ABSTRACT Goat milk powder is a nutritious and easy-to-store product that is highly favored by consumers. However, the presence of contaminating bacteria and their metabolites may significantly affect the flavor, solubility, shelf life, and safety of the product. To comprehensively and accurately understand the sanitary conditions in the goat milk powder production process and potential threats from bacterial contamination, a combination of Pacific Biosciences single molecule real-time sequencing and droplet digital PCR was used to evaluate bacterial contamination in seven goat milk powder samples from three dairies. Ten phyla, 119 genera, and 249 bacterial species were identified. Bacillus, Paenibacillus, Lactococcus, and Cronobacter were the primary genera. Bacillus cereus, Lactococcus lactis, Alkaliphilus oremlandii, and Cronobacter sakazakii were the dominant species. With droplet digital PCR, 6.3 × 104 copies per g of Bacillus cereus and 1.0 × 104 copies per g of Cronobacter spp. were quantified, which may increase the risk of food spoilage and the probability of foodborne illness and should be monitored and controlled. This study offers a new approach for evaluating bacterial contamination in goat milk powder and supplies a reference for the assessment of food safety and control of potential risk, which will be of interest to the dairy industry.

Abstract The objective of this study was to establish a method for detecting fatty acid esters of chloropropanols [including 2-monochloropropane-1,3-diol (2-MCPD) ester and 3-monochloropropane-1,2-diol (3-MCPD) ester] in infant milk powder using isotope dilution and GC-MS. The fat fraction in infant milk powder samples was extracted and treated with sodium methylate–methanol to cleave the ester bonds of the 2- and 3-MCPD esters. The resultant 2- and 3-MCPDs in the samples were purified with matrix solid-phase liquid–liquid extraction and derivatized with heptafluorobutyryl imidazole for GC-MS analysis. Standard samples of deuterium isotope–labeled 2- and 3-MCPD palmitic acid double esters and stearic acid double esters were used as the internal standards. We also detected 2- and 3-MCPD ester contents in 88 commercial samples of infant formula milk powder using this system. The detection system we established showed a good linearity of 2- and 3-MCPD ester contents in serially diluted standard solutions within the concentration range 25−1000 μg/L, with r2 > 0.9995 and an LOD of 30 μg/kg for both of the MCPD esters. The recoveries of the two MCPD esters spiked at 25, 50, 100, and 200 μg/kg in blank infant formula milk ranged from 98.2 to 110.5%, with RSDs <4.8%, suggesting good accuracy and reliability of this method. In the 88 commercial infant formula milk powder samples, the mean content of 2-MCPD ester was 41 μg/kg (0–52 μg/kg) and that of 3-MCPD was 185 μg/kg (0–316 μg/kg). Based on these results, we estimated the exposure levels of the two MCPD esters in infants fed on formula milk, and the results indicate potential health risks of consuming formula milk products contaminated by the two MCPD esters.

In this paper, a rapid and sensitive fluorescent aptasensor for the detection of aflatoxin M1 (AFM1) in milk powder was developed. Graphene oxide (GO) was employed to quench the fluorescence of a carboxyfluorescein-labelled aptamer and protect the aptamer from nuclease cleavage. Upon the addition of AFM1, the formation of an AFM1/aptamer complex resulted in the aptamer detaching from the surface of GO, followed by the aptamer cleavage by DNase I and the release of the target AFM1 for a new cycle, which led to great signal amplification and high sensitivity. Under optimized conditions, the GO-based detection of the aptasensor exhibited a linear response to AFM1 levels in a dynamic range from 0.2 to 10 μg/kg, with a limit of detection (LOD) of 0.05 μg/kg. Moreover, the developed aptasensor showed a high specificity towards AFM1 without interference from other mycotoxins. In addition, the technique was successfully applied for the detection of AFM1 in infant milk powder samples. The aptasensor proposed here offers a promising technology for food safety monitoring and can be extended to various targets.

The study was undertaken to evaluate some chemical parameters of powder milk available in Mymensingh town. Powder milk samples of six different brands namely Kwality, NIDO, Diploma, Anchor, Farmland & Starship were collected from local market. Powder samples were analyzed to know the chemical (PH, acidity, fat, protein, lactose, ash, moisture, solids-not-fat, total solids) characteristics. PH, acidity of all samples was almost similar to the cow milk  when the dried milk was reconstituted. PH ranged from 6.6-6.8, acidity from 0.10-0.15, fat from 26.198-27.89 g/100 g,  protein from 25.22-27.01 g/100g, lactose from 36.63-37.65 g/100g, ash from 5.34-5.48 g/100g, moisture from 3.36-4.48 g/100g, solids-not-fat from 67.99-70.07 g/100g, total solids from 95.51-96.63 g/100g, among the brands of powder milk. Statistical analysis showed significant variation for (PH and acidity) among different powder milk brands. chemical parameters in all brands of milk powder was found as per recommended standard. Abstract A collaborative study involving 19 laboratories was performed to validate motility enrichment on modified semisolid Rappaport-Vassiliadis (MSRV) medium for rapid detection of motile Salmonella in dried milk products. The MSRV method was compared with the AOAC culture method for detection of Salmonella in nonfat milk powder, whole milk powder, whey powder, casein powder, and butter milk powder. Samples were artificially inoculated with Salmonella at 2 levels of contamination. Un-inoculated control samples were included for each type of product. The sensitivity rates were 100% for the MSRV method and 99.0% for the AOAC culture method, while the specificity rate was 100.0% for both methods. Only for the samples of whey powder, which were inoculated with H2S negative S. tennessee, was there a significant difference in the proportion of samples positive by MSRV and the culture procedure. The MSRV method for detection of motileSalmonella in dried milk products has been adopted first action by AOAC INTERNATIONAL.
