This study sought to develop a validated reverse-phase high-performance liquid chromatography method for the quality control of the stem bark ingredients and its finished products and investigate the synergistic anti-inflammatory activity of the phytochemical constituents of C. regia stem bark. Fractionation and isolation of biomarkers were carried out by column chromatography on silica gel and monitored by thin-layer chromatography. The isolated biomarkers were characterized based on their melting points and extensive analysis of their spectroscopic data (IR, 1D and 2D NMR). The chromatographic separation was investigated and developed for the analysis of the biomarkers using µBondapakTM C18 (3.9×300 mm, 5 µm) as stationary phase. The mobile phase composition of 0.1 % trifluoroacetic acid as solvent A, and methanol as solvent B with gradient elution was finally selected. The carrageenan-induced edema in a 7-day-old chick model was used to assess the anti-inflammatory activity of the stem bark extract and compared to diclofenac sodium as a reference drug. Two compounds were successfully isolated and identified, as Regiafuran A (1) and 3,5,7,4’-Tetrahydroxy-2’-methoxyflavonol (2). The compounds were employed as biomarkers in the RP-HPLC method development. The developed method was validated and was successfully used to quantify the amount of 1 and 2 in the stem bark to be 0.224% ± 0.056%w/w and 0.354% ± 0.041%w/w respectively. The crude extract showed considerable anti-inflammatory activity compared to the reference drug, diclofenac. The method demonstrated acceptable levels of accuracy, precision, specificity, and robustness hence can be successfully adopted for routine quality control and standardization of the stem bark of C. regia. The stem bark of the plant exhibited significant anti-inflammatory activity.

Anogeissus pendula Edgew has been reported to be used in gastric disorder even though no attempt has been made to evaluate the same. The present study was designed to evaluate the hydroalcoholic extracts of stem bark and leaves for in vivo acute antiulcer and anti-inflammatory activities. Antiulcer activity was studied by pylorus ligation induced ulcers while anti-inflammatory activities was studied by carrageenan induced paw edema. The extract of stem bark at 200 and 400 mg/kg b. w., significantly (P<0.05) reduced ulcer index whereas extracts of both stem bark and leaves significantly (P<0.001) reduced paw diameter at the fifth hour. The hydroalcholic extract of stem bark of A. pendula possesses more significant antiulcer and anti-inflammatory activity than the hydroalcholic extract of leaves may be due to the presence of higher amount of phenolic compounds.

Summary Cussonia barteri Seem (Araliaceae) is a deciduous tree growing in savannah of Africa. Ethnomedicinally, it is used in Africa as an analgesic, anti-malarial, anti-inflammatory, anti-anaemic, anti-diarhoea, anti-poison, ani-pyschotic and anti-epileptic agent. This review provides a brief summary on the phytochemical screenings, ethnomedicinal and pharmacological applications of various parts of C. barteri. Leaves, stem bark and seed of C. barteri have been shown to be rich in saponins, flavonoids, phenols, sugars and alkaloids. Some of these constituents have been isolated and elucidated from C. barteri. Several compounds isolated from plant include triterpenes, saponins, polyenyne and quinic esters. Phytochemical constituents are also partly responsible for biological activities of C. barteri. Extracts and components isolated from the plant have demonstrated neuropharmacological, anti-larvicidal, anti-microbial, anti-inflammatory and antioxidant activities. Overall, the insights provided by this review reinforce the potential of C. barteri for drug development and create the need for further scientific probe of constituents of the plant with the aim of developing novel drug candidates.

Abstract Background: Holarrhena floribunda stem bark has anecdotal use in Ghanaian folk medicine for management of inflammatory conditions. This study was conducted to investigate the in vivo anti-inflammatory activity of the bark extract using models of acute inflammation in male Sprague Dawley rats, C57BL/6 mice and 36 ICR mice. Methods: A 70 % hydro-ethanol extract of the stem bark (HFE) was evaluated at doses of 5–500 mg/kg bw. Local anaphylaxis was modelled by the pinnal cutaneous anaphylactic test. Systemic anaphylaxis or sepsis were modeled by compound 48/80 or lipopolysaccharide, respectively. Clonidine-induced catalepsy was used to investigate effect on histamine signaling. Anti-oedematogenic effect was assessed by induction with carrageenan. Effects on mediators of biphasic acute inflammation were studied using histamine and serotonin (early phase) or prostaglandin E2 (late phase).Results: HFE demonstrated anti-inflammatory and/or anti-oedematogenic activity comparable to standard doses of aspirin and diclofenac (inhibitors of cyclooxygenases-1 and -2), chlorpheniramine (histamine H1-receptor antagonist), dexamethasone (glucocorticoid receptor agonist), granisetron (serotonin receptor antagonist) and sodium cromoglycate (inhibitor of mast cell degranulation). All observed HFE bioactivities increased with dose.Conclusions: The data provide evidence that the extract of H. floribunda stem bark has anti-anaphylactic and anti-oedematogenic effects; by interfering with signalling or metabolism of histamine, serotonin and prostaglandin E2 which mediate progression of inflammation. The anti-inflammatory and antihistaminic activities of HFE may be relevant in the context of management of COVID-19.
