Abstract The inhibition effect of selected amphiphilic quaternary alkyl-substituted ammonium salts on oxidation of the lecithin liposome membrane was studied, induced by ultraviolet light and Fe2+ ions with ascorbic acid added. The salts differed in alkyl chain length Ri having the following alkyl substitutents: R1 = C9H19; R2 = C10H21; R3 = C12H25; R4 = CH2OC14H29; R5 = CH2OC16H33. It was found that all the salts used induced inhibition in oxidation of the liposome membrane, both that induced by UV light and ferrous ions. The antioxidant activity of the salts studied, dependent on the chain length and concentration, can be represented by the relation R1 < R2< R3 < R4 < R5. A similar dependence was obtained when the hemolytic activity of compounds was studied. Activity of salts increase with alkyl chain length.

Lipid microspheres (LM), currently in clinical use as drug carriers, mainly consist of soybean oil as a core and lecithin as a surfactant. The purpose of our study wass to determine whether or not LM incorporation is receptor-mediated. U937 cells resuspended in a serum-free medium abundantly took up unmodulated LM. A binding study showed that U937 cells had a single binding site for LM (410 sites/cell at 24°C; 100 sites/cell at 4°C). Inhibition assays revealed that lecithin liposome, lysophosphatidylcholines, activated α2-macroglobulin, and HDL did not affect the binding of LM to U937 cells. VLDL strongly, and LDL and AcLDL moderately, inhibited the binding of LM to U937 cells. Ligand blotting analysis revealed that unmodulated LM in an apoprotein-free buffer directly bound to a 40 kDa protein in the cell membrane fraction. These results suggest that LM that is not modulated by any protein is incorporated by specific cells via receptor-mediated processes.Key words: lipid emulsion, drug delivery system, monocyte, free fatty acids.

The objective of the present work was to compare the effects of groups of tin and lead organometallic compounds and their mixtures with amphiphilic quaternary ammonium salts (QAS) on the process of calcium ion desorption from lecithin liposome membranes, as dependent on the properties of the hydrophilic and hydrophobic parts of QAS. In the investigations the method of radioactive labels was applied. Synergism and antagonism in the action of both groups of compounds were found. The effectiveness of the cooperation depended more on chain length of QAS compounds than on the size and polarity of their hydrophobic parts. The most effective of all compounds studied was a the mixture of benzyldimethylammonium chloride in a mixture with tripropyltin. Since the rate of calcium desorption proved to be a good measure of efficacy of biologically active surfactants, it seems that the conclusions reached in this paper may be useful for choosing compounds which are able to decontaminate the environment polluted with heavy metals.
