IN SITU DEGRADATION OF FEED INGREDIENTS, FERMENTATION PATTERN AND MICROBIAL POPULATION AS INFLUENCED BY RUMINAL AMMONIA CONCENTRATION

Effects of ruminal ammonia concentration on ruminal parameters and in situ degradation of feed ingredients were examined with nonlactating Holstein cows fed a complete mixed diet (70% barley silage and 30% concentrate mixture, dry matter (DM) basis). Ruminal ammonia concentrations of 16.3, 24.8 and 34.9 mg 100 mL−1 were achieved by continuous intraruminal infusions of NH4Cl. Ammonia concentrations in control animals were 11.2 mg 100 mL−1. Ruminal fluid pH decreased (P < 0.05) and blood urea N increased (P < 0.05) with NH4Cl infusion. NH4Cl infusion increased (P < 0.05) viable counts of proteolytic bacteria. Effective degradability of dry matter (EDDM) of concentrate ingredients and crude protein (EDCP) of soybean meal and fish meal were not affected by NH4Cl infusion. But EDCP of corn gluten meal was highest (P < 0.05) at ammonia concentration of 16.3 mg 100 mL−1 rumen fluid and EDDM and neutral detergent fiber (EDNDF) of barley silage decreased (P < 0.05) with increasing NH4Cl infusion. Results are interpreted to indicate that ruminal ammonia concentrations necessary for maximal bacterial growth may differ from concentrations required for maximal degradation of feedstuffs. However, decreased pH associated with NH4Cl infusion may have influenced the results obtained. Key words: Ammonia concentration, bacterial growth, in situ degradation

Abstract Six ruminally cannulated steers (average BW = 791 + 71 kg) were used in a replicated 3 × 3 Latin square experiment to determine the effects of roughage type on rumination, fiber mat characteristics, and rumen fermentation variables. Three roughages were included at 7% (DM basis) in a steam flaked corn-based diet; cotton burrs (CB), wheat silage (WS), or corn stalks (CS). Steers were fitted with a sensory collar to record rumination behaviors in 2-h intervals at the beginning of the experiment. Each 30-d period consisted of a 7-d of recovery, 14-d of diet adaptation, 7-d of rumination data collection (daily and bi-hourly average rumination), 1-d of rumen fluid collection, and 1-d of rumen evacuations. In situ degradation of individual roughages was determined for 4-d after period 3 evacuations. During rumen evacuations, ruminal contents were removed; the rumen fiber mat (RF) was separated from the liquid portion with a 2 mm sieve, weighed, and a subsample was dried. Data were analyzed using the MIXED procedure of SAS with steer as the experimental unit and roughage (CB, WS, and CS) as the main effect. Dry matter intake (DMI) was not different for CB and WS (P = 0.25) and greatest for steers consuming CS diet (P  &lt; 0.01). Roughage type did not influence the weight of the RF dry matter (%; DM; P = 0.92), RF weight (P = 0.69), or RF:DMI ratio (P = 0.29). Daily rumination (min/d) did not differ among roughages (P = 0.40), but min of rumination/kg of DMI was greatest for CS (18.0 min), min/kg of NDF was greatest for WS (89.8 min; P = 0.02), and min/kg of peNDF was greatest for CS (132.4 min; P  &lt; 0.01). Wheat silage had the greatest percentage of soluble DM and CB-R and CS-R (P  &lt; 0.01) had the greatest ruminal degraded DM fraction. Rumen fiber mat did not differ for roughages, although rumination min/kg of DMI and peNDF was greatest for steers consuming CS and WS. In situ degradation determined that CB-R and CS-R had the greatest percentage of ruminal degraded DM. Based on the objective of the experiment, roughage type did not influence daily rumination or fiber mat characteristics.

Four Hereford steers (471 ± 31 kg) fitted with ruminal cannulae were used in a 4 × 4 Latin square design to evaluate the effects and possible interactions between silage particle size and concentrate level on digestion and ruminal fermentation parameters. Diets were designed with two forage to concentrate ratios [low forage, 20:80, high forage, 50:50 on a dry matter (DM) basis], combined with two theoretical chop lengths of barley silage; short (SBS) = 4.7 mm and long (LBS) =18.8 mm. Results showed that steers fed diets with SBS consumed more DM than those fed LBS. An interaction (P < 0.05) between particle size and concentrate level was noted for molar proportion of acetate. At the higher concentrate level, feeding SBS diets reduced the molar proportion of acetate but increased the molar proportion of propionate. An interaction (P < 0.05) between particle size and concentrate level was noted for ruminal pH. When compared to LBS, the daily mean rumen pH was lower (P < 0.05) for steers fed SBS with 80% concentrate but not for those fed the 50% concentrate diet. Ruminal ammonia concentration was higher (P < 0.05) for those steers fed the 50% concentrate diets than for those fed the 80% concentrate diets. With high concentrate levels, SBS greatly reduced (P < 0.05) ammonia concentration. Passage rates for particulate matter and the retention time were decreased (P < 0.05) when SBS was fed. Feeding SBS reduced (P < 0.05) total tract digestibility of dry matter, crude protein (CP) and neutral detergent fiber (NDF) (65.5 vs. 68.5%, 66.5 vs. 73.4% and 39.4 vs. 45.9%), respectively. These results suggest that barley silage with a theoretical cut length of 18.8 mm can promote higher rumen pH, ammonia nitrogen, and reduced particulate passage rates. These changes in rumen fermentation parameters should favor optimum rumen function and digestion when high concentrate diets are fed. The yield and feeding value of barley (Hordeum vulgare 'Johnson'), oat (Avena sativa 'Calibre') and triticale(Triticosecale 'Carmen') silages were compared in a field study, a heifer growth trial (119 d in length, with four pens of eight Charolais cross heifers (average body weight 300 kg) per treatment), and a sheep digestibility trial (total fecal collection with eight sheep per treatment). The swathed and chopped plant material was ensiled in stack silos covered with plastic. Yields of dry matter (DM) and crude protein (CP) did not differ significantly (P > 0.05) among species. At harvest, oat and triticale had greater acid detergent fiber (ADF) levels than barley. After ensiling, triticale alone had greater neutral detergent fiber, ADF and acid detergent insoluble nitrogen levels than the other silages. The three silages did not otherwise differ in chemical composition. Heifer intakes of barley (6.06 kg d−1) and oat (5.70 kg d−1) silage DM were similar, and greater than for triticale (4.86 kg d−1). Average daily gain was greatest with barley (0.65 kg d−1), intermediate with oat (0.57 kg d−1) and least with triticale (0.49 kg d−1). Intake of silage DM by the sheep was greatest with barley silage (741 g d−1), intermediate with oat (617 g d−1) and least with triticale (434 g d−1), which appeared less palatable than the other silages. Dry matter and CP digestibilities were greatest for barley silage (64.2 and 71.6%, versus 58.3. and 61.1% for oat, and 58.8 and 65.4% for triticale, respectively). Oat silage had the lowest fiber digestibility. Sheep given triticale silage had lower rumen fluid volatile fatty acid concentrations, apparently due to low DM intake. Oat silage gave a greater molar proportion of acetate, a lower proportion of propionate, and a higher acetate to propionate ratio than either barley or triticale. Triticale silage produced a greater molar proportion of acetate and a lower proportion of butyrate than barley silage. Based on animal performance, it was concluded that barley silage was the preferred of the three cereal silages for growing cattle. Triticale produced a less acceptable silage due to poor palatability and low DM intake. Key words: Cattle, sheep, cereal silage, forage quality, digestibility

Abstract Five ruminally cannulated heifers were used in an incomplete 6 × 6 Latin square design to determine the effects of cereal silage (barley vs. corn), cereal grain (barley vs. corn vs. a 50:50 blend of barley and corn), and their interaction (S × G) on dry matter intake, ruminal fermentation, total tract digestibility, nitrogen balance, and in situ degradation. Corn silage (CS) or barley silage (BS) was included at 8% of dietary dry matter (DM). Within each silage source, diets contained (DM basis) either dry-rolled barley (BG; 86%), dry-rolled corn (CG; 85%), or an equal blend of barley and corn (BLEND; 85%). Periods were 25 d, with 5 d of dietary transition, 13 d of dietary adaptation, and 7 d of data and sample collection. Samples collected included feed and refusals, total urine and feces, and ruminal fluid. All data were analyzed using the Mixed model of SAS with the fixed effects of silage, grain, and the S × G. Dry matter intake (P ≥ 0.19) and mean ruminal pH (P ≥ 0.096) were not affected by the silage, grain, or S × G. Total short-chain fatty acid concentrations were greater for BLEND than BG or CG (grain, P = 0.003) and for CS (silage, P = 0.009) relative to BS. The molar proportion of acetate was greater for BS-BG and BS-CG (S × G, P &lt; 0.001), while molar proportion of propionate was greater for CS-BG (S × G, P &lt; 0.001) relative to other silage and grain source combinations. Rumen ammonia-N concentration was greater for CG than BG, or BLEND (grain, P &lt; 0.001), and greater for CS compared to BS (silage, P = 0.023). Apparent total tract digestibility of DM, organic matter, neutral detergent fiber, starch, and gross energy were greatest for BG (grain, P ≤ 0.035). Digestible energy content (Mcal/kg) was greater for BG (grain, P = 0.029) than CG and BLEND. Total nitrogen retention (g/d and % of intake) was greatest for CS-BG (S × G, P ≤ 0.033) relative to all other treatments. In situ degradation rates of DM, crude protein, and starch were greater for BG than CG (P ≤ 0.004). The potentially degradable fraction of DM, crude protein, and starch was greater for CG (P ≤ 0.031), while the undegradable fraction was greater for BG (P ≤ 0.046). For silage sources, CS had greater 24 h in situ DM digestibility (P = 0.009) and starch digestibility (24, 48, and 72 h incubations, P ≤ 0.034) relative to BS. Results suggest that while feeding a combination of CS and BG promotes propionate production and greater N retention; few other additive effects were observed.

The amaranth plants showed high potential feed value as forage for ruminants. An in-depth study of this plant, particularly in cattle, will help extend its utilization as an alternative protein and fiber feed source in cattle feeding. In this study, the nutrient compositions of three different species of amaranth, Amaranthus caudatus L., Amaranthus cruentus L., and Amaranthus hypochondriacus L.—two varieties for each species, A.ca 74, A.ca 91, A.cu 62, A.cu 66, A. hy 30, and A. hy 48—were evaluated. The in vitro technique was used to evaluate the fermentation characteristics such as total gas production, total volatile fatty acids (VFA) concentration, pH, and ammonia concentration of the rumen fluid. Moreover, the effective degradabilities of dry matter (EDDM) and crude protein (EDCP) of the amaranth forages were determined through in situ bag technique. The amaranth forages: A. caudatus, A. cruentus, and A. hypochondriacus showed better nutritive value than the locally produced forages in Chungcheong province of Korea. The CP of the amaranth ranged from 11.95% to 14.19%, and the neutral detergent fiber (NDF) and acid detergent fiber (ADF) contents ranged from 45.53% to 70.88% and 34.17% to 49.83%, respectively. Among the amaranth varieties, A. hypochondriacus 48 showed the most excellent ruminant feed nutrient quality (CP, 14.19%; NDF, 45.53%; and ADF, 34.17%). The effective degradabilities of dry matter (EDDM; 33–56%) and crude protein EDCP (27–59%) of the amaranth were lower compared to other studies, which could be due to the maturity stage at which the forages were harvested. Nonetheless, A. hypochondriacus 48 showed the highest EDDM (56.73%) and EDCP (59.09%). The different amaranth species did not differ greatly in terms of total VFA concentration or molar proportions, total gas production, or ammonia-N concentration. The high nutrient composition, and highly effective degradability of dry matter and crude protein, coupled with the favorable fermentation characteristics, suggest that the amaranth forages showed good to excellent feed quality for cattle.

Cereal grains are often subjected to physical processing before being fed to equids. However, little information is available on how physical processing of cereals affects degradation dynamics in equids. This experiment examines the effect of two physical processing methods (micronisation and extrusion) on in situ degradation of barley in the caecum of poniesThree caecally fistulated mature Welsh-cross pony geldings (approx. LW 270kg) were offered ad libitum grass hay plus minerals. Incubation bags (monofilament polyester 6.5 x 20cm, 41μm pores, 16mg/cm2 sample size) containing either unprocessed barley (UB), micronised barley (MB) or extruded barley (EB) were incubated in the caecum for fixed times according to both a forward (0, 2, 4, 6, 12, 8, 24, 48h) and reverse (48, 24, 8, 4, 12, 6, 2, 0h) incubation sequence. For each feedstuff residues from each time were bulked within pony and across incubation sequence for subsequent analysis of dry matter (DM) and starch (STC). Degradation profiles were fitted to the DM and STC disappearance data according to Ørskov and McDonald (1979).

Two adult male sheeps fitted with rumen cannula were used in two experiments to study the effects of synchronization of carbohydrate and nitrogen supply in sugarcane bagasse based total mixed ration (TMR) on in situ nutrient degradation. The first experiment was aimed to create three TMR with different synchronization index. Ingredient feedstuffs of TMR were evaluated for its in situ organic matter (OM) and nitrogen (N) degradation kinetics. On the basis of the OM and N degradation kinetics of feedstuffs, three sugarcane bagasse based TMR were formulated with synchronization indexes of 0.37; 0.50; and 0.63; respectively. The TMR had similar levels of crude protein (CP), total digestible nutrients, and neutral detergent fiber (NDF). In the second experiment, the three TMR with different synchronization index were evaluated for in situ degradability characteristics of OM, CP, NDF, and sulfur. The in situ degradation of OM in TMR were decreased (P<0.05) with the increasing of synchronization index. The higher synchronization index in TMR increased (P<0.05) CP degradation of CP. The NDF degradation decreased slightly by the alteration of synchronization index in TMR. The higher synchronization index in TMR reduced (P<0.05) in situ sulfur degradation, and this may not support to effects of synchronization of carbohydrate and nitrogen supply.

The degradation characteristics of dry matter (DM), protein, neutral detergent fiber (NDF) and acid detergent fiber (ADF) of corn–sunflower intercrop silage (CSS) and monoculture corn silage (CS) prepared from whole plants, harvested at two stages of maturity, were compared using eight mature rumen-cannulated steers. The degradation characteristics were determined by incubating the silages in nylon bags for up to 72 h in the rumen of animals fed the respective silages. The degradation characteristics determined for the silages were the soluble fraction, the insoluble but degradable fractions, the rate of degradation of the degradable fractions of silage DM and protein, and the in situ disappearance of NDF and ADF after 0.5 and 72 h of incubation. The rate of particulate passage from the rumen was determined using chromium-mordanted NDF of the four silages. Values were used to estimate effectively degraded DM and protein. The rates of DM and protein degradation were highest for late-cut CSS (6.3 and 6.0% h−1, respectively) and the least for late-cut CS (2.5 and 0.8% h−1, respectively). Averaged across stages of maturity, more (P < 0.05) DM and protein were effectively degraded with CSS (57.4 and 70.1%, respectively) than with CS (48.8 and 48.7%). Degradation of NDF in early-cut CSS was lower (P < 0.05) than in CS after 72 h of incubation. ADF disappearance from all of the silages after 72 h of incubation was similar. Ruminal degradation of DM and protein in CSS was greater than in CS, which may affect efficiency of utilization of CSS. <p>Para cuantificar la degradación <em>in situ </em>de tejidos vegetales y su relación con la composición química de especies forrajeras se seleccionaron láminas foliares de las gramíneas <em>Bouteloua repens </em>y <em>Bothriochloa pertusa </em>y foliolos de las leguminosas <em>Stylosanthes scabra</em>, <em>Desmodium barbatum </em>y <em>Tephrosia cinerea</em>. En el rumen se incuba­ron muestras de 10 mm de largo durante 0, 12, 24, 48 y 72 horas; para la lectura del área residual se digitalizaron ocho tejidos. A las 24 y 48 h de incubación <em>B. pertusa </em>presentó relaciones positivas entre la degradación de la epidermis adaxial (EA) y la FDN (R2= 90,2), entre el mesófilo y la DIVMS (R2=80,1), y entre los tejidos len­tamente degradables (TLD) y la FDA (R2= 83,9); y relaciones negativas entre EA y DIVMS (R2= –73,1), los TLD con la DEF (R2= –74,3), la EA con la PC (R2= –87,6), y el esclerenquima con la DIVMS y la PC (R2= –84,3 y R2= –90,8). Después de 72 horas de incubación las mayores áreas residuales en gramíneas fueron la estructura kranz entre 34,2% y 36,5%, el mesófilo entre 20,9% y 21,4%, el xilema de 11,5% a 17,1% y la EA entre 11,8% y 13,9%; no obstante el esclerénquima se relacionó estrechamente con indicadores de calidad nutricional. Las gramíneas obtuvieron mayores porcen­tajes de tejidos no degradables que las leguminosas; por lo tanto, la DIVMS y la DEF fueron menores. Entre las leguminosas, <em>S. scabra</em>, presentó la mayor digestibilidad de los tejidos rápidamente degradables, tanto en sequía como en lluvia; en la época seca se incrementó en 134,6% la degradación de estos tejidos. Adicionalmente, en esta especie el área residual de los taninos no presentó relación directa con la degra­dación del mesófilo. En leguminosas la tasa y extensión de la degradación de las epidermis se incrementaron marcadamente a mayor tiempo de incubación, presen­tándose diferencias entre especies, situación que no exhibieron las gramíneas. En <em>D. barbatum </em>la degradación de la EA se asoció con la DIVMS y la DEF, explicando en 77,4% y 72,95% estos porcentajes; de igual manera se reportaron relaciones negati­vas entre la degradabilidad de la epidermis y los porcentajes de FDN, FDA, lignina y taninos (R2= –0,76; R2= –0,79; R2= –0,53 y R2= –0,76, respectivamente).</p><p> </p><p><strong>Microbial <em>in situ </em>degradation of grasses and legumes leaf tissues and its realtionship with nutritional quality and precipitation</strong></p><p>A study was conducted to measure the <em>in situ </em>de- gradation of plant tissues and its relationship with forage chemical composition in leaves of the grasses <em>Bouteloua repens </em>and <em>Bothriochloa pertusa </em>and of the legumes <em>Stylosanthes scabra</em>, <em>Desmodium barbatum </em>and <em>Tephrosia cinerea</em>. Forage samples of 10 mm in length were incubated in the rumen for 0, 12, 24, 48 and 72 h and tissue residual area was determined by reading the residual area in eight digitalized leaf samples per forage. In <em>B. pertusa</em>, after 24 and 48 h of incubation there were positive relations between the degradation of adaxial epidermis (AE) and the neutral detergent fiber (NDF, R2 = 90.2), between mesophyll and in vitro dry matter degradability (IVDMD, R2 = 80,1), and between slowly degradable tissues (SDT) and the acid detergent fiber (ADF, R2 = 83.9); and negative relationships between AE and IVDMD (R2 = -73,1), between SDT and effective dry matter degradation (EDMD, R2 = -74,3), between AE and crude protein (CP, R2 = -87,6), and between sclerenchyma and IVDMD with CP (R2 = -84,3 and R2 = -90,8, respectively). After 72 h of incubation, the greater residual areas in grasses were the Kranz structure (34.2% to 36.5%), mesophyll (20.9% to 21.4%), xylem (11.5% to 17.1%) and AE (11.8% to 13.9%). The area of sclerenchyma was related close­ly to indicators of nutritional quality. Overall, the grasses had greater content of non-degradable tis­sues than the legumes; therefore, their IVDMD and EDMD were lower. Among the legumes, <em>S. scabra </em>showed the greater digestibility of rapidly degrada­ble tissues, both during the rainy and dry seasons. At the dry season, the degradation of these tissues increased by 134.6%. Additionally, in this legume, the content of tannins did not have a direct relation­ship with the degradation of mesophyll. The rate and extension of the degradation of epidermis in legumes, were noticeably increased with increasing length of incubation, existing differences between species, a situation which was not observed in the grasses. In <em>D. barbatum</em>, the degradation of the AE was positively associated with both the IVDMD and EDMD (R2 = 77,4% and 72,95%, respectively). On the other hand, the degradability of epidermis and the percentage of NDF had negative relations with ADF, lignin and tannins (R2 = -0,76; R2 = -0,79; R2 = -0,53 and R2 = -0,76, respectively).</p>

ABSTRACT The objective of this study was to evaluate the nutritive value of sugarcane silage with or without inoculation with P. acidipropionici or L. buchneri, over three fermentation periods. The experimental design was completely randomized in a 3 x 3 inoculant by fermentation period factorial arrangement (without inoculant, inoculant 1, inoculant 2; x three fermentation periods, 10, 60, 90 days). Values of pH, dry matter (DM), crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), hemicellulose (HEM) and lignin were determined and in situ DM degradability profiles were modelled for parameters a, b and c, potential degradation (A) and effective degradability (ED). The 90 day fermentation yielded a lower pH for both inoculants. There was an interaction between inoculant and fermentation period (P < 0.05) for DM content, with a reduction in silage DM without the additive at 90 days. The CP, HEM, ADF and lignin contents of sugarcane were not influenced by the treatments. The addition of P. acidipropionici provided the lowest NDF content at 10 days and presented a higher fraction a, potential degradation and ED. At 60 days, there was no variation in soluble fraction, the control silage showed a higher fraction b, higher potential degradation and ED. At 90 days of fermentation, L. buchneri silages presented a higher fraction a, degradation rate and DE and a higher b value was obtained in the silage without inoculant. Inoculants are effective in maintaining the silage DM content and nutritional value during prolonged fermentation periods.
