Application of salicylic acid induces systemic acquired resistance in tobacco. pchA and pchB, which encode for the biosynthesis of salicylic acid in Pseudomonas aeruginosa, were cloned into two expression vectors, and these constructs were introduced into two root-colonizing strains of P. fluorescens. Introduction of pchBA into strain P3, which does not produce salicylic acid, rendered this strain capable of salicylic acid production in vitro and significantly improved its ability to induce systemic resistance in tobacco against tobacco necrosis virus. Strain CHA0 is a well-described biocontrol agent that naturally produces salicylic acid under conditions of iron limitation. Introduction of pchBA into CHA0 increased the production of salicylic acid in vitro and in the rhizosphere of tobacco, but did not improve the ability of CHA0 to induce systemic resistance in tobacco. In addition, these genes did not improve significantly the capacity of strains P3 and CHA0 to suppress black root rot of tobacco in a gnotobiotic system.

The family of Mildew resistance Locus O (MLO) proteins is best known for its profound effect on the outcome of powdery mildew infections: when the appropriate MLO protein is absent, the plant is fully resistant to otherwise virulent powdery mildew fungi. However, most members of the MLO protein family remain functionally unexplored. Here, we investigateArabidopsis thaliana MLO3, the closest relative ofAtMLO2, AtMLO6andAtMLO12, which are the ArabidopsisMLOgenes implicated in the powdery mildew interaction. The co-expression network ofAtMLO3suggests association of the gene with plant defense-related processes such as salicylic acid homeostasis. Our extensive analysis shows thatmlo3mutants are unaffected regarding their infection phenotype upon challenge with the powdery mildew fungiGolovinomyces orontiiandErysiphe pisi, the oomyceteHyaloperonospora arabidopsidis, and the bacterial pathogenPseudomonas syringae(the latter both in terms of basal and systemic acquired resistance), indicating that the protein does not play a major role in the response to any of these pathogens. However,mlo3genotypes display spontaneous callose deposition as well as signs of early senescence in six-or seven-week-old rosette leaves in the absence of any pathogen challenge, a phenotype that is reminiscent ofmlo2mutant plants. We hypothesize that de-regulated callose deposition inmlo3genotypes is the result of a subtle transient aberration of salicylic acid-jasmonic acid homeostasis during development.

The presence study deals with powdery mildews in various cucurbits in Katsina city (Barhim Estate, Kofar Durbi, Kofar Sauri, Kofar Marusa and Low Cost), Nigeria. The finding shows that the areas infested with powdery mildew is one of the important disease of cucurbits. The Sphaerotheca fuliginea was identified to be the causal organism present on all observed cucurbits in the study. Highest frequency of disease was found in Kofar Sauri(79%) fallowed by Kofar Marusa (68%), Kofar Durbi (66%), Barhim Estate (65%) and the lowest frequency of occurrence of disease was found in Low Cost (55%).The intensity of the disease was moderate to severe in general but it was high in many fields, the area-wise variation was also noticed. On vegetables, the highest frequency of occurrence of powdery mildew disease was observed on L. cylindrica (76.4%) followed by C. moschata (60%), C. sativus (59.3%), C. vulgaris (53.9%) and lowest was found on C. melo (44.4%). The highest intensity of disease was found on C. moschata, followed by L. cylindrica, C. sativus, C. vulgaris and C. melo.
