Abstract The Calendula officinalis L. (Asteraceae) antimicrobial action has already been scientifically proved in other countries. The aim of this study was to assess the antibacterial action of the Callendula officinalis which is grown in Paraná - Brasil, under extraction conditions at ethanol 50%, with filtration and successive partitions with hexane, dichloromethane, ethyl acetate and butane, contrasting with strains Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC 25923, Staphylococcus epidermidis ATCC 12228 and Bacilus cereus, by the difusion technique in solid media by cavity and afterwards by the plaque dilution method. The results showed that some fractions of the extract inhibited the gram-prositive bacteria, with activity in contrast with Staphylococcus and Bacillus and non-existing in gram-negative, also indicating that the active ingredient responsible for the antibacterial action observed can be isolated from the Hexanic and DCM fractions under the extraction conditions proposed in this study.

There is a need for new antimicrobial systems due to increased global resistance to current antimicrobials. Pomegranate rind extract (PRE) and Zn (II) ions both possess a level of antimicrobial activity and work has previously shown that PRE/Zn (II) in combination possesses synergistic activity against Herpes simplex virus and Micrococcus luteus. Here, we determined whether such synergistic activity extended to other, more pathogenic, bacteria. Reference strains of methicillin-resistant Staphylococcus aureus (MRSA), Staphylococcus epidermidis, Escherichia coli, and Pseudomonas aeruginosa were cultured and subjected to challenge by PRE, Zn (II), or PRE + Zn (II), in time-kill assays. Data were obtained independently by two researchers using different PRE preparations. Statistically significant synergistic activity for PRE + Zn (II) was shown for all four bacterial strains tested compared to untreated controls, although the extent of efficacy and timescales varied. Zn (II) exerted activity and at 1 h, it was not possible to distinguish with PRE + Zn (II) combination treatment in all cases. PRE alone showed low activity against all four bacteria. Reproducible synergistic bactericidal activity involving PRE and Zn (II) has been confirmed. Potential mechanisms are discussed. The development of a therapeutic system that possesses demonstrable antimicrobial activity is supported which lends itself particularly to topical delivery applications, for example MRSA infections.

Įvadas. Akies infekcijos rizika dažnėja didėjant kontaktinių lęšių nešiotojų skaičiui. Kontaktinių lęšių tirpalai – priemonės, skirtos kontaktinių lęšių skalavimui, nešvarumų valymui, apnašų šalinimui, dezinfekcijai (mikroorganizmų sunaikinimui), drėkinimui bei laikymui. Kontaktinių lęšių tirpalų sudėtis nulemia skirtingas jų antimikrobines savybės. Tyrimo tikslas. Nustatyti kontaktinių lęšių skysčių antimikrobinį veikimą skirtinguose jų poveikio etaloninėms bakterijų kultūroms laiko intervaluose. Tyrimo medžiaga ir metodai. Mikrobiologinio tyrimo metu nustatytas kontaktinių lęšių tirpalų antimikrobinis poveikis bakterijoms – Staphylococcus aureus ATCC 25923, Staphylococcus epidermidis ATCC 12228, Escherichia coli ATCC 25922, Klebsiella pneumoniae ATCC 13883, Pseudomonas aeruginosa ATCC 27853, Bacillus cereus ATCC 11778. Bakterijų kultūros buvo veikiamos skirtingais kontaktinių lęšių tirpalais 0,5 val., 1 val., 2 val., 4 val., 6 val., 8 val. ir po to nustatytas likęs gyvybingų bakterijų kiekis tiriamajame tirpale. Rezultatai.

Abstract With the increase of general knowledge and the advancement of science and technology, antibacterial substances were used more than antibiotics. In our current study, the antibacterial virtues of CFO/BiOI nanocomposite were investigated due to its high importance on Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Bacillus cereus. MIC, MBC , Disk Diffusion and IC50 tests Cefalotin (CF), Amoxicillin (AMX), Gentamicin (GM), Trimethoprim-sulphamethoxazole (SXT) and Ceftriaxone (CRO) antibiotics in concentration 30W, 10i, 10t , 25h and 30 were used to find the antibacterial properties of the synthesized nanocomposite, respectively. For the synthesis of nanocomposites polyethylene glycol (PEG) and sulfonic acid was used as a solvent. It is noteworthy that the synthesis was performed by heat dissolution method without the presence of surfactant. Also, various techniques such as X-Ray Diffraction(XRD), Scanning Electron Microscope (SEM), High resolution mapping and Energy Dispersive X-ray spectroscopy (EDAX) have been used to determine the properties of produced nanocomposites. SEM test results showed that the formed nanoparticles were globular and their size was limited area of 22 to 34 nm. The results showed CFO / BiOI nanocomposite exhibits strong significant biological activity against Bacillus cereus. The results of MBC (Minimum Bactericidal Concentration) and MIC (Minimum Inhibition Concentration) tests for CFO/BiOI nanocomposites on bacteria were examined in the range of 0.12-0.48 mg/ml and 0.06 to 0.24 mg/ml respectively. According to the results, the minimum IC50 value was determined at a concentration of 0.061 mg/ml. On the other hand, the most resisting and susceptible bacteria in this method were Pseudomonas aeruginosa and Bacillus cereus, respectively. These findings are identical to those of a prior study on CoFe2O4 nanoparticles antibacterial properties. MBC of the nanocomposites, 50 µl from all the tubes that showed no obvious bacterial growth were distributed on BHI agar plates and incubated for 24 h at 37 ◦C. The MBC endpoint is defined as the lowest concentration which killed 98% of the bacterial population.
