Selection, characterisation and evaluation of potential probiotic Lactobacillus spp. isolated from poultry droppings

Aim of the present study was to characterise and evaluate probiotic potential of lactobacilli isolated from indigenous poultry. Lactobacilli were isolated from poultry droppings and identified by genus specific polymerase chain reaction and 16S rRNA gene sequencing. Isolates were characterised in vitro by their ability to tolerate low pH and bile salts, phytase activity, antimicrobial activity, antibiotic susceptibility profile, and autoaggregation and coaggregation with poultry gut pathogens. In vivo evaluation of selected isolates was done by their effect on the body weight gain and immune response of broiler chicks. Total of 90, one-day old chicks, were randomly divided in 9 groups and given selected lactobacilli alone and in combinations (108 cfu/bird, daily) from day 7 to day 35. Body weight gain and humoral immune response to New Castle Disease Virus (NDV) vaccine were determined weekly. Three lactobacilli isolates (SMP52, SMP64 and SMP70) were selected as potentially probiotic bacteria on the basis of in vitro characterisation and identified as Lactobacillus crispatus, Lactobacillus casei and L. crispatus, respectively. Chicks supplemented with ‘SMP52’, ‘SMP64’, ‘SMP70’ and ‘SMP64+SMP70’ and a commercial probiotic product (Protexin) showed significantly higher mean weight gain per bird (1,584±35.2, 1,629±30.6, 1,668±34.7, 1,619±29.5 and 1,576±31.7 g/bird, respectively) as compared to negative control group (1,394±26.7 g/bird), on day 35. SMP 70 also showed significantly higher geometric mean titre against NDV vaccine at day 21 as compared to negative control. It is concluded that L. crispatus SMP52, L. casei SMP64 and L. crispatus SMP70 are potential probiotic candidates which alone or in different combinations may increase body weight of broilers.

Background and Aim: Steviol glycosides extracted from the leaves of Stevia rebaudiana Bertoni have been of much consideration recently because of their beneficial effects on health, raising the possibilities for improving farm animals' health. Although some studies on stevia's dietary effect on body weight gain are available, few studies have been conducted to evaluate stevioside supplementation on immune response in broilers. This experiment aimed to analyze how a stevia-based sweetener can affect broiler chickens' growth performance and humoral response. Materials and Methods: In this experiment, one hundred and twenty 1-day-old Cobb-line broiler chicks fed with commercial starter/grower diets were included in three groups and supplemented with stevia-based sweetener at levels 0, 80, and 160 ppm, respectively. Chickens were weighed on day 0 and every 7 days for the next 6 weeks. Chicks were then immunized on days 10 and 24 with a Newcastle and infectious bronchitis vaccine and blood sampled on days 7, 24, and 35. Serologic assays were performed to detect specific antibody levels. Results: The body weight means and body weight gain on day 42 were found to be significantly higher in birds from the group fed with 80 ppm of stevia-based sweetener than those in the control group and slightly higher than those in the group supplemented with 160 ppm of stevia-based sweetener. Likewise, on day 35, antibodies against the Newcastle disease virus were higher in the treatment groups. Immune response to infectious bronchitis virus vaccination was not statistically different among the three groups through the experiment. Conclusion: Stevia-based sweetener at 80 ppm in commercial-based diets improved body weight gain and immune response in broiler chickens at the market age.

Poultry farming holds a special place in ensuring the products that the consumers demand, it provides the population with essential food products,such as eggs and meat,that contain vital micro and macronutrients, proteins, lipids, and vitamins. Therefore, the issues of rational, economically feasible feeding of meat poultry, namely broiler chickens, are an urgent task. It is also essential to find effective methods of their application in order to correct the natural resistance and immune and biological reactivity of birds. The purpose of this research is to study the effect of the biological stimulant-Nucleostim on the growth and development of chickens, hematological, and immunological parameters of the blood of birds.This Biostimulant is a purified bovine spleen extract containing at least 1 mg / ml of low molecular weight peptides (nucleotides and nucleosides) formed as a result of autolysis, using dry whey and diatomite as fillers. Onthe application ofNucleostim, the gain in live weight of chickens was increased by 9.7%. At the end of the experiment, the livability of the chicks of the experimental group treated with Nucleostimcame up to 88%, compared with the 72% of the control group. The use of biostimulant had a stimulating effect on the liver of chickens confirmed by the research results presented in the article, as well as contributed to the development of the thymus in the setting of general dystrophy. Thus, it improved chicklivability and increased body weight gain. The biological stimulant-Nucleostim as an adaptogenic, anabolic, and immunostimulatory agent is promising for finding new drugs that improve the health and productivity of poultry.

The objective of this study was designed to test whether supplementation of the diet with arginine (Arg) or glutamine (Gln) or their combination influences the production, organ weights and humoral immune response of broilers. A total of 432 one-day-old male Ross 308 broiler chickens were divided into 6 treatment groups: control, Arg-0.5%, Arg-1%, Gln-0.5%, Gln-1% and Arg-0.5%+Gln-0.5%. Drinking water and feed were provided ad libitum. On day 18 of the experiment 50% of chickens in each treatment group were immunised with bovine serum albumin. Ten and 21 days after immunisation blood samples were collected to determine the anti-albumin IgY titre, interleukin 6 (IL6) and interferon gamma (IFNG) and to measure the weight of the liver, spleen, bursa of Fabricius and thymus. Arg or Gln supplementation of the diets influenced neither the production nor the organ weights until 18 days of age. Between 18 and 39 days of age both Arg (0.5% and 1%) and Arg + Gln supplementation improved the feed conversion ratio (FCR) by 3.7%, 6.3% and 4.9%, respectively, while Gln-1% worsened it by 15%. Immunisation slightly (−0.79%) depressed the body weight gain of broilers fed the control diet, which was significantly improved by both Arg (0.5 or 1%) and Arg + Gln supplementation. Immunisation increased the weight of the spleen, bursa and thymus and decreased that of the liver. Supplementation with 1% Gln depressed (−5.13%) the body weight gain of the immunised chickens but strongly stimulated the immune response. Supplementations with Arg and Gln did not influence the IL6 and IFNG level of the blood; however, on day 10 after immunisation these two parameters showed a negative correlation with each other. Regarding production, organ weights and immunity, Arg supplementation should be recommended in the grower phase, while Gln supplementation can be useful in pullets raised for egg production, where a good immune response to vaccinations is an important factor.

The current study examined the attenuating influence of dietary carrot pomace powder (CaPP) on hypercholesterolemia and various oxidative stress-associated with biochemical parameters in hypercholesterolemic rats. Thirty two male albino rats weighing 110±10 g were divided into four groups, the first group received the basal diet only and served as (negative control), the second group received the hypercholesterolemic diet and served as positive control, the other groups received hypercholesterolemic diet supplemented with 10%, 20% CaPP for six weeks. The obtained results revealed that groups supplemented with 10% and 20% CaPP significantly decrease total lipid, total cholesterol, triglycerides, low density lipoprotein cholesterol, liver enzymes: alanine aminotransferase, aspartate aminotransferase compared to positive and negative groups. Organs weight, body weight gain significantly decreased compared with positive control. Moreover dietary carrot pomace powder can used to reduce the body weight and reducing hypercholesterolemic complications. In addition, dietary carrot pomace powder serves to improve the blood picture and to reduce the blood glucose level in hypercholesterolemic rats and could use in obese people for body loss. Data of kidney function (Urea) record an increase in CaPP 20% level (26.9±2.96) but this increase was non significant with the negative control group (26.6±3.1).

Background: Cryptosporidiosis caused by Cryptosporidium spp. is a zoonotic disease and is the most prevalent pathogens worldwide and leads to severe diarrhoeal diseases and affects the immunological status of the individual. Thus, the study was undertaken to examine the anti-cryptosporidial efficacy of curcumin in comparison with ethanolic extract of curcuma longa in immunocompromised mice infected with oocysts isolated from cattle calves of Jammu region and identified as Cryptosporidium parvum using nested PCR on small subunit ribosomal ribonucleic acid (SSU rRNA) gene. Methods: Two hundred female Swiss albino mice were equally divided into ten groups. Group I were kept as a healthy control, group II were immunocompromised, group III were immunocompromised and infected, group IV animals were immunocompromised, infected and treated orally with nitazoxanide. Animals in groups V to VII were immunocompromised, infected and treated with ethanolic extract of C. longa @ 4, 6 and 8 mg/kg/day/os respectively whereas groups VIII to X were immunocompromised, infected and treated with pure salt of curcumin @ 4, 6 and 8 mg/kg/day/os respectively for 5 successive days. Thus, mean oocysts per gram faeces, body weight gain and histopathological changes were measured in different groups. Result: Administration of curcumin as a therapeutic agent @ 8 mg/kg body weight for five days resulted in higher percent mean oocyst reduction of 74.03% and improved body weight gain in experimentally infected mice. Histopathological changes showed that treatment with oral curcumin (group X) in animals had minimal and improved intestinal lesions as compared to animals treated with C. longa (group VII). Altogether, curcumin showed promising anticryptosporidial effects under in vivo conditions and deserves further exploration.

The research aimed at investigating whether there was the massage effect on the growth and development of the infants of 3 – 4 months old in the working area of PHC Kanjilo, Barombong District, Gowa Regency.  This was a quasy experiment research with the pretest-posttest method on the treatment and control groups. Sample consisted of 38 subjects who were selected by the purposive sampling technique. The samples were divided into 2 group, namely the treatment group ( n=19) and control group (n=19) by matching the same age and sex in the two groups. The variables measured were the body weight gain, height gain, development indicator change. The research used the statistic test of Wilcoxon signed rank test, Mann Whitney U test, McNemar test with the  significance level of 0.05. The research result indicates that after the massage has been carried out for 30 days, there is the significant change particularly on the body weight gain variable with the  value of p = 0,008 (<0.05) and the development indicator change variable with the value of p = 0.05.  While for the height, the change is not significant between the infants who are given the massage and are not given the massage with the value of p = 0,091 (> 0.05), likewise, the development indicator change is not significant on the infants who are not massaged (p=0,317).

The aim was to investigate how blueberry husks and/or mixtures of probiotic strains (Lactobacillus crispatusDSM16743,L. gasseriDSM16737 andL. plantarumDSM15313 (LABmix), orBifidobacterium infantisDSM15159 and DSM15161 (BIFmix)) affect colonic fermentation, caecal counts of lactobacilli, bifidobacteria andEnterobacteriaceae, body weight gain, and blood concentrations of carboxylic acids (CA) and ammonia in rats. Dietary fibres in blueberry husks were fermented to 61 % in colon, and the elevated faecal excretion of fibre and protein contributed to the high faecal bulking capacity (1·3). The caecal pool of CA was higher in rats fed blueberry husks than the fibre-free control (P < 0·05), and the propionic acid proportion was higher in the distal colon than in the control group (P < 0·05). Probiotics lowered the caecal amount of CA when added to blueberry husks (P < 0·001), while the propionic acid proportion was higher with LABmix (P < 0·01) than blueberry husks only. The propionic acid and butyric acid concentrations in blood were higher in rats fed blueberry husks and probiotics than those fed blueberry husks only (P < 0·01), implying that the absorption of these acids was facilitated by the bacteria. The caecal counts of lactobacilli, bifidobacteria andEnterobacteriaceaewere lower in rats fed blueberry husks than the control diet (P < 0·05). The body weight gain was partly influenced by the caecal tissue and contents weights, and BIFmix decreased the ammonia concentration in blood (P < 0·05). We conclude that colonic fermentation is differentially affected by dietary fibre and probiotics, which may be of importance when developing foods with certain health effects.

Introduction: Nimesulide is a potent anti-inflammatorywith rapid and long-lasting effects, but also with a high riskof hepatotoxicity. Objective: This work aimed to preventnimesulide-induced hepatotoxicity through the associationof nimesulide with a hepatoprotective agent. Materials andMethods: First, we tested three hepatoprotective agents:N-acetylcysteine, L-carnitine, and Gingko biloba extract inan in vitro hepatic cell model. Both N-acetylcysteine and G.biloba showed promisor results. We selected N-acetylcysteineto continue the studies in an animal model. In vivo study wasperformed using male Wistar rats divided in 4 groups: control,nimesulide (100mg/kg/day), nimesulide (100mg/kg/day) +N-acetylcysteine (100mg/kg/day) and N-acetylcysteine alone(100mg/kg/day). Treatments were given by gavage, daily, for15 days. Results: Animals receiving nimesulide alone showedlower body weight gain compared to control. Body weightgain in the nimesulide + N-acetylcysteine group was higherthan nimesulide alone, evidencing lower toxicity. However,the body weight gain of the nimesulide + N-acetylcysteinegroup was still lower than the control animals. Animals treatedwith nimesulide alone presented an increased relative mass ofheart, liver, and spleen and significant hepatic damage seen inmicroscopy when compared to other groups. N-acetylcysteineco-administered with nimesulide prevented the increasedheart mass, but the same was not true with liver and spleen.Conclusions: This work evidence partial protection elicitedby the association of N-acetylcysteine and nimesulide againstnimesulide-induced hepatotoxicity.

The aim of this study was to analyse the effects of different daily intakes of rapeseed bee pollen on the growth and biochemical blood serum indicators in male and female rats. A total of 40 clinically healthy male and female Wistar rats were randomly divided into four groups. In the control group (C) rats were fed a standard complete diet; in the experimental groups standard diets were supplemented with different doses of bee pollen. Treatment group T1 was given standard diet with the addition of bee pollen at a 0.3% concentration; in group T2 the addition was 0.5%; and in group T3 it was 0.75%. The experimental period lasted for 90 days. A significant effect (P < 0.05) of bee pollen on the body weight gain and feed conversion ratio of female rats was found. Significantly (P < 0.05) higher cholesterol concentration in blood serum of male rats was found in the groups with bee pollen addition (groups T2 and T3) compared to the control group. Lower triglyceride serum content in all female experimental groups (T1 and T3) was observed in comparison to the control. Higher serum cholesterol content in the experimental female rats was detected; significant differences were analysed in groups T1 and T3 compared to the control female group. Rapeseed bee pollen at concentrations of 0.5 and 0.75% positively affected the body weight gain of female rats, however, with higher feed consumption (P < 0.05). Rapeseed bee pollen reduced the triglycerides serum content in female rats and increased the cholesterol serum content in male and female rats (P < 0.05).

A study was conducted to determine the performance, carcase and gastrointestinal tract (gut) characteristics of broilers fed diets supplemented with dry peppermint (<I>Mentha piperita</I> L.) or thyme (<I>Thymus vulgaris</I> L.) leaves, which are among the alternative growth promoters. In the study, 312 one-week-old broilers (Ross-308) were used. There were 3 dietary treatments, each consisting of 4 replications (13 males and 13 females in each replication). The control group was fed basal diet, while the peppermint and thyme groups were fed diets supplemented with 0.2% peppermint or thyme (w/w) as menthol and thymol (70 mg per kg diet) sources, respectively. From 7 days to 35 days of age, the body weight gain was higher (<I>P</I> < 0.05) in broilers fed the peppermint-supplemented diet compared to the control, but the effect of peppermint on body weight gain disappeared at 42 days of age. Feed intake, feed to gain ratio, carcase weight, carcase yield, and the relative weights of the edible inner organs and whole gut, and the relative length of the whole gut were not significantly affected by peppermint and thyme contents. The peppermint or thyme leaves increased (<I>P</I> < 0.05) the abdominal fat pad at 42 days of age. Thus, the dry peppermint leaves had a higher growth promoting efficacy than the dry thyme leaves at an early stage of broilers’ life, but an increase in abdominal fat should be taken into account for carcase quality and processing.
