ASCORBIC ACID METABOLISM IN SWINE. THE EFFECTS OF FREQUENCY OF FEEDING AND LEVEL OF SUPPLEMENTARY ASCORBIC ACID ON SWINE FED VARIOUS ENERGY LEVELS

The effects of supplementary ascorbic acid and frequency of feeding on the performance of 3-wk-old, castrated, male swine fed 140, 240 and 340 kcal/kg W0.75 of metabolizable energy (ME) was investigated using a 3 × 3 × 2 factorial experiment. Frequency of feeding had no influence on the growth rate within any one energy group. Supplementary ascorbic acid up to the level of 1,000 mg per day produced improvement in growth rates at all levels of ME intake. The improvement in rate of growth appeared not to be due to water retention because there were no differences in creatinine excretion rates (per kg of body weight per day) between any of the groups. Ascorbic acid supplementation at the rate of 1,000 mg/day resulted in an increased proportion of the label from glucose-U-14C incorporated into the saponifiable fraction of an adipose tissue biopsy from the group fed a daily ration of 140 kcal ME/kg W0.75 in five equal amounts during the day. Supplementary ascorbic acid appeared to have no other effects on lipid metabolism. Excretion of ascorbic acid was significantly increased when there was an increased ME intake and an increased level of vitamin supplementation. These observations confirm an earlier report that the ascorbic acid available to the swine is a function of the energy level in the diet. It is suggested that observations (vide supra) made in ME-deprived animals may be due to decreased amounts of ascorbic acid physiologically available to the pig.

Supplying sufficient nutrients, such as dietary energy and protein, has a great effect on the growth and rumen development of ruminants. This study was conducted to evaluate the effects of dietary energy and protein levels on growth performance, microbial diversity, and structural and physiological properties of the rumen in weaned lambs. A total of 64 two-month-old Hu lambs were randomly allotted to 2 × 2 factorial arrangements with four replicates and with four lambs (half male and half female) in each replicate. The first factor was two levels of dietary metabolizable energy (ME) density (ME = 10.9 MJ/Kg or 8.6 MJ/Kg), and the second factor was two levels of dietary crude protein (CP) content (CP = 15.7% or 11.8%). The trial lasted for 60 days. A low dietary energy level restrained the growth performance of lambs (p < 0.05). The ruminal concentration of acetate and the ratio of acetate to propionate increased but the propionate concentration decreased significantly with the low energy diet. However, the rumen morphology was not affected by the diet energy and protein levels. Moreover, a low energy diet increased ruminal bacterial diversity but reduced the abundance of the phylum Proteobacteria (p < 0.05) and genus Succinivibrionaceae_uncultured (p < 0.05), which was associated with the change in ruminal fermentation phenotypes. By indicator species analysis, we found three indicator OTUs in the high energy group (Succinivibrionaceae_uncultured, Veillonellaceae_unclassified and Veillonellaceae_uncultured (p < 0.01)) and two indicator OTUs in the low energy group (Bacteroidales_norank and Lachnospiraceae_uncultured (p < 0.01)). In conclusion, these findings added new dimensions to our understanding of the diet effect on rumen microbial community and fermentation response, and are of great significance for establishing the optimal nutrient supply strategy for lambs.

Abstract Superoxide dismutase 1 suppresses oxidative stress within cells by decreasing the levels of superoxide anions. A dysfunction of the ovary and/or an aberrant production of sex hormones are suspected causes for infertility in superoxide dismutase 1-knockout mice. We report on attempts to rescue the infertility in female knockout mice by providing two antioxidants, ascorbic acid and/or coenzyme Q10, as supplements in the drinking water of the knockout mice after weaning and on an investigation of their reproductive ability. On the first parturition, 80% of the untreated knockout mice produced smaller litter sizes compared with wild-type mice (average 2.8 vs 7.3 pups/mouse), and supplementing with these antioxidants failed to improve these litter sizes. However, in the second parturition of the knockout mice, the parturition rate was increased from 18% to 44–75% as the result of the administration of antioxidants. While plasma levels of progesterone at 7.5 days of pregnancy were essentially the same between the wild-type and knockout mice and were not changed by the supplementation of these antioxidants, sizes of corpus luteum cells, which were smaller in the knockout mouse ovaries after the first parturition, were significantly ameliorated in the knockout mouse with the administration of the antioxidants. Moreover, the impaired vasculogenesis in uterus/placenta was also improved by ascorbic acid supplementation. We thus conclude that ascorbic acid and/or coenzyme Q10 are involved in maintaining ovarian and uterus/placenta homeostasis against insults that are augmented during pregnancy and that their use might have positive effects in terms of improving female fertility.

Abstract A meta-analysis was conducted to determine the standardized ileal digestible (SID) lysine (Lys) recommendations for 11–150 kg PIC pigs housed under commercial conditions. Data from 29 trials dating from 2013 to 2020 utilizing 48,338 pigs were recorded in a database. Number of pens/treatment and pigs/trial ranged from 9 to 16 and 12 to 25, respectively. Sire lines were PIC 337 in 25 trials, PIC 327 in 3 trials, and PIC TR4 and 327 in 1 trial. Dam lines were PIC Camborough in 18 trials and PIC Camborough 29 in 9 trials. The SID Lys to calorie ratio curves were built for both metabolizable energy (ME) and net energy (NE) systems using the feed ingredient composition in NRC (2012) for energy levels. The response variables average daily gain (ADG) and feed efficiency (G:F) were analyzed using generalized linear and non-linear mixed models with heterogeneous variance (Gonçalves et al., 2016). Each treatment mean within a trial (n = 288) was considered the experimental unit and each trial was used as a random effect. The models were developed for mixed gender pigs and the PIC 337 growth curves were used to estimate the recommendations for barrows and gilts. There was no evidence for an interaction between sire lines or dam lines and treatment (P &gt; 0.10). The SID Lys to calorie recommendations are based on the average for ADG and G:F (Table 1). At these levels, approximately 100% of maximum ADG and 99.4% of maximum G:F are achieved. The NE to ME ratio that results in a similar SID Lys recommendation as a percentage of the diet ranged from approximately 0.72 to 0.74. The updated biological requirements for PIC pigs remained similar compared to previous PIC recommendations (Gonçalves et al., 2017). However, the requirement estimates have been adjusted for late nursery and late finishing phases.

This study was carried out to evaluate the energy levels in the diet to obtain better performance rates and quality of eggs from laying hens in the second production cycle. One hundred and eighty Bovans Goldline laying hens with 62 weeks of age were used during four 28-day periods. A completely randomized experimental design was used with four metabolizable energy levels (2,650, 2,725, 2,800, 2,875 and 2,950 kcal/kg), each with six replicates of six birds. The energy level of diet did not affect the weight of the egg, yolk, albumen and eggshell, the percentages of yolk, albumen and eggshell, yolk color and egg specific gravity. Feed intake, egg production, egg mass and feed conversion per egg mass and per dozen eggs increased significantly with increasing levels of metabolizable energy. Feed intake decreased linearly as the energy level in the diet increased. The metabolizable energy levels showed a quadratic effect on egg production, egg mass and feed conversion per egg mass and per dozen eggs. The metabolizable energy level of 2,830 kcal/kg was the most appropriate to promote better performance and quality of eggs from laying hens in the second production cycle.
