ABSTRACT The humicolous actinomycete Streptomyces coelicolor routinely adapts to a wide variety of habitats and rapidly changing environments. Upon salt stress, the organism is also known to increase the levels of various compatible solutes. Here we report the results of the first high-resolution metabolomics time series analysis of various strains of S. coelicolor exposed to salt stress: the wild type, mutants with progressive knockouts of the ectoine biosynthesis pathway, and two stress regulator mutants (with disruptions of the sigB and osaB genes). Samples were taken from cultures at 0, 4, 8, and 24 h after salt stress treatment and analyzed by liquid chromatography-mass spectrometry with an LTQ Orbitrap XL mass spectrometer. The results suggest that a large fraction of amino acids is upregulated in response to the salt stress, as are proline/glycine-containing di- and tripeptides. Additionally we found that 5′-methylthioadenosine, a known inhibitor of polyamine biosynthesis, is downregulated upon salt stress. Strikingly, no major differences between the wild-type cultures and the two stress regulator mutants were found, indicating a considerable robustness of the metabolomic response to salt stress, compared to the more volatile changes in transcript abundance reported earlier.

The Brain

The cerebral vasculature ensures proper cerebral function by transporting oxygen, nutrients, and other substances to the brain. Distribution of oxygenated blood throughout the neuroaxis takes place at the level of the circle of Willis (CW). While morphologic and functional alterations in CW arteries and its main branches have been reported in cerebrovascular and neurodegenerative diseases, accompanying changes in protein expression profiles remain largely uncharacterized. In this study, we performed proteomics to compile a novel list of proteins present in mouse CW arteries and its ramifications. Circle of Willis arteries were surgically removed from 6-month-old wild-type mice, proteins extracted and analyzed by two proteomics approaches, gel-free nanoLC-mass spectrometry (MS)/MS and gel-based GelLC-MS/MS, using nanoAcquity UPLC coupled with ESI-LTQ Orbitrap XL. The two approaches helped maximize arterial proteome coverage. Six biologic and two technical replicates were performed. In all, 2,188 proteins with at least 2 unique high-scoring peptides were identified (6,630 proteins total). Proteins were classified according to vasoactivity, blood–brain barrier specificity, tight junction and adhesion molecules, membrane transporters/channels, and extracellular matrix/basal lamina proteins. Furthermore, we compared the identified CW arterial proteome with the published brain microvascular proteome. Our database provides a vital resource for the study of CW cerebral arterial protein expression profiles in health and disease.

e16083 Background: In the prostatic epithelium endothelin-1 (ET-1), fibroblast growth factor-2 (FGF2) and hepatocyte growth factor (HGF) are involved in cell proliferation and inhibition of apoptosis. The aim of this study is to investigate whether neuropeptides such as ET-1 and growth factors such as FGF and HGF may constitute an alternate pathway to intracellular steroidogenesis. Methods: PC3 cells, an androgen independent prostate cancer cell line were cultured in RPMI 1640, fully supplemented with FBS 10% and in serum free conditions. The cells were incubated with ET-1(10nM, 3 hrs incubation), FGF-2 (10 ng/ml, 3 hrs incubation) and HGF(33 ng/ml, 3 hrs incubation), and then progesterone, 17-OH progesterone and aldosterone were measured in cell lysates by LC-MS using LTQ Orbitrap XL MS 2.5.5 SP1 (Thermo-Fisher) equipped with Accela AS 2.2.1, Accela pump. The data were analysed by Xcalibur 2.1.0 Software. Results: In baseline conditions PC3 cells produce no progesterone but high amounts of 17-OH progesterone and small amounts of aldosterone. Upon stimulation with endothelin there was a significant increase in progesterone, significant decrease in 17-OH progesterone and a very significant increase in aldosterone. Similarly stimulation with both HGF and FGF resulted in significant decreases in 17-OH progesterone and increases in aldosterone. Conclusions: Our results imply that in androgen deprivation conditions intracellular steroidogenesis can still take place by stimulation via seven membrane and tyrosine kinase receptors, contributing in part to a steroid dependent phenotype in addition to a steroid independent one, thus implying the potential for new therapeutic interventions.

Η παρούσα διδακτορική διατριβή περιλαμβάνει τη μελέτη της χημικής σύστασης και βιολογικής δράσης πέντε αυτοφυών taxa Sideritis: δύο taxa S. raeseri Boiss & Heldr ssp. raeseri που συλλέχθηκαν στο Συρράκο Ιωαννίνων και το Κελλάρι Μελλισουργών Άρτας, δύο taxa S. scardica Griseb που συλλέχθηκαν στο Βούρινο -Ροδιανής Κοζάνης και την Κρυσταλοπηγή Φλώρινας, ένα taxon S. perfoliata L. ssp perfoliata που συλλέχθηκε στην Κορυφή Τρικάλων. Γίνεται συγκριτική προσέγγιση μεταξύ των taxa Siderits, καθώς και με ένα εμπορικό σκεύασμα πράσινου τσαγιού (C. sinensis, γνωστό για το μεγάλο εύρος ερευνών και την πληθώρα δράσεών του), τσάι Κεϋλάνης. Ειδικότερα, πραγματοποιήθηκαν εκχυλίσεις με τρεις διαλύτες διαφορετικής πολικότητας (μεθανόλη, νερό και εξάνιο). Στα ξηρά εκχυλίσματα:•έγινε έλεγχος ως προς την απόδοση επί τοις εκατό της εκχύλισης σε στερεό για κάθε φυτικό είδος,•έγινε έλεγχος ως προς την περιεκτικότητα σε ολικά φλαβονοειδή και ολικά φαινολικά, καθώς και ως προς την αντιοξειδωτική δράση με τη μέθοδο του DPPH˙ και σύγκριση με την αντιοξειδωτική δράση των πρότυπων ουσιών Quercetin, Vitamin C και Troxol,•έγινε ποιοτικός και ποσοτικός προσδιορισμός με τη χρήση πρότυπων ουσιών με υγρή χρωματογραφία συζευγμένη με φασματομετρία μάζας υψηλής διακριτικής ικανότητας και ακρίβειας UHPLC/LTQ-Orbitrap XL•έγινε έλεγχος ως προς την βιολογική δράση σε καρκινικές κυτταρικές σειρές και σύγκριση με φυσιολογικά κύτταρα.Τα αποτελέσματα βρέθηκαν παρόμοια μεταξύ των taxa Sideritis, ενώ παρατηρήθηκαν διαφορές μεταξύ αυτών και του C. Sinensis. Συγκεκριμένα, τα taxa Sideritis παρουσίασαν τη μεγαλύτερη περιεκτικότητα σε ολικά φλαβονοειδή σε υψηλής πολικότητας διαλύτες (μεθανόλη, νερό). Αντίθετα στο C. sinensis η μεγαλύτερη περιεκτικότητα σε ολικά φλαβονοειδή βρέθηκε στο διαλύτη εξάνιο. Τέλος, συγκρίνοντας τα αποτελέσματα IC50 που βρέθηκαν για τα εκχυλίσματα με αυτά των πρότυπων ουσιών, όλα τα μεθανολικά και υδατικά εκχυλίσματα χαρακτηρίζονται ως μέτριας αντιοξειδωτικής δράσης, με εξαίρεση το μεθανολικό εκχύλισμα του C. Sinensis που εμφάνισε ισχυρή αντιοξειδωτική δράση, όλα δε τα εξανικά εκχυλίσματα χαρακτηρίζονται ασθενούς αντιοξειδωτικής δράσης. Κατά τον ποιοτικό και ποσοτικό προσδιορισμό, προέκυψαν παρόμοια αποτελέσματα για τα taxa Sideritis και διαφορετικά για το C. Sinensis. Για τις ουσίες που βρέθηκαν εκατέρωθεν (taxa Sideritis & C. Sinensis) τα επίπεδα συγκέντρωσης στο C. Sinensis είναι πολλαπλάσια των taxa Sideritis.Τέλος, ελέγχθηκε η κυτταροτοξικότητα in vitro, έναντι τεσσάρων κυτταρικών σειρών και συγκρίθηκε με το cis-Diamminadichloroplatinum (cis-platin). Οι κυτταρικές σειρές είναι: HeLa, Α549, LS174 και η σειρά MRC5. Τα αποτελέσματα συγκρινόμενα με cis-DDP υπολείπονται της κυτταροτοξικής δράσης τους. Όμως, θα μπορούσαν να αξιολογηθούν ως ενδιαφέροντα τα αποτελέσματα του εξανικού εκχυλίσματος του S. perfoliata L. ssp.perfoliata, το οποίο παρουσίασε τη μεγαλύτερη δραστικότητα έναντι και των τριών καρκινικών σειρών ακόμα, και από αυτή του C. Sinensis, ενώ εμφάνισε επιλεκτική δραστικότητα για τη φυσιολογική κυτταρική σειρά. Ενδιαφέρον παρουσίασαν τα αποτελέσματα για τη δραστικότητα των μεθανολικών και υδατικών εκχυλισμάτων των άλλων δυο taxon Sideritis στην καρκινική σειρά κυττάρων, HeLa.
