Introduction: Pentavalent antimony is the first choice drug for leishmaniasis in dog. Leishmaniasis has a complex pathogenesis and it manifests various clinical signs, some of which are often similar to those associated with the toxicity induced by antimonial treatment. Among the reasons for this toxicity, also a general problem of drug’s quality has been reported. Methodology: The general and local tolerability of two commercially available meglumine antimoniate based veterinary products was evaluated in 12 healthy dogs, 6 receiving Antimania (Fatro, Italy) and 6 receiving Glucantime (Merial, Spain), following repeated subcutaneous administrations of therapeutic doses for 14 days. Results: Individual and mean values of haematological and biochemical parameters in both groups remained in the physiological range, with no considerable differences within the two groups. The general tolerability of the drugs was also supported by clinical observations and physical examination of the dogs throughout the whole study period. Only slight local reactions at the injection sites, that spontaneously disappeared, were observed for both products starting from 12-84 hours after the administration. The pharmacokinetic parameters indicated no antimony accumulation. Conclusions: These results suggest that meglumine antimoniate administered at the recommended dosage regimen is well tolerated by healthy dogs, and that there is no significant difference between the two tested products.

ABSTRACT: This study aimed to determine the lethal concentration and the structural and ultra-structural effects caused by the formalin exposure on juveniles of Arapaima gigas. Ninety fish (60.1± 2.5g and 20.2±0.9cm) were exposed to 0, 22, 44, 66, 88 and 110mg L-1 in order to determine the lethal concentration (LC50-96h) that was 36.4mg L-1 of formalin. Sublethal effects were evaluated using histopathological analysis on the gills and assessment of behavioral alterations and clinical signs. The LC50 of formalin for 24, 48 and 72h was 88.3, 64.7 and 56.8mg L-1 respectively. Clinical signs and behavioral changes were found: erratic swimming, lethargy, crowding on the water surface, loss of hydrodynamic equilibrium, spasms and agonistic confrontation, which were observed only at 88 and 110mg L-1. The histological alteration index (HAI) showed that 66, 88 and 100mg L-1 presented significant difference (p<0.05) in relation to unexposed fish, indicating that moderate damage to the gills of fish exposed to formalin had occurred. The mean values of alteration (MVA) for 22, 44, 66, 88 and 110mg L-1 were 1.14, 1.29, 1.51, 1.53 and 1.60 respectively, and differences in this index were only observed with 110 mgL-1 of formalin. It is therefore possible to conclude that sublethal concentrations of formalin (22.0mg L-1) did not compromise the health of juveniles of A. gigas. Finally, concentrations greater than to LC50-96h may be carefully used for short-term exposure, since the MVA for all concentrations tested only indicated localized lesions that did not compromise gills functionality of exposed fish.

ABSTRACT: The influence of the proximal tibial angles in the cranial cruciate ligament (CCL) rupture in dogs is still controversial, and little is known regarding this topic in cats. The aim of this study was to evaluate and compare the angles of the proximal portion of the tibia in dogs and cats with and without CCL rupture. Retrospective and prospective radiographs of the stifle joints were obtained and divided into four groups. Group 1 was composed of 70 stifle joint images of dogs without orthopedic disorders (healthy dogs), group 2 had 70 stifle joint images of dogs with CCL rupture, group 3 had 50 stifle joint images of cats without orthopedic disorders (healthy cats) and group 4 had 25 stifle joint images of cats with CCL rupture. Radiographs were taken with the stifle joint in the mediolateral projection, positioned at the angle of hind limb support. Between the two groups of dogs evaluated, the dogs with CCL rupture had statistically greater tibial plateau angle (TPA) compared with healthy dogs. No difference was shown in relation to the TPA between healthy cats and cats with CCL rupture. In relation to the patellar ligament angle by tibial plateau method the values for the healthy dogs were significantly higher than those for the CCL ruptured dogs. Similarly, healthy cats had significantly higher mean values than cats with CCL rupture. In the patellar ligament angle by common tangent method there was no significantly difference between the two groups of dogs. Between the two groups of cats, animals with CCL rupture had statistically higher mean values than healthy cats. In general, the groups of dogs showed higher mean values than the groups of cats. For the patellar ligament insertion angle (PLIA) healthy dogs showed a significantly higher mean than dogs with CCL rupture. There was no significant difference between the groups of cats. In conclusion, the TPA and the PLIA possibly influence the etiology of CCL rupture in dogs but not in cats. The low patellar ligament angle measured by common tangent method may favorably influence the reduced incidence of CCL rupture in cats.

ABSTRACT A comparative in vivo pharmacokinetic study of florfenicol was conducted in 18 crossbred pigs infected with Actinobacillus pleuropneumoniae following intravenous (i.v.), intramuscular (i.m.), or oral (p.o.) administration of a single dose of 20 mg/kg. The disease model was confirmed by clinical signs, X rays, pathohistologic examinations, and organism isolation. Florfenicol concentrations in plasma were determined by a validated high-performance liquid chromatography method with UV detection at a wavelength of 223 nm. Pharmacokinetic parameters were calculated by using the MCPKP software (Research Institute of Traditional Chinese Veterinary Medicine, Lanzhou, China). The disposition of florfenicol after a single i.v. bolus was described by a two-compartment model with values for the half-life at α phase (t 1/2α), the half-life at β phase (t 1/2β), the area under the concentration-time curve (AUC0-∞), and the volume of distribution at steady state (V ss) of 0.37 h, 2.91 h, 64.86 μg · h/ml, and 1.2 liter/kg, respectively. The concentration-time data fitted the one-compartment (after i.m.) and two-compartment (after p.o.) models with first-order absorption. The values for the maximum concentration of drug in serum (C max), t 1/2α, t 1/2β, and bioavailability after i.m. and p.o. dosing were 4.00 and 8.11 μg/ml, 0.12 and 3.91 h, 13.88 and 16.53 h, and 122.7 and 112.9%, respectively, for the two models. The study showed that florfenicol was absorbed quickly and completely, distributed widely, and eliminated slowly in the infected pigs, and there was no statistically significant difference between the pharmacokinetic profiles for the infected and healthy pigs.

In canine leishmaniosis (CanL), kidneys are affected in virtually all dogs. Treatment of CanL is limited in Europe to meglumine antimoniate and miltefosine. This study evaluated the pharmacological, toxicological, and pathological effects of both drugs in healthy beagle dogs. Four male and four female dogs were divided into two groups. The animals in Group 1 were administered an oral solution of 2% of miltefosine at 2 mg/kg b.w. once a day, for twenty-eight days. The animals in Group 2 were administered a preparation of meglumine antimoniate at 100 mg/kg b.w. subcutaneously once a day for twenty-eight days. After treatment, all dogs were followed-up for a further twenty-eight days. Dogs were observed daily and clinically examined ten times throughout the study. On days -1 and 55 a renal biopsy was performed on all dogs and analyzed by light microscopy, immunofluorescence, and electron microscopy. All the examinations failed to demonstrate any lesions in the miltefosine-treated dogs. Conversely, all the meglumine antimoniate–treated dogs demonstrated severe tubular damage, characterized by tubular cell necrosis and apoptosis. In conclusion, although no clinical signs of renal disease were evident, the use of meglumine antimoniate in the pharmacological treatment approach of CanL-affected dogs should be carefully considered.

Rheumatoid factor (RF) is an important marker of many autoimmune diseases. Presence of RF is characteristic for rheumatoid arthritis and other inflammatory diseases of various underlying aetiologies. The aim of our study was to introduce and standardise a convenient assay for RF-IgM detection in canine serum and to test it on a group of selected dogs. ELISA was chosen as a method of analysis due to its high specificity, selectivity and optimal quality in clinical use. For standardised method evaluation we examined serum samples from 80 dogs classified into 4 groups as follows: I - dogs positive in the ANA test (ANA+) suffering from orthopaedic or complex diseases; II - dogs negative in the ANA test (ANA-) suffering from orthopaedic or complex diseases; III - dogs negative in the ANA test (ANA-) suffering from chronic inflammatory diseases; IV - control healthy dogs. Significant difference between mean values of RF-IgM in respective groups was detected. Dogs from groups I and II showed the highest mean values. These were significantly higher than the values in dogs from groups III and IV and the differences between the last mentioned two groups were also significant. Our data suggest that the introduced method is suitable for use in the veterinary laboratory practice.

ABSTRACT A comparison has been made between (A), a relatively simple method for the measurement of aldosterone secretion rate, based on paper chromatography and direct densitometry of the aldosterone spot and (B) a more elaborate isotope derivative method. The mean secretion rate in 9 normal subjects was 112 ± 26 μg per 24 hours (method A) and 135 ± 35 μg per 24 hours (method B). The »secretion rate« in one adrenalectomized subject after the intravenous injection of 250 μg of aldosterone was 230 μg per 24 hours (method A) and 294 μg per 24 hours (method B). There was no significant difference in the mean values, and correlation between the two methods was good (r = 0.80). It is concluded that the densitometric method is suitable for clinical purposes as well as research, being more rapid and less expensive than the isotope derivative method. Method A also measures the urinary excretion of the aldosterone 3-oxo-conjugate, which is of interest in many pathological conditions. The densitometric method is obviously the less sensitive and a prerequisite for its use is an aldosterone secretion of 20—30 μg per 24 hours. Lower values are, however, rare in adults.

Implant-supported prostheses hygiene and peri-implant tissues health are considered to be predictive factors for successful prosthetic rehabilitation. Therefore, the purpose of this study was to evaluate the effectiveness of brushing associated with oral irrigation measured as biofilm-removing capacity, maintenance of healthy oral tissues, and patient satisfaction. A randomized, crossover clinical trial was conducted with 38 patients who wore conventional maxillary complete dentures and mandibular overdentures retained by the O-ring system. The patients were instructed to use the following hygiene methods for 14 days: mechanical brushing [MB (brush and dentifrice - Control)]; and MB with oral irrigation [WP (Waterpik - Experimental)]. Biofilm-removing capacity and maintenance of healthy oral tissues were evaluated by the Modified Plaque Index (PI), Gingival Index (GI), Probing Depth (PD), and Bleeding on Probing Index (BP) recorded at baseline and after each method. In parallel, patients answered a specific questionnaire using a Visual Analogue Scale after each hygiene method. Data were analyzed by Friedman and Wilcoxon tests (α=0.05). The results showed significantly lower PI, GI, PD, and BP indices after application of the hygiene methods (P&lt;0.001) than those observed at baseline. However, no significant difference was found between MB and WP. The satisfaction questionnaire responses to both methods showed high mean values for all questions, with no statistically significant difference found between the answers given after the use of MB and WP (P&gt;0.05). The findings suggest that WP was effective in reducing PI, GI, PD, and BP indices and provided a high level of patient satisfaction.

O trabalho teve por objetivo verificar diferenças entre rendimento do concentrado proteico e proteína bruta da folha da mandioca (Manihot esculenta Crantz), obtidos a partir de três variedades comerciais: branca, cascuda e vermelha. As manivas foram plantadas seguindo o delineamento experimental inteiramente casualizado com três repetições. Nas comparações entre as variedades, considerou coletas escalonadas pós-plantio, realizadas aos 12, 14 e 16 meses. O concentrado proteico foi obtido a partir da farinha das folhas inteiras e submetido ao método de termo - coagulação ácido e a proteína bruta pelo método padrão AOAC. Os dados foram submetidos a análise de variância e comparados pelo teste de Tukey - 5% de probabilidade. Os resultados não indicaram diferença significativa entre as médias obtidas para rendimento de concentrado proteico. Para a variável porcentagem de proteína bruta a variedade Branca foi a que apresentou maior valor, com média de 46,25%, seguida pela Cascuda e Vermelha, 44,52% e 37,30%, sucessivamente. Conclui-se que outros estudos devem avaliar condições que possam influenciar no teor de proteína foliar, como clima e solo, e, avaliar os níveis de ácido cianídrico de cada variedade, indicando qual das três é a melhor para a extração do concentrado proteico das folhas. Palavras chaves: Manihot esculenta Crantz, variedades comerciais, concentrado proteico, proteína bruta. ABSTRACT: The study aimed to assess the differences between income protein concentrate, crude protein of cassava leaf (Manihot esculenta Crantz), obtained from three commercial varieties: white, red and cascuda. The cuttings were planted following the completely randomized design with three replications. Comparisons between the varieties considered after planting staggered collections, held on 12, 14 and 16 months. The protein concentrate was obtained from flour of whole sheets and subjected to the term method - acid coagulation and crude protein by AOAC standard method. The data were submitted to ANOVA and Tukey test - 5% probability. The results indicated no significant difference between the mean values obtained for protein concentrate income. For the variable percentage of crude protein White variety showed the highest, with an average of 46.25%, followed by cascuda and Red, 44.52% and 37.30%, successively. We conclude that further studies should evaluate conditions that may affect the leaf protein content, such as climate and soil, and evaluate the hydrocyanic acid levels of each variety, indicating which of the three is the best for the extraction of protein concentrate from leaves Key words: Manihot esculenta Crantz, commercial varieties, protein concentrate, crude protein.

Introduction: Atrophic vaginitis is a common problem in postmenopausal women and results from decreased levels of blood estrogen. It is associated with symptoms of itching, burning, dyspareunia, and postmenopausal bleeding. The present study evaluated the effects of fenugreek extract on atrophic vaginitis. Materials and Methods: This randomized controlled clinical trial was performed on 60 postmenopausal women in Ardabil, Iran, in 2018. The participants were selected using block randomization with the allocation ratio 1:1. Those in the intervention group received 0.5g (the applicator filled to the half-full mark) fenugreek vaginal cream 5% twice a week for 12 weeks. The control group received conjugated estrogens vaginal cream at the dose of 0.625 mg (the applicator filled to the half-full mark) containing 0.3 mg of conjugated estrogens. Atrophic vaginitis was evaluated before and after the treatment through clinical examination, clinical signs, and measurement of Vaginal Maturation Index (VMI). Findings: After the 12-week intervention and modification of the baseline score, the mean (standard error) score for atrophic vaginitis signs was 3.100 (1.43-4.75). This difference was statistically significant in intragroup comparison and in favor of the control group in intergroup comparison (p=0.001). VMI was less than 49% in 86.7% and 46.7% of the participants in the intervention and control groups, respectively. This was a significant difference in favor of the control group (p=0.001). Conclusion: The results of this study showed that total fenugreek extract could be effective in treating signs of atrophic vaginitis, but it was not as effective as ultra-low-dose estrogen.

Ruminal acidosis is an important clinical emergency in small ruminants. In this study, eight healthy farm goats and 24 goats presented at TVCC of the college of Veterinary Sciences and A.H., Anand with clinical signs of ruminal acidosis and having rumen liquor pH below 6 were examined for alterations in the ruminal fluid and serum biochemical parameters. Among various rumen fluid parameters evaluated, the mean values of rumen fluid pH decreased significantly (4.71±0.11 vs. 6.90±0.10), while sediment activity time (46.67±1.20 vs. 24.50±0.78 min) and methylene blue reduction time (29.50±0.73 vs. 10.03±0.27 min) increased significantly in acidotic goats. The normal greenish, aromatic viscous color, odour and consistency of rumen fluid of healthy goats also changed to milky grey/creamy, sour/pungent watery in acidotic goats. The rumen protozoal activity decreased to nil in acidotic goats as compared to the healthy goats. Among various serum biochemical constituents, the mean values of glucose (92.43±1.37 vs. 74.13±1.83 mg/dl), BUN (26.49±0.47 vs. 22.63±1.19 mg/dl), serum creatinine (01.01±0.02 vs. 00.83±0.02 mg/dl) and albumin (03.22±0.03 vs. 03.05±0.05 g/dl), ALT (56.75±1.55 vs. 27.88±1.14 IU/L) and AST (93.25±1.82 vs. 54.00±1.75 IU/L), increased significantly, while there was significant decrease in serum calcium (09.09±0.14 vs. 10.29±0.08 mg/dl) in acidotic goats. The mean values of alkaline phosphatase (IU/L) in acidotic goats increased non-significantly from the base values of healthy goats.
