Aim. The aim of the study was a comparative assessment of total phenolic content and total flavonoid content in in vitro plants, regenerated plants, plants grown in a growth chamber, and tissue culture of several genotypes of Deschampsia antarctica. Methods. In vitro culture, Folin-Ciocalteu method, spectrophotometry, high-performance liquid chromatography. Results. The total content of phenolic compounds and total flavonoid content was determined in the samples of three D. antarctica genotypes: G/D12-2a (2n=26), DAR12 (2n=26+0–3B) and Y66 (2n=36–39). The content of these biologically active compounds was the highest in in vitro plants and it was higher than in plants grown in a growth chamber. The highest content of BAC was found in DAR12 in vitro plants (16.50 mg of ferulic acid equivalent and 21.26 mg of rutin equivalent per g of dry weight, respectively). The regenerated plants did not differ significantly in the content of BAC from the original in vitro plants. In tissue culture, the content of BAC was lesser. One- and two-year-old tissue cultures did not differ significantly in the content of phenolic compounds and flavonoids. Conclusions. The relatively high content of phenolic compounds and flavonoids in in vitro plants and in regenerated plants indicates that in vitro cultivated D. antarctica plants can be a promising raw material for production of valuable BACs. Objective: The present study evaluates the antioxidant and α-amylase inhibitory activity of leaf and bark extracts of Phyllanthus indofischeri with methanol and water as solvents. In addition to this, the total phenolic content and total flavonoid content was determined.Methods: The total phenolic and total flavonoid content of the extracts was determined by folin ciocaletus reagent method and aluminium chloride colorimetric method respectively. The antioxidant and α-amylase inhibitory activity were measured by various assays, including α, α-diphenyl-ẞ-dipicryl-hydrazyl (DPPH) free radical scavenging, 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) radical scavenging, superoxide radical scavenging, total antioxidant capacity by phosphomolybdate method and porcine pancreatic α-amylase inhibitory assay. The IC50 values were calculated and compared with standards such as gallic acid, ascorbic acid and α-acarbose.Results: The results illustrated that all the extracts of Phyllanthus indofischeri exhibit significant antioxidant and α-amylase inhibitory activity. Among the extracts, methanolic leaf extract showed high levels of activity followed by bark water extract.Conclusion: Phyllanthus indofischeri extracts had shown antioxidant and α-amylase inhibitory activity. On the basis of these results, Phyllanthus indofischeri can be used as a natural antioxidant and hypoglycemic agent against various disorders related to oxidative stress; and the isolation of bioactive compounds was warranted.

In this work the spray drying of the fresh stevia leaves aqueous extract without encapsulating agents was carried out. The effect of the inlet air temperature (160-200 ºC) and the feed flow rate (2-3 kg/h) on the total phenolic content, the total flavonoid content and the antioxidant capacity were evaluated using Folin-Ciocalteau, aluminum chloride and DPPH methods respectively. The inlet air temperature had a significant effect on all parameters evaluated that showed a decrease when increasing the inlet air temperature, the feed flow rate had a significant effect on total flavonoids content and the antioxidant capacity, by increasing the feed flow rate the inhibition of the DPPH radical decreased and the total flavonoid content increased. The treatment at 160 ºC and 3 kg/h retained highest total flavonoid content and the antioxidant capacity. Keywords: Phenolic compounds, stevia, spray drying, antioxidant capacity

Establishment of in vitro cultures from Astragalus glycyphyllos, determination of biomass and analysis of total flavonoids, rutin and camelliaside A were performed. To increase flavonoid production various combinations of plant hormones and light/dark regimen were investigated. Suspension cultures with exogenous quercetin were evaluated for possible increase in flavonoid production. Shoots, calli and suspensions were successfully established. Rutin and camelliaside A were proved in highest amount in shoots. Calli, cultivated on modified G48 medium, with double amount of Ca2+ and Mg2+, achieved higher total flavonoid content (2.37 and 2.03 mg/g DW). Suspensions cultures, cultivated on modified G48 medium with 10, 20 and 30 mg/mL quercetin achieved higher total flavonoid content (0.09, 0.10 and 0.13 mg/mg DW). Biotransformation of quercetin to isoquercitrin was achieved. The highest concentration of isoquercitrin (56.73 ng/mg DW) was observed on suspensions cultures cultivated on modified G48 medium, with 20 mg/mL quercetin.

BACKGROUND: The number of obesity cases is still increasing worldwide and has reached an epidemic scale. Plants are known to have a protection role in the development of obesity, however their antioxidant and anti-obesity activities have not widely known. This study was conducted to assess the in vitro antioxidant and anti-obesity activities of different types of freeze-dried fruits cultivated in Indonesia, especially Canarium sp., Averrhoa bilimbi L. and Malus domestica.METHODS: Total phenolic content of freeze-dried fruits was identified by Folin-Ciocalteu method, while the total flavonoid content was measured by aluminium chloride colorimetric assay. To assessed the antioxidant activity, 2,2-diphenyl 1-picrylhydrazyl (DPPH) scavenging activity assay and 2,2’-azino-bis (3-ethylbenzothiazoline6-sulphonic acid) (ABTS) reducing activity assay were performed. The α-amylase and lipase inhibitory activity assay were performed to assess the anti-obesity activity. For comparison, hydroxycitric acid (HCA) compound was also assessed with DPPH, ABTS, α-amylase and lipase assays.RESULTS: A. bilimbi had the highest total phenol content (6.35 µg GAE/mg), meanwhile M. domestica had the highest total flavonoid content (2.06 µg QE/mg). A. bilimbi also showed the highest antioxidant activity both in DPPH and ABTS assay, with inhibitory concentration (IC50)=279.99 µg/mL and 631.78 µg/mL, respectively. The freeze-dried M. domestica had the highest anti-α-amylase activity (IC50=258.85 µg/mL), while Canarium sp. had the highest anti-lipase activity (IC50=118.66 µg/mL).CONCLUSION: Freeze-dried fruits demonstrate in vitro benefits toward obesity. A. bilimbi has potent antioxidant activity and is beneficial against obesity-related adverse health effect by relieving oxidative stress. M. domestica and Canarium sp. hamper the fat accumulation by reducing the carbohydrate absorption and dietary lipid.KEYWORDS: antioxidant, anti-obesity, Canarium sp., Averrhoa bilimbi L., Malus domestica, hydroxycitric acid

Background: Chickpea (Cicer arietinum L.), a self-pollinating legume being cultivated globally as a rich source of vegetarian protein. It plays an important role in human feed and nutritional security, especially in agricultural-based communities. Chickpea has higher bioavailability of protein, good sources of polyphenols and flavonoids. Besides their nutritional value, chickpea seeds contain various phenolic compounds. Phenolic compounds are of particular interest due to their contribution to the seed colour, sensory characteristics and several biological properties. Flavonoids are one of the main groups of phenolic compounds found in grain legumes. Desi and Kabuli chickpeas are being used worldwide and there are few studies where both desi and kabuli chickpeas seed flour for above properties with respect to physiological traits has been reported. Methods: The present investigation has been formulated to compare popular chickpea genotypes of desi and kabuli types for biochemical parameters viz., protein content, amino acid, total flavonoid content, total phenolic contents and RSA as well as two physiological traits i.e., chlorophyll content and leaf area index. The experiment consisted of 44 genotypes () grown in Randomized Block Design with row to row distance of 30 cm, in two replications during Rabi 2018-19. Result: The average crude protein content in desi and kabuli chana varied from 18.2% (Dollar variety) to 26.7% (JG315) and total phenolic content (TPC) ranged 1.22 to 0.74 mg/g. Total Flavonoid content (TFC) varied from 0.39 (ICCV-2) to 0.61 mg/g (JAKI-9218) with mean value of 0.47. Radical scavenging activity (RSA) in chickpea genotypes ranged from 36.2 to 49.5% with mean value of 40.86%. Total amino acid significantly correlated with TPC and TFC and TPC significantly correlated with TFC at 5% significant level.

Abstract Background: The petals of Rosa species are used in the food industry and various traditional medicinal products, but few studies exist on the phytochemical composition and antioxidant activity of petals of Rosa L. species grown in Iran. Objective: Phytochemical characteristics and antioxidant activity and some phenolic compounds of petals of six Rosa L. species were studied. Methods: Total phenolic content, total flavonoid content, and antioxidant activity were determined using Folin–Ciocalteu reagent, aluminum chloride method, ferric-reducing antioxidant power (FRAP), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assays, respectively. An HPLC system was used for quantitative analysis of phytochemical compounds. Hierarchical cluster analysis (HCA) and principal component analysis (PCA) were performed among the variables analyzed using Minitab software. Also, heat maps were used to visualize phytochemical characteristics and antioxidant activity in each species using GraphPad Prism software. Results: The amount of total phenol content, total flavonoid content, and antioxidant activity were in the range of 25.13–52.01 mg gallic acid equivalents/g dry weight (DW), 0.61–0.82 mg quercetin equivalents/g DW, 11.47–20.93 µmol Fe++/g DW (FRAP), and 31.66–74.44% (DPPH), respectively. The p-coumaric acid (647.28 μg/g DW) and chlorogenic acid (24.37–135.23 μg/g DW) were found to be the most abundant phenolic compounds in the extracts of rose petals. The HCA and PCA revealed three distinct categories of species based on phytochemical composition and antioxidant activity. Conclusions: These results showed that phytochemical characteristics of different rose species widely correlated with species type and are promising sources of natural antioxidants beneficial for use in the food or pharmaceutical industries. Highlights: Iran is one of the main centers for genetic diversity of Rosa L. The petals of Rosa species are used in the food industry and various traditional medicinal products, but few studies exist on the phytochemical composition and antioxidant activity of petals of Rosa L. species grown in Iran. Antioxidant activity and phytochemical compound of Six Rosa L. species petals grown in Iran were studied. Phenolic compounds in petals of Rosa were analyzed by HPLC. The color parameters, amount of total phenolic, total flavonoids, antioxidant activity and some individual phenolic compounds were significantly variable amongst Rosa species.

Objective: This study investigated the activity of the extracts and the isolated compound on its potential in vitro antidiabetic, anti-inflammatory and potential anti-cancer effect, total flavonoid content against alpha-glucosidase enzyme inhibition and on macrophage respectively from Andrographispaniculata (Burm. F) wall. Ex Nees. Methodology: The isolation of the constituents was done using column while the in vitro anti-inflammatory and antidiabetic was done using nitric oxide and α-glucosidase enzyme inhibition assay while anticancer assessment was done performed using cell viability on various human hepatocellular carcinoma cell) and Chang liver (normal cell line) were determined by 3-(4,5-dimethylthiazolzyl-2yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. Results: Although, the total flavonoid content was low, ethyl acetate extract indicated the highest total flavonoids content and it’s statistically different from methanol, ethanol: water (1:1 v/v) and aqueous extracts. Ethyl acetate extracts exhibited the highest percentage inhibition (29.8 %) against nitric oxide scavenging activity (NaNO2) compared to other extracts. Also, the isolated crystals showed a significant inhibition against NaNO2. Moreover, the ethyl acetate extract showed the highest percentage inhibition of α-glucosidase enzyme with optimal concentration of 950μg/ml for 50 % inhibition (IC50) while the other three extracts (methanol, 50 % ethanol: water (1:1 v/v) and aqueous) indicated activity below 50 % inhibition which might be due to total flavonoid content. The potential anti-cancer effect indicates that both the methanol extract and crystals (AP02 and AP03) may have the same compound. Conclusion: The compound isolated might be Andrographolide and the activity might be due to flavonoid content for the extract.

The objective of this study was to determine the optimal value of factors affecting the extraction of phenolic compounds from sugar apple seeds. The effects of solvent nature (ethanol, methanol and water), solvent concentration (40, 60,80 and 100%), solid-to-solvent ratio (2.5/100, 5.0/100, 7.5/100 and 10 mg/100 mL ), extraction temperature ( 40, 50, 60 and 70 ºC ) and extraction time (60, 120, 180 and 240 min) were investigated. Total phenolic content and total flavonoid content were used for the determination of phenolic compounds of sugar apple seeds extract. Experimental results showed that all the examined parameters had statistically significant (p < 0.05) effects on phenolic compound extraction. The optimal extraction conditions were as follows: extracting solvent 80% ethanol, sample-to-solvent ratio 5 g/100 mL, extraction contact time 120 min and extraction temperature 60 ºC with values of 356.92 ± 9.27 mgGAE/100g DW, 240.03 ± 6.16 mgQE/100g DW for the total phenolic content and the total flavonoid content, respectively.
