The aim of the present study was to determine the frequency of different allergies in horses from central Poland. Allergic diseases are common in horses all over the world. Common conditions that have been associated with immunoglobulin E (IgE)-mediated allergy in horses include insect bite hypersensitivity (IBH), urticaria and recurrent airway obstruction (RAO). A variety of serological assays are available to measure serum levels of allergen-specific IgE antibodies. They can provide supportive evidence for a diagnosis of equine allergic dermatitis associated with environmental allergens. This study was performed on clinically healthy, Polish noble half-breed horses. In this study, four groups of allergens were identified as commonly inducing allergic reactions: American house dust mites (D. farinae), cultivated plants (rape), English plantain (Plantago lanceolata) and biting midges (Culicoides nubeculosus). More than 50% of horses showed positive reactions to these groups of allergens. The factor that contributed most to allergy occurrence among horses was American house dust mites (D. farinae). In recent years, efforts have been made to identify new allergens and to better understand the specific pathogenesis of allergic diseases in animals. The development of allergic disease diagnostics has been facilitated by the availability of diagnostic methods along with enhanced awareness of allergic diseases in veterinary practice. Allergic disorders in horses are ultimately gaining the attention they deserve thanks to the fact that veterinary allergology has become a dedicated veterinary discipline.

Background and Objectives: The evolution of allergic rhinitis to asthma is a part of “atopic march”. The aim of this study was to analyze possible predictive markers for asthma occurrence in patients with allergic rhinitis to house dust mites (HDM). Materials and Methods: Fifty-eight patients with persistent allergic rhinitis (PAR) were included. The clinical, biological evaluation and fractionated exhaled nitric oxide (FeNO) measurement were performed at enrolment. The patients were clinically evaluated after one year to determine asthma occurrence. Results: The severity of rhinitis symptoms, levels of total immunoglobulin E (IgE), ICAM-1, VCAM-1, E-selectin and IL-6, but not IL-8 and TNF-α were higher in patients with allergic rhinitis who developed asthma compared to non-asthmatics, but the differences were not significant to considered them as predictive factors for asthma occurrence. The risk of asthma was independently influenced by patients aged over 30 years ((OR-3.74; CI95% 0.86–16.31; p = 0.07), a duration of allergic rhinitis over 12 months ((OR-4.20; CI95% 0.88–20; p = 0.07) and a basal FeNO over 28 parts per billion (pbb) ((OR-18.68; CI95% 3.79–92.05; p < 0.001). Conclusion: Clinical and biological parameters may predict asthma occurrence in patients with persistent allergic rhinitis to HDM. Adult patients with a longer duration of rhinitis symptoms and a high level of FeNO have a greater risk to develop asthma.

Abstract HDM (House dust mites) are important environmental trigger factors of airway allergic diseases, the allergens of HDM were detected in the human gut mucosa, which induces local inflammation and increases intestinal permeability. This study tests a hypothesis that house dust mites contribute to the development of OVA (ovalbumin)-induced food allergy. The serum levels of IgE against HDM in patients with food allergy were detected with UniCAP100 (Pharmacia, Uppsala, Sweden); the HDM-induced/the OVA-induced mouse model of food allergy was developed. Difference between 2 groups was determined by Student t test or ANOVA if more than two groups. Compared to the healthy controls, the patients with food allergy have higher levels of serum IgE against HDM. Compared to food allergy alone groups, the levels of IgE against HDM in food allergy with asthma or allergic rhinitis groups were increased significantly. In mouse models, we found that HDM/OVA induced allergy-like symptoms, lower body temperature, and lower body weight. The levels of IgE, IgG1, mMCP-1 (mouse mast cell protease-1), IL-4 and IL-5 in the HDM and HDM + CT (cholera toxin) groups were higher than the control groups, and the levels of IgE, IgG1, IL-4 and IL-5 in the HDM, OVA and HDM + OVA groups were higher than the control groups. The pathology of intestinal tract in the HDM and HDM + CT/the HDM, OVA and HDM + OVA groups were more severe and exhibited more eosinophils than the control groups. Moreover, the prior exposure to HDM induced intestinal barrier dysfunction, and facilitated the development of intestinal allergy in mice. Based on above data and previous researches, we put forward that HDM exposure enhances subsequent responses to OVA-induced food allergy.

The growth of allergic diseases dictates the need to develop new forms of therapeutic allergens with high immunogenic and low allergenic activity. For many years, our laboratory has been developing drugs for the diagnosis and treatment of house dust mites (HDM) allergies. The purpose of this study is to summarize the results of the five-year development of therapeutic preparations of HDM allergens. During this period, we obtained the following forms of therapeutic allergens: a granular sublingual dosage form of a mixed allergen from Dermatophagoides pteronyssinus (Der.p) and Dermatophagoides farinaе (Der.f) and a succinylated monomeric HDM allergoid Der.p. The physicochemical and immunobiological properties of the obtained preparations were studied by methods: electrophoresis in PAGE in the presence of SDS-sodium, micropoint immunoblot, ELISA, inhibition of the binding reaction of allergen-specific IgE in the sera of patients. The research results showed that the obtained preparations have a reduced allergenic and increased immunogenic activity in comparison with native extracts. The created forms of mite allergens can be further used to treat patients sensitized to HDM of the genus Dermatophagoides.

Sensitisation to mites is frequent in atopic dogs. The main mite genus involved in canine atopic dermatitis is Dermatophagoides . The importance of storage mite allergens in dogs has been controversial. The aim of this study was to evaluate the sensitisation rates against storage mites ( Lepidoglyphus destructor and Tyrophagus putrescentiae ) and house dust mites ( Dermatophagoides farinae and D. pteronyssinus ) in atopic dogs from Galicia, a highly humid and temperate region of Spain, using a FcɛRIα-based immunoglobulin E (IgE) in vitro test. The study was performed on 95 dogs suffering from atopic dermatitis and presenting detectable specific serum IgE levels: 91.6% of the dogs tested positive for storage mites, whereas sensitisation to house dust mites was detected in 87.4%. These results indicate the importance of storage mites in this specific geographic area.

Abstract: In house dust are a lot of house dust mites that found in damp houses, cotton mattress, pillows, bolsters, and other home furnishings. Sources of dust containing house dust mites are most in the bedroom  especially cotton mattress. Data from distric health centers Paal 2 found that there are many people who suffer from asthma and allergic diseases, coupled with the humid air in Manado City to allow the development of house dust mites in the area. The purpose of this research is to determine the species and density of house dust mites in urban Perkamil Paal 2 districts Manado city. This research method is descriptive survey to obtain data on the species and density of house dust mites by simple random sampling method. The results found 5 species of house dust mites are Dermatophagoides spp, Acarus spp, Glycyphagus destructor, Tarsonemus spp and Cheyletus spp in bedrooms and living room. House dust mite densities obtained an average of 2,33 in bedroom and 2,07 in living room. The conclusion is house dust mite Dermatophagoides spp most commonly found in bedrooms and living room. Sumber debu yang mengandung tungau debu rumah terbanyak adalah debu kamar tidur terutama debu di kasur kapuk. Dari data Puskesmas Kecamatan Paal 2 didapatkan bahwa masih banyak masyarakat yang menderita asma dan penyakit alergi, ditambah lagi dengan udara Kota Manado yang lembab sehingga memungkinkan terjadinya perkembangan tungau debu rumah di daerah tersebut. Tujuan penelitian ini untuk mengetahui jenis dan kepadatan tungau debu rumah di Kelurahan Perkamil Kecamatan Paal 2 Kota Manado. Metode penelitian ini secara survei deskriptif untuk mendapatkan data tentang jenis dan kepadatan tungau debu rumah dengan metode simple random sampling. Hasil yang didapat ditemukan 5 jenis tungau debu rumah yaitu Dermatophagoides spp, Acarus spp, Glycyphagus destructor, Tarsonemus spp dan Cheyletus spp di ruang tidur dan ruang tamu. Kepadatan tungau debu rumah didapatkan rata-rata 2,33 di ruang tidur dan 2,07 di ruang tamu. Kesimpulan yang diambil yaitu tungau debu rumah Dermatophagoides spp paling banyak ditemukan di ruang tidur maupun ruang tamu. Kata kunci: jenis, kepadatan, tungau debu rumah

Abstract: House dust mite are insects that can cause allergies in susceptible people. Population of dust mites in the home depends on factors such as the level of the sea, an area with a longer summer than rainy season, a variety of animals in the house, the house is dirty and a lot of dust, as well as the temperature and humidity of the area. Manado city has several of the factors. Purpose of the study is to investigate the people’s behavior of the population of house dust mites in Perkamil village sub-district of Paal 2 Manado city. Result obtained was 34,86% which shows that people’s knowledge about house dust mites are still lacking. The result of attitude is 97,09% indicated public attitudes towards the prevention and eradication of house dust mite is good. And result of the public action showed 75,96% of the community action efforts to prevent and eradicate house dust mites is enough. Necessary education from health care workers about house dust mites and allergic diseases, prevention and eradication, so that the knowledge that still low will be better and the action that will be good enough. Populasi tungau debu di dalam rumah bergantung pada faktor – faktor, seperti  tinggi rendahnya rumah dari permukaan laut, daerah dengan musim panas yang lebih panjang dari musim hujan, adanya berbagai macam binatang di dalam rumah, rumah yang kotor dan banyak debu, serta suhu dan kelembaban daerah tersebut. Kota Manado memiliki beberapa faktor tersebut. Tujuan penelitian adalah untuk mengetahui perilaku masyarakat terhadap populasi tungau debu rumah di Kelurahan Perkamil Kecamatan Paal 2 Kota Manado. Hasil yang didapatkan adalah 34,86% menunjukkan pengetahuan masyarakat Kelurahan Perkamil mengenai Tungau Debu Rumah masih kurang. Untuk sikap didapatkan hasil 97,09% menunjukkan sikap masyarakat terhadap upaya pencegahan dan pemberantasan Tungau Debu Rumah sudah baik. Dan untuk tindakan masyarakat didapatkan hasil 75,96% menunjukkan tindakan masyarakat terhadap upaya pencegahan dan pemberantasan Tungau Debu Rumah sudah cukup. Perlu dilakukan penyuluhan dari petugas kesehatan tentang Tungau Debu Rumah dan penyakit Alergi, cara pencegahan dan pemberantasan, agar pengetahuan masih rendah akan menjadi lebih baik lagi dan tindakan masyarakat yang sudah cukup akan menjadi baik. Asthma is a chronic life-threatening disease of worldwide importance. Although allergic asthma and related atopic conditions correlate strongly with immune sensitization to house dust mites, it is unclear why antigens from mites provoke such powerful allergic immune responses. We have characterized the protease activity of Der p I, the group I protease allergen of the house dust mite Dermatophagoides pteronyssinus, and here report that it cleaves the low-affinity immunoglobulin (Ig) E Fc receptor (CD23) from the surface of human B lymphocytes. Der p I selectively cleaves CD23 and has no effect on the expression of any other B cell surface molecules tested. We speculate that this loss of cell surface CD23 from IgE-secreting B cells may promote and enhance IgE immune responses by ablating an important feedback inhibitory mechanism that normally limits IgE synthesis. Furthermore, since soluble CD23 is reported to promote IgE production, fragments of CD23 released by Der p I may directly enhance the synthesis of IgE. alpha 1-Antiprotease, a pulmonary antiprotease, is also shown to inhibit the cleavage of CD23 by Der p I. This may be significant in the etiopathogenesis of asthma, because other indoor pollutants associated with asthma are known to potently inhibit this antiprotease. These data suggest that the proteolytic activity of Der p I, the group I allergen of the house dust mite D. pteronyssinus, is mechanistically linked to the potent allergenicity of house dust mites. Furthermore, inhibition of Der p I by alpha 1-antiprotease suggests a mechanism by which confounding factors, such as tobacco smoke, may act as a risk factor for allergic asthma.
