CHIP COLOR, SUGAR CONTENT AND YIELD OF SUMMER POTATOES DURING GROWTH IN SOUTHERN ONTARIO

Tubers of five summer potato cultivars were monitored weekly for chip color, reducing sugars, sucrose, and marketable yield for 4 wk commencing 3 July, about 80 d after planting, in 1985 and 1986 at Harrow and Simcoe, Ontario. Atlantic and Conestoga tubers generally contained the lowest sugar levels, were the only cultivars to produce acceptable colored chips by 3 July, and had lighter chip color than all cultivars, even lighter than the standard cultivar, Superior. Conestoga yields by 3 July (about 13 t ha−1) exceeded those of Superior (about 10 t ha−1), but Atlantic required 7–10 d more growth to produce comparable yields. Atlantic and especially Conestoga may be more suitable than Superior for use as main summer chipping cultivars in southern Ontario.Key words: Solanum tuberosum L., cultivar, chip color, sugar content, marketable yield, maturity

Uncovering the genetic basis and optimizing the late blight tolerance trait in potatoes (Solanum tuberosum L.) are crucial for potato breeding. Late blight disease is one of the most significant diseases hindering potato production. The traits of late blight tolerance were evaluated for 284 potato cultivars to identify loci significantly associated with the late blight tolerance trait. Of all, 37 and 15 were the most tolerant to disease, and 107 and 30 were the most susceptible. A total of 22,489 high-quality single-nucleotide polymorphisms and indels were identified in 284 potato cultivars. All the potato cultivars were clustered into eight subgroups using population structure analysis and principal component analysis, which were consistent with the results of the phylogenetic tree analysis. The average genetic diversity for all 284 potato cultivars was 0.216, and the differentiation index of each subgroup was 0.025–0.149. Genome-wide linkage disequilibrium (LD) analysis demonstrated that the average LD was about 0.9 kb. A genome-wide association study using a mixed linear model identified 964 loci significantly associated with the late blight tolerance trait. Fourteen candidate genes for late blight tolerance traits were identified, including genes encoding late blight tolerance protein, chitinase 1, cytosolic nucleotide-binding site–leucine-rich repeat tolerance protein, protein kinase, ethylene-responsive transcription factor, and other potential plant tolerance-related proteins. This study provides novel insights into the genetic architecture of late blight tolerance traits and will be helpful for late blight tolerance in potato breeding.

The use of colored tubers of Solanum tuberosum L. is growing worldwide due to their health benefits and attractive color. The positive health effects of purple-fleshed tubers are a result of anthocyanins and various phenolic compounds. The aim of this study was to evaluate and compare variety Blue Congo and its cross-breeds of Desiree and Granola to yellow-fleshed tubers. The concentration of total phenols, anthocyanins, sugars, and mineral elements were evaluated in all tubers. The results showed differences between all tested materials, with largest differences in sugar content. Moreover, the results confirmed the preservation of health improving compounds of Blue Congo when cross-bred with yellow-fleshed tubers. The total phenolic content and anthocyanin concentrations of all analyzed tubers were above the comparison yellow ones.

Prior to Agrobacterium-mediated genetic transformation in vitro regeneration protocol was established for three potato cultivars (Alfa, Cambray Rosa Morelos and Atlantic) grown in Mexico using leaf, node and internodal explants. Regeneration protocol was developed with or without the intervention of callus. Two potato cultivars, namely, Cambray Rosa Morelos and Alpha were transformed using Agrobacterium tumefaciens strain LBA4404 harboring binary plasmid pBI121 containing the GUS and nptII genes. GUS histochemical assay and PCR analysis were conducted on rooted shoots grown in media without hormones but supplemented with antibiotics. Quality evaluations in potatoes are of necessity to meet the strict demands of the chip processing industry. Important parameters assessed include specific gravity, dry matter content, chip color, reducing sugars, and glycoalkaloids. This study was designed with the purpose of identifying specialized potato clones with acceptable qualities for processing chips, in comparison with two selected control varieties, Dubaek and Superior. As a result, high dry matter and specific gravity were observed for three potato clones, and the quantified ά-solanine levels ranged from 0.15 to 15.54 mg·100 g−1 fresh weight (FW). Significant variations (p < 0.05) in reducing sugar levels were observed in clones stored at different temperature conditions. After reconditioning of the tubers at 22 °C for 21 days, a significant drop in reducing sugar levels was recorded. In addition, fried chips for each potato clone were evaluated, and the color measured on the basis of the Snack Food Association (SFA) chip color score standard. Reconditioned tubers exhibited much lighter and better chip color compared to their counterparts cold-stored at 4 °C. This study observed that for quality processing of potato chips, clones with combined traits of high dry matter, low levels of glycoalkaloids and reducing sugars, and acceptable chip color should be used as raw materials.

Over a 4-yr period (1974–1977) the effect of metribuzin at three rates and times of application was evaluated on four potato (Solanum tuberosum L.) cultivars commonly grown on Prince Edward Island. Metribuzin applied preemergence and early postemergence had no adverse effect on marketable and total yields of Irish Cobbler, Netted Gem, Kennebec, and Sebago cultivars. Delaying metribuzin application until late postemergence resulted in significantly reduced marketable yields of Irish Cobbler and Sebago but not Netted Gem and Kennebec when compared with preemergence and early postemergence treatment. Total yields with late postemergence treatment were significantly less than those with the preemergence treatment for all four cultivars and significantly less than early postemergence treatment with Irish Cobbler and Sebago.

Biochemical variability between thirteen European and five Czech potato (Solanum tuberosum L.) cultivars grown in theCzechRepublic was studied by soluble protein, isoesterase, and isoperoxidase electrophoretic patterns. It was confirmed that cultivar differences in protein polymorphism can be revealed by applied electrophoretic patterns. It was shown that the different character of protein and isozyme profiles required different approaches to their evaluation. For complex patterns such as electrophoretic soluble protein spectra, it is more convenient to use the evaluation of their absorbance profiles and for simpler profiles of isozymes the evaluation based on the presence or absence of a&nbsp;band in a&nbsp;definite position (simple matching) should be used. In spite of the complexity of tetraploid disposition of analysed cultivars, the results suggested higher similarity of profiles between relative cultivars and they also indicated the existence of higher similarity between cultivars from the same breeding firm. &nbsp;
