In situ rumen disappearance of low and high fiber canola meal relative to regular canola meal was determined following 12 h of rumen incubation. In situ amino acid disappearance was similar in low and high fiber and regular canola meal except for threonine and glutamate which was lower (P < 0.05) in high fiber canola meal. A study was conducted to determine amino acid composition and ruminal degradability of linseed meal (LSM) relative to canola meal (CM). Linseed meal had higher (P < 0.05) arginine, isoleucine, and phenylalanine and less (P < 0.05) lysine than CM. Except for histidine, methionine, and threonine, ruminal escape values of essential amino acids were higher (P < 0.05) for LSM than CM. Key words: Linseed meal, amino acids, ruminal degradability

A study was conducted to determine the level and digestibilities of amino acids and the level of glucosinolates in toasted and non-toasted canola meal samples produced in commercial crushing plants in western Canada. A total of 26 non-toasted and 31 toasted canola meals samples were analyzed for apparent ileal amino acid digestibilities using broiler chickens between 21 and 28 d of age. Commercially toasted canola meal contained 5.6 g 16 g-1 N (range 5.3–5.9) of Lys with a digestibility ranging from 65.5 to 85.7% (mean = 78.0%). Non-toasted canola meal contained 6.0 g 16 g-1 N (range 5.7–6.3) Lys with digestibility values ranging from 87 to 92% (mean = 89.7%). Toasted canola meal had an average total digestible amino acid content (not including Tyr and Trp) of 69.6 g 16 g-1 N (range 56.6–75.6) while non-toasted canola meal had 77.6 g 16 g-1 N (range 73.1–82.0). The levels of aliphatic glucosinolates in the toasted (6.16 μmol g-1 meal, range 1.2–12.5) and non-toasted meals (10.5 μmol g-1 meal, range 7.8–15.1) were low and likely of nonutritional consequence. In conclusion, non-toasted canola meal contains higher levels of digestible amino acids than conventional toasted canola meal and therefore consideration should be given to modifying commercial systems to produce a non-toasted meal. The rumen degradability and postruminal digestion of rumen escape dry matter (DM), nitrogen (N) and metabolically essential amino acids (EAA) for canola meal (CM) from five different processing plants (samples 1–5) and one sample of soybean meal (SBM) were compared. Rumen degradability was estimated by incubating sample-filled small nylon bags in situ in the rumen of steers for 0, 4, 8, 12, 16 and 30 h. Postruminal digestion was estimated using a sequence of ruminal in situ incubations for 0, 4, 8, 12 and 16 h, in vitro incubation in an acid-pepsin solution and a mobile nylon bag technique distal to the abomasum. The rumen escape of nutrients for CM1 was consistently lower compared to other CM samples, while CM4 resulted in the highest escape of nutrients at 30 h. An estimate of maximum degradability of EAA measured at 30 h differed among CM samples, with the rumen escape of EAA being CM4 > CM2 = CM5 ≥ CM1 = CM3 > SBM. The DM, N and EAA content of mobile bag residues was relatively uniform among CM samples, particularly after 12 and 16 h of ruminal fermentation, but was higher for CM than for SBM, probably due to a low digestibility of CM hulls. The rumen degradability of CM from different processing plants differed and as a consequence the postruminal digestion of nutrients in CM differed. Three rumen-cannulated steers were used in three separate trials wherein canola meal (CM) was incubated in situ for 0.1, 4, 8, 12, 16 and 36 h in Dacron bags. Disappearance parameters were estimated for dry matter (DM), nitrogen (N) and individual amino acids (AAs). Effective degradabilities (EDs) were estimated at rumen particulate outflow rates of 0.08, 0.05 and 0.02 h−1. An ED for DM that was lower than that of most AAs indicated there was a limit to its degradability. In contrast, the ED of N was higher than that of 11 of 17 AAs; the disappearance parameters for N were a poor indicator of those estimated for individual AAs. The concentrations of AAs in escape CM estimated at the specified rumen outflow times, compared with non-incubated CM, were higher for all AAs except cystine, proline and glutamate, whose concentrations were lower, and histidine and arginine, whose concentrations did not differ. Compared with non-incubated CM, in escape CM there was a 33% reduction in concentration of glutamate, a 28% enrichment of tyrosine, a lower enrichment, ranging from + 14 to + 23%, for valine, isoleucine, threonine, phenylalanine, serine, aspartate and alanine, and little change in concentration for other AAs. Although the concentrations of AAs in rumen escape CM differed from that in non-incubated CM, those concentrations in escape CM were relatively uniform among outflow rates of 0.08, 0.05 and 0.02 h−1. A simple and efficient method for the synthesis of N,N’-orthogonally protected imide tethered peptidomimetics is presented. The imide peptidomimetics were synthesized by coupling the in situ generated selenocarboxylate of Nα-protected amino acids with Nα-protected amino acid azides in good yields.

Dog livers were perfused in situ for periods up to 6 hr with dog blood recycled through a pump-oxygenator. An amino acid mixture was administered for 90 min. Concentrations of amino acids were determined at intervals of 30 min or more. Rates of uptake and metabolism were calculated. After the start of perfusion, there is a fall in most plasma amino acid concentrations and a reciprocal rise in liver amino acids. Addition of amino acids causes a sharp rise in plasma amino acids. There is a rapid uptake of most of the amino acids by liver, although the concentrations of amino acids in liver fail to rise appreciably. Notable exceptions are valine, leucine, and isoleucine. Uptake of amino acids stimulates: a) an increase in the rate of synthesis of urea which ultimately accounts for 90% of the metabolized amino acids; b) a net synthesis of ornithine; and c) net noncatabolic metabolism of amino acids which may in part be protein synthesis. The results support the view that the liver temporarily stores a part of ingested amino acids as proteins, and subsequently makes them available to other organs.

Amino acid transport by juxtamedullary (JM) nephrons and its relationship to transport by superficial cortical (SC) nephrons and to function of vasa recta and collecting ducts were examined in vivo and in situ by free-flow micropuncture of Henle's loops, collecting ducts, and vasa recta and by continuous microinfusion of Henle's loops in exposed rat papillae. Fractional deliveries (FDs) of six neutral amino acids, two acidic amino acids, and taurine to tips of Henle's loops of JM nephrons could be substantially below those to early distal loops of SC nephrons, indicating that reabsorption before loop tips could be greater in JM than in SC nephrons. FDs to collecting ducts lower than to JM loop tips suggested reabsorption distal to loop tips. This was confirmed by continuous microinfusion of ascending limbs of Henle's loops. Distal site of reabsorption is unknown, but amino acids may move passively out of the thin ascending limb and be recycled into vasa recta and descending limb. Recycling of amino acids was supported by high FDs to tips of Henle's loops (sometimes greater than 1.0), higher concentrations in ascending than in descending vasa recta at same papilla level, and high mean concentrations in vasa recta.

SUMMARYFour Holstein steers (297 ± 56 kg of body weight (BW)) fitted with duodenal cannula and rumen catheter, were housed in metabolism cages and used in a 4 × 4 Latin Square trial to evaluate the effect of both protein source andAcacia mearnsiitannin extract on digestibility, ruminal fermentation, rumen microbial protein synthesis, N utilization and on duodenal flow of individual amino acids. The diet was offered at restricted amount of 25 g of dry matter (DM)/kg BW and consisted of maize silage plus concentrate, in a proportion of 0·7:0·3 (DM basis) respectively. Concentrate was formulated with either soybean meal or canola meal as protein source, with or without 50 g/kg ofA. mearnsiitannin extract (i.e. 15 g/kg of total dietary DM). There was no effect of protein source on most variables. The apparent and true organic matter (OM) digestibilities, as well as neutral detergent fibre (NDF) digestibility were negatively affected by tannin extract inclusion without, however, affecting digestible OM intake. The amount of nitrogen (N) excreted in faeces increased whereas the urinary N excretion decreased in tannin extract treatments. No interaction time × treatment was detected for any rumen variable and no treatment effect was observed for rumen fluid pH and reducing sugars concentration. Rumen fluid concentration of ammonia N was lower for the canola meal plus tannin extract treatment. Rumen concentration ofα-amino compounds was not affected by tannin extract but was higher when canola meal was the protein source. The duodenal flow of OM, total N,α-amino N and non-ammonia non-microbial N increased with tannin extract inclusion, whereas the duodenal flow of microbial N was not affected by treatment. For both protein sources, the amount of most individual amino acids flowing to the duodenum increased due to tannin extract addition. In conclusion, the dietary inclusion of 15 g/kg DM of tannin extract fromA. mearnsiiimproved the amino acid supply independently of whether the protein source was canola meal or soybean meal, without affecting the amino acid profile, to steers fed maize silage plus concentrate, with a minor but significant impact on OM digestibility.

Extensive incorporation of amino acid carbon into tissue protein was accompanied by high protein turnover and recycling rates of amino acid carbon.
