INFLUENCE OF MANGANESE CONCENTRATION IN NUTRIENT SOLUTION ON THE GROWTH AND ELEMENTAL CONTENT OF THE "KATAHDIN" POTATO PLANT

Hoagland’s solutions containing nine levels of manganese ranging from 0 to 100 ppm were applied as a drench tri-weekly to potato plants (Solanum tuberosum L. cv. Katahdin) growing in perlite. Although no deficiency symptoms were observed during the 48-day growing period, varying degrees of toxicity were associated with concentrations of 25.0 ppm Mn and higher in nutrient solution. The tissue concentration necessary to induce toxicity symptoms in foliage appeared to fall between 134 and 426 ppm Mn. Solution concentrations in excess of 25 ppm Mn caused a significant reduction in dry weight of top, root and rhizome portions, and in rhizome number. Increasing Mn treatments generally influenced elemental tissue content as follows: K and Mn increased, Mg decreased, Al and Ca decreased at and above 50 ppm and 75 ppm Mn, respectively, whereas P increased at 50 ppm Mn and above; B, Cu and Mo were unaffected and although N, Fe and Zn contents were somewhat erratic, they were not significantly different at 100 ppm Mn from the control (0.50 ppm Mn).

Tujuan penelitian ini adalah mengidentifikasi toksisitas beberapa fungisida nabati dalam mengendalikan penyakit Layu Fusarium (F. oxysporum) pada tanaman kentang (Solanum tuberosum L.) secara In Vitro. Metode Penelitian yang digunakan yaitu metode umpan beracun. Data dianalisis dengan Rancangan Acak Lengkap (RAL) dengan Analisis Varian (ANAVA) yang dilanjutkan dengan menggunakan metode BNT (Beda Nyata Terkecil). Hasil Penelitian, diperoleh nilai toksisitas fungisida nabati tertinggi yaitu pada ekstrak daun sirsak dengan nilai HR (69,44%), kategori berpengaruh, ditandai dengan diameter koloni 2,75 cm (100ppm) dan yang terendah toksisitasnya yaitu pada ekstrak daun jeruk purut dengan nilai HR (49,81%), kategori cukup berpengaruh ditandai dengan diameter koloni 3,75 cm (25ppm). Semakin tinggi konsentrasi yang diujikan maka semakin tinggi toksisitas dari fungisida nabati yang diberikan.Kata Kunci: fungisida nabati, Fusarium oxysporum, tanaman kentang, In Vitro Abstract Potato plants (Solanum tuberosum L.) is one of the horticulture plants which often decreases in terms of production and productivity, due to the attack of wilt, one of which is caused by Fusarium oxysporum. The purpose of this study was to determine the toxicity of several biofungicides in controlling Fusarium wilt (F. oxysporum) in potato plants (Solanum tuberosum L.) in Vitro. The research method used was the In Vitro method with the poison bait method. Data were analyzed by Completely Randomized Design with Variant Analysis (ANAVA), followed by the BNT method. The results showed that the highest biofungicide toxicity value was soursop leaf extract with HR values (69.44%), influential categories, characterized by colony diameter 2.75 cm (100ppm) and the lowest toxicity, namely in kaffir lime leaf extract with a value of HR (49.81%), quite influential category was characterized by colony diameter of 3.75 cm (25ppm). The higher the concentration tested, the higher the toxicity of the biofungicide given.Keywords: biofungicides, Fusarium oxysporum, Potato Plants, In Vitro.

The aim of this study is to determine the effects of different concentrations and combinations of the phytohormones, 1-naphthaleneacetic acid (NAA), and 6-benzylaminopurine (BAP): M1 (0.5 mg / l +1 mg / l), M2 (1 mg / l + 0.5 mg / l) , M3 (2 mg / l +2 mg / l), M4 (0.5 mg / l + l mg / l, NAA), M5 (1.0 mg / l + l mg / l , NAA), and M6 (2.0 mg / l + l mg / l, NAA). This study was carried out in dark condition on callus induction of potato plants (Solanum tuberosum L.) cultivars from potato tuber bud so as to demonstrate the role of light. The callus initiation begins after 7 days of incubation for all studied media. After two months of incubation, the better development of callus was noted in Spunta variety by using medium M1, M2, M3, and M6. The calluses took a compact structure of brown-white color for both varieties with a callus induction rate of 20- 40%. This was collected with kondor variety for M2 and (M3, M4, M5) media respectively and 10-30% for M4 (M1, M2, M3) for Spunta variety also. The highest fresh weight was recorded on M2 medium with 0.26g for Kondor variety and 0.93g for Spunta variety.

Penelitian ini bertujuan untuk mengetahui tingkat populasi jamur tanah di lahan pertanaman kentang (S. tuberosum L.). Sampel tanah diambil di perkebunan tanaman kentang Desa Pinasungkulan Kecamatan Modoinding yang banyak menggunakan pestisida. Penelitian ini dilakukan dengan mengisolasi jamur pada tanah di pertanaman kentang di Desa Pinasungkulan, Kecamatan Modoinding, Kabupaten Minahasa Selatan dengan pengenceran bertingkat dan dilanjutkan dengan menghitung jumlah koloni dengan metode Total Plate Count (TPC). Dari hasil isolasi diperoleh 6 famili dengan 11 jenis jamur tanah yang memiliki koloni terbesar pada isolat KJ1 sebesar 71 koloni dan terendah pada isolat KJ8, KJ10, dan KJ 11 masing-masing sebesar 2 koloni.Kata kunci: Tanaman kentang, jamur tanah, tingkat populasi, Total Plate Count (TPC) ANALYSIS OF SOIL FUNGUS POPULATION LEVELS  IN POTATO PLANTS (Solanum tuberosum L.) BASED ON TOTAL PLATE COUNT (TPC) METHODSABSTRACTThis study aimed to determine the level of soil fungus population on potato planting land (S. tuberosum L.). Soil samples were taken at a potato field plantation in Pinasungkulan Village, Modoinding Subdistrict, which used many pesticides. This research was carried out by isolating fungi on soil in potato plantations in Pinasungkulan Village, Modoinding District, South Minahasa Regency with multilevel dilution and continued by counting the number of colonies using the Total Plate Count (TPC) method. The result showed that the isolation obtained 6 families with 11 types of soil fungi that have the largest colony in KJ1 isolates of 71 colonies and the lowest in isolates KJ8, KJ10, and KJ 11 of 2 colonies respectively.Keywords: Potato plants, soil fungi, population level, Total Plate Count (TPC)

The localization of cysteine proteinase PLCP-2 was investigated in potato plants (Solanum tuberosum L.) cultivar Désirée by electron microscopy. Healthy and PVY<sup>NTN</sup> infected potato plants were grown in vitro on media with or without a supplement of jasmonic acid. We had already shown that PLCP-2 is present in leaves, stems, tips of shoots and tips of roots of healthy and PVY<sup>NTN</sup> infected plants. It was detected in various cell types in protein bodies in vacuoles, in cytoplasm and in cell walls. There were significantly larger amounts of PLCP-2 in plants grown on medium with a supplement of jasmonic acid in both healthy and virus infected plants. More protein bodies in vacuoles were found in plants grown on medium with addition of jasmonic acid.

Growth measurements of potato (Solanum tuberosum L.) cvs. Norland (NL), Denali (DN), and Kennebec (KN) were taken from 21-day-old plantlets grown in vitro. Studies were conducted in a growth chamber, with nodal explants grown in culture tubes with loose-fitted Magenta 2-way caps containing Murashige and Skoog salts with either 0, 1, 2 or 3% sucrose. The cultures received either 100 or 300 μmol m-2 s-1 photosynthetic photon flux (PPF), and the growth chamber was maintained at either 400 or 4000 μmol mol-1 CO2. All cvs. showed significant increases in growth on 0% sucrose media at 4000 μmol mol-1 CO2, indicating an autotrophic response. At 400 μmol mol-1 CO2, all cvs. showed an increase in total plantlet dry weight (DW) with increasing sucrose under both PPF levels. Within any sucrose treatment, the highest total DW for all cvs. resulted from 300 μmol m-2 s-1 PPF and 4000 μmol mol-1 CO2. At 4000 μmol mol-1 CO2, shoot DW declined with sucrose above 2% for DN and sucrose above 1% for NL at both PPF levels, suggesting that high sucrose levels may hinder growth when CO2 enrichment is used.

Seven potato genotypes were evaluated on field, with a random block design with four repetitions, and an experimental plot of three rows measuring five m long. The variables analyzed were morphological, physiological, agroclimatic and yield, by calculating Correlations and Principal Components Analysis (PCA), in attempt to find the degree of association between variables, and which variables contributed to the variation in the yield. The most correlated variables in the yield were plant height, the dry weight of leaves, and chlorophyll content. The PCA permitted a better screening between variables, such as photosynthesis, water use efficiency. When considering all variables together, the best varieties were Norteña, Mondial and Atlantic, found to be superior to the regional genotype (Alpha).
