Lemon processing generates thousands of tons of residues that can be preserved as flours by thermal treatment to obtain phenolic compounds with beneficial bioactivities. In this study, the effect of different drying temperatures (40, 50, 60, 70, 80, 90, 100 and 110 °C) on the Total Phenolic Content (TPC), antioxidant and antimicrobial activities of phenolic compounds present in Citrus. lemon (L.) Burn f waste was determined. Identification and quantification of phenolic compounds were also performed by UPLC-PDA and UPLC-ESI-MS analysis. Eriocitrin (19.79–27.29 mg g−1 DW) and hesperidin (7.63–9.10 mg g−1 DW) were detected as the major phenolic compounds in the flours by UPLC-PDA and confirmed by UPLC-ESI-MS. Antimicrobial activity determined by Minimum Inhibitory Concentration (MIC) against Salmonella typhimurium, Escherichia coli and Staphylococcus aureus was observed. Accordingly, a stable functional flour as a source of bioactive phenolic compounds obtained from lemon residues at 50 °C may be produced as a value-added product useful in various industrial sectors.

Background: Hydnum repandum, Cantharellus cibarius, Ramaria fennica, Boletus edulis, and Craterellus cornucopioides are all wild edible mushroom species from the Kastamonu province. The aim of this study was to investigate antioxidant properties and amino acid contents of these mushrooms. Materials and Methods: The mushrooms were analyzed for their free amino acid compositions using a high performance Amino Acid analyzer (Biochrom 30). Also, antioxidant activity and total phenolic concentrations of five different mushroom species were studied by using spectrophotometric methods. Results: The mushrooms contained 17 amino acids (g/100 g): Glutamic acid (2.56–1.11), Alanine (1.49–0.54), Arginine (1.62–6.77), Aspartic acid (1.45–0.81), Leucine (1.08–0.64), Methionine (1.05–0.06), Valine (1.05–0.66), Lysine (1.01–0.57), Serine (0.68–0.38), Cysteine (0.88–0.11), Isoleucine (0.61–0.39), Glycine (0.82–0.35), Threonine (0.82–0.44), Phenylalanine (0.66–0.42), Proline (0.60–0.47), Tyrosine (0.58–0.38), and Histidine (0.48–0.22). The total free amino acid (TAA) contents ranged from 169.2 mg/g in Boletus edulis to 89.1 mg/g in Hydnum repandum. These five different mushroom species contain eight essential amino acid species except tryptophan. The antioxidant activity of mushroom extracts was expressed as the percentage of DPPH radical inhibition and IC50 values (mg/mL). The percentage of inhibition ranged from 2.38% to 88.05% and IC50 values ranged from 0.03 to 13.98 mg/mL. The total phenolic content ranged from 0.66 to 7.74 mg/g of dry mushroom extract, expressed as gallic acid equivalents. Conclusion: Methanolic extract of Boletus edulis showed the highest phenolic content and strong antioxidant activity. As a result, the significant linear correlation between the values for the total phenolic content and antioxidant activity of mushroom extracts was corroborated in all of the investigated mushroom species, with the exception of Craterellus cornucopioides.

The molecular mass of the galactose—H+ symport protein GalP, as its histidine-tagged derivative GalP(His)6, has been determined by electrospray MS (ESI-MS) with an error of <0.02%. One methionine residue, predicted to be present from the DNA sequence, was deduced to be absent. This is a significant advance on the estimation of the molecular masses of membrane-transport proteins by SDS/PAGE, where there is a consistent under-estimation of the true molecular mass due to anomalous electrophoretic migration. Addition of a size-exclusion chromatography step after Ni2+-nitrilotriacetate affinity purification was essential to obtain GalP(His)6 suitable for ESI-MS. Controlled trypsin, trypsin+chymotrypsin and CNBr digestion of the protein yielded peptide fragments suitable for ESI-MS and tandem MS analysis, and accurate mass determination of the derived fragments resulted in identification of 82% of the GalP(His)6 protein. Tandem MS analysis of selected peptides then afforded 49% of the actual amino acid sequence of the protein; the absence of the N-terminal methionine was confirmed. Matrix-assisted laser-desorption ionization MS allowed identification of one peptide that was not detected by ESI-MS. All the protein/peptide mass and sequence determinations were in accord with the predictions of amino acid sequence deduced from the DNA sequence of the galP gene. [ring-2-13C]Histidine was incorporated into GalP(His)6in vivo, and ESI-MS analysis enabled the measurement of a high (80%) and specific incorporation of label into the histidine residues in the protein. MS could also be used to confirm the labelling of the protein by 15NH3 (93% enrichment) and [19F]tryptophan (83% enrichment). Such MS measurements will serve in the future analysis of the structures of membrane-transport proteins by NMR, and of their topology by indirect techniques.

Objective: Aegle marmelos (L.) Correa has been widely used in Indian traditional medicine and has many reported pharmacological activities. The aim of this research was to formulate solid lipid nanoparticles (SLNs) of Aegle oil (AO) that enhanced the beneficial antimicrobial activity of the oil. Methods: The chemical composition of Aegle leaf essential oil was analysed by GC-MS. Additionally, a phytochemical study of A. marmelos methanolic leaf extract was conducted using Folin-Ciocalteu colorimetric assay for determination of total phenolic content as well as ultra-performance liquid chromatography coupled to tandem mass spectrometry (UPLC-ESI-MS-MS) analyses for identification of individual components. Six formulations of AO-loaded SLNs (AO-SLNs) were prepared by a double emulsification method. The particle size, zeta potential (ZP), polydispersibility index (PDI) and drug encapsulation efficiency (EE) of the SLNs were determined. The morphology of the SLNs was observed by transmission electron microscopy (TEM). The antimicrobial activity of AO and AO-SLNs was assessed using disc diffusion method. Results: Thirty-two compounds were identified in the Aegle oil, of which Δ-carene and α-phellandrene were the most abundant (48.14% and 34.14%, respectively). The estimated total phenolic content was 968mg gallic acid equivalents (GAE)/g, while UPLC-ESI-MS-MS led to the tentative characterization of thirteen metabolites. The SLNs showed ZP, PDI and EE 125 ± 0.22nm, –37.85, 0.282, and 92%, respectively. AO and AO-SLNs showed significant antimicrobial activity, and the SLNs could sustain the release of AO from their gel vehicles. Conclusion: Our results provide evidence for the application of AO-SLNs in topical and transdermal delivery systems.

AbstractExtrusion-cooking of plant materials may enhance antioxidant activity and improve health benefits. Selected antioxidant polyphenols in extruded corn gruels enriched with different amounts of linden flowers were determined by LC-ESI-MS/MS and quality characteristics were determined.Phenolic content increased with Tiliae inflorescentia addition and was not decreased by high-temperature extrusion. Linden flower incorporation into instant gruels should be limited to 10% to retain acceptable sensory properties.

Abstract Biostimulants are commonly used in horticulture, primarily to increase yield quantity and quality, as well as plant tolerance to stress. The purpose of the study was to determine the effect of amino acid-based biostimulants and amino acids in combination with Ascophyllum nodosum filtrate, applied during the growing season on the phenolic content in broccoli heads both after harvest and after long storage in a cold store. The seedlings were planted in mid-July, and heads were harvested from September to October in each year of the study. Ascophyllum nodosum filtrate at a concentration of 1% was used during seedling production. The plants were watered with biostimulants twice, in the fourth and fifth week after sowing. They were also sprayed with amino acid-based biostimulants (1.5 dm3 ha-1) three times, i.e. after the second, fourth and sixth week after planting. The biostimulants were not used in the control treatment. After harvest, the heads were stored for three weeks in a cold store at 1-2°C and 95% RH. Chemical analyses of total phenolic, caffeic, ferulic and sinapic acids, and of quercetin and kaempferol were carried out on samples of fresh broccoli and after the first, second and third week of storage. It was found that the application of biostimulants resulted in a significant increase in the total phenolic content, sinapic acid content, as well as quercetin content. Both in the control treatment and in the plants treated with the biostimulants, the concentration of all the tested phenolic compounds increased with the duration of storage.
