Background: Relapse is a common problem in substance-use disorders. Accumulating evidence has shown that diazepam can reduce the abuse potential of some drugs. Objectives: We investigated the effects of diazepam during and after the withdrawal period on the reinstatement of conditioned place preference (CPP) induced by a single dose of methamphetamine in male Wistar rats. Methods: Three groups of rats were trained to acquire methamphetamine CPP. The rats in the control group received, respectively, saline and methamphetamine (1 mg/kg, daily, ip) in the extinction and priming phases. Two other groups (one and two) had a similar process, but the rats in group one received diazepam (3 mg/kg, daily, ip) in the extinction phase and those in group 2 received a single dose of diazepam just before the priming phase. The CPP was checked daily during all processes. Results: We showed that a 3-day period injection of methamphetamine (1 mg/kg) resulted in a positive conditioning score on the first day after conditioning. Also, 3 mg/kg of diazepam daily in the extinction period shortened the withdrawal period for one day and extinction happened on the 9th and 10th days. Diazepam also reduced the conditioning score at the reinstatement phase (P < 0.01). Furthermore, diazepam injection (3 mg/kg) before the methamphetamine priming dose did not have any significant effect on the reinstatement of drug-seeking. Conclusions: This study demonstrated that administration of diazepam during the extinction period was associated with reduced maintenance and reinstatement of methamphetamine-induced CPP, while the single dose of diazepam could not prevent the reinstatement of methamphetamine.

This study aimed to evaluate the liver, kidney damage caused by S. typhimurium and to estimate the oxidative damage in association with this bacteria. A highly virulent isolates of S. typhimurium were obtained from the department of internal and preventive medicine/ College of Veterinary Medicine/ University of Baghdad. A twenty five local rabbits of both genders with age range (2-4 months) weeks old were used for this study, the rabbits were divided randomly into five groups each group contains 5 rabbits :- group 1: drenched orally with 5 ml of normal saline and consider as control group, group 2: were drenched orally with (5 ml) suspension which contain (5��109 CFU) of Salmonella typhimurium and regarded as infected group, group 3 were drenched orally with (5 ml) suspension which have (5��109 CFU) of Salmonella typhimurium then treated with a single dose of gentamicin alone at 0.05ml/kg (5mg/ml) orally after presence of signs (after 24hrs. post inoculation), group 4 were drenched (5 ml) suspension having (5��109 CFU) of Salmonella typhimurium then treated with a single dose of Ca-EDTA alone at 40mg/kg orally after presence of signs (after 24hrs. post inoculation) and group 5 were drenched (5 ml) suspension that contain (5��109 CFU) of Salmonella typhimurium then treated with a single dose of combined gentamicin at 0.05ml/kg (5mg/ml) orally after presence of signs (after 24hrs. post inoculation) and Ca-EDTA 40mg/kg after presence of signs (after 24hrs. post inoculation).The results of biochemical profile showed a significant increase (p less than 0.05) in ALT, creatinine and urea levels in infected group as compared with control group, while, the treated groups especially group 5 showed a significant improvement in ALT, Urea and creatinine levels which returned to relative normal levels as compared with infected group after 96hrs. post treatment. Also, the results of oxidative stress showed a significant increase in the levels of MDA in G2, G3, G4 and G5 after 48 hrs. post treatment, while the level of GSH showed a significant decrease in the level at 48hrs., both were returned to relative normal levels after 96hrs.post treatment especially in group 5.In conclusion, S. typhimurium can causing liver and kidney damage which is manifested by increase ALT, Urea and Creatinine. Also, MDA and GSH is increased due to salmonellosis.

Abstract The aim of this study was to evaluate the effect of a lower dose of parathyroid hormone- PTH (1-34) on osteogenic potential of bone healing around titanium implants inserted into the tibia of rats. A blind parallel study was conducted in 45 adult male Wistar rats. Each rat received one titanium implant (4.5 x 2.2 mm) and was randomly assigned to receive subcutaneous injections, three times/week for 30 days, of the following treatments: group 1 - 40 µg/kg of PTH (1-34) (n=15); group 2 - 2 µg/kg of PTH (1-34) (n=15) and; group 3 - only the vehicle required for hormone dissolution (n=15). Thirty days after surgery, the animals were sacrificed and specimens containing the implant and the surrounding bone were removed and processed for non-decalcified sections. The sections were evaluated according to the following histometric parameters: proportion of mineralized tissue (PMT) adjacent to the implant threads (500 µm band); bone filling within the limits of the threads (BF) and; bone-to-implant contact (BIC). For the cortical region, both hormone dosages (groups 1 and 2) promoted better results, for all parameters, when compared to control group (p<0.05). Similar results were observed for the BF parameter in the cancellous region (p=0.0394). Therefore, systemic administration of PTH (1-34) stimulates bone formation around titanium implants, even at low doses.

Abstract: Infertility is an inability of couples to reproduce after carrying out sexual intercourses for at least a year without using any contraceptives. There are 10-15% couples worldwide who experience infertility problems and almost one half of them is on men. Spermatozoa qualities including concentration, motility, and morphology are used as an indicator for men’s fertility. The utilization of herbal medicine (i.e. Eurycoma longifolia Jack extracts) is now being an alternative way to improve fertility among the community. This study was carried out to find the effects of Eurycoma longifolia Jack on spermatozoa qualities. This experimental study was conducted to nine male wistar rats (Rattus norvegicus) weighing from 200-250 grams, aging from 12-15 months. These nine adult rats were divided into 3 groups of 3 rats each. 400 and 600 mg/kg/day of Eurycoma longifolia Jack extracts were orally administrated to group 1 and 2 respectively, while group 3 were treated as control group. After 50 days, the animals of group 1, 2 and 3 were sacrificed. As a result, there is an improvement in qualities of spermatozoa which are statistically significant (p<0,05) in group 2. Thus, provide the beneficial effects of 600mg/kg Eurycoma longifolia Jack extracts on the qualities of spermatozoa.Keywords: pasak bumi, spermatozoaAbstrak: Infertilitas adalah ketidakmampuan pasangan untuk dapat hamil setelah satu tahun berhubungan intim tanpa menggunakan kontrasepsi. Kualitas spermatozoa meliputi konsentrasi, motilitas dan morfologi spermatozoa merupakan salah satu indikator fertilitas pada pria. Penggunaan tanaman herbal atau yang lebih dikenal jamu telah menjadi pengobatan alternatif di masyarakat. Tanaman herbal yang dimaksud adalah pasak bumi. Penelitian ini bertujuan untuk mengetahui efek dari esktrak pasak bumi (Eurycoma longifolia Jack) terhadap kualitas spermatozoa. Penelitian ini menggunakan 9 ekor tikus wistar jantan (Rattus norvegicus) dengan berat badan 200-250 g berumur 12-15 bulan. Sembilan ekor wistar dibagi menjadi tiga kelompok, dan satu kelompok terdiri dari tiga ekor tikus wistar. Penelitian ini dilaksankan selama 50 hari. Esktrak pasak bumi (Eurycoma longifolia Jack) diberikan dengan dosis 400mg/kgBB per hari pada perlakuan 1 dan dosis 600 mg/kgBB per hari pada perlakuan 2. Setelah 50 hari hewan coba pada kelompok kontrol, perlakuan 1 dan perlakuan 2 diterminasi. Hasil penelitian memperlihatkan terjadi peningkatan kualitas spermatozoa yang signifikan secara statistik (p<0,05) pada perlakuan 2 terhadap kelompok kontrol. Hasil tersebut menunjukan bahwa Esktrak pasak bumi (Eurycoma longifolia Jack) pada dosis 600 mg/kgBB dapat meningkatkan kualitas spermatozoa.Kata kunci: pasak bumi, spermatozoa

The objective of the present study was to evaluate the superovulatory response and embryo production in beef donors using 8 twice-daily injections of FSH or an alternative protocol in which the last 4 FSH injections were replaced with a single injection of eCG. In Exp. 1, 12 mature Bonsmara donor cows, with a body condition score between 3 and 4 (1 to 5 scale) were superstimulated twice every 46 days in a crossover design (i.e. in each experiment all cows received 2 treatments and the 2 treatments were equally represented in each replicate). On Day 0 a.m., all donors received an intravaginal device with 1.2g of progesterone (Diprogest 1200®, Zoovet, Santa Fe, Argentina), along with 50mg of progesterone i.m (Progestar®, Zoetis, Buenos Aires, Argentina) and 5mg of oestradiol-17β (17βOestradiol®, Rio de Janeiro, Argentina) IM. On Day 4 a.m., the superstimulatory treatments were initiated and donors in the control group received 8 applications of FSH (Folltropin-V, Vetoquinol, Lure, France), IM (total dose: 300mg NIH-FSH-P1) in a twice-daily decreasing dosage schedule over 4 days (i.e. 60, 60, 50, 50, 30, 30, 10, and 10mg, respectively). Donors in the FSH+800 eCG group received only the first 4 applications of FSH (i.e. 60, 60, 50 and 50mg, respectively) and on Day 6 a.m. they received 800IU of eCG (Novormón®, Zoetis) IM in a single dose. All donors received 500μg of cloprostenol (Ciclase DL®, Zoetis) IM on Day 6 a.m. and p.m., and the intravaginal devices were removed on Day 7 a.m. All cows also received 100μg of gonadorelin acetate (GnRH, Gonasyn gdr, Zoetis) on Day 8 a.m. and were inseminated with frozen-thawed semen from 2 bulls 12 and 24h later. On Day 15, ova/embryos were collected and evaluated according to the IETS standards. The data were analysed by GLMM (Infostat, 2018). In Exp 2., 18 Bonsmara donors with similar conditions as those in Exp. 1 were superovulated twice in a crossover design. Cows in both groups received similar treatments to those in the FSH+eCG treatment of Exp. 1, except that the total dosage of FSH was 200mg (i.e. 60, 60, 40, and 40mg, respectively) and the eCG given on Day 6 a.m. was either 600IU (group 1) or 800IU (group 2). In Exp 1., the FSH (control) group had a higher (P&lt;0.01) number of fertilized oocytes, but there were no differences in the other end points evaluated (Table 1). In Exp. 2, no differences were found between FSH+800 eCG and FSH+600 eCG groups in any of the parameters evaluated. In conclusion, the replacement of the last 4 injections of FSH by a single dose of eCG decreases the number of treatments required in a superovulation program without negatively affecting the production of transferable embryos. PURPOSE: To investigate the effect of enemas containing probiotics and budesonide on the colonic mucosa in experimental colitis. METHODS: Fifty male Wistar rats with experimental colitis induced by 10% acetic acid enema were randomized to five groups (10 rats each) according to the treatment: group 1 - saline solution, group 2 - budesonide (0.75 mg/kg/day), group 3 - probiotics (1mg/day), group 4 - probiotics plus budesonide, and group 5 - control, with not-treated rats. The following variables were studied: body weight, macroscopic and microscopic score of the colonic mucosa, and DNA content of the mucosa. RESULTS: All animals lost weight between the beginning and the end of the experiment (280+ 16 mg versus 249+21 mg, p< 0.001). There was no significant difference among the groups in relation to both the macroscopic and histological score. The budesonide + probiotic group showed higher DNA content than control group (1.24+ 0.15 versus 0.92+ 0.30 mg/100mg of tissue, p=0.01). CONCLUSION: Budesonide in addition to probiotics enhance the mucosal trophism in experimental colitis.

AbstractThe aim of this study was to evaluate the cardioprotective effects of kolaviron (KV), a biflavonoid fromForty-two male Wistar rats (180–200 g) were randomly divided into seven groups of six rats each. Group 1 served as control; group 2 received ISO (85 mg/kg subcutaneously); groups 3, 4 and 5 received ISO and KV1 [100 mg/kg orally (p.o.)], KV2 (200 mg/kg, p.o.) and QUE (25 mg/kg, p.o.), respectively; and groups 6 and 7 received QUE and KV2, respectively.Administration of ISO caused significant (p<0.05) elevation of serum creatine phosphokinase, lactate dehydrogenase, alkaline phosphatase, alanine aminotransferase and aspartate aminotransferase by 2.2-, 1.9-, 2.1-, 1.9- and 1.7-fold, respectively, relative to controls, with a concomitant decrease in cardiac activities of these enzymes. Administration of ISO led to significant decrease (p<0.05) in the levels of cardiac superoxide dismutase, catalase, glutathioneKV protects against ISO-induced cardiotoxicity in vivo, suggesting its usefulness as a possible chemoprophylactic agent against cardiotoxic drugs.

OBJECTIVE: Surgical scar tensile strength may be influenced by several factors such as drugs, hormones and diet. The purpose of the present study was to determine the influence of a shrimp-enriched diet on the tensile strength of rat scars. METHODS: Forty male Wistar rats were submitted to a 4 cm dorsal skin incision and the wounds were sutured with 5-0 nylon interrupted suture. The animals were divided into two groups: Group 1 (control) received a regular diet, and Group 2 (experimental) received a shrimp-enriched diet. The two diets contained the same amounts of proteins, lipids and carbohydrates. The rats in each group were divided into two subgroups according to the time of assessment of the scar tensile strength: subgroup A, studied on the 5th postoperative day, and subgroup B, studied on the 21st postoperative day. RESULTS: The tensile strength of the scar on the 5th postoperative day was lower in the animals that received the shrimp-enriched-diet (303.0, standard error of mean= 34.1) than in the control group (460.1, SEM = 56.7) (p<0.05). CONCLUSION: A shrimp diet reduces the tensile strength of the scar. The next step of this study will be to clarify the mechanism in which shrimp affects tensile strength.

Introduction: Monosodium glutamate (MSG) is one of the most widely employed food enhancers. Although the umami compound, controversy persists regarding the effects of MSG intake on body weight. Chronic MSG intake may result in excessive body weight gain and obesity. Consumption of MSG result in organ damage, cardiovascular disease, oxidative stress, and also risk factors for obesity. This study aims to determine the effect of oral MSG on obesity in adult male Wistar rats (Rattus norvegicus).Methods: This true experimental study used the post-test control group design. Twenty-four adult male Wistar rats were randomly divided into four groups: control (received distilled water), Group 1 (MSG 0.378 mg/gr BW), Group 2 (0.756 mg/gr BW) and Group 3 (1.512 mg/gr BW). The obesity parameter was obtained by the Lee index. Kruskal-Wallis test follows by Mann-Whitney test were used to compare the Lee index between groups.Results: Lee’s index mean for each group was 358.4%, 314.1%, 287.8%, and 320.9%, respectively. The Kruskal Wallis test showed a significant difference in the Lee index between groups (p = 0.043). A follow-up test using Mann-Whitney found a significant difference between group 2 and the control group (p = 0.043, p <0.05). The mean of Lee index of group 2 was 70.51% lower than the control group.Conclusion: This study concluded that Lee index was not increased in MSG-treated rats than in the control group after oral MSG intervention for 21 days.

Citrullus lanatus thumb (Cucurbitaceae) commonly called watermelon is widely consumed in this part of the world as food and medicine. This study was carried out to examine the effect of methanolic extract of watermelon (Citrullus lanatus) rind on prednisolone suppressed immunity in male wistar rats. A total of 20 male wistar rats weighing 150-294g were used in 4 groups with five rats each. Group 1, the control group was given distilled water and feed, Group 2 was given 200 mg/kg body weight of methanolic extract of watermelon rind, Group 3 rats were given 2.5 mg/kg body weight of prednisolone and Group 4 rats were given 2.5 mg/kg body weight of prednisolone and 200 mg/kg body weight of methanolic extract of watermelon rind. Prednisolone and the methanolic extract of watermelon rind were administered orally for a period of 30 days. Blood samples were collected by cardio puncture from the rats for white blood cell (WBC), lymphocyte, and granulocyte and monocyte counts at the end of the experiment.  The data were statistically analysed using one-way ANOVA (Analysis of variance). Data were considered significant at p<0.05. The results obtained showed that methanolic extract of watermelon rind caused a significant increasen in immune function of rats when compared with the control and immune suppressed rats.

Background: Panoramic and cephalometric radiography is very important for diagnosis, treatment plan, and evaluation of orthodontic treatment results. Panoramic and cephalometric radiography are frequently performed at the same time, causing DNA damage and chromosome aberration. Purpose: This study aims to analyse the effect of X-ray exposure in panoramic and cephalometric radiography on micronuclei cell numbers. Methods: Laboratory-based analytical study with 60 healthy-male Wistar rats weighing 200–300 grams divided into 6 treatment groups (n=10). The control group: without radiographic exposure, the treatment group 2: using panoramic radiographic exposure followed by cephalometric, and the treatment group 3: using panoramic radiographic exposure and 24 hours later performed cephalometric radiographic. The unit of analysis was the polychromatic erythrocytes of mice cell, were examined 24 hours and 48 hours after irradiation had been finished. The polychromatic erythrocytes were examined using May-Gruenwald-Giemsa staining and 100x magnification under a microscope with 2000 cells per view. Data obtained were analysed using the SPSS 20 version software. The mean and standard deviations were calculated for each clinical parameter, and a one‐way ANOVA statistical test of significance was used. Statistical significance was set at p<0.05. Results: The analysis showed a significant increase (p<0.05) in the number of micronucleus in groups that used panoramic radiographic exposure followed by cephalometric. Conclusion: X-ray radiation can increase the number of micronucleus in polychromatic erythrocyte cells in rats.
