In the present investigation, nutritional, anti-nutritional and antioxidant potential of Cucumis sativus forma hardwickii was evaluated for the first time. The results revealed that leaf and fruit of Cucumis sativus f. hardwickii were rich source of ash, carbohydrate, fat, fibre and proteins. In addition, appreciable amounts of Vitamin C and E were also observed. Considerable antinutritional factors in leaf and fruit were noted which can be reduced by different processing methods. In sequential solvent extraction, methanol found to be the best in the extraction of antioxidants, which was supported with the highest antioxidant activities. Among all ¬¬the extracts, it was noted that leaves extracted with methanol had higher amount of phenols and flavonoids than fruit. In addition, the potent radical scavengers mainly phenolics were detected using RP-HPLC where tannic acid, gallic acid and hydroxybenzoic acid were also quantified in leaf and fruit. Superior antioxidant potential may be due to the presence of other bioactive compounds present in fruit. Additional research on bioactive properties of this plant is needed to explore this neglected wild cucumber for nutraceutical purpose and as a functional food.

Cucurbitaceae is a family of health-promoting plants due to their compounds with beneficial effects. The aim of this study was to analyze, for the first time, the chemical composition, the antioxidant activity and the metal chelating properties of fruit juices obtained from four different species of the Cucurbitaceae family cultivated in Romania, namely Momordica charantia, Cucumis metuliferus, Benincasa hispida and Trichosanthes cucumerina. The samples of juice were analyzed by high-performance liquid chromatography (HPLC) and all the four species displayed high levels of the two triterpenes, oleanolic and ursolic acids, and also in phenolic compounds, including catechin, (−)-epicatechin and gallic acid. The juices demonstrated significant antioxidant activity against the free radical 2,2-diphenyl-1-picrylhydrazyl (ranging from 20 to 95%,), a good iron binding ability (ranging from 7.45 ± 0.28% to 86.95 ± 0.97%) and also promising antioxidant potential against the ABTS radical (ranging from 4.97 to 32.60 μETx/mL juice). Our findings raise interesting questions for further research on Cucurbitaceae fruit juices and, consequently, their very good antioxidant potential suggests these fruits should be further explored for their protective effect against oxidative damage. This is the first time the chemical composition and antioxidant activities of fruit juices from these four Romanian Cucurbitaceae varieties have been investigated.

Methylated flavonols form a special group with modulating biological activities in comparison with kaempferol and quercetin. The present study isolated ten compounds including two kaempferol methyl ethers: 5-hydroxy-3,7,4′-trimethoxyflavone (1), 3-hydroxy-5,7,4′-trimethoxyflavone (6); four quercetin methyl ethers: retusin (5-hydroxy-3,7,3′,4′-tetramethoxyflavone) (4), 3,5-dihydroxy-7,3′,4′-trimethoxyflavone (5), 3,4′-dihydroxy-5,7,3′-trimethoxyflavone (7), and 3,5,7,3′,4′-pentamethoxyflavone (9); β-sitosterol (2); 5-hydroxy-1-(4′-hydroxyphenyl)eicosan-3-one (3); p-hydroquinone (8); and vanillic acid (10) from the rhizomes and fruit of Amomum koenigii J.F.Gmel. (Zingiberaceae). Their structures were determined by MS, NMR, and X-ray spectroscopic techniques. Among the methylated flavonols, 1, 4–7, and 9 were isolated for the first time from the rhizomes, while 1, 4, and 5 were isolated from the fruit. Compounds 2, 3, 7, 8, and 10 were reported for the first time from the species. Three main methylated flavonols 1, 4, and 5 were quantitatively analyzed in the rhizomes of A. koenigii by RP-HPLC-DAD; their contents were determined to be 1.81% (1), 1.38% (4), and 1.76% (5). The antimicrobial assay against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Staphylococcus aureus, Aspergillus niger, Fusarium oxysporum, Candida albicans, and Saccharomyces cerevisiae and antioxidant DPPH scavenging test were performed for the isolated methylated flavonols.

We have investigated potentials of flowers of Gardenia jasminoides as a source for antioxidant-rich functional food and nutritional supplement. To this end, chemical composition and the scavenging activities of the flower extracts against 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid), O2-⁃ and ⁃OH were investigated. G. jasminoides flowers were found to be rich in hydrocarbons, terpenoids, phenolics, alcohols, aldehydes, fatty acids, ketones, and esters that are known to be free radical scavengers. Furthermore, extracts of G. jasminoides flowers exhibit scavenging activity for DPPH, ABTS, O2-⁃, and ⁃OH. Therefore, G. jasminoides flowers offer potentials for development of antioxidant-rich functional food and nutritional supplement.

Oil extracted from Persea Americana seed was assayed for its physiochemical properties and antioxidant potential using various standard methods. The oil content of the seed was found to be < 10%. Brownish-red color oil was liquid at room temperature, with specific gravity of 0.91±0.02 g/mL. Other physiochemical parameters determined were; acid value (4.51±0.08 mgKOH/g), %FFA (2.26±0.08), peroxide value (2.40±0.57 mgO2/Kg), ester value (31.26±0.03 mgKOH/g), saponification value (35.76±0.07 mgKOH/g) and iodine value (23.5±0.07). The results of the antioxidant activities of the seed oil showed that the flavonoid content (80.00±1.41 mgQE/g) was ~10 folds higher than the phenolic content (8.27±0.06 mgGAE/g). The DPPH radical scavenging value was found to be 51.54±0.25% with an IC50 value of 4.68±0.02 mg/mL and reducing power with an average absorbance of 0.85±0.01 and an IC50 value of 0.001±0.02 mg/mL. Gallic acid showed better antioxidant activities than the oil studied. The results obtained in this study showed that Persea Americana seed oil has nutritional, industrial as well as medicinal potentials.

Berberis lyceum Royle has such pronounced medicinal values that it is used as cure of many diseases and has exhibited great therapeutic effects among the local communities throughout the world. The present research was carried out to evaluate the quantitative ethnobotanical status and phytochemical analysis of B. lyceum. Regular field trips were arranged to the study area (Shangla District) in August 2017 to October 2019 and interviews with 100 residents (age range: 30 to 50 years) were conducted. The approach adopted for ethnobotanical data was semi-scientific as the inhabitants were not aware about the modern names of some diseases and therefore physician prescriptions were also consulted. Ethnobotanical data were examined using relative frequency of citation and % use value. The % use value of B. lyceum indicates that the people of District Shangla mostly used it for curing of different diseases. In spite of tremendous uses the plant still survived in this area mainly due to the non-accessibility for humans of the mountain tops; otherwise, increase in anthropogenic activities even in these hilly areas poses a threat of the eradication of this plant. To correlate the folkloric uses with its phytochemical composition, HPLC (high performance liquid chromatography) analysis was performed and a total of six phenolic compounds (quercetin, chlorogenic acid, berberine, rutin, mandellic acid, and hydroxy benzoic acid) were identified in its root. As most of the health complications are correlated to oxidative stress therefore in vitro antioxidant activity were also performed using DPPH (2, 2- diphenyl-1-picrylhydrazyl) and ABTS (2, 2′-azinobis-3-ethyl benzo thiazoline -6- sulfonic acid) assays. The observed antioxidant potential may most probably be due to berberine and chlorogenic acid that were present in highest concentration in the analyzed extract. The effectiveness of the selected plant as remedy for a number of diseases (that were pointed out by the local community) may be due to its phytochemical composition especially berberine and chlorogenic acid as oxidative stress is the root cause of many diseases. The plant extract exhibited high antioxidant potential (DPPH IC50 = 165µg/mL; ABTSIC50 = 110µg/mL) in relation to the detected concentration of berberine and chlorogenic acid. It can be inferred from experimental results that the ethnopharmacological efficacy of this endangered species may be due to its phytochemical composition and antioxidant activities. This case study helps to revive the importance of B. lyceum in local communities and emphasizes the need for its conservation.

Seven flavonoids were isolated from the butanol fraction of the methanolic extract of the aerial parts of Cynanchum acutum L. (Asclepiadaceae). All of which have been isolated for the first time from the genus Cynanchum. Their structures were established as quercetin 3-O-β-galacturonopyranoside (1), quercetin 7-O-β-glucopyranoside (2), tamarixtin 3-O-β-galacturonopyranoside (3), kaempferol 3-O-β-galacturonopyranoside (4), 8-hydroxyquercetin 3-O-β-galactopyranoside (5), tamarixtin 3-O-α-rhamnopyranoside (6), and tamarixtin 7-O-α-arabinopyranoside (7) on the basis of their chromatographic properties, chemical and spectroscopic data. The major isolated flavonoids 1, 2 and 3 were found to exhibit significant antioxidant and antidiabetic activities (by measuring blood glucose and insulin levels). This is the first report about the antioxidant and antidiabetic activities of compounds 1 - 3.

Abstract Background: The composition and sequence of amino acids have a prominent influence on the antioxidant activities of peptides. Objective: A series of isolation and purification experiments was conducted to explore the amino acid sequence of antioxidant peptides, which led to its antioxidation causes. Methods: The degreased apricot seed kernels were hydrolyzed by compound proteases of alkaline protease and flavor protease (3:2, u/u) to prepare apricot seed kernel hydrolysates (ASKH). ASKH were separated into ASKH-A and ASKH-B by dialysis bag. ASKH-B (MW < 3.5 kDa) was further separated into fractions by Sephadex G-25 and G-15 gel-filtration chromatography. Reversed-phase HPLC (RP-HPLC) was performed to separate fraction B4b into two antioxidant peptides (peptide B4b-4 and B4b-6). Results: The amino acid sequences were Val-Leu-Tyr-Ile-Trp and Ser-Val-Pro-Tyr-Glu, respectively. Conclusions: The results suggested that ASKH antioxidant peptides may have potential utility as healthy ingredients and as food preservatives due to their antioxidant activity. Highlights: Materials with regional characteristics were selected to explore, and hydrolysates were identified by RP-HPLC and matrix-assisted laser desorption ionization-time-of-flight-MS to obtain amino acid sequences.

Purpose The purpose of this study is to evaluate the content of bioactive pigments in coloured callus of Azadirachta indica and to understand the correlation between the callus colours with their bioactive constituents, antioxidant properties and cytotoxicity. These assessments will yield valuable insight into the use of in vitro-derived pigments for possible use as functional natural colourants. Design/methodology/approach In this study, the authors have successfully developed a protocol to produce leaf-derived callus of various colours with enhanced content of bioactive pigments in A. indica through plant tissue culture. Comparative analysis of the pigments content (chlorophyll, carotenoid, phenolics and anthocyanins) in the coloured callus was conducted, followed by evaluation of its bioactive properties. The antioxidant properties against 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radicals, ferric reducing antioxidant power and cytotox activities of the coloured callus extracts were also reported. Findings Callus of various colours were successfully produced in A. indica through plant tissue culture, and their valuable pigment content and bioactivity were evaluated. The green callus contained the highest amount of anthocyanin, followed by brown and cream callus. The total anthocyanin contents in both the green and brown callus was more than two-fold higher than that in cream callus. Contrasting observation was obtained for total phenolic content (TPC), where the TPC of cream callus was significantly higher than that in brown callus. Nevertheless, the green callus also exhibited the highest TPC. Green callus also contained the highest amount of total chlorophyll and carotenoid, as well as exhibited the highest antioxidant potential, and was found to be the only extract with active cytotox activity against SKOV-3 cells. Correlation analysis revealed that the excellent bioactivity exhibited by the coloured extracts was strongly correlated with the bioactive pigments present in the callus. Research limitations/implications The major bioactive compounds identified in the methanolic extracts of A. indica coloured callus are anthocyanins, phenolics, chlorophylls and carotenoids. Future research work should include improvements in the extraction and identification methods, which may lead to detection and determination of other compounds that could attribute to its bioactivity, to complement the findings of the current study. Practical implications This analysis provides valuable information on the application of plant tissue culture as an alternative source for sustainable production of major pigments with medicinal benefits in A. indica for possible use as functional natural colourants. Originality/value A comparative study on bioactive pigment production in coloured callus from A. indica leaves and its antioxidant potential and cytotoxicity is original. To the best of the authors’ knowledge, this is the first report detailing a comparative evaluation on the production of coloured callus in A. indica and its relative biochemical composition and bioactive properties.

The objective of this study to examine phytochemical analysis of sonchus oleraceus l, (asteraceae) (moliata) extracts which were extracted by four solvents, petroleum ether, chloroform, methanol and distill water and to assess their potential antibacterial against four standard bacteria gram-positive and gramnegative (bacillus subtilis staphylococcus aureu, salmonella typhi, and pscudomona aeruginosa) and antioxidant activities. (moliata) extracts using cold maceration method to extract sonchus oleraceus l, (asteraceae) locally known as (moliata) extracts, where dpph assay and paper disc diffusion assay were employed to evaluate the antioxidant and antibacterial activities respectively. The results showed that tannins, triterpenes, a sterols, flavonoids and cardiac glycosides compounds were present in the sonchus oleraceus l, (asteraceae) lives (moliata) extracts. The methanol extract of leaves showed high antioxidant potential (77± 0.03 % ) and chloroform showed low antioxidant potential activity (4±0.09) compare with propyl gallate as standard (93± 0.010 %) against dpph. Chloroform extract was effective and give moderate zone of inhibition against staphylococcus aureus. Methanol extract was effective and showed moderate zone of inhibition against staphylococcus aureus, bacillus subtilis, salmonella typhi and pseudomonas aeruginosa.

Background: Young ricegrass (Oryza sativa L.) can be introduced as one of functional food product since sprouts have been much interested in this era due to their high nutritive values. Bio-fortification of selenium is one strategy to enhance plant bioactivity. However, the level of selenium used is varied among species of plants, hence, the proper level needs to be explored.Objective: To investigate the influence of selenium bio-fortification on nutritional compositions, bioactive compounds content and anti-oxidative properties of young ricegrass.Methods: Sodium selenite ranging 0, 10, 20, 30 and 40 mg Se/L has been hydroponically bio-fortified into ricegrass then grown for 8 d and investigated the changes of growth characteristics, selenium content, accumulation of bioactive compounds and anti-oxidative properties.Results:  Results revealed that selenium bio-fortified exogenously increased the accumulation of selenium in ricegrass by 529% at 40 mg Se/L treatment without negatively changes in leaves biomass at the day of harvesting. However, root part weight slightly decreased when increased selenium level. Selenium at concentration of 10 and 20 mg Se/L can stimulate the production of phenolic compounds and antioxidant activities in young ricegrass as measured by DPPH, ABTS, FRAP and chelating assay. Conversely, higher level of selenium fortification reduced the accumulation of phenolics in ricegrass may due to pro-oxidant expression.Conclusion: Selenium bio-fortification can be used as a useful technique to improve quality of ricegrass plantation. 10 mg Se/L treatment was an ideal to trigger the synthesis of phenolics which exhibited high antioxidant activities. While, 40 mg Se/L treatment was ultimate for the production of Se plant foods.Keywords: Antioxidant activities; Bio-fortification; Ricegrass; Selenium
