Several new heterocyclic systems based on a hydroxybenzofuro[2,3-b]pyridine building block were prepared. This benzofuropyridine is easily available from the Meerwein reaction of benzoquinone and a heterocyclic diazonium salt, followed by reduction and cyclization. Electrophilic substitution and further condensations give polycyclic systems, including oxazolo- and chromeno-fused analogues.

Two new conjugated microporous polymers, TPB-B-CMP and TPA-B-CMP, with β-diketone-boron difluoride dye as the key building block were designed and successfully prepared. They were further employed as efficient metal-free heterogeneous photocatalysts.

3-Amino-6-iodoacridine (10), 3,6-diiodoacridine (11) and 9-amino-2-ethoxy-6-iodoacridine (14) were prepared by iodide ion substitution of the corresponding diazonium salt whereas 3,6-diamino-4,5-diiodoacridine (12) and 6,9-diamino-2-ethoxy-5-iodoacridine (13) were prepared by direct iodination with iodide ion in the presence of chloramine-T. The latter reaction proceeded in relatively high yield and has been used for the synthesis of high specific activity 125I-labelled compounds (12), (13). The 1H and 13C N.M.R. spectra of (10)-(14) and model compounds indicate higher electron density at C4(C5) than at C2(C7) in 3(6)-amino-substituted acridines in agreement with the observed pattern of electrophilic substitution.

Reaction of 2.2-diethoxy-1-ethenediazonium salt la with primary amines 5 yields 1-alkyl-1 H1,2,3-triazoles 7. With hydrazide 8 diazonium salt la reacts completely differently. The thus formed 2′-[2-phenyl-4H-1,3,4-oxadiazine-5(6H)-ylidene]benzohydrazide (9) can be annellated under elimination of water or transformed by reaction with phosgene, thionyl chloride and phthalic acid dichloride respectively to give the heterocycles 10-13.

As a part of our ongoing synthetic quest to expand the frontiers of contemporary medicinal chemistry, we now report an expedient synthesis of a potentially useful bicyclo[1.1.1]pentane building block, 3-(pyrazin-2-yl)bicyclo[1.1.1]pentane-1-carboxylic acid. This report also showcases the application of this motif as a “spacer” probe in a biological study.
