Association of class I bovine lymphocyte antigens with profitability and lifetime yields in the Holstein breed

Ninety-four Holstein cows from the Agriculture and Agri-Food Canada herd at Ottawa were serologically typed for the BoLA-A locus. Of the 37 serologically defined specificities, only 10 were present in Holstein cows. A gene substitution model, which included the effect of BoLA alleles and the breeding value of the sire as a covariant, was used to evaluate the association between these genes and lifetime yields and profitability. Allele A14 was significantly (P < 0.05) associated with increased lifetime milk, protein and lactose yields as well as greater milk value and annualized discounted net returns. Therefore, the potential may exist for increasing the lifetime yields and profitability in dairy cattle using genes of the BoLA-A locus. Key words: BoLA, lifetime yields, profitability, Holstein

Ninety progeny tested Canadian Ayrshire bulls were serologically typed for the BoLA-A locus to determine the association of these alleles with production traits. After exclusion of bulls carrying alleles that occurred at frequencies lower than 2% or whose production proofs were not available, records from 78 bulls remained for analysis. A gene substitution model, which included the effects of BoLA alleles and the breeding value of the sire of the bull as covariate, was used to evaluate the additive effects of BoLA alleles of bulls (ETAs) on the production traits of their daughters. Allele W17 was significantly associated with ETA for milk, fat and protein yields. Allele CA96 was associated with significant increases in ETA for fat yield. There was no significant association of BoLA alleles with either ETA for fat or protein percentage. Key words: BoLA, production traits, Ayrshire

Score productive longevity of dairy cattle is an important scientific and practical importance. It is known that among genetic factors, significant effects of linear affiliation of their parents. It is demonstrated that effective breeding subject to strict compliance with the screening system, selection and evaluation of animal breeding values reported by domestic and foreign authors. The purpose of the work was to study the influence of linear membership on productive longevity and lifetime milk productivity of Holstein cattle of European breeding. Studies carried out on cows Holstein breed using data management system dairy cattle breeding "Orsek". Found that targeted breeding work and selection in a herd of imported European cattle breeding Holstein using information systems selection bulls MAR company CRI meets the latest requirements of the ICAR and Subcommittee on the unified international assessment of manufacturers (Interbull) contributed to a change in the number of separate lines among the cows first generation. This has led to an increase in the percentage of manufacturers lines daughters Valiant 1650414.73, Starbuc  352790.79, as well as Cheef 1427381.62. on the basis of the desired economic-useful signs. It is established that the value of productive longevity of cows depending on linear conditioning averaged 3.1 ... 3.9 lactations. By this measure, the daughter of Starbuck's lines were superior animals Holstein breed other lines by the number of lactations: Chevalier – 0.1 or 2.6% (td = 0.25; P ≤ 0.95); Bell – 0.5 or 14.7% (td = 0.95; P ≤ 0.95); Valiant – at 0.8 or 25.8% (td = 1.52; P ≤ 0.95); Eleveyshn – 0.8 or 25.8% (td = 1.91; P ≤ 0.95); Cheef – 0.5 or 14.7% (td = 1.27; P ≤ 0.95). It has been proven that milk productivity and milk quality of the imported Holstein cows and their offspring of domestic generation attributable to linear. The highest lifetime yield of Holstein breed cows had Starbuck lines, which outnumbered their import contemporaries other lines: Chevalier – 2818.7 kg or 8.2% (td = 0.46; P ≤ 0.95); Bell – 3404.5 kg or 10.1% (td = 0.61; P ≤ 0.95); Valiant – 5553.8 kg or 17.5% (td = 1.26; P ≤ 0.95); Eleveyshn – 7368.9 kg or 24.6% (td = 1.78; P ≤ 0.95); Cheef – 5500.5 kg or 17.3% (td = 1.38; P ≤ 0.95). The father's genotype had a significant impact on the quality of the milk. It should be noted that cows Starbuck's line differed relatively high fat content in the milk. By this measure the superiority of animals this line over imported cows other lines: Chevalier – 0.5% (td = 2.38; P ≥ 0.95); Bell – 0.2% (td = 1.38; P ≤ 0.95); Valiant – 0.6% (td = 2.78; P ≥ 0.95); Eleveyshn – 0.4% (td = 3.23; P ≥ 0.99); Cheef - 0.2% (td = 1.44; P ≤ 0.95). However, large protein content in milk cows belonging to different lines of Bella, who on this indicator had superiority over animals other lines: Chevalier – 0.2% (td = 1.03; P ≤ 0.95); Starbuck – 0.1% (td = 0.59; P ≤ 0.95); Valiant – 0.5% (td = 2.31; P ≥ 0.95); Eleveyshn – 0.2% (td = 1.85; P ≤ 0.95); Cheef – 0.1% (td = 1.15; P ≤ 0.95). It should be noted that only the cows first generation Valiant lines and Eleveyshn marked positive relationship between life and the content of cows in milk fat and protein (r = +0.17 ... +0.40), which may indicate the possibility of holding breeding work to improve these symptoms. So in the breeding work it is advisable to use manufacturers of prominent lines that combine high breeding value on productivity of daughters with improving effect on the duration of their use. The results of the research show that the effective use of well-known manufacturers and imported Holstein breed brood in breeding work with dairy cattle of Ukraine. However, shortening the productive use of animals in conditions of industrial technology should serve as a stimulus to study and find ways to solve this problem.

A related Holstein breed has recently been used to improve domestic dairy cattle and create highly productive herds by purchasing semen from breeding bulls, as well as a large number of heifers and calves of foreign breeding. The purpose of the work is a comparative assessment of the productive qualities of Black Pied and Holstein cows of foreign breeding. Purebred Holstein cows differ from animals of Black Pied breed in milk yield per lactation. Cows of both breeds have fairly high suitability for machine milking. The priority remains with the animals of the Holstein breed of foreign selection. In terms of milking intensity and udder volume, they reliably surpass Black Pied cows at P≤0.05 - P≤0.01, respectively, in terms of indicators. Holstein cows were also the best in the udder index. Correlation coefficients between the productive qualities, as well as between the quantitative indicator of productivity and the qualitative indicators of the suitability of cows for machine milking, were the same or did not differ significantly. A positive correlation was established between milk yield and the duration of the service period from 0.1 to 0.05, depending on the breed.

Abstract. Introducing resistance to mastitis into breeding programmes for dairy cattle seems to be one of the possible methods for limiting the increasing number of mastitis cases. Looking for candidate genes for genetic markers is one of the strategies. The BoLA–DRB3 alleles have considerable promise as potential mastitis marker. The objective of the study was to evaluate relationships between two BoLA–DRB3 alleles (BoLA–DRB3.2*16 and BoLA-DRB3.2*23) and estimated breeding value (EBV) for somatic cell count (SCC) in Polish dairy cattle. A total of 525 Polish Holstein cows in two experimental farms were evaluated for test-day SCC. The polymorphism of BoLA–DRB3 gene was identified using the MPT-PCR method. The variance components for test-day SCC were estimated by the REML method using animal model. The breeding value of SCC was calculated according to the BLUP method. Linear contrast between model-adjusted breeding values of SCC for all animals was used to test for differences between genotypes. In Polish Holstein cows population BoLA allele DRB3.2*23 was associated with increase of EBV for SCC in milk. It was observed increase of EBV for SCC from cows carrying this allele comparing to cows carrying BoLA–DRB3.2*16 (p ≤0.07) or other alleles (p ≤0.01).

To establish the duration of a severe feed restriction necessary to reversibly decrease milk production in high-producing Holstein cows, milk, plasma and urine parameters were assessed every 6 h during a 24-h restriction and 18-h refeed period. The 24-h restriction decreased yields of milk, protein and lactose by more than 30% without inducing ketosis or impairing milk production post-trial. According to the current “Instruction” used in dairy cattle selection and breeding in the Republic of Kazakhstan, bulls-producers of dairy breeds are assessed according to the their offspring quality based on the principle of “peer daughter”. This means that the phenotypic indicators of the daughters of the tested bulls are compared with the corresponding indicators of their peers. In European countries with developed dairy cattle breeding, as well as in Canada, the USA, etc., to ensure a reliable forecast of the genetic value of individuals (primarily, bulls-producers), use is made of the best linear unbiased forecast method (BLUP method). This method implies that the breeding value of producers is determined by the deviation values of the development of traits of the examined animal from its average values in the population. Especially urgent area is the research aimed at improving breeding programs, including assessing the breeding value of bulls-producers of dairy breeds using BLUP methods based on the productive qualities of the mass of dairy cattle in the Republic of Kazakhstan. The research material included the data on the phenotypic indicators of the milk productivity of first-calf cows (the amount of milk yield, the content of fat and protein in milk, the yield of milk fat and protein) of the Holstein black-motley dairy cattle breed, obtained from the information and analytical database of the Republic of Kazakhstan for 2016–2017. It was found that when evaluating according to the official “Instruction”, 16 sires out of 256 bulls (6.2%) got the stud category in 2016, 14 sires (9.2%) out of 152 bulls in 2017, and – 30 sires of 249 bulls (12.0%) over the cumulative period. The results of the conducted research prove that the use of the classic “Instructions” in dairy cattle breeding has lower efficiency (by 42.8–90.0%) as compared with the assessment of the breeding value of bulls based on the BLUP method.The selection of sire bulls into breeding groups based on the “peer daughter” methodology is not reliable enough and rather ineffective. Comparing the results of assessing the breeding qualities of sire bulls, obtianed using two methods in all compared periods (2016, 2017, 2016–2017), the authors established a clear superiority of the BLUP method over the current Instruction used in the Republic of Kazakhstan.

The main goal of dairy breeds selection should be improving breeding and productive qualities of animals under modern conditions. The majority of farms, using native breeds to produce milk, has created optimal conditions for keeping and feeding, selection and matching, growing of replacements etc. Further improvement of created native dairy breeds for economically useful traits occurs at total use of purebred Holstein bulls (semen) of foreign selection. In order to realistically assess milk productivity (milk yield, fat content in milk and fat yield) of Ukrainian Black-and-White and Red-and-White Dairy cows should be conducted a comparative analysis of Holstein cows under the same conditions of feeding and keeping. It was established that Ukrainian Red-and-White Dairy cows were characterized by the highest milk yields for 305 days of all lactations, taken into account, the among three investigated breeds. Their milk yield during the first lactation was 5933 kg of milk, during the second – 6393 kg, the third – 6391 kg and during higher lactation – 6650 kg. Ukrainian Black-and-White Dairy cows were second by milk yield (except for the second lactation), during the first lactation – 5932 kg of milk, the third – 6462 kg and higher – 6541 kg, and Holstein cows were third, during the first lactation – 5794 kg of milk, the second – 6381 kg, the third – 6335 kg and higher – 6469 kg. The fat content was almost the same and varied within 3.49-3.58% in milk of Ukrainian Red-and-White Dairy cattle, 3.50-3.60% in milk of Ukrainian Black-and-White Dairy cattle and 3.50-3.56% in Holsteins’ milk. The difference between the breeds was within 0.01-0.04%. All the investigated breeds had predominance in fat yield for three lactations over standards of these breeds: Ukrainian Red-and-White Dairy cows from 75.1 to 93.4 kg, Ukrainian Black-and-White Dairy cows – 75.1-89.0 kg respectively and Holstein cows – 41.9-60.2 kg. It was found different level of positive correlation between milk yield and fat yield in all the cases and high correlation (r = 0.604-0.921, P < 0.001) in five cases (41.7%) Negative correlation coefficients indicate that selection of animals to higher milk yield in the herd will decrease the second trait – fat content in milk. Positive and highly significant correlation between milk yield and fat yield indicates that selection of cows in the herd to higher milk yields will increase fat yield. It was revealed that bulls were among the factors impacted the milk productivity (milk yield, fat content, fat yield) of three investigated breeds. So, the force (η²x) of father’s impact on milk yield was15.4-47.9%, fat content – 22.0-43.4% and fat yield – 14.9-47.7% taking into account a lactation and a breed. The force of lines impact (η²x) was second; it was on milk yield 6.1-24.5%, fat content – 4.1-17.1 and fat yield – 5.8-23.5%. The force of breeds impact (η²x) was last; it was on milk yield 0.3-2.9%, fat content – 0.2-0.3% and fat yield – 0.6-2.7%. So, the comparative studies of milk productivity of Ukrainian Red-and-White and Black-and-White Dairy cattle with Holsteins indicate that under similar conditions of feeding and keeping, these native breeds can compete with Holstein cattle. The milk yield for 305 days of higher lactation was 6650 kg of milk in Ukrainian Red-and-White Dairy cows, 6541 kg in Ukrainian Black-and-White Dairy cows and 6469 kg in Holsteins. It was found the inverse correlation r = -0.025-0.316 between milk yield and fat content in milk in most cases. Selection and matching of animals in the herd should be carried out simultaneously on these traits. It was found positive repeatability of milk yields between the first and second, the third and higher lactations (rs = 0.036-0.741), indicating the reliability of forecasting increase in milk productivity during the next lactations in all herd. Bulls have the greatest impact (η²x) on milk productivity among the factors taken into account: milk yield – 15.4-47.9%, fat content in milk – 22.0-43.4% and fat yield – 14.9-47.7%.

От породы коров и ее особенностей зависят эффективность использования кормов, качество и технологические свойства молока. Материалом исследований служили коровы чернопестрой голштинской и айрширской пород. Для повышения качества сенажа из козлятника восточного использовали микробиологический препарат. В готовом корме с биоконсервантом содержание сухого вещества было выше на 3,66, обменной энергии на 8,04, сырого протеина на 10,86, переваримого протеина на 13,59. Улучшение качества корма оказало положительное влияние на метаболические процессы в рубце подопытных коров. Независимо от технологии приготовления сенажа коровы айрширской породы лучше переваривали, по сравнению с голштинами, сухое вещество на 2,062,91 (Р0,05), органическое на 1,212,04 (Р0,05), сырой протеин на 2,222,49 (Р0,05), БЭВ на 0,921,36, но при этом голштины лучше переваривали сырой жир на 2,732,78 (Р0,05) и сырую клетчатку на 1,551,86. В результате внесения в сенаж биоконсерванта Силостан и улучшения переваримости корма у коров опытных групп значительно улучшился химический состав и технологические свойства молока. Очень важно для сыроделия, что в молоке коров, получавших в рационе сенаж с биоконсервантом, повышается массовая доля казеина на 2,42,7, а доля сывороточных белков и фракции казеина, которые не свертываются под действием сычужного фермента, наоборот, снижается на 1,41,5 и на 0,30,9, соответственно. Уменьшается продолжительность свертывания молока сычужным ферментом, снижаются потери сухого вещества с сывороткой, и увеличивается выход казеинового сгустка. Плотность казеинового сгустка повышается в молоке голштинских коров на 14,3, айрширских на 5,7 и составляет, соответственно, 1,92 и 2,96 г/см2.The efficiency of feed use, the quality, as well as technological properties of milk depend on cow breed and its characteristics. Blackandwhite Holstein and Ayrshire breeds were used as the material for research. Microbiological preparation was used to increase the quality of halyage made of Eastern galega. The content of dry substance in the readymade feed was 3.66 higher, the metabolizable energy 8.04 higher, raw protein 10.86 higher, and digestible protein 13.59. Improvement of feed quality positively influenced the metabolic processes in the rumen of experimental cows. Without regard to the technology of halyage preparation, cows of Ayrshire breed showed betterdigestion of dry substance by 2.06 2.91 (P0.05) than Holstein cows, organic substance by 1.21 2.04 (P0.05), raw protein by 2.22 2.49 (P0.05), nitrogenfree extractive fraction by 0.92 1.36, but Holstein breed showed better digestion of raw fat by 2.73 2.78 (P0.05) and raw fiber by 1.55 1.86. Chemical composition and technological properties of milk improved significantly as a result of addition of Silostan biopreservative to the halyage and improvement of feed digestibility in cows from test groups. The fact that the mass percentage of casein is increased by 2.4 2.7, and the share of serum proteins and ycasein fractions which do not coagulate under the influence of enzyme rennet is inversely decreased by 1.4 1.5 and 0.3 0.9, accordingly, is very important for cheese making. The duration of milk coagulation with rennet is decreased, the loss of dry substance with serum is decreased, and the yield of casein coagulate is increased. Density of casein coagulate is increased in the milk of Holstein cows by 14.3, Ayrshire cows by 5.7, and equals to 1.92 and 2.96 g/cm2, accordingly.

Abstract NFκB1/p105 is the critical member of the NFκB family which can suppress inflammation, ageing, and cancer when p50/p50 homodimer is formed. Currently, the research about the role of NFκB1/p105 during cow mastitis is limited. Here, we analyzed the correlation of six single-nucleotide variants of the NFκB1 gene with somatic cell count, milk yield, milk fat content, and milk protein content in 547 Chinese Holstein cows, and explored the mRNA expression profiles of the NFκB family and ubiquitin ligases (βTrCP1, βTrCP2, KPC1, KPC2) in LPS-induced bovine mammary epithelial cells (MECs) by transcriptome-Seq. The association analysis showed that cows with SNV2-TT and SNV6-CC in the NFκB1 gene had significantly higher milk protein content (P &lt; 0.05), while cows with SNV5-TT in the NFκB1 gene had significantly lower somatic cell score (SCS), but CC genotype at SNV5 locus was not detected in our Holstein cows. The transcriptome-Seq results demonstrated the mRNA expression of NFκB1 was increased and peaked at 4 h post-induction, while the mRNA expressions of both KPC1 and BCL3 that promote the anti-inflammation function of NFκB1/p105 were decreased in LPS-induced bovine MECs. TNFAIP3, an inhibitor of both degradation and processing of p105 precursor, was markedly increased by more than 3 folds. Furthermore, bta-miR-125b which targets at the 3ʹUTR of TNFAIP3 was reduced by 50%. These results indicated that SNV5-TT of the NFκB1 gene with lower SCS may be an anti-mastitis genotype that could cope with infection more efficiently in Chinese Holstein cows. In addition, the anti-inflammation role of NFκB1/p105 seemed to be inhibited in LPS-induced-bovine MECs because the formation of the p50/p50 homodimer was arrested. This study provides a new perspective to understand the inflammatory mechanism in dairy mastitis.
