The toxicity of lead acetate (L. A.) concerned to public health disruptor due to its persistence in the environment and it has the adverse influence on the human and animal health as well. It causes physiological,biochemical, and neurological dysfunctions in humans. Histologically it has a negative effect on the liver which is considered one of the major target organs where acts as detoxification machine by elimination the toxic substance from the blood in rich with it.  As well as it affects kidneys that are the two of the most filtering organs. Therefore the present study was aimed to investigate the histopathological effect of L.A. on liver and kidney tissues in male rats. Twenty male rats involved in the study were equally and randomly divided into two groups each of them involved 10 animals. Group I (castrated rats) and Group II (control) each group received 80mg/L of lead acetate dissolved in one liter distilled water by drinking for 15 days. Histological sections showed some alterations including abnormal architecture, cell degeneration, nuclear degeneration, hyperchromatic hepatocytes, immune cells, degeneration in tubules, dilation in sinusoids, dilation in central vein of liver increased bowman's space glomerular atrophy degeneration of tubular cells in liver and kidney tissues of rats in castrated rats from control group. But the size of degenerated tissue was more severe in castrated male rats. It was concluded that the castration process could produce a hypogonadism and decreased testosterone which owns many receptors in kidney and liver may produce adverse influence with L.A. administration.

Albino rats were made hypervitaminotic A by feeding 30,000 iu of vitamin A daily by mouth; the effect of this hypervitaminosis was compared with a similar pair-fed group. Food consumption and body weight gain were reduced. Study of liver and kidney slices shows that the latter manifest no significant increase in oxygen consumption in presence of succinate or acetate. There is slight increase in O2 consumption in brain homogenates. Liver homogenate shows 15.2% inhibition with succinate and 5.6% with ascorbate oxidation. Liver homogenate shows 19.3% inhibition of succinate dehydrogenase activity by Thunberg technique with methylene blue indicator. It is concluded that hypervitaminosis A inhibits liver respiration by affecting the dehydrogenase, or any immediate step following it, and the cytochrome c-cytochrome oxidase end of the succinoxidase system is little affected.

Background and Objectives. Environmental pollution with the different Aluminum (Al) containing compounds has been increased. Liver and kidney are two vital organs targeted by Al accumulation. The aim of this study was to assess the possible protective and curative effects of Lepidium sativum Linn (LS) against Al-induced impairment of liver and kidney in albino rat and to explore the mechanism behind this effect. Materials and Methods. This experimental animal-based study included fifty albino rats divided into five groups, the control, LS-treated (20 mg/kg), AlCl3-treated (10 mg/kg), AlCl3 then LS, and AlCl3 plus LS-treated, simultaneously for 8 weeks. At the end of the experiment, hepatic and renal functions as well as the biomarkers of antioxidants activities were assessed in the serum. Both liver and kidney were dissected out and histopathologically examined. Results. This study showed that administration of AlCl3 caused a significant (p<0.05) reduction in rats body weight. It significantly increased serum AST, ALT, ALP, bilirubin, urea, and creatinine levels and decreased total protein and albumin. AlCl3 significantly reduced enzymatic (catalase), nonenzymatic (reduced glutathione), and ferric reducing antioxidant power (FRAP) in the serum. Histopathologically, it induced necrosis and degeneration of hepatocytes, glomeruli, and renal tubules. Administration of LS after or along with AlCl3 significantly restored the serum biomarkers of liver and kidney functions to their near-normal levels and had the ability to overcome Al-induced oxidative stress and preserved, to some extent, the normal hepatic and renal structure. The coadministration of LS had a superior effect in alleviating Al-induced changes. Conclusion. Exposure to AlCl3 induced a set of functional and structural changes in the liver and kidney of rats evident through both biochemical and histopathological assessment. The antioxidant activity of LS seeds mediated a protective and curative effect of LS against such changes. Further study through a rigorous clinical trial to prove LS activity on human is recommended.

<p><strong>Objective: </strong>The aim of the present study was to investigate the daily effect of pre and post treatment of carob aqueous extract (600 mg/kg b. wt.); on rats exposed to were exposed to water pipe smoke (10 mg/kg; 15 minutes) for 2, 4, 6 and 8 w.</p><p><strong>Methods: </strong>Rats were divided into 6 groups; 1<sup>st</sup> group was served as a control. Rats of the 2<sup>nd</sup> group were administered amiodarone (an antiarrhythmic medication used as a model for lung toxicity) at a dose level of 30 mg/kg. The rats of 3<sup>rd</sup> and 4<sup>th</sup> groups were exposed to water pipe smoke and treated with carob extract respectively. However, the two remaining groups (5<sup>th</sup> and 6<sup>th</sup>) were the pre-and post-treatment groups with carob extract respectively.</p><p><strong>Results: </strong>Amiodarone and water pipe smoke induced a significant increase in serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP); on the contrary, induced a significant decrease in serum total protein and albumin. In addition; serum creatinine and urea showed a significant increase; as well as; amiodarone and water pipe smoke increased the oxidative stress (malondialdehyde/glutathione; MDA/GSH) in lung tissue homogenate. While; carob extract gavage showed protective and curative effects in liver and kidney functions; in addition; it decreased MDA level and increased GSH level significantly.</p><p><strong>Conclusion: </strong>Finally, we can conclude that the carob aqueous extract treatment revealed ameliorative effects on rats were exposed to water pipe smoke.</p>

The Impact

Sepsis is one of the most common causes of death among hospitalized patients. Activity of xanthine oxidase (XO), a reactive oxygen species-producing enzyme, is known to be elevated in septic patients. Our aim was to investigate the possible protective role of XO inhibitor, febuxostat (FEB), in a rat model of sepsis-induced liver and kidney injures. Adult male albino rats were divided into four groups ( n = 12 each): sham control, sham + FEB, cecal ligation and puncture (CLP), and CLP + FEB groups. FEB (10 mg/kg per os (p.o.)) was given once daily for 2 days and 30 min prior to laparotomy with CLP. CLP was associated with a high mortality rate accompanied by significant liver and kidney injuries indicated by elevated serum alanine aminotransferase, aspartate aminotransferase, urea, and creatinine levels and confirmed by histopathological tissue injury. Moreover, there was an increase in neutrophil gelatinase-associated lipocalin, uric acid, malondialdehyde, and nitric oxide levels and with decreased superoxide dismutase activity and total antioxidant capacity. In addition, CLP caused increased expression of the inflammatory markers tumor necrosis factor alpha, interleukin 1beta protein levels, and nuclear factor kappa B immunoexpression. Finally, CLP operated rats exhibited an upregulation in the apoptotic mediators, caspase 3, and P-C-Jun N-terminal kinases (JNK) proteins. FEB treatment of CLP rats caused a significant improvement and normalization in all measured parameters. Moreover, FEB amerliorates degenerative histopathological changes and improves the overall survival rate. In conclusion, FEB exhibited a protective effect in sepsis-induced liver and kidney injuries most probably through its anti-inflammatory, antioxidant, and antiapoptotic properties and attenuating JNK signaling pathway secondary to its XO enzyme inhibitory activity.
