AbstractLaser microsurgery is a powerful tool for neurobiology used to ablate cells and sever neurites in-vivo. We compare a relatively new laser source to two well-established designs. Rare-earth-doped mode-locked fibre laser that produce high power pulsed radiation recently gained popularity for industrial uses. Such systems are manufactured at high standards of robustness and low maintenance requirements typical of solid-state lasers. We demonstrate that an Ytterbium-doped fibre femtosecond laser is comparable in precision to other femtosecond lasers, but with added reliability. It is more precise and can lesion deeper in tissue than a solid-state nanosecond laser. These advantages are not specific to the model system ablated for our demonstration, namely neurites in the nematode C. elegans, but are applicable to other systems and transparent tissue where a precise submicron resolution dissection is required.

This paper proposes a new optical angle measurement method in the optical frequency domain based on second harmonic generation with a mode-locked femtosecond laser source by making use of the unique characteristic of the high peak power and wide spectral range of the femtosecond laser pulses. To get a wide measurable range of angle measurement, a theoretical calculation for several nonlinear optical crystals is performed. As a result, LiNbO3 crystal is employed in the proposed method. In the experiment, the validity of the use of a parabolic mirror is also demonstrated, where the chromatic aberration of the focusing beam caused the localization of second harmonic generation in our previous research. Moreover, an experimental demonstration is also carried out for the proposed angle measurement method. The measurable range of 10,000 arc-seconds is achieved.

Deep brain stimulation (DBS) is a neurosurgery technique widely used in movement disorders, although its mechanism of action remains unclear. In fact, apart from the stimulation itself, the mechanical insertion of the electrode may play a crucial role. Here we aimed to distinguish between the insertional and the DBS effects on brain glucose metabolism. To this end, electrodes were implanted targeting the medial prefrontal cortex in five adult male Wistar rats. Positron Emission Tomography (PET) studies were performed before surgery (D0) and seven (D7) and nine days (D9) after that. DBS was applied during the 18FDG uptake of the D9 study. PET data were analysed with statistical parametric mapping. We found an electrode insertional effect in cortical areas, while DBS resulted in a more widespread metabolic pattern. The consequences of simultaneous electrode and DBS factors revealed a combination of both effects. Therefore, the insertion metabolic effects differed from the stimulation ones, which should be considered when assessing DBS protocols.
