Reduction of colony diameter using two DMI-fungicides and one strobilurine fungicide was evaluated in the <i>Septoria tritici</i> population. In an <i>in vitro</i> assay, 57 strains of <i>S. tritici</i> originating from different parts of the Czech Republic were grown on agar plates containing different concentrations of fungicides. The mean EC<sub>50</sub> values of strains were analysed for flusilazole (Capitain), metconazole (Caramba) and one strobilurine fungicide: azoxystrobin (Amistar). ED 50 values for metconazole were low. There were highly significant correlation in reaction of pathogen strains to metconazole and flusilazole. Some isolates showed significant higher level of resistance to azoxystrobin. The data will be used in a continuing survey of resistance development in the <i>S. tritici</i> population originating from the territory of the Czech Republic.

The baseline sensitivity of 55 isolates of Sclerotinia sclerotiorum, collected from oilseed rape in 6 regions of the Czech Republic, to selected fungicides was determined during the period 2013–2015. One single-component fungicide – Horizon (tebuconazole), and four multicomponent fungicides – Pictor (boscalid, dimoxystrobin), Efilor (boscalid, metconazole), Prosaro 250 EC (prothioconazole, tebuconazole), and Propulse (fluopyram, prothioconazole), were chosen as these are commonly used locally. The effect of each fungicide on the in vitro pathogen radial mycelial growth and EC<sub>50 </sub>values for the respective fungicides were determined. The following MIC values were estimated; for the fungicides Horizon 250 EW, Efilor, and Propulse the mean MIC values ranged between 0.125 and 0.250 µl/ml, for Prosaro 250 EC ranged between 0.0625 and 0.125 µl/ml, and for Pictor ranged from 0.00781 to 0.01562 µl/ml. No strains of S. sclerotiorum resistant to the tested fungicides were detected and the growth of all isolates was fully inhibited at concentrations corresponding to their registered dose rates. The highest fungicidal efficacy on the collected S. sclerotiorum isolates was recorded for Pictor, followed by Prosaro 250 with an EC<sub>50</sub> value 0.05856 µl/ml and then the remaining fungicides Propulse, Efilor, and Horizon 250 EW (EC<sub>50</sub> values 0.07277, 0.07221, and 0.08519 µl/ml, respectively).

Cercospora leaf spot (CLS), caused by the fungal pathogen Cercospora beticola, is the most important foliar pathogen of sugar beet worldwide. Extensive reliance on fungicides to manage CLS has resulted in the evolution of fungicide resistance in C. beticola worldwide, including populations in the Czech Republic. One important class of fungicides used to manage CLS is the sterol demethylation inhibitors (DMI). The aim of our study was to assess DMI resistance in C. beticola from the Czech Republic and elucidate the molecular basis of DMI resistance in this population. A total of 50 isolates were collected in 2018 and 2019 from the major sugar beet growing regions of the Czech Republic and assessed for in vitro sensitivity to the DMI fungicides propiconazole, prochloraz, and epoxiconazole. These analyses identified three strains that exhibited 50% effective concentration (EC50) values > 1.0 μg mL–1 against respective fungicides, which were therefore considered resistant. In contrast, strains that exhibited lowest EC50 values were considered sensitive. To explore the molecular basis of resistance in these three strains, the cytochrome P450-dependent sterol 14α-demethylase (Cyp51) gene was sequenced. Sequence analysis identified a Y464S mutation in all three resistant strains. To assess whether Cyp51 gene expression may play a role in DMI resistance, selected strains were grown in vitro with and without fungicide treatment. These analyses indicated that Cyp51 gene expression was significantly induced after fungicide treatment. Thus, we conclude that Y464S point mutation along with induced Cyp51 gene overexpression is likely responsible for resistance against DMI fungicides in C. beticola from the Czech Republic.

Araneofauna of vineyards is relatively known in Central Europe but we have a lack of knowledge about araneofauna which occur directly on the vine plants. Our investigation was focused on spiders which live on vine plants, especially on the vine plants trunks. We investigated spiders in six vineyards in southern Moravia (Šatov, Mikulov, Popice, Morkůvky, Nosislav and Blučina). Vineyards were under different soil management, traps were placed on different parts of particular locality (terraced and plain) and all localities were under integrated pest management. We employed two types of cardboard traps for spider collecting during whole vegetation season. Altogether, we collected 21 spider species which belong to seven families. The most important species was Marpissa nivoyi (Lucas, 1836), which is mentioned in the Red List as vulnerable (VU) and Sibianor tantulus (Simon, 1868) which had unknown distribution in the Czech Republic. The other very interesting result is that the most common species is myrmecomorph Synageles venator (Lucas, 1836), which is scarcely recorded in such huge numbers as we documented in our study.

ABSTRACT Combination therapy may enhance imipenem/cilastatin/relebactam’s (I/R) activity against Pseudomonas aeruginosa and suppress resistance development. Human-simulated unbound plasma concentrations of I/R at 1.25 g every 6 h (h), colistin at 360 mg daily, and amikacin at 25 mg/kg daily were reproduced alone and in combination against six imipenem-nonsusceptible P. aeruginosa isolates in an in vitro pharmacodynamic model over 24 h. For I/R alone, the mean reductions in CFU ± the standard errors by 24 h were −2.52 ± 0.49, −1.49 ± 0.49, −1.15 ± 0.67, and −0.61 ± 0.10 log10 CFU/ml against isolates with MICs of 1/4, 2/4, 4/4, and 8/4 μg/ml, respectively. Amikacin alone also resulted in 24 h CFU reductions consistent with its MIC, while colistin CFU reductions did not differ. Resistant subpopulations were observed after 24 h in 1, 4, and 3 I/R-, colistin-, and amikacin-exposed isolates, respectively. The combination of I/R and colistin resulted in synergistic (n = 1) or additive (n = 2) interactions against three isolates with 24-h CFU reductions ranging from −2.62 to −4.67 log10 CFU/ml. The combination of I/R and amikacin exhibited indifferent interactions against all isolates, with combined drugs achieving −0.51- to −3.33-log10 CFU/ml reductions. No resistant subpopulations were observed during I/R and colistin combination studies, and when added to amikacin, I/R prevented the emergence of amikacin resistance. Against these six multidrug-resistant P. aeruginosa, I/R alone achieved significant CFU reductions against I/R-susceptible isolates. Combinations of I/R plus colistin resulted in additivity or synergy against some P. aeruginosa, whereas the addition of amikacin did not provide further antibacterial efficacy against these isolates.

AbstractBackground:We investigated time trends in age, gender, growth hormone (GH) dose and height standard deviation score (SDS) in children with GH deficiency (GHD), born small for gestational age (SGA) or with Turner syndrome (TS) starting GH treatment.Methods:Data were obtained from children enrolled in the NordiNet®International Outcome Study (IOS) between 2006 and 2015 in the Czech Republic, France, Germany, Serbia and Montenegro (all indications), and Switzerland and the UK (GHD only). Trends were analyzed by linear regression. Patients were divided by age into early-, medium- or late-start groups in three different time periods.Results:Approximately one-third of children starting treatment for GHD were girls, with no apparent increase in proportion over time. The mean baseline age for starting treatment decreased significantly (p<0.001) for both GHD and SGA in the Czech Republic and Germany. In the other countries studied, over 40% of children started treatment for GHD and SGA late (girls >10, boys >11 years) between 2013 and 2015. The mean baseline GH doses were largely within recommended ranges for GHD and SGA, but below the lowest recommended starting dose for TS in almost every year since 2011 except in France.Conclusions:Approximately one-third of children starting treatment for GHD were girls. Between 2013 and 2015, more than 40% of children started treatment for GHD and SGA late except in Germany and the Czech Republic. TS patients received below-recommended doses. These results highlight the need for earlier identification of short stature in children, particularly girls, and for dose optimization in TS.

The black walnut (Juglans nigra L.), an introduced species into the Czech Republic, is planted for its production of valuable timber. A systematic study of the black walnut growth rate at different localities and consequently the creation of standard volume tables under Central European conditions therefore appear to be of high relevance. The aim of our study was to reveal the black walnut growth pattern in its pure stands in two riparian forests along the Morava and Svratka River (Str&aacute;žnice locality and Židlochovice locality, respectively). To describe and to compare relationships among diameter at breast height (dbh), tree height and age, we used a forest management plan and measured 573 and 670 trees in pure sample stands at Str&aacute;žnice and Židlochovice, respectively. We found out that the measured mean DBH and mean height were consistently higher in Str&aacute;žnice, however the relationships of DBH to height, age to DBH, and age to height showed the same pattern at both localities. The mean heights of trees were 16.3, 24.1, 28.7, 31.9, and 34.5 m in 20, 40, 60, 80, and 100 year age classes and were consistently taller in comparison with other, mainly European, regions. Therefore our results suggest the high potential of black walnut in timber production in riparian forests in the Czech Republic, mainly in the South Moravian region.

The efficacies of the new strobilurin fungicide kresoxim-methyl for the protection of apple leaves from infection by baseline populations of Venturia inaequalis were uniform across five major apple growing regions in North America. The mean ED50 value determined for 25 populations was 0.35 μg ml-1, with values ranging from 0.11 μg ml-1 to 0.75 μg ml-1. The mean level of scab control achieved at the kresoxim-methyl dose of 4 μg ml-1 was 93%. For one of the five orchards sampled in each region, kresoxim-methyl sensitivities of germinating conidia were determined. Sensitivities of 250 isolates were broadly distributed, with ED50 values ranging from 0.003 μg ml-1 to 0.14 μg ml-1 and a mean of 0.02 μg ml-1. This broad range of in vitro sensitivities was not reflected for the in vivo efficacy of kresoxim-methyl in the protection of apple leaves from scab infections. The discrepancy between in vivo and in vitro sensitivities implies that in vivo tests are more useful for the monitoring of kresoxim-methyl sensitivities of orchard populations. Because it can be expected that only isolates resistant under both test conditions will be prone to future selection, such isolates will contribute to increased frequencies of the least sensitive isolates described in this baseline study. Testing of in vitro isolate sensitivities will, therefore, provide an additional tool in the monitoring of kresoxim-methyl resistance development.

In 2014, 18 hospitals in the Czech Republic participated in a survey of the incidence of Clostridium difficile infections (CDI) in the country. The mean CDI incidence was 6.1 (standard deviation (SD):7.2) cases per 10,000 patient bed-days and 37.8 cases (SD: 41.4) per 10,000 admissions. The mean CDI testing frequency was 39.5 tests (SD: 25.4) per 10,000 patient bed-days and 255.8 tests (SD: 164.0) per 10,000 admissions. A total of 774 C. difficile isolates were investigated, of which 225 (29%) belonged to PCR ribotype 176, and 184 isolates (24%) belonged to PCR ribotype 001. Multilocus variable-number tandem repeat analysis (MLVA) revealed 27 clonal complexes formed by 84% (190/225) of PCR ribotype 176 isolates, and 14 clonal complexes formed by 77% (141/184) of PCR ribotype 001 isolates. Clonal clusters of PCR ribotypes 176 and 001 were observed in 11 and 7 hospitals, respectively. Our data demonstrate the spread of two C. difficile PCR ribotypes within 18 hospitals in the Czech Republic, stressing the importance of standardising CDI testing protocols and implementing mandatory CDI surveillance in the country.

In 2001&ndash;2004 virulence of the wheat leaf rust population in the Czech Republic was studied on Thatcher near-isogenic lines with Lr1, Lr2a, Lr2b, Lr2c, Lr3a, Lr9, Lr10, Lr11, Lr15, Lr17, Lr19, Lr21, Lr23, Lr24, Lr26 and Lr28. Samples of leaf rust (180 in total) were obtained from different parts of the Czech Republic. Resistance genes Lr9 and Lr19 were effective to all tested isolates like in the previous years. Unlike the previous years virulence on Lr24 and Lr28 was observed. Relatively effective were also Lr1, Lr2a, Lr24, Lr28 and Lr2b. Other Lr genes were defeated by the majority of the tested samples. Our results transformed to the numbers of physiologic races indicate that race 61SaBa prevailed like in the previous years, followed by races 61, 2, 12SaBa, 2SaBa, 14, 77SaBa, 12, 57, 6, 53, 53SaBa, 77, and 14SaBa. Twenty-one winter wheat cultivars registered in 2001&ndash;2004 were tested with 8 leaf rust isolates. Out of them 15 showed resistance at least to one rust isolate. High resistance recorded in field trials for cvs Batis and Ilias, which were susceptible to all tested rust isolates at the seedling stage, demonstrates the importance of field (partial) resistance. &nbsp;
