3 monovarietal wines obtained in two consecutive years in Jidvei Winery Romania were investigated in respect of total polyphenols, total flavonoids and antioxidant activity before and after blending for sparkling wine obtaining. The three varieties were Chardonnay, Pinot Noir and Royal Maiden (Feteasca Regala). Total polyphenols were determined with Folin-Ciocalteu reagent, total flavonoid content with sodium nitrite, aluminum chloride and sodium hydroxide and antioxidant activity using Radical Scavenging Activity and Ferric Reducing Antioxidant Power methods. All mentioned parameters were determined by UV-VIS absorption spectrometry. The determined total polyphenolic content were within the interval 307.21-727.43 mgGAE/l wine for simple varieties and within 405.44-472.74 mgGAE/l for blends and final product (sparkling wine). Total flavonoid content ranged between 16.74 and 242.46 mgQE/l wine in simple varieties and between 38.48 and 80.18 mgQE/l for blends and sparkling wine. Antioxidant activity was correlated with total polyphenols and total flavonoids.

ABSTRAK Bahan alam dapat dijadikan bahan obat baru karena mengadung metabolit sekunder. Di Indonesia terdapat lebih kurang 30.000 jenis tumbuh-tumbuhan yang mengandung metabolit sekunder, lebih kurang 7.500 jenis diantaranya termasuk tanaman berkhasiat obat. Salah satunya adalah tanaman kasturi (Mangifera casturi Kosterm.). Tanaman M. casturi merupakan tumbuhan khas Kalimantan Selatan yang hanya dimanfaatkan oleh masyarakat untuk dikonsumsi, tidak untuk pengobatan. Penelitian ini bertujuan untuk menentukan kadar flavonoid total dan aktivitas antioksidan dari ekstrak etanol daun M. casturi. Penelitian ini bersifat non-eksperimental. Sampel yang digunakan adalah daun M. casturi yang berasal dari Kabupaten Banjar, Kalimantan Selatan. Penentuan Kadar flavonoid total dilakukan secara spektrofotometri UV-Vis dengan pereaksi kompleks AlCl3 sedangkan aktivitas antioksidan ditentukan dengan metode DPPH (2,2-difenil-1-pikrilhidrazil). Hasil penelitian diperoleh kadar flavonoid total ekstrak etanol daun M. casturi sebesar 9,31 ± 0,08 %b/b dan aktivitas antioksidan dengan nilai IC50 sebesar 34,558 ppm sehingga termasuk dalam kategori antioksidan yang sangat aktif. Kata kunci: Antioksidan, DPPH, Flavonoid Total, Mangifera casturi. ABSTRACT Natural resources can be used as the new medicine ingridients because it has second metabolite. In Indonesia, there are more than 30.000 kinds of plants that contain second metabolite, more or less than 7.500 kinds of those are medicinal plants. One of those plants is Kasturi (Mangifera casturi Kosterm.). M. casturi is a typical plant from South Kalimantan that be used as a food not as a medicine. The purpose of this research is to determine the total of flavonoid content and anti-oxidant activity from ethanol extract of M. casturi leaves. This study is a non-experimental research. Sample which used in this research is M. casturi Leaves from Banjar Region, South Kalimantan. The research for total Flavonoid content is done by UV-Vis spectrophotometric with AlCl3 reagent complex while the anti-oxidant activity is determined by DPPH (2,2-difenil-1-pikrilhidrazil) method. The result of this research are extract of M. casturi leaves obtains 9,31 ± 0,08 %b/b of total flavonoid and the antioxidant activity result with IC50 value is 34,558 ppm, so it can be categorized as a very active anti-oxidant. Diabetes mellitus is an important social and medical problem, as it causes the development of dangerous complications that lead to disability and mortality. This disease is characterized by a multi-vector pathogenesis that requires a comprehensive approach to treatment. Due to the use of mixtures of medicinal plants in the treatment of diabetes, it is possible to cover all aspects of the development of this disease and its complications, because the combination of different medicinal plants contain more biologically active substances that affect all parts of the pathogenetic mechanism of diabetes mellitus and its complications. Plant biocompounds have a wide range of pharmacological action and various mechanisms of influence on the development of diabetes and diabetic angiopathies (inactivation of the antioxidant defense system, activation of lipid peroxidation and the development of oxidative stress). In addition, the niche of the pharmaceutical market of Ukraine with phytomixtures is almost empty. The aim of the work was to study the total flavonoid content in some herbal mixtures, which have established hypoglycemic, hypolipidemic and antioxidant activity in previous studies in vivo The study of the total flavonoid content in herbal samples was carried out by UV-spectrophotometry with the recording of the absorption spectrum of complexation product of flavonoids with aluminum chloride (λmax 415 nm) in terms of rutin. According to the results of UV-spectrophotometric determination, it was found that the total flavonoid content in the herbal mixture No 3 was 1.47 ± 0.04%, in the herbal mixture No 4 – 1.78 ± 0.04%, in the herbal mixture No 7 – 1.29 ± 0.02%, in the herbal mixture No 13 – 1.16 ± 0.02% and in the herbal mixture No 19 – 3.62 ± 0.06% in terms of rutin. It was found that the best flavonoid content (3.62 ± 0.06%) was found in the plant mixture № 19, which contains nettle leaves, dandelion roots, blueberry leaves, rose hips and peppermint herb. Phytochemical studies may indicate a correlation between the content of flavonoids and pharmacological properties of the studied mixtures, because the functional hydroxyl groups of the latter cause strong antioxidant activity by neutralizing free radicals, chelation of heavy metal ions, increasing the activity of antioxidant protection products, which has been confirmed in previous in vivo and in vitro studies.

Theobjective of thisstudyis to investigate the phenotypic and biochemicalparameters of selectedsweet cherry (Prunus avium) from middle region of Morocco. Themain biochemical composition, contentsof total polyphenols, total flavonoids, total anthocyanins and antioxidant activity, were measured in the fruits of four sweet cherry cultivars (ʻBurlat’, ʻVan’, ʻNapoleon’andʻCerisette’) grown in two locations (“Laanoceur”and “Toufselt”) in the MiddleAtlas. The free radical scavenging activity was evaluated spectrophotometrically using 1,1‒diphenyl‒2‒picrylhydrazine (DPPH)and ABTS assay.The fruit weight, and pulppercentagewerestudied and found to range between 237–329 g, 38.14–42.22%, 39.21–44.36g, and 52.27–57.48%, respectively.The total phenolic and total anthocyanin content rangedfrom305.99 and 306.67 mg EqGal/100g D.W, total flavonoid contents were within the range of 481.73-517.67 mgeqRE/100g D.W, and total anthocyanin contents were between 1.09 and 2.89 mg Eqcyanidin 3-glucoside/100g D.W. Antioxidant activity ranged from 17.18 to 18.11 mg EqTrolox/100g D.W for DPPH assayand from 27.97 to 29.60 mg EqTrolox/100f D.W for ABTS method.The highest values of total anthocyanin content and antioxidant activity (DPPH) were recorded in ʻBurlat’. The highest value of total flavonoid content was found in ʻCerisette’. Cherries from “Laanoceur”and “Toufselt” locations are characterized by similar biochemical composition and antioxidant activity, except for total anthocyanin content that shows slightly elevated values in “Laanoceur”. The close correlation between total phenolic contents and antioxidant activities (r2=0.73) show that antioxidant activity of cherry fruit depends on total polyphenols.

An experiment was conducted to determinate the phytochemical properties (chlorophyll-a, chlorophyll-b, carotenoids, antioxidant activity, total phenolic and flavonoid content) in ten medicinal plants namely Syal Kata (Argemone mexicana L.), Akanda (Calotropis gigantea (L.) Dryand.), Dumur (Ficus carica L.), Chalmugra (Gynocardia odorata R.Br.), Kata kachu (Lasia spinosa (L.) Thwaites), Shetodrone (Leucas aspera (Wild.)), Khona (Oroxylum indicum (L.) Kurz), Reri (Ricinus communis L.), Ghat kachu (Typhonium trilobatum (L.) Schott) and Bazna (Zanthoxylum rhetsa DC.) for ethno-botanical uses. The young fresh leaves were harvested and were subjected to methanolic (95%) extract. Total phenolic contents were analysed by using Folin - Ciocalteau method where gallic acid was used as standard. Total phenolic content varied from 94.53 mg GAE/100 g FW (C. gigantea) to 484.88 mg GAE/100 g FW (L. aspera). Total flavonoid contents were performed by using Quercetin as standard. Total flavonoid content varied from 137.25 mg QUE /100 g FW (C. gigantea) to 334.27 mg QUE/100 g FW (G. odorata). Antioxidant activity of these extracts was performed by using DPPH free radical scavenging assay. Total anti-oxidant capacity varied from IC50 value 35.37 mg/mL (A. mexicana) to 90.47 mg/mL (F. carica) where ascorbic acid is used as standard. Results indicated that among the ten medicinal plants phenolics rich in L. aspera, flavonoids rich in G. odorata, antioxidant activity rich in A. mexicana. Z. rhetsa leaf rich in chlorophyll-a content (311.67 mg/100 g FW), L. aspera leaf rich in chlorophyll-b content (157 mg/100 g FW), Z. rhetsa rich in chlorophyll- (a+b) content as 439 mg/100 g FW, A. mexicana leaf rich in carotenoids content as 96 mg/100 g FW.

Medicinal plants are a source for a wide variety of natural antioxidants. In the study reported here, we have conducted a comparative study between two medicinal plants roots having the same geographic origin and growing in the same natural conditions. The present study is designed to evaluate the radical scavenging activity, total phenol content (TPC) and total flavonoid content (TFC) of the MeOH roots extracts of Asphodelus microcarpus and Asphodeline lutea. The antioxidant activity of the extracts was examined by DPPH method. Total phenol and total flavonoid quantities of the samples were determined spectrophotometrically using Folin-Ciocalteu and AlCl3 reagents respectively. The roots in both plants exhibited the free radical scavenging property (IC50= 0.30, 0.54 mg/ml) for A. microcarpus and A. lutea, respectively compared to that of the positive control BHT (IC50= 0.017 mg/ml). As assumed, the amount of total phenolics was (17.90, 13.02 mg GAE/ g dry weight for A. microcarpus and A. lutea root extracts, respectively. Roots extract of Asphodelus microcarpus were the richest in total phenolic compounds and also it has been found to be rich in flavonoids (14.60 mg rutinoside/g dry weight). To our knowledge, this is the first report on the antioxidant activity of A. microcarpus and A. lutea root from Syrian origin and our findings suggest the possibility of using the roots as a novel source of natural antioxidant agent for the food and pharmaceutical industries
