Molecular evidence for sympatric taxa within Pemphigus betae (Hemiptera: Aphididae: Eriosomatinae)

AbstractMitochondrial DNA cytochrome c oxidase subunit 1 (DNA barcode) and nuclear microsatellite flanking region sequences were used to analyse populations of putative “sugarbeet root aphid”, Pemphigus betae Doane, from sites in Alberta, Canada. Three sympatric genotypes were revealed, identified as P. betae, P. populivenae Fitch, and an undetermined third species. All three genotypes formed morphologically indistinguishable galls on the same set of cottonwood (Populus L., Salicaceae) host species, often on the same tree. Gall morphology is frequently used to identify Pemphigus species. Our results indicate that this practice may be unreliable for these three taxa at least.

This paper reports four new species of the primitively segmented spider genus Songthela from Chongqing Municipality, China, based on morphological characters of both males and females: S. jinyun sp. nov., S. longbao sp. nov., S. serriformis sp. nov. and S. wangerbao sp. nov. We also provide the GenBank accession codes of mitochondrial DNA barcode gene, cytochrome c oxidase subunit I (COI), for the holotype of four new species for future identification.

Restriction fragment length polymorphism (RFLP) markers were used to study genetic variation in the basidiomycete fungus Phellinus weirii (Murr.) Gilbertson, the cause of laminated root rot of conifers. In an initial study, three isolates each from the Douglas-fir type and the cedar-type biological species were surveyed with 12 restriction enzymes and 20 random, mitochondrial, and nuclear-ribosomal gene probes. The two biological species were distinct with most probe–enzyme combinations (91%). Variation within biological species was detected for the random and ribosomal DNA probes but not for the mitochondrial DNA probes. In a subsequent study 65 probe–enzyme combinations (13 × 5) that had detected variation within the Douglas-fir type biological species were used to analyze 27 isolates derived from six infection centers, two host species, and two geographic areas in western Oregon. Infection centers differed from one another in numerous probe–enzyme combinations but were nearly genetically uniform within. Isolates from the two host species, Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) and mountain hemlock (Tsuga mertensiana (Bong.) Carr.), showed few RFLP differences. Initiation of infection centers, and subsequent vegetative or basdiospore initiated immigration, appear to be rare events. Key words: ribosomal DNA, mitochondrial DNA, RFLP, root rot, conifer.

Samia ricini (Wm. Jones) and Samia cynthia (Drury) (Lepidoptera: Saturniidae) have been used as traditional sources of food as well as silk-producing insects. However, the phylogenetic relationship between the two silkworms remains to be addressed. In this study, the mitochondrial cytochrome c oxidase subunit 1 (COI) gene sequences corresponding to DNA barcodes from 13 Samia species were analysed, and a DNA barcode-based phylogenetic framework for these Samia species was provided. Phylogenetic analysis showed that multiple individuals of a species could be clustered together. Our analysis revealed a close relationship among Samia yayukae Paukstadt, Peigler and Paukstadt, Samia abrerai Naumann and Peigler, Samia kohlli Naumann and Peigler, Samia naessigi Naumann and Peigler, Samia naumanni Paukstadt, Peigler and Paukstadt, and Samia kalimantanensis Paukstadt and Paukstadt. The mixed clustering relationship and low Kimura-2-parameter (K2P) genetic distance (0.006) between individuals of S. ricini and Samia canningi (Hutton) indicated that the cultivated silkworm S. ricini was derived from the non-cultivated silkworm S. canningi. The remote phylogenetic relationship and high K2P genetic distance (0.039) indicated that S. ricini and S. cynthia are distinct species, thus providing solid molecular evidence that they had entirely independent origins. The relationships between S. kalimantanensis and S. naumanni and between S. cynthia and Samia wangi Naumann and Peigler, as well as the potential cryptic species within S. abrerai were also discussed. This is the first study to assess the DNA barcodes of the genus Samia, which supplements the knowledge of species identification and provides the first molecular phylogenetic framework for Samia species.

Isostenosmylus Krüger, 1913 is the richest genus of Osmylidae of the Neotropical region, with 17 described species so far, which are distributed mainly in the Andean region and in the South of Brazil and Paraguay. A new remarkable Colombian species of Isostenosmylus—I. ammirabilis sp. nov.—is herein described and illustrated. DNA barcode of mitochondrial gene cytochrome c oxidase subunit I (COI) for this species is also provided. Taxonomic keys for the genus are updated.

En este estudio se analizan dos fragmentos del gen de la citocromo c oxidasa subunidad I (COX1) del ADN mitocondrial de 61 individuos del género Rhynchophorus colectados en la Región de Murcia a fin de determinar su procedencia. El análisis filogenético del fragmento 1 de las muestras de la Región de Murcia conjuntamente con las secuencias disponibles en GenBank indica que los individuos corresponden a la especie Rhynchophorus ferrugineus.Las secuencias de Murcia se colapsan en un único haplotipo (H8 mediterráneo) que aparece dentro del clado de R. ferrugineus. De los análisis filogeográficos se infiere que el origen de los individuos de Murcia es Egipto. Adicionalmente, se examinó una región contigua del COX1 (fragmento 2) en la que las secuencias se colapsaron en dos haplotipos. In this research two fragments of the cytochrome c oxidase subunit I (COX1) gene of the mitochondrial DNA were analyzed in 61 individuals of the genus Rhynchophorus collected in the Region of Murcia with the aim of determining their origin. Phylogenetic analysis of fragment 1 of the samples collected in the Region of Murcia together with the available sequences in GenBank, indicated that these individuals correspond to the species R. ferrugineus. Sequences from Murcia collapsed into the H8 Mediterranean haplotype, which cluster into the R. ferrugineus clade. Phylogeographic analysis shows that the origin of the individuals collected in the Region of Murcia is Egypt. Additionally, a contiguous fragment of COX1 (fragment 2) was analyzed and the sequences collapsed into two haplotypes.

SUMMARYGenetic variation was examined in the anoplocephalid cestode Progamotaenia festiva, from Australian marsupials, in order to test the hypothesis that P. festiva, is a complex of sibling species and to assess the extent of host switching reported previously based on multilocus enzyme electrophoresis (MEE). Polymerase chain reaction (PCR)-based single-strand conformational polymorphism (SSCP) was used for the analysis of sequence variation in the cytochrome c oxidase subunit 1 (cox1) gene among 179 specimens of P. festiva (identified based on morphology and predilection site in the host) from 13 different host species, followed by selective DNA sequencing. Fifty-three distinct sequence types (haplotypes) representing all specimens were defined. Phylogenetic analyses of these sequence data (utilizing maximum parsimony and neighbour-joining methods) revealed 12 distinct clades. Other heterologous species, P. ewersi and P. macropodis, were used as outgroups and the remaining bile-duct inhabiting species, P. diaphana and P. effigia, were included in the analysis for comparative purposes. The latter 2 species were nested within the clades representing P. festiva. Most clades of P. festiva identified were restricted to a single host species; one clade primarily in Macropus robustus was also found in the related host species M. antilopinus in an area of host sympatry; another clade occurring primarily in M. robustus occurred also in additional kangaroo species, M. rufus and M. dorsalis. High levels of genetic divergence, the existence of distinct clades and their occurrence in sympatry provide support for the hypothesis that P. festiva represents a complex of numerous species, most of which, but not all, are host specific. Three distinct clades of cestodes were found within a single host, M. robustus, but there was no evidence of within-host speciation.

Abstract A recent phylogenetic survey of the New World orioles (genus Icterus; Omland et al. 1999) suggested that the Baltimore Oriole (I. galbula) and the Black-backed Oriole (I. abeillei) are sister taxa. That survey examined mitochondrial DNA (mtDNA) from a single representative of each species in the genus. Here, we examine mtDNA sequences from 15 Black-backed and 20 Baltimore Orioles. The two species appear to be very recently diverged, with average sequence divergences for both cytochrome b (cyt b) and the control region indicating a probable late Pleistocene split. Despite this very recent divergence, there is one fixed base-pair difference between the species in cyt b and another in the control region, suggesting that one or both species have undergone a bottleneck during or since speciation. This molecular evidence of recent divergence suggests that male plumage differences between Black-backed and Baltimore Orioles evolved very rapidly. Especiación Reciente entre Icterus galbula y Icterus abeillei Resumen. Un estudio filogenético reciente de los orioles del Nuevo Mundo (género Icterus; Omland, et al. 1999) sugiere que Icterus galbula e I. abeillei son taxa hermanos. En aquel estudio se examinó el ADN mitocondrial de un sólo representante de cada especie del género. En este estudio examinamos secuencias de ADN mitocondrial de 15 individuos de I. abeillei y 20 de I. galbula. Las dos especies parecen haber divergido recientemente con una separación promedio de las secuencias nucleotídicas en citocromo b y la región de control que indica que la divergencia ocurrió probablemente a fines del Pleistoceno. A pesar de haber divergido tan recientemente, existe una diferencia fija de un par de bases en la secuencia nucleotídica entre las dos especies en citocromo b y otra diferencia fija en la secuencia nucleotídica de la región de control, lo cual sugiere que una o ambas especies han sufrido un efecto de cuello de botella desde o durante el proceso de especiación. Esta evidencia molecular de divergencia reciente sugiere que los elementos del plumaje en los machos evolucionaron muy rápidamente entre I. abeillei e I. galbula.

Undescribed females representing four morphological types were found in a collection of adult Diamesa from about 5000 m altitude in Rongbuk, Tibet. Short DNA sequences of cytochrome oxidase subunit 2 were used to associate two single males in the material with conspecific females. Diamesa solhoyi n.sp. and Diamesa aculeata n.sp. are described. The complete type material and additional specimens have been deposited in the Insect Collection at the Institute of Zoology, Academia Sinica, Beijing (IZAS). The sequences are deposited in Genbank with accession numbers AM051227–AM051233.
