The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a major worldwide pest of cruciferous vegetables (Talekar and Shelton 1993). Male and female adults of this species can be accurately identified by visible differences in their external genitalia. According to Robertson (1939), the sex of pupae can also be identified, but, in our experience, this approach is more difficult. For some types of research, determination of larval sex would be useful. For example, one could determine sex-specific responses of larvae to a treatment by observing mortality, development rate, or behavior of male and female larvae separately. In studies requiring hybrid mating between different strains of diamondback moth (Tabashnik et al. 1992), one could isolate groups of male larvae separately from female larvae to avoid mating within a strain. Some lepidopteran larvae can be sexed by the gonads being visible through the cuticle of the fifth abdominal segment or by the position of external pits marking the location of the genital discs in the eighth and ninth abdominal segments (Stehr 1987). But, because of their small size, it is not practical to sex diamondback moth larvae by external pits and we have found no studies in which male and female larvae were identified by gonadal form. Herein we report that third- and fourth-instar larvae of diamondback moth can be sexed efficiently and reliably.

Abstract Using a sensitive RT-QPCR assay, we analyzed the regulatory effects of sex and different dosage compensation mutations in Drosophila. To validate the assay, we showed that regulation for several genes indeed varied with the number of functional copies of that gene. We then confirmed that dosage compensation occurred for most genes we examined in male and female flies. Finally, we examined the effects on regulation of several genes in the MSL pathway, presumed to be involved in sex-dependent determination of regulation. Rather than seeing global alterations of either X chromosomal or autosomal genes, regulation of genes on either the X chromosome or the autosomes could be elevated, depressed, or unaltered between sexes in unpredictable ways for the various MSL mutations. Relative dosage for a given gene between the sexes could vary at different developmental times. Autosomal genes often showed deranged regulatory levels, indicating they were in pathways perturbed by X chromosomal changes. As exemplified by the BR-C locus and its dependent Sgs genes, multiple genes in a given pathway could exhibit coordinate regulatory modulation. The variegated pattern shown for expression of both X chromosomal and autosomal loci underscores the complexity of gene expression so that the phenotype of MSL mutations does not reflect only simple perturbations of genes on the X chromosome.

Gender determination of the human remains can be very challenging, especially in the case of incomplete ones. Herein, we report a proof-of-concept experiment where the possibility of gender recognition using Raman spectroscopy of teeth is investigated. Raman spectra were recorded from male and female molars and premolars on two distinct sites, tooth apex and anatomical neck. Recorded spectra were sorted into suitable datasets and initially analyzed with principal component analysis, which showed a distinction between spectra of male and female teeth. Then, reduced datasets with scores of the first 20 principal components were formed and two classification algorithms, support vector machine and artificial neural networks, were applied to form classification models for gender recognition. The obtained results showed that gender recognition with Raman spectra of teeth is possible but strongly depends both on the tooth type and spectrum recording site. The difference in classification accuracy between different tooth types and recording sites are discussed in terms of the molecular structure difference caused by the influence of masticatory loading or gender-dependent life events.

In the eyes of male and female flies a quantitative determination of the tetrahydropterin, the corresponding dihydro-compound (Sepiapterin) and of xanthommatin from 0—5 days after hatching was made. The increase of xanthommatin almost stops 24 hours after hatching; about 13 μg are present after that time. The tetrahydropterin, after a drastic increase during the first 24 hours, remains at a level of about 1,1 — 1,2 µg. In contrast the dihydrocompound which is nearly absent at the time of hatching is deposited on the eye-granules between the third and fifth day. At that time, especially in the male the dihydropterin/tetrahydropterin ratio is increased. Starvation during the first 24 hours has essentially no effect on the amount of xanthommation whereas the amount of pterins is markedly reduced.

AbstractThe determination of phenolic acids’ and flavonoids’ content in daily food rations (DFRs) of a selected group of male and female students and the development of chromatographic conditions is the primary goal of the study. The presence of 7 phenolic components were confirmed in the prepared extracts from all diets reconstructed within a period of 3 years. The highest concentrations were determined for hesperidin (124 and 55.6 mg for women and men, respectively), naringin (47.6 mg in female and 37 mg in male diets) and chlorogenic acid (19.7 and 19.8 mg for women and men). The antioxidant potential of the daily food rations, measured with a DPPH test, was higher for women (range 47.1‒78.8%) than for men (range 34.5‒78.0%) and was found to strongly correlate with the total phenolics content of the samples (Folin-Ciocalteu test) (correlation coefficient 0.90).

(1) Background: Antimicrobial resistance represents an urgent health dilemma facing the global human population. The development of novel antimicrobial agents is needed to face the rising number of resistant bacteria. Ultrashort antimicrobial peptides (USAMPs) are considered promising antimicrobial agents that meet the required criteria of novel antimicrobial drug development. (2) Methods: Alapropoginine was rationally designed by incorporating arginine (R), biphenylalanine (B), and naproxen to create an ultrashort hexapeptide. The antimicrobial activity of alapropoginine was evaluated against different strains of bacteria. The hemolytic activity of alapropoginine was also investigated against human erythrocytes. Finally, synergistic studies with antibiotics were performed using the checkerboard technique and the determination of the fractional inhibitory index. (3) Results: Alapropoginine displayed potent antimicrobial activities against reference and multi-drug-resistant bacteria with MIC values of as low as 28.6 µg/mL against methicillin-resistant S. aureus. Alapropoginine caused negligible toxicity toward human red blood cells. Moreover, the synergistic studies showed improved activities for the combined conventional antibiotics with a huge reduction in their antimicrobial concentrations. (4) Conclusions: The present study indicates that alapropoginine exhibits promising antimicrobial activity against reference and resistant strains of bacteria with negligible hemolytic activity. Additionally, the peptide displays synergistic or additive effects when combined with several antibiotics.

A general account of the internal reproductive organs and the external genitalia and their development is given. The ‘penis lobes’ develop earlier than the ‘valvae lobes’, and independently of them. The tegumen is the modified ninth tergite. The tenth segment is visibly distinguished into a tergal and sternal part in the pupal stages, and the anal tube passes between the two sclerites. The uncus and the gnathos are dorsal and ventral processes respectively of the tenth segment. The anellus lobes develop as lateral processes of the ninth sternite on either side of the penis. The vasa deferentia during larval life do not extend beyond the eighth abdominal segment and lie in a latero-ventral position. They meet the extensions from the ectodermal ‘ductus ejaculatorius duplex’ during the last larval stadium. From the earliest caterpillar stage there exists a pair of ectodermal ducts formed by the differentiation of the epidermis on the ninth sternite. Towards the end of larval life they divide by constriction to form the accessory glands and the ‘ductus ejaculatory duplex’. At this stage they extend on either side to meet the vasa deferentia. The vesiculae seminales develop by distension from the region of junction between the vasa deferentia and the proximal portion of the ejaculatory duct. The ‘ductus ejaculatorius simplex’ arises as an ectodermal invagination between the pair of ‘penis lobes’ during the final larval instar. It is established that, with the exception of the vasa deferentia, all the remaining elements in the efferent genital system are derived from the ectoderm.

The present study was conducted to determine the early sex in turkeys by observation of the differences in body weight between male and female birds. A total of 30-day old black color unsexed poults having almost similar body weight at hatching were considered for the experimentation and housed at the Poultry Farm of Bangladesh Agricultural University, Mymensingh-2202, Bangladesh. All birds were reared up to 12 weeks of age under intensive management with supplementation of commercial broiler starter and grower feeds. Birds were reared under similar management conditions. Significantly higher (p<0.01) body weight was attained in male poults (104g/bird) than the female (90g/bird) at the end of 1st week of age. Similarly, at the end of 2nd week of age higher (p<0.01) body weight attained by male poults (198.31g/bird) than the female (162.13g/bird). At the end of 3rd weeks of age male poults attained higher (p<0.01) body weight (307.23g/bird) than the female (251.33g/bird). After 4 weeks of rearing, male turkeys attained significantly higher (p<0.01) live body weight (424.46g/bird) than the female turkeys (347.87g/bird). The weekly average body weight gains of male and female birds were 94.18g/bird and 76.5g/bird, respectively. Thus, the male and female birds were successfully identified on the basis of differences in their body weight. Weekly feed intake for both the male and female birds was also increased with their age. Up to 4 weeks of age, both the male and female poults consumed same amount of feed (753.46g/bird). The FCR of male and female poults differed non-significantly in 1st, 3rd and 4th week. On the contrary, in 2nd week of age the FCR of male poults (1.60) was significantly lower (p<0.01) than female (2.11). Survivability was 100% up to 4th week of age irrespective of sex of the poults. The birds were reared up to 12 weeks of age until to confirm their sex by observation of the phenotypic appearance. Results of the phenotypic observation of male and female birds correspondence hundred percent accuracy with the results obtained in body weight based differences between male and female birds. It is therefore concluded that farmers can identify male or female poults as early as first week of age on the basis of body weight differences. Progressive Agriculture 31 (3): 218-226, 2020

Abstract With the upcoming medical Cannabis regulation, quality control methods on raw material will be required. Besides testing for contaminants and potency, there are also pharmaceutical and forensic interests in the determination of the terpene profile in different strains of Cannabis as complementary identification methods. A simple non-destructive HS-SPME GC-MS method was used to identify the terpene content in twelve Cannabis samples, four of them were of the hemp type (Harle-tsu), seven from various marihuana types and one of the intermediate type. They all were previously analyzed by HPLC to determine the potency (THC and CBD content). Spectral library matching was used to identify the terpenes compounds. Thirty terpenes compounds were detected, nine of them were present in all Cannabis samples and used to find their terpene profile: α-pinene, β-pinene, β-myrcene, D-limonene, terpinolene, linalool, caryophyllene, α-bergamotene and humulene. Three of them, caryophyllene, α-pinene and β-myrcene were found as larger components in most of samples. A principal components analyses (PCA) was performed. The four hemp type samples showed two different profiles, two samples showed caryophyllene as main component and the others two with β-myrcene as such. The marihuana type samples showed wider profiles with no clear patterns at all, which is not surprising because of the low number of samples. The simple methodology shows viable to set the terpenes profile for analyses of raw Cannabis material. Suitability for differentiation between different sorts of types needs more studies, with increasing numbers of samples.

Abstract Objective: This study investigated the antibacterial activity of Ilex paraguariensis extracts against 32 different strains of non-typhoidal Salmonella (NTS) through the determination of the Minimum Inhibitory Concentration (MIC), Mutant Prevention Concentration (MPC), Mutant Selection Window (MSW), and the detection of virulence genes by multiplex PCR assays. Results: The MIC values of Ilex paraguariensis against Salmonella spp. strains varied between 0.78 mg/ml and 6.25 mg/ml with a MIC 90 of 3.12 mg/ml. The highest MPC in this study was 48 mg/ml yielding a Mutant Selection Window of 41.75 mg/ml. The MSW values of the remaining strains varied between 1.56 and 8.87 mg/ml. Genes of pathogenicity detected in Salmonella spp. isolates were most commonly the stn, sdiA, invA, sopB, invH, and sopE genes. The antibacterial activity of Yerba Mate extracts was not affected by the antimicrobial resistance patterns or pathogenicity genes expressed. More work is needed to identify the active antibacterial compound(s) responsible for the antibacterial activity.

The assassin bug tribe Metapterini belongs to the subfamily Emesinae (Hemiptera: Heteroptera: Reduviidae). Morphologically, it is characterized by the conspicuous basal process of the posteroventral series in the foreleg and the presence of wing polymorphism, with a high proportion of the genera with micropterous or apterous species. Here, the male and female ectodermal genitalic structures are documented for ten genera and twenty-three species of Metapterini, including eight species of the speciose genus Ghilianella Spinola, 1850. Descriptions and digital macrophotographs are provided for abdominal segment 8, pygophore, parameres, and phallus of the male, and for tergite 8, tergite 9, gonocoxae, gonapophyses, gonoplac, and bursa copulatrix of the female. The asymmetric male genitalia within Emesinae are discussed. From this morphological documentation sixty six phylogenetic characters are coded, presented as a data matrix and analyzed cladistically, and their potential usefulness for resolving relationships among Metapterini is discussed.
