  Objective: The inhibitory properties of successive extracts from Dioscorea bulbifera (Dioscoreaceae) tubers have been evaluated for the presence of phytochemical constituents and antimicrobial efficacy against multidrug-resistant (MDR) clinical isolates was evaluated.Methods: The tuber of D. bulbifera was oven dried and extracted successively with n-hexane, chloroform, methanol, ethanol, and water. The antimicrobial potential of successive extracts against MDR isolates was studied by agar well-diffusion method. Qualitative phytochemical analysis was performed.Results: Qualitative phytochemical analysis demonstrated the presence of steroids, flavonoids, cardiac glycosides, saponins, and reducing sugars in almost all the extracts tested. Anthraquinones, phlobatanins, and tannins were not reported in any extracts tested. The in vitro antimicrobial activity of various solvents and water extracts of D. bulbifera was further investigated against ten MDR bacteria and three fungi, respectively. Aqueous and chloroform extracts were found to be more potent being capable of exerting significant inhibitory activities against the majority of the isolates such as Escherichia coli, Acinetobacter sp., Salmonella paratyphi, Klebsiella pneumoniae, and Candida albicans. The highest inhibitory activity was observed for K. pneumoniae with wide inhibition zone diameters (17 ± 0.15 mm), followed by E. coli 1(13 ± 0.11) mm, and Acinetobacter sp. (11 ± 0.12).Conclusion: Based on the present study, the extracts of D. bulbifera tubers have shown excellent activity against MDR microbial cultures tested. Further study is recommended for clinical evaluation, of the efficacy of crude extract in herbal medicine that can serve as a base for the development of novel potent drugs and phytomedicines.

Los aceites esenciales son compuestos orgánicos importantes en el sector farmacéutico debido a sus propiedades antimicrobianas, volviéndose una alternativa de solución para determinadas fitopatologías. En el presente estudio se evaluó in vitro la actividad inhibitoria de aceites esenciales de Curcuma longa y Zingiber officinale contra Colletotrichum sp. obtenidas a partir de cultivos de Dioscorea rotundata en Toluviejo (Sucre, Colombia). Concentraciones de 3,000; 5,000; 8,000 y 10,000 mg/L de extracciones de aceite (método de hidrodestilación asistida por microondas) fueron aplicados a cultivos in vitro del hongo. Además, se empleó un testigo absoluto y controles positivo y negativo. Los aceites fueron caracterizados por cromatografía de gases acoplado a espectrometría de masa. La actividad inhibitoria se realizó por medio de la prueba de siembra en superficie (porcentaje de índice de inhibición). Los mayores valores de índice antifúngico se observaron a 10,000 mg/L similar al obtenido con el control positivo por benomil. De la caracterización química de los aceites esenciales el metabolito secundario que se presentó con mayor rango fue el monoterpeno 3-Thujeno y el citral. Los aceites esenciales de C. longa y Z. officinale generaron actividad inhibitoria contra el crecimiento de Colletotrichum sp., y por lo tanto pueden ser empleados para reducir la antracnosis.

AbstractMany populations of high-mountainous relic dioecious willows in Central Europe only consist of female individuals and are thus limited in their reproductive potential. We completed micropropagation experiments with shoot apexes and nodal segments of common and endangered willow (Salix) species, which can help to reintroduce autochthonous genotypes to their natural sites. Until recently, cultivation of green young shoot apexes of S. alba and S. lapponum showed the highest percentage of regeneration. We successfully applied the two-times-sterilisation due to high contamination of natural explants. The OK medium was the most efficient culture medium. In vitro propagation of willows with unisexual catkins, anther and ovule cultures were tested and optimised. Isolated anthers were cultivated on selected media and then microcallus and calluses of S. caprea and calluses of S. viminalis were formed on the A medium. Among various tested and optimised media for the ovule culture, the CP medium was the most efficient one. In this case, only the microcalluses of S. viminalis were observed. We developed biotechnological procedures that can be useful in conserving fragmented populations of high-mountainous willows.
