Background : Bacillus species represent a rich source of new bioactive metabolites that can combat diseases. Methods: Bacillus strain was isolated from the marine sponge Spongia officinalis and routinely maintained on marine broth. The bacteria strain was identified as Bacillus 2011SOCCUF3 using 16S rDNA sequencing. The strain was cultured on Tryptone Casein Oat Soluble Starch (TCOATSS) media with continuous agitation for 4 days. The fermented broth was centrifuged, and the supernatant was mixed with 10% (w/v) of adsorbent resin (XAD-7HP and XAD-16N, 1:1) and shaken continuously at a reduced speed for 7 h; and the resin was collected by filtration through sintered glass funnel and washed with MilliQ water, and then eluted with methanol to obtain the extract. The extract was evaporated in vacuo at reduced temperature and pressure to obtain the dry extract. The dry extract was purified by vacuum liquid chromatography, eluting with methanol in acetone gradient. The in vitro antimicrobial and antioxidant activities were investigated using the agar-well diffusion, DPPH scavenging and the phosphomolybdate methods respectively. Results: The extract and fractions showed good antimicrobial activities with minimum inhibitory concentration range of <1.0 mg/mL. The extract and fractions also exhibited good antioxidant activities with their IC50 values been comparable to the standard. Conclusion: Thus, a novel Bacillus strain isolated from the marine sponge (Spongia officinalis) obtained from Cortiou and Riou, France, exhibited promising antimicrobial and antioxidant activities.

Omani (Dhofari) frankincense is world famous for its high quality and fragrance. The oleo-gum resin obtained from the tree of Omani species i.e., Boswellia sacra (local name: luban) is widely used by the natives. The aim of the study was to determine the presence of phytochemicals in chloroform and ethanol extracts of B. sacra resin and to evaluate the in vitro cytotoxic, antimicrobial- and antioxidant-activities. The ethanol and chloroform extracts of Hojari luban were obtained in good yield. The extracts were subjected to phytochemical screenings, quantification of total phenolics (TP) and evaluation of in vitro biological activities viz. antioxidant, antibacterial and cytotoxic potential. The bioactivities of four boswellic acids, the major constituents of the oleo-gum resin, were also predicted with the help of PASS and CLC-pred software. Both the extracts showed similar level of TP (14.67 mg/g and 14.79 mg/g in terms of gallic acid equivalent) but polar fraction was noted to be more active against E. coli. A moderate antioxidant activity was shown by the resin extracts however their cytotoxic activity was observed to be quite remarkable at 1000 μg/l which could be attributed to their high boswellic acid content. Results of in silico PASS studies showed boswellic acids to possess an array of useful biological activities including chemopreventive activity. Further, CLC-pred software revealed that only keto boswellic acid is expected to exert antineoplastic activity against pancreatic carcinoma cell lines, however all the four major boswellic acids are likely to be active against non-tumor embryonic lung fibroblast (MRC5). Objective: To screen the antimicrobial activity Of Callyspongia Diffusa (Marine Sponge) Associated Endophytic Bacterial Strains.Methods: We have isolated endophytic bacterias CDB-1 and CDB-2 from marine sponge Callyspongia diffusa and identified as Pseudomonas taiwanensis strain and Lysinibacillussphaericus strain respectively by the phylogenetic analysis. Fractions of CDB-1 and CDB-2 were screened for in vitro and in vivo antimicrobial activities against pathogenic bacteria and mycobacterium tuberculosis H37 RV strain by Minimum Inhibitory Concentration (MIC) method.Results: The lowest MIC against Kleibesella pnumoniae, Escherichia coli and Enterococcus feacalis was found to be 0.2 µg/ml and 0.4 µg/ml respectively for CDB-2. A significant antifungal activity was observed against Candida albicans (0.2-0.8 µg/ml) and Aspergillus niger (0.2-0.4 µg/ml). Further, Chloroform fraction of CDB-1 and ethyl acetate fraction of CDB-2 have shown significant anti-tubercular activity against the tested organism with MIC of 6.25µg/ml. This was supported by in vivo antimicrobial activity against K. Pneumonia infection in mice and least haemolytic activity against erythrocytes was observed. Compared to chloramphenicol.Conclusion: In this study, we have reported the marine natural species offer a rich source of bioactive metabolites that can exploit to develop novel, useful and potential therapeutic agents.

Humans and plants are experiencing different kinds of diseases. Many of these diseases are mainly caused by microorganisms like fungi, bacteria and viruses that adversely affect their hosts. Bioactive compounds produced by bacteria can be used to treat the infections caused by these microorganisms. Free radicals that can cause serious damage to the cell can be repressed by antioxidants that are produced by bacteria as secondary compounds. The current case study was therefore planned to investigate the antimicrobial and antioxidant potentials of the organic and aqueous extracts of Bacillus clausii KP10 isolated recently by our group from hot springs of Tatta Pani, Poonch, Azad Kashmir. The antimicrobial activities were assessed using well diffusion method against two Gram positive bacteria; B. subtilis, and S. aureus, and two Gram negative bacteria; E. coli and P. multocida and three fungal strains; A. niger, A. parasiticus and A. ustus. Antioxidant activities of the extracts were done by DPPH free radical scavenging assay. In our experiments the maximum zones of inhibition were shown by ethanolic extract against P. multocida (21 mm), S. aureus (13 mm) and methanol soluble fraction showed zone of inhibition against A. ustus (21 mm) as compared to the standard ciprofloxacin for bacteria and as antifungal agents with terbinafine. In our studies n-hexane fraction of B. clausii KP10 showed maximum 88.67% antioxidant activity. Finally, results were analyzed statistically through analysis of variance (ANOVA). Hence it is proved that the bioactive fractions obtained from B. clausii KP10 are good source of antimicrobials and antioxidants and they can be used as medicines in future for the respective treatments.

This is the first report on the antioxidant and antifungal activities of callus cultures from Inula crithmoides L. (Asteraceae). Callus cultures were initiated from leaf sections, on initial culture MS basal medium supplemented with various concentrations of 2,4-D (2,4-dichlorophenoxyacetic acid), NAA (1-naphthaleneacetic acid) and IBA (indole-3-butyric acid) and a 72% survival was achieved. Significant differences between the various auxins used as phytohormones on callus growth were found. Maximum callusing was noticed on the leaf explants grown on MS basal medium supplemented with 1 mgL–1 2,4-D. Subsequently the antioxidant and antimicrobial activities of the methanol extract from calli were investigated. Antioxidant studies suggested that the methanol extracts of dark-grown and light-grown callus were able to reduce the stable free radical 2,2-diphenyl-1-picrilhydrazyl (DPPH). In the inhibition against lipid peroxidation, extracts of dark-grown callus showed the strongest effect with IC50 values better than those of the standards. The methanol extract of callus cultures had significant antifungal activity only against two of the fungi tested: Alternaria solani and Phytophthora cryptogea. Against all the other tested fungi, the I. crithmoides calli extracts showed fungistatic activity.

Dialium angolense is used in Bagira for its various medicinal properties particularly in the management of infectious diseases. In this study, the methanol and aqueous extracts of leaves and fruits were evaluated for their in vitro antioxidant and antimicrobial properties and their in vivo toxicity on Cavia porcellus. The major phytochemical classes of extracts were screened using standard in-tube reactions. The antimicrobial study was tested on Candida albicans, Escherichia coli, Salmonella typhi, Staphylococcus aureus and Streptococcus pneumoniae using agar well diffusion and dilution methods, while the antioxidant activity was evaluated by a DPPH assay. For the acute toxicity study, animals (6/group) were orally given in a single dose 5000, 1000 or 15000 mg of extract/kg body weight (BW) then observed for 14 days. In sub-acute toxicity assays, 150 or 300 mg/kg BW/day were orally given, and animals observed for 28 days. Total phenolics and total flavonoids contents ranged 1.19 to 1.61 mg GAE.g-1 and 0.45 to 1.01 mg QEg-1, respectively. The extracts presented antioxidant activity with IC50 ranging 4.9 to 6.9 µg/mL. The minimal inhibitory concentration (MIC) on tested strains ranged from 1.9 to 500 µg/mL with the aqueous extract of fruits as a most active extract: MIC=1.9 µg/mL on E. coli and C. albicans. No signs of toxicity were noted during the acute and sub-acute toxicity assessments, suggesting a maximal tolerate doses (MDT) and LD50 > 15000 mg/kg BW. This study highlights the antioxidant and antimicrobial activities of Dialium angolense and suggests that further studies be directed towards the isolation of active compounds.

Objective: The objective of this study was undertaken to estimate the total phenolic contents (TPCs), in vitro antioxidant of different solvent extracts of M. oleifera leaves, oral acute toxicity and LD50 determination of the 85% methanolic extract as well as the chromatographic isolation and identification of the extract constituents.Methods: The antioxidant activity of different solvent extracts of Moringa oleifera leaves were estimated using three antioxidant assays and the total phenolic contents (TPCs) were also evaluated using Folin-Ciocalteu’s assay. The n-BuOH extract undergoes further chromatographic isolation owing to the high antioxidant activity using 2, 2'-diphenyl-1-picrylhydrazyl radical (DPPH) method, which resulted in the isolation of seven compounds.Results: The results showed that the TPCs values of the tested extracts were varied from 309.52 to 43.28 mg gallic acid equivalent/g dry extract. The reducing power antioxidant activities (RPAA) were 0.434, 0.402, 0.395, 0.149, 0.143 and 0.124, while the total antioxidant capacity (TAC) values were 316.43, 203.35, 181.56, 86.70, 76.62 and 50.83 mg ascorbic acid equivalent/g dry extract; for n-BuOH, EtOAc, 85% MeOH, H2O, CH2Cl2, and pet. ether extracts, respectively. The oral acute toxicity study of the 85% methanol extracts of M. oleifera and M. peregrina revealed that; their LD50 values were 3458.3 and 4125 mg/kg respectively, thus the two plants could be classified as slightly toxic in the scale of Hodge and Sterner which reflected their nutrient values as edible plants. The isolated compounds were identified on the basis of their 1H and 13C-NMR spectra as; cis-p-coumaric acid 4-O-(2'-O-β-D-apiofuranosyl)-β-D-glucopyranoside (1), chlorogenic acid (2), niazirin (3), 3,4-dihydroxy-β-phenylethoxy-O-α-L-rhamnopyranosyl-(l→2)-α-L-rhamnopyranosyl-(1→3)-4-O-caffeoyl-β-D-glucopyranoside (4), gallic acid (5), taxifolin (6), and benzyl-carbamo-thioethionate (7).Conclusion: The M. oleifera leaves showed promising antioxidant activities and slightly toxic behavior.

Objectives: In vitro biological activities of methanolic extracts of five Turanecio species have been studied. Materials and Methods: The phenolic compositions of the extracts were evaluated by the Folin- Ciocalteu assay and by HPLC analysis. Antioxidant activities were determined with two in vitro assays namely, DPPH (2,2-diphenyl-1-picrylhydrazyl) scavenging assay and phosphomolybdenum assay. The antimicrobial activities of the extracts were evaluated against 13 microorganisms. Results: T. hypochionaeus var. hypochionaeus was showed the highest DPPH inhibition with 88.84% at 100 μg·mL-1. All of the extracts were exerted high total antioxidant activities 128.00- 243.13 mg AAE g-1) and seem to be a promising source of natural antioxidants. The phenolic contents in the extracts varied from 26.17 to 60.99 mg·g-1 as gallic acid equivalent. Chlorogenic acid, caffeic acid and p- coumaric acid were the predominant constituents. The methanolic extracts revealed promising antibacterial activities against most bacteria. No activity was recorded against yeasts tested. Conclusion: The polyphenolic constituents appear to be responsible, at least in part, for the extract’s activities. The present study confirms that tested Turanecio species contains significant source of phenolics have antioxidant and antimicrobial activities and may have therapeutic potential.

Nettle (Urtica dioica L), as a plant rich in biologically active compounds, is one of the most important plants used in herbal medicine. Studies have shown that this plant has antioxidant, antiplatelet, hypoglycemic and hypocholesterolemia effects. In this study, we characterized three Alternaria endophytic fungi isolated from their host U. dioica. We hypothesized that these endophytic fungi can produce new bioactive metabolites, which may possess the bioactive property with potential application in the medical and pharmaceutical industries. The antibacterial activity was evaluated against reference and isolated strains, including Methicillin-Resistant Staphylococcus aureus. A wide range of antimicrobial activities similar to those measured in nettle leaves was detected especially for Alternaria sorghi. Furthermore, the highest antioxidant activity detected with DPPH free radical scavenging was measured for A. sorghi and nettle leaves ethyl acetate extracts. In addition, whereas catalase activity was similar in the three isolated fungi and nettle leaves, total thiol content and superoxide dismutase activity were significantly higher in leaves. A. sorghi showed the best activities compared to other isolated fungi. The characterization and further production of bioactive compounds produced by this endophyte should be investigated to fight bacteria and especially those that develop drug multi-resistance.

The utilization of medicinal plants to treat infectious disease is a common practice in developing countries worldwide. The present study was aimed at evaluating the crude extracts of Ephedra gerardiana (root and stem) with different chemicals for antioxidant and antimicrobial (fungal and bacterial) potential. The results revealed that the ethyl acetate fractions of E. gerardiana (root and stem) have significant free radical scavenging potential with values 2.96±0.39 and 2.73±0.84 while n-butanol and aqueous fractions showed IC502.69±0.26 and 3.44±0.69 µg/ml in stem. Furthermore, crude extract and fractions also revealed promising antibacterial activities against all tested microbial strains while aqueous fraction showed no activities against Bacillus subtilis, Kleibsiella pneumoniae, and Pseudomonas aeruginosa. Interestingly, all crude extracts and fractions were nonactive against fungal strain, Aspergillus niger and Aspergillus flavus, as compare to control. In summary, the Ephedra gerardiana (root and stem) extract and fraction possess antioxidant activities, which might be helpful in preventing or slowing the progress of various oxidative stresses, suggested to be a strong pharmaceutical agent.

The leaves of African pumpkins (Momordica balsamina L.) are a commonly consumed traditional vegetable. They are a good source of polyphenolic antioxidants and carotenoids, which are, however, affected by cooking or digestion. We investigated the effect of household cooking methods (stir-frying or boiling) on the changes in bioactive metabolites, antioxidant capacity, release and accessibility of β-carotene and also inhibition of inhibitory activity against α-amylase and α-glucosidase enzymes during in vitro digestion of African pumpkin leaves compared to the raw leaves. Compared to boiled or raw leaves, stir-frying improved the availability of bioactive metabolites at the gastrointestinal phase. Quercetin 3-galactoside and rhamnetin 3-O-glucoside (marker compounds) discriminated the stir-fried leaves from raw leaves and boiled leaves after digestion. Stir-frying improved the release and accessibility of β-carotene and enhanced the antioxidant activities compared to boiling. Dialysable fractions of stir-fried leaves exhibited the greatest inhibitory activity against α-amylase and α-glucosidase enzymes compared to the raw and boiled leaves, as well as acarbose. Stir-frying, therefore, is recommended for use in household cooking to benefit consumers by increasing the intake of phenolics and β-carotene.
