Introducción: las infecciones por Staphylococcus spp. multirresistentes están asociadas con una mayor morbimortalidad de los pacientes afectados. Objetivo: caracterizar fenotipos de resistencia a meticilina, macrólidos y lincosamidas de 50 aislados de Staphylococcus spp. provenientes de pacientes de un centro hospitalario en la ciudad de Valledupar (Colombia). Materiales y métodos: las pruebas de susceptibilidad a meticilina, eritromicina y clindamicina se realizaron por los métodos de microdilución en caldo y difusión en agar. La resistencia a meticilina se tamizó por la técnica de dilución en agar y la resistencia inducible a clindamicina por la prueba D. Resultados: los aislados de Staphylococcus fueron obtenidos principalmente de heridas (58%) y orinas (12%) y en las áreas de consulta externa (40%), cirugía (24%) y urgencias (10%). Staphylococcus aureus se aisló en un 68%, seguido de Staphylococcus epidermidis (14%), Staphylococcus lugdunensis (8%), Staphylococcus saprophyticus (4%), Staphylococcus haemolyticus (4%) y Staphylococcus hominis (2%). La resistencia a meticilina se encontró en el 36% de los aislados de Staphylococcus aureus y el 8% de los estafilococos coagulasa negativos. Se evidenciaron cinco fenotipos de resistencia; el fenotipo con sensibilidad a eritromicina y clindamicina fue el más frecuente (54%), seguido del de resistencia a ambos antibióticos (14%). La resistencia inducible a clindamicina fue del 12%, encontrándose en el 8% de aislados de Staphylococcus aureus y el 4% de los de Staphylococcus epidermidis. Conclusiones: la prueba D es esencial para detectar el fenotipo de resistencia inducible a clindamicina en aislados de Staphylococcus spp. y evitar su administración frente al inminente fracaso terapéutico.

Abstract A retrospective analysis of microbiology data from general infections was performed. From 105 isolates strains of Staphylococcus spp. 36 (34,28%) were Staphylococcus epidermidis, 33 (31,42%) Staphylococcus aureus, 21 (20%) Staphylococcus haemolyticus, and 15 (14,28%) were Staphylococcus hominis. Results: S. hominis isolates were predominantly resistant to betalactamins (93,33 %) and even Imidazole (60 %). 53,33 % of strains were resistent to aminosides and 33,33 % to Ciprofloxacin. All strains (100%) were sensitive to Vancomycin, but also all were susceptibile to Quinupristin-Dalfopristin. A high percentage of S. hominis were sensitive to Moxifloxacin, Linezolid (93,33 %), and to teicoplanin (86,67%). Discussion: S. hominis is a member of skin normal flora, but all strains of S. hominis were isolated from generalized infection with a high rate of mortality.

Rápidos, portáteis e acessíveis, os aparelhos celulares foram incorporados às atividades e rotinas diárias da população. Diante de tal intimidade, esses aparelhos também passaram a se constituir em um meio de crescimento para agentes microbiológicos comensais e acidentais. Medidas de controle como a adoção de boas práticas de higiene são imprescindíveis para a diminuição da carga microbiológica e, por consequência, decréscimo dos riscos de transmissão destes agentes ao homem, principalmente em imunodeprimidos. Dessa forma, o presente trabalho objetivou identificar os micro-organismos presentes em aparelhos celulares de acadêmicos e professores da Universidade de Cuiabá (UNIC), Campus Primavera do Leste – MT, através de amostragens obtidas de 24 aparelhos de telefones celulares de voluntários. Foram realizadas coletas para cada um dos aparelhos celulares disponibilizados pelos voluntários, através de um swab de transporte. Este foi deslizado na parte externa do aparelho, cobrindo toda a área da tela, conectores e alto-falante e, em seguida, inseridos em meio Stuart e remetidos ao laboratório. Do total de amostras coletadas, 19 (79,1%) apresentaram crescimento bacteriano em meio de cultura nutritivo. Os micro-organismos isolados foram Staphylococcus aureus em seis amostras (31,6%), Staphylococcus lugdunensis em outras seis (31,6%), Staphylococcus epidermidis em três (15,8%), Staphylococcus spp. em três (5,8%) e Escherichia coli em uma (5,2%) das amostras. Diante dos resultados obtidos se pôde constatar que os aparelhos celulares podem atuar como meio de multiplicação microbiológica, assim se deve adotar medidas preventivas de higiene e antissepsia das mãos e destes aparelhos, a fim de evitar a proliferação e veiculação de micro-organismos por meio destes. Palavras-chave: Telefone Celular. Staphylococcus aureus.  Escherichia coli. AbstractFast, portable and affordable, mobile phones have been incorporated into the daily activities and routines of the population. Faced with such intimacy, these devices have also become a growth medium for commensal and accidental microbiological agents. Control measures such as the adoption of good hygiene practices are essential to reduce the microbiological load and, consequently, decrease the risks of transmission of these agents to humans, especially in immunosuppressed ones. Thus, the present work aimed to identify the microorganisms present in cell phones of academics and professors of the University of Cuiabá (UNIC), Campus Primavera do Leste - MT, through samples obtained from 24 cell phones of volunteers. The samples  were collected for each of the mobile devices made available by the volunteers through a transport swab. This was slid on the external part of the device, covering the entire display area, connectors and speaker, then inserted into Stuart medium and sent to the lab. Of the total samples collected, only 19 (79.1%) showed bacterial growth in nutrient culture medium. Microorganisms isolated from growth media were Staphylococcus aureus in six samples (31.6%), Staphylococcus lugdunensis in six (31.6%), Staphylococcus epidermidis in three (15.8%), Staphylococcus spp. in three (5.8%) and Escherichia coli in one (5.2%) of the samples. Given the results obtained, it can be seen that cell phones can act as a means of microbiological multiplication, so preventive measures of hygiene and antisepsis of  hands should be adopted, in order to prevent the proliferation and spread of microorganisms through them . Keyword: Cell Phone. INTRODUÇÃO: Com a evolução da tecnologia e a disponibilização de telefonia móvel tornou-se cada dia mais comum estes aparelhos fazerem parte da rotina da população e também dos profissionais de saúde inclusive em seus ambientes de trabalho. Apesar dessa tecnologia trazer benefícios, pesquisadores têm associado a presença de patógenos de origem hospitalar em dispositivos de profissionais de saúde, o que torna susceptível a possíveis infecções e o carreamento desses agentes. Como o telefone móvel está em uso rotineiro, sendo visto inclusive em postos de enfermagem, surge à indagação e a necessidade de avaliarmos se há colonização destes aparelhos e quais agentes.   OBJETIVO: Verificar se há colonização de telefones móveis dos profissionais de saúde (enfermagem) e se há alguma rotina de higienização dos celulares e se há adesão a higienização de mãos.   METODOLOGIA: Foi aplicado um questionário aos profissionais de enfermagem de um hospital geral do interior de Minas Gerais, a amostra constou de 50 servidores, após foi coletado Swab umedecidos em caldo BHI (Brain Heart Infusion) dos telefones celulares deste profissionais. Após os swabs foram  inoculados nos meios sólidos Ágar Sangue Colúmbia (Ágar MacConkey (meio seletivo e diferencial para detecção de bacilos entéricos Gram negativos, fermentadores ou não da lactose, em amostras diversas) e Ágar Chocolate (utilizado para o cultivo de microrganismos exigentes, embora cresçam neste meio quase todos os tipos de microrganismos. (meio nutricionalmente rico, não seletivo, geralmente utilizado para o isolamento e cultivo de microrganismos fastidiosos e não fastidiosos de uma variedade de materiais clínicos e não clínicos, possibilitando a verificação da atividade hemolítica das colônias). Foram  incubadas em estufa a 35ºC ± 1ºC por 24 a 48 horas e após realizado a leitura do crescimento microbiológico.   RESULTADOS: Após a coleta do material e análise das mesmas em laboratório, observado que houve crescimento em 33 amostras (66%), dentre os microorganismos identificados nas culturas temos: Staphylococcus aureus, Staphylococcus epidermides, Staphylococcus spp, Staphylococcus lugdunenis, Staphylococcus haemolyticus, Staphylococcus saprophyticus, Bastonetes gram negativos, Bastontes gram positivos e Candida sp.   CONCLUSÃO: Conclui-se que há colonização dos aparelhos celulares, que podem proporcionar a propagação de agentes infecciosos e atuar diretamente na disseminação de microorganismos no ambiente hospitalar.

Abstract Objectives To determine the epidemiological cut-off values (ECOFFs) of norvancomycin for Staphylococcus aureus, Staphylococcus epidermidis, Staphylococcus haemolyticus and Staphylococcus hominis. Methods We collected 1199 clinical isolates of Staphylococcus species from five laboratories located in four cities in China. MICs and inhibitory zone diameters of norvancomycin were determined by broth microdilution and the disc diffusion method, separately. ECOFFs of norvancomycin for four species were calculated by ECOFFinder software following EUCAST principles. Methicillin and vancomycin resistance genes (mecA/mecC and vanA/vanB/vanC/vanD/vanE) were screened for by PCR in all isolates. Pearson correlation and χ2 test were used to calculate the correlation of MICs and inhibition zone diameters, and MICs and resistance genes, respectively. Results MICs of norvancomycin for all strains from five laboratories fell in the range of 0.12–2 mg/L. ECOFFs of norvancomycin were determined to be 2 mg/L for S. epidermidis and S. haemolyticus and 1 mg/L for S. aureus and S. hominis. A weak correlation was observed between MIC values and zone diameters for S. haemolyticus (r = −0.36) and S. hominis (r = −0.26), while no correlation was found for S. epidermidis and S. aureus. The mecA gene was detected in 63.1% of Staphylococcus, whereas no isolate carried mecC, vanA, vanB, vanC, vanD or vanE. ECOFFs of norvancomycin were not correlated with mecA gene carriage in Staphylococcus species. Conclusions ECOFFs of norvancomycin for four Staphylococcus species were determined, which will be helpful to differentiate WT strains. The correlation of MICs and zone diameters of norvancomycin was weak in Staphylococcus species.

Objetivo: Caracterizar os fatores de virulência e o perfil de suscetibilidade antimicrobiana de isolados de Staphylococcus spp. provenientes de amostras de jaleco e estetoscópio dos profissionais de saúde, e das maçanetas das portas da enfermaria e Unidade de Terapia Intensiva do Hospital Universitário Sul Fluminense. Material e Métodos: As amostras de estetoscópio, maçaneta e jaleco, foram coletadas com“swab” estéril umedecido em solução salina e foram colocados em caldo Infusão de Cérebro e Coração Bovino (BHI) e incubados por 24-48h a 37ºC para o crescimento. Foram feitas as identificações necessárias para caracterização dos fatores de virulência e o perfil de suscetibilidade antimicrobiana de isolados de Staphylococcus spp. no laboratório de Microbiologia da Universidade Severino Sombra. Resultados: Foi obtido um total de 59 Staphylococcus spp, identificados como Staphylococcus aureus (n=6) e Staphylococcus coagulase negativos (n=53). Do grupo coagulase-negativo foram identificados 34 isolaods de Staphylococcus epidermidis e 19 de Staphylococcus saprophyticus. Em relação aos fatores de virulência, 76,3% isolados foram produtores de “slime”. A hemólise foi detectada em 50,9% dos isolados, destes 35,6% apresentaram hemólise total e 15,3% a hemólise parcial. O sinergismo hemolítico foi positivo em 59,3% dos isolados sendo que destes, 13 não apresentaram hemolisinas. De acordo com o perfil de suscetibilidade, 96,6% foram resistentes à penicilina. A gentamicina foi o antibiótico que apresentou menor percentual de resistência (15,3%). Conclusão: Os jalecos, estetoscópios e maçanetas das portas, podem ser considerados como um veículo potencial para transmissão de micro-organismo dentro do ambiente hospitalar. Synthetic antimicrobials have a negative impact on food quality and consumer health, which is why natural antimicrobials are urgently needed. Coagulase-negative staphylococci (CoNS) has gained considerable importance for food poisoning and infection in humans and animals, particularly in biofilms. As a result, this study was conducted to control the CoNS isolated from food samples in Egypt. CoNS isolates were selected on the basis of their antibiotic susceptibility profiles and their biofilm-associated behavior. In this context, a total of 29 different bacteriophages were isolated and, in particular, lytic phages (6 isolates) were selected. The host range and physiological parameters of the lytic phages have been studied. Electron microscopy images showed that lytic phages were members of the families Myoviridae (CoNShP-1, CoNShP-3, and CoNSeP-2 isolates) and Siphoviridae (CoNShP-2, CoNSsP-1, and CoNSeP-1 isolates). CoNShP-1, CoNShP-2, and CoNShP-3 were found to be virulent to Staphylococcus haemolyticus, CoNSsP-1 to Staphylococcus saprophyticus and CoNSeP-1 and CoNSeP-2 to Staphylococcus epidermidis. Interestingly, the CoNShP-3 exhibited a typical polyvalent behavior, where not only lysis CoNS, but also other genera include Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Staphylococcus aureus (VRSA), Bacillus cereus and Bacillus subtilis. In addition, CoNShP-3 phage showed high stability at different temperatures and pH levels. Indeed, CoNShP-3 phage showed an antibiofilm effect against Staphylococcus epidermidis CFS79 and Staphylococcus haemolyticus CFS43, respectively, while Staphylococcus saprophyticus CFS28 biofilm was completely removed. Finally, CoNShP-3 phage demonstrated a high preservative efficacy over short and long periods of storage against inoculated CoNS in chicken breast sections. In conclusion, this study highlights the control of CoNS pathogens using a polyvalent lytic phage as a natural antibacterial and antibiofilm agent from a food safety perspective.

The analysis of 16S rRNA gene sequences has been the technique generally used to study and confirm the identification and taxonomy of staphylococci. However, bacterial species cannot always be distinguished from each other using cultural methods. Thus,   clinical samples were collected from 190 cases only 31 positive for staphylococcal infections with Urinary Tract Infection, Wounds, Burns, Otitis media, diarrhea infections, were applied for microbiological analysis which include: cultures on Manitol salt agar and  HiCrome UTI Agar medium all the isolates gave positive golden yellow and identify as Staphylococcus spp. DNA was extracted from Staphylococcus spp and the 16srRNA gene were amplified by using specific primer, then sequencing of nucleic acid of genes was performed by machine is AB13730XL, Applied Biosystem, Macro gen company, the DNA sequencing results of flank sense of 16srRNA gene from 31 strains of Staphylococcus  was confirm the identification into species level: Staphylococcus haemolyticus, Staphylococcus aureus, Staphylococcus epidermidis And   Staphylococcus sciuri. Analysis of the sequences appeared that there two substitution(Transversion, Transition) in the Staphylococcus aureus strains with  Sequence ID LC090540.1 location at Range of nucleotide from 4 to 636, 100% compatibility with NCBI while no substitution appeared in the  Staphylococcus haemolyticus strains which have the sequence ID LN998078.1, 99% compatibility with NCBI also the sequence ID KR812401.1 which related to the strain Staphylococcus sciuri not appeared any substitution after sequencing analysis. Types of substitution detected in partial 16srRNA gene in Staphylococcus epidermidis strains 13  Transversion  and 5 transition substitution location at range of nucleotide from 6 to 1026 have the  Sequence ID KF575160.1 compared with data obtained from Gene Bank, these finding lead to conclusion, our assay allows rapid and confirm the detection to avoid possibility of misidentification of Staphylococcus species based on cultural analysis, the study aimed to  propose the partial sequencing of the gene as an alternative molecular tool for the analysis of Staphylococcus species and for decreasing the possibility of misidentification. New submission of local Iraqi Staphylococcus clinical isolated during the current study show successfully record of four isolate Staphylococcus sciuri, Staphylococcus epidermidis, Staphylococcus aureus and Staphylococcus haemolyticus with GenBank accession number: KY938530.1 ,KY938529.1, ,KY938528.1, and  GenBank: KY938527.1respectivelly.

ABSTRACT In staphylococci, methicillin (meticillin) resistance (MR) is mediated by the acquisition of the mecA gene, which is carried on the size and composition variable staphylococcal cassette chromosome mec (SCCmec). MR has been extensively studied in Staphylococcus aureus, but little is known about MR coagulase-negative staphylococci (MR-CoNS). Here, we describe the diversity of SCCmec structures in MR-CoNS from outpatients living in countries with contrasting environments: Algeria, Mali, Moldova, and Cambodia. Their MR-CoNS nasal carriage rates were 29, 17, 11, and 31%, respectively. Ninety-six MR-CoNS strains, comprising 75 (78%) Staphylococcus epidermidis strains, 19 (20%) Staphylococcus haemolyticus strains, 1 (1%) Staphylococcus hominis strain, and 1 (1%) Staphylococcus cohnii strain, were analyzed. Eighteen different SCCmec types were observed, with 28 identified as type IV (29%), 25 as type V (26%), and 1 as type III (1%). Fifteen strains (44%) were untypeable for their SCCmec. Thirty-four percent of MR-CoNS strains contained multiple ccr copies. Type IV and V SCCmec were preferentially associated with S. epidermidis and S. haemolyticus, respectively. MR-CoNS constitute a widespread and highly diversified MR reservoir in the community.

Latar belakang. Pasien di unit perawatan intensif (UPI) anak (pediatric intensive care unit/PICU) merupakan pasien dengan penyakit kritis sehingga sebagian besar pasien menggunakan antibiotik. Profil resistensi bakteri akan membantu untuk pemilihan terapi empirik yang akan digunakan pada suatu unit.Tujuan. Untuk mendapatkan profil bakteri di PICU RSUD Koja berdasarkan hasil kultur, Metode. Hasil kultur yang dilakukan pada seluruh pasien yang masuk ke PICU pada periode Mei 2015 – April 2016 pada hari kerja pertama dikumpulkan dari departemen patologi klinik RSUD KojaHasil. Didapatkan 486 hasil kultur, 410 di antaranya tidak menunjukkan pertumbuhan kuman. Hasil positif terdapat pada 64 bakteri Gram-positif dan 12 Gram-negatif. Bakteri Gram positif terbanyak ditemukan adalah Staphylococcus hominis (19), Staphylococcus epidermidis (18), Staphylococcus haemolyticus (13), dan Staphylococcus aureus (9). Sementara itu, bakteri Gram negatif terbanyak ditemukan adalah Salmomella typhi (6) dan Acinetobacter baumanii (2). Pada kelompok bakteri Gram positif, antibiotik yang paling sensitif adalah vancomisin (95,2%), gentamisin (68,3%), cotrimoxazole (44,4%), cefotaxime (31,7%), dan ceftriaxone (31,7%). Pada kelompok bakteri Gram negatif, antibiotik yang paling sensitif adalah meropenem (84,6%), cotrimoxazole (84,6%), amikasin (61,5%), gentamisin (53,8%), and cefepime (46,2%).Kesimpulan. Vankomisin merupakan antibiotik yang paling poten untuk digunakan. Cefotaxime dan gentamicin kami pilih untuk digunakan sebagai antibiotik empirik di unit kami, dengan meropenem sebagai lini berikutnya. Vankomisin kami gunakan hanya pada kasus khusus.
