving when a strong undercooling is achieved. The polymer produced showed hysteresis behaviour with thermal absorbing/releasing components depending on the salt concentration of the polymer solution.

Abstract Growth and biological conditions of Messastrum gracile were evaluated to compare the effect of photoautotrophic and mixotrophic cultivation on the increase of biomass production and chemical conditions cultured in macrophyte and commercial culture media. The growth rate (k) of M. gracile was different in the culture media, higher in mixotrophic cultivation for Lemna minor culture medium, whilst to Eichhornia crassipes and NPK culture media were higher in photoautotrophic cultivation. Mean lipid contents in photoautotrophic cultivation were 8.2% biomass dry weight, whereas they reached 19% biomass dry weight in mixotrophic cultivation. Protein contents were below 48% biomass dry weight in photoautotrophic cultivation and 30% biomass dry weight in mixotrophic cultivation. Messastrum gracile cultured in macrophyte culture media (E. crassipes and L. minor) and NPK culture medium provided satisfactory results with regard to lipid and protein contents in mixotrophic and photoautotrophic cultivations, respectively. Lipid and protein contents in alternative media were higher or similar to the CHU12 commercial culture medium.

Background and Aims: Orchids in Mexico are mainly threatened by deforestation, changes in land use, illegal trade, deficiencies in environmental policy and legislation, and a lack of community participation in the conservation of their forests. Erycina hyalinobulbon is an endemic twig epiphyte orchid with a short life cycle and with large flowers in relation to its size, for which it has been harvested from its wild populations. The objectives of this work were to evaluate the in vitro development of E. hyalinobulbon in culture media with organic supplements, to compare sucrose vs. N’Joy Stevia® as a carbon source for its initial stages of development, and to evaluate the development of its seedlings in media enriched with plant growth regulators (PGR).Methods: For the sowing of seeds, PhytamaxTM and MS medium at 30% of its basal salts were used in combination with organic supplement (coconut milk, pineapple puree and banana puree), along with the Phy medium used as control. In order to measure the effect of sucrose vs. N´Joy Stevia® as a carbon source, these two treatments were used, with the PhytamaxTM medium. To evaluate the development of seedlings with PGR, three treatments were tested: the 100% PhytamaxTM control, 30% PhytamaxTM with 1.166 ml/l of Maxi-grow and the medium Chiu.Key results: PhytamaxTM medium added with banana promoted germination by 9.3%, being a low cost and easy production option. Ninety days after sowing, N´Joy Stevia® as a carbon source promoted germination by 8%. The best development of the seedlings was registered in the medium PhytamaxTM without PGR.Conclusions: With this study, it was possible to develop an accessible in vitro propagation system for E. hyalinobulbon, in order to sustainably manage it and favor its conservation.

A biomassa de microalgas tem apresentado potencial para produção de biodiesel, contudo a viabilidade do cultivo de microalgas depende de fonte de nutrientes de baixo custo. O presente estudo objetivou utilizar o chorume como fonte de nutrientes para microalgas. Os experimentos foram conduzidos visando avaliar a obtenção da biomassa microalgal, conversão de lipídios e rendimento em ésteres metílicos de ácidos graxos, para os meios de cultivos utilizando 5%, 12% e 20% de chorume, com concentrações de 0,02, 0,05 e 0,08 g N. L-1 e para meio de controle contendo 1% de, Nitrogênio (N), Fósforo (P) e Potássio (K), na concentração de 20 g L-1, 5 g L-1 e 20 g L-1, respectivamente. A microalga utilizada neste trabalho foi a de classe Chlorophyceae e família Coccomyxaceae. Os resultados demonstraram que o meio com concentração de 12% de chorume obteve melhores resultados, possibilitando alcançar 1,19 g de biomassa, conversão de 108,15 mg g-1 de lipídios e conteúdo de ésteres de 410,77mg g-1, a microalga utilizada apresentou ainda predominância dos ácidos graxos palmítico e oleico, apresentando baixa quantidade de ácidos graxos saturados o que pode fornecer ao combustível, resistência ao frio. E tais aspectos demonstraram que o chorume pode ser uma fonte promissora de nutrientes para o cultivo das microalgas estudadas. AbstractMicroalgae biomass has presented potential for biodiesel production, however the viability of microalgae cultivation depends on low cost nutrient source. The present study aimed to use leachate as a source of nutrients for microalgae. The experiments were conducted to evaluate the microalgal biomass, lipid conversion and yield in fatty acid methyl esters, for the culture media using 5%, 12% and 20% leachate, with concentrations of 0.02, 0.05 and 0.08 g N. L-1 and for control medium containing 1% Nitrogen (N), Phosphorus (P) and Potassium (K), at a concentration of 20 g L-1, 5 g L-1 and 20 g L-1, respectively. The microalgae used in this work was Chlorophyceae class and Coccomyxaceae family. The results showed that the medium with a concentration of 12% of leachate obtained better results, allowing to reach 1.19 g of biomass, conversion of 108.15 mg g-1 of lipids and esters content of 410,77 mg g-1. The microalgae used also presented predominance of palmitic and oleic fatty acids, presenting low amount of saturated fatty acids which can provide the fuel with cold resistance. And these aspects demonstrated that the leachate can be a promising source of nutrients for the cultivation of the studied microalgae.
