AbstractThe pink stem borer (PSB) (Sesamia nonagrioides Lefèbvre, 1827; Lepidoptera: Noctuidae) is the main insect pest of maize (Zea mays L., Poaceae) in the Mediterranean area. Maize varieties partially resistant to PSB have been identified, but few studies have documented the mechanisms of resistance involved. The objectives of this research were to study the levels of leaf and sheath antibiosis of a maize population, EPS12, and determine whether antibiosis improved over the selection process for less stalk tunneling. A hybrid check and two Bacillus thuringiensis (Bt) hybrids were also examined. Several bioassays were carried out in a control chamber to evaluate antibiosis of maize leaves and sheaths against PSB larvae. Leaf antibiosis was significantly higher for EPS12 than for the hybrid check and sheath antibiosis was low for all non-Bt hybrids studied. We conclude that leaf and sheath antibiosis was not significantly improved during the selection process. Future efforts should look for other possible mechanisms of resistance, such as stem rind antibiosis.

RNA interference (RNAi) is a transforming technology with high potential for practical applications in biology, including specific and safe insect pest control. For developing RNAi-based pest-control products no general recommendations exist and the best strategy needs to be determined for each insect pest separately on a case-by-case basis. In this research, the potential of silencing the genes encoding the subunits of the ecdysone receptor complex, EcR and Ultraspiracle (USP) by RNAi was evaluated in the corn borer, Sesamia nonagrioides, using different delivery approaches and targeting different developmental stages. In conjunction with our previous research it is demonstrated that prepupae are sensitive to RNAi triggered by dsRNA injection and that feeding of dsRNA-expressing bacteria throughout S. nonagrioides’ larval life can lead to limited developmental malformations with no potent insecticidal results. Our results, consistent with previous studies, indicated a great fluctuation of exogenous RNAi effectiveness in the Lepidopteran species, suggesting that further factors should be taken into consideration in order to expand this very promising field into the ‘’RNAi-resistant’’ insect species.

Spodopteraexigua, a multifeeding insect pest, has developed a high level of resistance to chlorantraniliprole, which is a benzoylurea insecticide that targets the ryanodine receptors (RyRs). Herein, the resistant strain (SE-Sel) and sensitive strain (SE-Sus) were obtained by bidirectional screening for six generations. The potential oviposited eggs and oviposition rate of the SE-Sel strain were dramatically lower than those of the SE-Sus strain; on the contrary, the weights of prepupae and preadult were significantly increased. As a post-mating response, the higher number of non-oviposited eggs in the SE-Sel strain was caused by a lower mating rate. In addition, the expression levels of vitellogenin (SeVg) and its receptor (SeVgR) in the SE-Sel strain were consistently lower than those in the SE-Sus strain. An RyRI4743M mutation, contributing to the resistance to chlorantraniliprole, was located in the S3 transmembrane segments and might have affected the release of calcium ions; it led to the upregulated expression of the neuropeptide SeNPF and its receptor SeNPFR, and the mating and oviposition rate were significantly recovered when the SeNPF was knocked down though RNA interference (RNAi) in the male adult of the SE-Sel strain. Moreover, the expression of the juvenile hormone-binding proteins SeJHBWDS3 and SeJHBAN in the male adult of the SE-Sel strain was significantly decreased, which proved the existence of a fitness cost from another angle. Therefore, these results indicate that the fitness cost accompanied by chlorantraniliprole resistance in S. exigua may be related to the decrease in mating desire due to SeNPF overexpression.

Shorea robusta Gaertn. f. (Dipterocarpaceae), commonly known as sal, is one of the most important timbers of India both ecologically and economically. It is of Indian origin, widely distributed in central and north India and constitutes an important ecosystem, which provides cool and calm environment rich in biodiversity. This tree species has a highest number of insect fauna among the forest trees. Of about 346 insects recorded on sal, about 155 species of insects belonging to the order Lepidoptera (105), Coleoptera (31), Thysanoptera (9), Hemiptera (4), Orthoptera (4), Ephemeroptera (1) and Isoptera (1) are associated with living tree. The major insect pests of sal are, ‘seed and seedling borer’ Pammene theristis Meyrick (Lepidoptera: Eucosmidae), feeding on seed while they are on the tree; Sitophilus rugicollis Casey (Coleoptera : Curculionidae), damage seed either on the forest floor or during storage; white grubs such as Holotrichia spp. (Coleoptera : Scarabaeidae), which feed on the root system of the plants in nurseries and beetles feed on the foliage; defoliators such as Caviria sericea Moore (Lepidoptera : Lymantriidae), Lymantria mathura Moore (Lepidoptera : Lymantriidae), Paectes subapicalis Walker (Lepidoptera : Noctuidae), Trabala vishnou Lefebvre (Lepidoptera : Lasiocampidae), which feed on the leaves; stem borer such as Hoplocerambyx spinicornis Newman (Coleoptera : Cerambycidae) and sap sucker, such as Drosicha stebbingi Green (Hemiptera : Coccidae). Termites, such as Coptotermes curvignathus Holmgren (Isoptera : Rhinotermitidae) and timber borers, such as Sinoxylon anale and S. crassum (Coleoptera : Bostrychidae), damage sal timber in dying, felled or stored logs in depots.
