Background: The loop diuretic drug furosemide is widely used for the treatment of edema in various conditions, such as pulmonary, cardiac and hepatic edema, as well as cardiac infarction. Furosemide, due to its poor water solubility and low bioavailability after oral administration of conventional dosage form, is categorized as class IV in the biopharmaceutical classification system. Objective: In the case of furosemide, this release profile is responsible for various physiological problems, acute diuresis being the most serious. This adverse effect can be circumvented by the modified release of furosemide from tablet formulations compared to those forms designed for immediate release. Method: In this report, a D-optimal combined experimental design was applied for the development of furosemide containing bilayer and compression coated tablets, aiming at lowering the drug’s burst release in the acidic environment of the stomach. A D-optimal combined design was selected in order to include all requirements in one design with many levels for the factors examined. The following responses were selected as the ones reflecting better criteria for the desired drug release: dissolution at 120 min (30-40%), 300 min (60-70%) and 480 min >95%. The new formulations, suggested by the Doptimal combined design, incorporated different grades of Eudragit ® polymers (Eudragit® E100 and Eudragit® L100-55), lactose monohydrate and HPMC K15M. The dissolution profile of furosemide from these systems was probed via in vitro dissolution experiments in buffer solutions simulating the pH of the gastrointestinal tract. Results: The results indicate that the use of Eudragit® E100 in conjunction with lactose monohydrate led to 21.32-40.85 % drug release, in the gastric medium, in both compression-coated and bilayer tablets. This is lower than the release of the mainstream drug Lasix® (t=120 min, 44.5% drug release), implying longer gastric retention and drug waste minimization. Conclusion: Furosemide’s release in the intestinal environment, from compression coated tablets incorporating Eudragit® L100-55 and HPMC K15M in the inner core or one of the two layers of the bilayer tablets, was delayed, compared to Lasix®

The research in this paper optimized the extraction technique of lentinan with ultrasonic assistant method on the basis of hot water extraction technique, and investigated the promoting function of ultrasound to polysaccharides extraction. Extraction condition was selected by means of orthogonal experimental design, four factors and three levels L9(34), after key elements were respectively chosen through single factor experiments. Comparison between optimal extraction parameters of two method, hot water extraction technique and ultrasonic assistant extraction technique, showed decreased extraction temperature and significantly shortened extraction time, which existed in the second means, improved the extraction efficiency. Lentinus edodes polysaccharide extracted with ultrasonic assistant technique, the extraction rate and polysaccharide content percentage increased 6.22% and 8.66% respectively, comparative to which extracted with hot water extraction technique.

AbstractThe effectiveness of pressurized hot-water extraction conditions for obtaining galactoglucomannans (GGMs) from Pinus halepensis suitable for applications like coatings and films packaging was investigated. For this purpose, high molar masses with high yields are required, presenting a serious challenge for hot-water extraction processes. The extraction of GGMs was carried out in an accelerated solvent extractor (ASE) and the isolation was performed by precipitation in ethanol. Three temperatures in the range 160–180 °C and five extraction times 5–90 min were tested in order to optimize extraction parameters of GGMs, avoiding thermal and chemical degradation in hot-water. Total dissolved solids (TDS) were determined gravimetrically after freeze-drying and weight average molar masses (Mw) were determined by high-performance size exclusion chromatography (HPSEC). Total non-cellulosic carbohydrates were determined by gas chromatography (GC) after acid methanolysis. Free monomers were additionally analyzed by GC. Lignin in water extracts was measured by an ultraviolet (UV) method. Acetic acid was determined after alkaline hydrolysis of acetyl groups and analyzed by HPSEC. The main parameters influencing the extraction processes of the GGMs, namely, extraction time and temperature were studied. Optimal extraction parameters of GGMs were identified at 170 °C and 20 min extraction time, with average Mw of extracted fraction of 7 kDa leading to a GGM yield of approximately 56 ${\text{mgg}}_{\text{o}.\text{d}.\text{m}}^{-1}$, corresponding to 6% on dry wood basis.

In this study, the effect of barley malt extract at two brix levels (74 and 79 °Bx) and three ratios of malt extract/honey (65:35, 70:30 and 75:25) on the flow behavior properties of honey–malt spread at three temperature levels (35 ℃, 45 ℃ and 55 ℃) was investigated. Time-dependent behavior data of the spread samples were appropriately fitted to the Weltman, first-order stress decay with a zero stress value and first-order stress decay with a non-zero stress value models. Also, the Power-law, Herschel–Bulkley, Casson and Bingham models were used for curve fitting the time-independent behavior data. Regarding the R2 and root mean square error coefficients, the first-order stress decay with a non-zero stress value and Herschel–Bulkley models were selected as the suitable models to describe the flow behavior of samples. The results for time-dependent properties showed that spread samples exhibit a thixotropic behaviour, as the viscosity for all samples decreased with increase in shearing time at a constant shear rate of 50 s−1.
