Abstract Background To investigate the antinociceptive and anti-inflammatory activities of aqueous extract of a blended mixture of dried leaves of Ocimum gratissimum and Psidium guajava, a traditional analgesic drug polyherbal (TADP) used as a remedy for pain-related conditions. Methods Antinociceptive activity of TADP (100, 200 and 400 mg/kg) was evaluated in the hot plate test and acetic acid-induced nociception in mice while the anti-inflammatory was evaluated in carrageenan-induced paw oedema in rats. Levels of nitrite, myeloperoxidase, glutathione and malondialdehyde were assayed in carrageenan-induced paw tissue. Results TADP (200 and 400 mg/kg) significantly prolong the latency time in the hot-plate test. TADP (100–400 mg/kg) produced a dose-dependent significant inhibition of the acetic-acid induced abdominal constriction. The antinociceptive activity of TADP in the presence of naloxone and atropine was not reversed whereas yohimbine and glibenclamide significantly reversed it. TADP (100, 200 and 400 mg/kg) significantly reduced the swelling in the carrageenan-induced oedema model and also produced a reduction in the nitrite and myeloperoxidase level. TADP (400 mg/kg) significantly reduced malondialdehyde concentration and increase glutathione level in the carrageenan-induced rat paw. TADP significantly decrease the number of cellular infiltrates in the histopathological assessment. Conclusion These results indicate that polyherbal product containing blended leaves of Ocimum gratissimum and Psidium guajava possess antinociceptive and anti-inflammatory properties, hence represents a promising alternative remedy in inflammation-induced pain.

Various parts of psidium guajava linn are used in ethnomedicine in controls and treatments of different disorders such as liver, heart, and blood related disorders [11]. This research work investigate qualitative phytochemical composition of ethanol leaf extract of psidium guajava linn and its effect on some hematological parameters in albino rats. Qualitative phytochemical analysis of the extract was performed by standard procedures. A total of 20 adult male albino rats were used, they were randomly placed into 5 groups (A, B, C, D and E) each group containing 4 rats groups A, B, C, and D were administered, via oral intubation, the extract at doses of 200, 400, 600, and 800 mg/kg body weight respectively of the extract for 7 consecutive days. Phytochemical screening shows that the extract was rich in tannins anthraquinones, and flavonoid while alkaloids, saponins, terpenes and glycoside were slightly present. There was a decrease in the average body weight, physical activities, and feed and water intake of the rats in the test groups relative to the control hemoglobin concentrate, total white blood cell and packed cell volume recorded in the groups  administered the extract were significantly (p<0.05) higher than in the control this effect on  the haemotological parameters was found to be dose dependent. The findings of this study suggest that the aqueous extract of the fruit plants may be useful in control and treatment of blood related disorders.  These useful applications may be as a result of the phytochemical present in the extract.

There is a trend to use medicinal plants for primary medical care or as dietary supplements; however, the safety of many of these plants has not been studied. The objective of this work was to determine the toxic effect of the aqueous extract of Calea ternifolia (C. zacatechichi), known popularly as “dream herb” in vivo and in vitro in order to validate its safety. In vivo, the extract had moderate toxicity on A. salina. In vitro, the extract induced eryptosis of 73% at a concentration of 100 μg·mL−1 and it inhibited CYP3A by 99% at a concentration of 375 μg/mL. After administering 8.5 mg/kg of C. ternifolia to rats, we found a reduction in platelets and leukocytes and an increase in urea and the liver enzymes alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP). Histological analysis showed spongiform changes in the proximal tubules of renal tissue and a lymphoid infiltrate in liver tissue. This plant is used in the treatment of diabetes, and it is commercialized as a dietary supplement in several countries. Our results show renal and hepatic toxicity; therefore, more profound research on the toxicity of this plant is needed.
