The use of immunogold-silver staining (IGSS) to enhance label penetration and Localization for immunocytochemistry or in situ hybridization utilizing a variety of metallic salts has been documented. In this morphological study, the effects of silver acetate, silver lactate and silver nitrate were evaluated for immunogold-labeling of a trial natriuretic peptides (ANP) in rat right atria.Eight Wistar Kyoto retired breeders were sedated with pentobarbital, perfused with either 4% paraformaldehyde (LM) or Karnovsky's fixative (EM), and right atria were dissected, processed, embedded in paraffin or epon, respectively and sectioned according to conventional methods. For light microscopy, an indirect IGSS method according to Hacker (3) was performed. Paraffin sections on glass slides were washed in ddH2O, immersed in Lugol's iodine, washed in ddH2O and treated with 2.5% aqueous sodium thiosulfate for 20 sec. After additional washes in ddH2O and TBS-0.1% fish gelatin, 10% normal goat serum (PBS with 1% BSA) was applied for 20 min before an overnight incubation at 4°C with a polyclonal α-ANP primary antibody (Peninsula Labs, 1:1000 in TBS/BSA).

A method for measuring both dissolved ozone (DO3) concentration and UV absorbance was developed adopting ultraviolet (UV) absorption method (JWWA, 1993) using sodium thiosulfate (Na2S2O3) solution for removing residual ozone in ozonated water. A DO3 monitor based on this method was tested. This method was proven to be effective from experimental results. The performance of the monitor was examined with continuous ozonated water. As a result, the monitor performed stably during about 2 months, so that both DO3 concentration and UV absorbance in the ozonated water could be accurately measured. Therefore, the authors have proposed the new aquatic control system with this monitor for ozonation.

The elimination of flooding and lowering of the groundwater table after large-scale river regulation allow deep penetration of soils by plant roots, soil fauna, and microorganisms, thus creating favorable conditions for advanced pedogenesis. Although the changes of the morphology and properties of agriculturally used drained alluvial soils in Central Europe have been well characterized, studies in riparian forests remain insufficient. An analysis of 21 profiles of forest soils located on the Holocene river terrace (a floodplain before river regulation and embankment) in SW Poland confirmed a noticeable pedogenic transformation of soil morphology and properties resulting from river regulation. Gleyic properties were in most profiles replaced with stagnic properties, testifying to a transition from dominant groundwater supply to precipitation-water supply. The development of a diagnostic mollic and cambic horizons, correlated with the shift in soil classification from Fluvisols to Phaeozems, and in the majority, to Cambisols, demonstrated a substantial change in habitat conditions. The transformation of alluvial soils may result in an inevitable modification of forest management in the river valley, including quantitative alteration in species composition of primarily riparian forests.
