<p>Present study was designed to investigate the effects of 200 mg/ml solvent/Kg of body weight of <em>Bauhinia variegata’s</em> leaf extract on neuromuscular coordination, locomotory and exploratory behavior of adult female albino mice. Six week old female albino mice were used as experimental animals. Mice were either orally supplemented with 200 mg/ml/Kg of body weight <em>Bauhinia variegata’s</em> leaf extract or with saline solution [0.9%] for 7 days. Behavioral observations were made by applying a series of neurological tests [Rota rod, Elevated plus maze, Light and dark box, Open field and Novel object test]. Exploratory and locomotory behavior during Open Field test was improved in leaf extract treated mice as they had more means speed [P = 0.01], rotations [P = 0.001], clockwise rotations [P = 0.004] and anti-clockwise rotations [P = 0.001] than control mice. <em>Bauhinia variegata’s</em> leaf extract treated mice had higher transition frequency [P = 0.002] and spent more time in light compartment during light dark box test than saline treated controls. <em>Bauhinia variegata’s</em> leaf extract treated mice covered more distance [P = 0.033] with higher speed [P = 0.028] compared to control mice during Elevated Plus Maze test. <em>Bauhinia variegata’s</em> leaf extract improves the locomotory and exploratory behavior in adult female albino mice. Leaf extract of <em>Bauhinia variegate </em>can be further explored to be used as a therapeutic agent for the treatment of neurological ailments.</p>

Background: Puberty is a developmental transition in which an estrogenic surge occurs, mediating the release of xenoestrogens, like aluminium. Aluminium’s effect on anxiety in rodents at the different developmental stages is inconsistent. Aims: This study aimed at investigating the effect of the metalloestrogenic property of aluminium on anxiety-like behavioral changes in prepubertal and young adult female rats. Objective: Considering this aim, our objective was to evaluate the anxiety-like behavior by the elevated plus maze in prepubertal and young adult female rats with or without acute exposure to aluminium. Methods: To address this property of aluminium, 5mg/Kg body weight (Al-5) and 10 mg/Kg body weight (Al-10) of aluminium was administered intraperitoneally to female rats at two developmental stages, prepubertal (PP; n = 8 for each dose) and young adult (YA; n = 6 for each dose) for two weeks. Post-treatment, three days behavioral assessment of the rats was done employing elevated plus maze. Results: Reduced escape latency was seen in Al-5, Al-10 pre-pubertal rats, and Al-5 young-adult rats on day 3. A significant reduction in open arm time was seen in the Al-5 young-adult rats. Aluminium treatment in the pre-pubertal rats reduced their head dipping and grooming. Reduced sniffing, head dipping, and stretch-attended posture in the treated young-adult female rats showed that they had impaired risk-taking tendency. Conclusion: Differential effect on the anxiety-like behavior in the pre-pubertal and young-adult female rats might be due to the metalloestrogenic property of aluminium, acting differently on the two age groups.

Background: Solanum aethiopicum (L.), family Solanaceae, is known as garden eggs. The fruit is used in the treatment of insomnia, diabetes and constipation. Objective: The aim of this study was to evaluate anxiolytic-like activity of fractions obtained from crude methanol extract of Solanum aethiopicum fruit. Method: Acute toxicity testing was conducted according to the OECD guidelines 420 via oral and intraperitoneal routes (ip). n-Hexane (HF), chloroform (CHF), ethyl-acetate (EAF), n-butanol (BF) and residual aqueous fraction (RAF) at doses of 25, 50 and 100 mg/kg ip were experimented using the open field, elevated plus maze, staircase, light dark box and hole-board tests. Results: Results: In open field test, there was statistically significant increase in frequency of central square entry by EAF 25mg/kg, 50mg/kg and 100mg/kg and RAF 25mg/kg, 50mg/kg and 100mg/kg all at p<0.05 compared to distilled water (D/W) group. Elevated plus maze test showed statistically significant increases in open arm entry and duration by CHF 25mg/kg, RAF 25mg/kg and 50mg/kg again at p<0.05. Also, in the staircase test, statistically significant decrease in frequency of rearing with no effect on step climbing was observed by RAF 25mg/kg (p< 0.05) compared to D/W. Light and dark box test produced increased light box entry and duration by EAF 25mg/kg, RAF 25mg/kg and 50mg/ kg at p<0.05. Furthermore, the hole -board test showed statistically significant increases in number of head dips by EAF 50mg/kg and 100mg/kg as well as RAF 25mg/kg, 50mg/kg and 100 mg/kg at p<0.05. Conclusion: The fractions obtained from Solanum aethiopicum fruits possesses anxiolytic-like activity.

Abstract ObjectiveAnxiety behavior is regulated by different neurotransmitter systems. There has been no direct relationship between endocannabinoid and cholinergic systems on anxiety in previous studies. This study investigated the effects of each of these systems separately and simultaneously using Donepezil (Cholinesterase inhibitor) and URB-597 (endocannabinoid degrading enzyme inhibitor) on anxiety-like behavior. MethodEighty-eight male mice were divided into eleven groups (n=8) including control (saline), diazepam (0.3 mg /kg), URB-597 (0.1, 0.3, or 1 mg /kg), donepezil (0.5, 1 or 2 mg/kg) and the combination of the two drugs at low, medium and high doses. All treatments were injected intraperitoneally 30 minutes before the elevated plus maze test. ResultsSeparate administration of URB597, donepezil or diazepam increased the number and time spent of open arms compared to the control group. Concurrent administration of URB and donepezil at low, medium and high doses did not change the number of open arms entries compared to the control group, but they reduced the number of entries to the closed arms. ConclusionsThese results suggest that strengthening any cholinergic or endocannabinoid system has anxiolytic effect similar to diazepam. However, the interaction of these two systems has fewer anxiolytic effects compared to the effects of each alone. It seems that these drugs alone may represent a strategy for the treatment of anxiety disorders.

Obesity is characterized by massive adipose tissue accumulation and is associated with psychiatric disorders and cognitive impairment in human and animal models. However, it is unclear whether high-fat diet (HFD)-induced obesity presents a risk of psychiatric disorders and cognitive impairment. To examine this question, we conducted systematic behavioral analyses in C57BL/6J mice (male, 8-week-old) fed an HFD for 7 weeks. C57BL/6J mice fed an HFD showed significantly increased body weight, hyperlocomotion in the open-field test (OFT) and Y-maze test (YMZT), and impaired sucrose preference in the sucrose consumption test, compared to mice fed a normal diet. Neither body weight nor body weight gain was associated with any of the behavioral traits we examined. Working memory, as assessed by the YMZT, and anxiety-like behavior, as assessed by the elevated plus maze test (EPMT), were significantly correlated with mice fed an HFD, although these behavioral traits did not affect the entire group. These results suggest that HFD-induced obesity does not induce neuropsychiatric symptoms in C57BL/6J mice. Rather, HFD improved working memory in C57BL/6J mice with less anxiety, indicating that an HFD might be beneficial under limited conditions. Correlation analysis of individual traits is a useful tool to determine those conditions.

The present work was carried out to assess the Onosma bracteatum anxiolytic and antidepressant properties. Swiss albino mice (male) were fed orally with hydroalcoholic extract at different doses 50, 100, and 200 mg 1 hour prior to test with the standard diazepam and fluoxetine. Anxiolytic and antidepressant activities were evaluated by using open field, elevated plus maze, force swimming, and tail suspension test. Results of open field test showed an increase in number of line crossing as well as number of rearing in dosage-dependent design. Although results of elevated plus maze test evidently showed antianxiety effect of O bracteatum by increasing the time spent in open arms along with decreasing the time spent in closed arms in dosage-dependent way. For the evaluation of antidepressant effect, O bracteatum diminished the immobility time and expanded mobility time in forced swim model in dosage-dependent way. Likewise, O bracteatum expanded time span of mobility along with diminished immobility time in tail suspension method in dosage-dependent way. Outcome demonstrated that plant at the dose of 200 mg/kg body weight showed significant potential which was similar to that standard diazepam and fluoxetine. Hence, O bracteatum may be used as potent natural psychotherapeutic agent against the mental disorders.

Background: The objective of this study was to evaluate the attenuation of anxiety on acute administration of aqueous extract of Terminalia belerica fruit pulp (AETBFP) by using elevated plus maze test and dark and light arena models.Methods: Thirty Swiss albino mice were divided into five groups, Group I received vehicle (1% Gum acacia suspension, 3ml/kg, orally), Group II received standard drug Diazepam (1mg/kg, orally) and Group III, IV and V received AETBFP 9, 18 and 36 mg/kg, orally respectively. In elevated plus maze test, the mouse was placed on the central platform facing towards open arm. The percentage of time spent and frequency of entries and number of rears in open arm was counted for a period of 5 min. In dark and light arena, the time spent, number of entries and number of rears in light arena was counted for a period of 5 min. The mean±SEM values were calculated for each group. The data was analyzed using one-way ANOVA followed by Dunnet’s multiple comparison tests; p< 0.05 was considered as statistically significant.Results: Significant (p<0.05) reduction in anxiety was noted in experimental animals when given at a dose of AETBFP (36mg/kg), where number of entries and duration of stay in open arm and light arena increased in elevated plus maze and light and dark arena respectively when compared with control animals.Conclusions: Our study reveals that AETBFP at a dose of 36mg/kg has significant attenuation of anxiety in Swiss albino mice.

Anxiety disorder is one of the most common mental ailments exhibited by human. It is characterized by motor tension, sympathetic hyperactivity and apprehension and vigilance syndromes. A part from very few chemical remedies available like benzodiazepine and serotonin modulators, not much treatment options are at hand that safely and effectively alleviate anxiety. The ethanol extract of the fruits of plant Limonia acidissima was evaluated for its anti-anxiety activity in swiss albino mice at dose 200mg/kg and 400mg/kg body weight. Anti-anxiety was assessed by using elevated plus maze (EPM) and light and dark model methods. The ethanolic extract exhibit anxiolytic effect in experimental mice. So it is supports the use of Limonia acidissima as anxiolytic agents. Further investigation should be made to elucidate the active constituent of responsible for the activity.
