Peningkatan produksi dan reproduksi ternak akan optimal jika penyediaan pakan yang memadai dan pengendalian penyakit dilakukan secara efektif. Gangguan penyakit pada ternak dapat menurunkan produktivitas ternak. Penyakit yang biasanya menyerang ternak sapi adalah penyakit cacing. Penyakit ini tidak mengakibatkan kematian pada ternak, namun menyebabkan kerugian yang sangat besar karena menurunkan berat badan dan daya produktivitas hewan. Biji labu kuning telah lama digunakan sebagai antihelmintik. Penentuan efektivitas biji labu kuning sebagai obat cacing, maka dilakukan penelitian dengan memasukkan cacing sebanyak 10 ekor ke dalam cawan petri yang berisi infusa biji labu kuning dengan konsentrasoi yang berbeda, yaitu (P1): konsentrasi kandungan infusa biji labu kuning 25/100 ml, (P2): konsentrasi kandungan infusa biji labu kuning 50/100 ml, dan (P3): konsentrasi kandungan infusa biji labu kuning 75/100 ml. Hasil penelitian menunjukkan bahwa dengan konsentrasi infusa labu kuning sebesar 75% mampu membunuh semua cacing fasciola yang  ada. Konsentrasi yang lebih rendah menunjukkan angka kematian sebesar 47% dan 77%. Dengan demikian dapat direkomendasikan bahwa penggunaan infusa labu kuning sebesar 75/100 ml.

Kegunaan labu kuning di Indonesia masih sebatas daging buah yang dapat diolah menjadi panganan seperti kue basah, kolak dan sayur berkuah. Secara empiris, biji labu kuning telah digunakan untuk mengatasi cacingan. Penelitian ini dilakukan untuk mengetahui mortalitas cacing gelang (Ascaridia galli) dalam ekstrak etanol biji labu kuning (Cucurbita moschata Duchesne). Penelitian ini menggunakan 25 ekor Ascaridia galli yang dibagi menjadi 5 kelompok, kelompok I kontrol negatif menggunakan larutan NaCl fisiologis, kelompok II kontrol positif menggunakan larutan pirantel pamoat 0,5 %, kelompok III, IV dan V berturut-turut menggunakan 25 mg/ml, 50 mg/ml dan 100 mg/ml ekstrak etanol biji labu kuning. Parameter penelitian ini ditentukan dengan melihat persentase nilai skor pasca inkubasi 12 jam, 24 jam, dan 36 jam. Skor 3 diberikan apabila seluruh tubuh Ascaridia galli bergerak, skor 2 diberikan jika hanya sebagian tubuh Ascaridia galli bergerak, skor 1 jika Ascaridia galli diam tetapi masih hidup, dan skor 0 apabila Ascaridia galli mati. Hasil uji in vitro dengan perlakuan 25 mg/ml ekstrak etanol biji labu kuning menyebabkan kematian 3 ekor Ascaridia galli  atau 60% pasca inkubasi 36 jam, sedangkan ekstrak etanol biji labu kuning dengan perlakuan 50 mg/ml, 100 mg/ml dan kelompok kontrol positif mengakibatkan kematian 4 ekor Ascaridia galli atau 80% pasca inkubasi 36 jam. Dari hasil penelitian disimpulkan bahwa ekstrak etanol biji labu kuning (Cucurbita moschata Duchesne) dosis 25 mg/ml, 50 mg/ml, dan 100 mg/ml secara in vitro dalam waktu 36 jam mampu mengakibatkan mortalitas Ascaridia galli. The use of yellow pumpkin in Indonesia is still limited to fruit meat that can be processed into snacks such as soggy cakes, porridge and vegetable soup. This research was conducted to determine the mortality of Ascaridia galli in ethanol extract of yellow pumpkin seeds (Cucurbita moschata Duchesne). This study used 25 Ascaridia galli which were divided into 5 groups, group I was negative control using physiological NaCl solution, group II was positive control using 0.5% pirantel pamoate solution, group III, IV and V respectively used 25 mg / ml, 50 mg/ml and 100 mg/ml ethanol extract of yellow pumpkin seeds. The parameters of this study were determined by looking at the percentage of post-incubation scores 12 hours, 24 hours, and 36 hours. A score of 3 is given if the whole body of Ascaridia galli moves, a score of 2 is given if only part of the body of Ascaridia galli moves, a score of 1 if Ascaridia galli is still but still alive, and a score of 0 if Ascaridia galli dies. In vitro test results with 25 mg/ml ethanol extract of pumpkin seeds caused 3 deaths of Ascaridia galli or 60% after incubation for 36 hours, while ethanol extract of yellow pumpkin seeds treated with 50 mg / ml, 100 mg/ml and positive control group resulting in the death of 4 Ascaridia galli or 80% after 36 hours incubation. From the results of the study concluded that the ethanol extract of yellow pumpkin seeds (Cucurbita moschata Duchesne) doses of 25 mg / ml, 50 mg / ml, and 100 mg / ml in vitro within 36 hours can lead to Ascaridia galli mortality.

Pumpkin seeds are commonly consumed in Greece. Although Cucurbita moschata is locally grown in Lemnos and is traditionally used in pumpkin pies, the seeds are currently discarded after consumption of the fruit flesh. The aim of the present study was to investigate the nutritional functionality of pumpkin seeds from Cucurbita moschata grown in Lemnos.Cucurbita moschatas’ seeds, raw or roasted, were appropriately extracted and the results are presented for raw versus (vs) roasted seed extracts. The phenolic content was expressed as μg gallic acid/g of seeds according to Folin-Ciocalteau assay (370.3 ± 19.1 vs 551.0 ± 22.0). Antioxidant capacity was expressed as equivalent amount for 50% scavenging in mg of seeds for DPPH (50.03 ± 5.91 vs 25.82 ± 6.77) and ABTS (17.85 ± 0.77 vs 12.77 ± 0.76) assays, and as μmol of trolox/g of seeds for FRAP (1.19 ± 0.05 vs 2.50 ± 0.23) and CUPRAC (2.13 ±0.11 vs 3.25 ± 0.06) assays. Anti-inflammatory/anti-thrombotic and anti-diabetic activities were expressed as mg of seeds for 50% inhibition of platelet activating factor (0.62 vs 0.15) and as μg of seeds for 25% inhibition of alpha-glycosidase (40.0 vs 61.0) activities respectively. Moreover, anti-atherogenic activity was expressed as the % increase in lag time of human plasma oxidation (62.7 versus 163.2)Raw and roasted pumpkin seed extracts exert anti-oxidant, anti-thrombotic/anti-inflammatory, anti-atherogenic and antidiabetic activities. Cucurbita moschata seeds may represent a novel opportunity for development of functional foods, with a local interest in Lemnos that would contribute also to the regional public health improvement.

Fruit flies (Diptera: Tephritidae) are the primary pests of horticultural crops worldwide, causing both direct and indirect damage to the fruit production, including Anastrepha grandis (Macquart, 1846), which exclusively infests Cucurbitaceae fruit. Dose-response tests were used to examine the effects of gamma radiation exposure on 24 – 48 h old eggs and third instar larvae, both in vitro and in Atlas squashes (Cucurbita moschata). The following physicochemical properties of Atlas squashes exposed to gamma radiation were evaluated: titratable total acidity, sugar, pH, external colouration, and texture. It was found that under in vitro conditions, a 20.0 Gy dose of gamma radiation was able to prevent larval eclosion, but a 200 Gy dose was necessary to prevent emergence after the exposure of third instar larvae. In Atlas squashes, 200 and 250 Gy doses of gamma radiation were able to prevent the emergence of A. grandis adults from eggs and larvae, respectively. The radiation doses estimated by Probit 9 to prevent emergence from squashes infested with eggs and third instar larvae were 257.13 Gy and 270.25 Gy respectively. The use of gamma radiation did not significantly affect the physicochemical properties of Atlas squashes, which suggested that gamma radiation may represent a potentially useful technique for the quarantine control of A. grandis in cucurbits.

There are risks involved in the production and exportation of fruit fly hosts due to the possible spread of tephritid pests during distribution. Anastrepha grandis attacks cucurbit fruits and is considered an A1 quarantine pest in many countries. The objective of this study was to evaluate the effect of hot water treatment on the eggs and larvae of A. grandis in vitro, and on ‘Atlas’ squash (Cucurbita moschata). The eggs and third-instar larvae of A. grandis were exposed to hot water at temperatures of 42.0, 44.0, 46.0, 46.5, 47.0, 47.5, 48.0, 49.0 and 50.0 (± 0.5) °C for durations of 0 (control), 10, 20, 30 and 60 minutes. Water temperatures of at least 44 °C affected the in vitro larval eclosion of A. grandis during all exposure times. No adults were obtained when in vitro A. grandis larvae were treated at 49 °C and 50 °C at all exposure times and, 48 °C for 30 and 60 minutes. No adults were obtained when squashes infested with A. grandis eggs or larvae were treated at temperatures of 49 °C and 50 °C during any exposure time, as well as subjected to 48 °C for 20 minutes. Anastrepha grandis larvae were slightly more susceptible to hydrothermal treatment than eggs in squashes. Hot water treatment applies at a temperature of 48 °C for 20 minutes is an effective phytosanitary treatment for squashes cv. Atlas infested with eggs and larvae of A. grandis.

Pumpkin is considered a healthy and functional food. The consumption of pumpkins and pumpkin-based foods has been shown to confer several beneficial effects on human health due to their antioxidant capacity and terpenoid content. Consequently, this study aimed to characterize the in vitro antioxidant capacity (using FRAP and ABTS assays), terpenoid profile (using an untargeted lipidomics approach via high-resolution UHPLC-Orbitrap mass spectrometry), and carotenoid content (by HPLC-DAD) in pumpkin fruit pulp from accessions differing for species (11 Cucurbita maxima and 9 Cucurbita moschata), cultivar, and origin, belonging to a Serbian breeding collection. These accessions are candidates for inclusion within programs intended to improve pumpkin fruit quality. The results obtained in this work allowed us to highlight the best marker compounds, discriminating both the region of accession collection or breeding (“origin”) and the plant species. Furthermore, our findings have helped to identify the most suitable antioxidant-rich varieties to select for national breeding programs for improving human health. These findings provide valuable information to the overall current understanding of the potential health benefits of pumpkins and the discriminant triterpenoids underlying the C. maxima and C. moschata accessions investigated here, which include those of Serbian and non-Serbian origin.

Background: Echinococcus granulosus parasite causes a zoonotic disease which is important for public and veterinary health. Since pumpkin seeds (Cucurbita sp.) are used as traditional vermifuge in Iran, they may be a potential herbal anthelmintic. Methods: This study was designed in 2016 to evaluate the in vitro scolicidal effect of Cucurbita moschata seeds form northern part of Iran. Hydroalcoholic and petroleum ether extracts were prepared by maceration and soxhlet respectively. Both extracts with four different concentrations (100, 10, 1, 0.1 mg/ml) were incubated against protoscoleces in 5, 15, 30 and 60 min. Results: Maximum mortality was 16% with 1% hydroalcoholic extract in 60 min. The highest mortality with organic extract was 4% with 10% concentration in 60 min (P=0.015). Conclusion: Since highest mortality was 16%, the extract did not reach to LD50 (50% mortality). Therefore, the potency of the total extract is not sufficient as potential scolicidal drug.

Carotenoids are a widespread group of fat-soluble pigments, and their major nutritional importance comes from their pro-vitamin A activity and their antioxidant capacity. In this study, two different pumpkin cultivars (Cucurbita maxima, also named `Delica’ and Cucurbita moschata, also known as `Violina’) from the southern Po Delta area were investigated in terms of carotenoid content and the influence of food processing on compositional changes and carotenoid bioaccessibility. Quali- and quantitative determination of carotenoids in sample extracts were performed on a C30 column by means of an online coupled HPLC-UV/Vis-APCI-MS/MS technique. The identification of separated compounds was tentatively achieved by merging (i) chromatographic data, (ii) UV-Vis spectra, and (iii) MS/MS fragmentation spectra. The chromatographic profiles for the two cultivars showed qualitative differences. Two major carotenoids were considered for quantification purposes and further investigations: lutein and β -carotene. Quantification of target carotenoids was performed with external calibration through analytical standards. The concentration of lutein and β -carotene was higher in C. maxima than in the other variety, C. moschata. Carotenoids are susceptible to degradation (isomerization and oxidation) during food processing (i.e., cooking), and the concentration of lutein and β -carotene were monitored in oven-cooked and steam-cooked pumpkins. The steam-cooking process was superior in terms of limiting carotenoid loss. A complete functional profile of pumpkins as a source of carotenoids was gained with the evaluation of their in vitro bioaccessibility and their bioavailability after intake during human digestion. Bioaccessibility of lutein and β -carotene were estimated by an in vitro static digestion model that involved salivary, gastric, and duodenal phases. Bioaccessibility values progressively increased from the salivary to the duodenal phase for both pumpkin varieties and cooking methods. Bioaccessibility of lutein was always lower than β -carotene for both cultivars and for both cooking methods. Bioaccessibility values for lutein and β -carotene changed from 1.93% to 2.34% vs. 4.94% and 8.83% in the salivary phase, from 2.7% to 4.63% vs. 7.83% and 15.60% in the gastric phase, and from 10.04% to 13.42% vs. 25.81% and 35.32% in the duodenal phase. For both target compounds, bioaccessibility in the duodenal phase was more than twice the gastric values, and it underlined that the type of cooking did not influence release from the initial matrix.
