Although it is widely agreed that the cerebellum is necessary for learning and consolidation of new motor tasks, it is not known whether adaptation to kinematic and dynamic errors is processed by the same cerebellar areas or whether different parts play a decisive role. We investigated arm movements in a visuomotor (VM) rotation and a force field (FF) perturbation task in 14 participants with cerebellar degeneration and 14 age- and gender-matched controls. Magnetic resonance images were used to calculate the volume of cerebellar areas (medial, intermediate, and lateral zones of the anterior and posterior lobes) and to identify cerebellar structure important for the two tasks. Corroborating previous studies, cerebellar participants showed deficits in adaptation to both tasks compared with controls ( P < 0.001). However, it was not possible to draw conclusions from the performance in one task on the performance in the other task because an individual participant could show severe impairment in one task and perform relatively well in the other (ρ = 0.1; P = 0.73). We found that atrophy of distinct cerebellar areas correlated with impairment in different tasks. Whereas atrophy of the intermediate and lateral zone of the anterior lobe correlated with impairment in the FF task (ρ = 0.72, 0.70; P = 0.003, 0.005, respectively), atrophy of the intermediate zone of the posterior lobe correlated with adaptation deficits in the VM task (ρ = 0.64; P = 0.015). Our results suggest that adaptation to the different tasks is processed independently and relies on different cerebellar structures.

A human arm, up to the wrist, is often modelled as a redundant 7 degree-of-freedom serial robot. Despite its inherent nonlinearity, we can perform point-to-point reaching tasks reasonably fast and with reasonable accuracy in the presence of external disturbances and noise. In this work, we take a closer look at the task space error during point-to-point reaching tasks and learning during an external force-field perturbation. From experiments and quantitative data, we confirm a directional dependence of the peak task space error with certain directions showing larger errors than others at the start of a force-field perturbation, and the larger errors are reduced with repeated trials implying learning. The analysis of the experimental data further shows that a) the distribution of the peak error is made more uniform across directions with trials and the error magnitude and distribution approaches the value when no perturbation is applied, b) the redundancy present in the human arm is used more in the direction of the larger error, and c) homogenization of the error distribution is not seen when the reaching task is performed with the non-dominant hand. The results support the hypothesis that not only magnitude of task space error, but the directional dependence is reduced during motor learning and the workspace is homogenized possibly to increase the control efficiency and accuracy in point-to-point reaching tasks. The results also imply that redundancy in the arm is used to homogenize the workspace, and additionally since the bio-mechanically similar dominant and non-dominant arms show different behaviours, the homogenizing is actively done in the central nervous system.

Abstract Systematic perturbations in motor adaptation tasks are primarily countered by learning from sensory-prediction errors, with secondary contributions from other learning processes. Despite the availability of these additional processes, particularly the use of explicit re-aiming to counteract observed target errors, patients with cerebellar degeneration are surprisingly unable to compensate for their sensory-prediction error deficits by spontaneously switching to another learning mechanism. We hypothesized that if the nature of the task was changed—by allowing vision of the hand, which eliminates sensory-prediction errors—patients could be induced to preferentially adopt aiming strategies to solve visuomotor rotations. To test this, we first developed a novel visuomotor rotation paradigm that provides participants with vision of their hand in addition to the cursor, effectively setting the sensory-prediction error signal to zero. We demonstrated in younger healthy control subjects that this promotes a switch to strategic re-aiming based on target errors. We then showed that with vision of the hand, patients with cerebellar degeneration could also switch to an aiming strategy in response to visuomotor rotations, performing similarly to age-matched participants (older controls). Moreover, patients could retrieve their learned aiming solution after vision of the hand was removed (although they could not improve beyond what they retrieved), and retain it for at least 1 year. Both patients and older controls, however, exhibited impaired overall adaptation performance compared to younger healthy controls (age 18–33 years), likely due to age-related reductions in spatial and working memory. Patients also failed to generalize, i.e. they were unable to adopt analogous aiming strategies in response to novel rotations. Hence, there appears to be an inescapable obligatory dependence on sensory-prediction error-based learning—even when this system is impaired in patients with cerebellar disease. The persistence of sensory-prediction error-based learning effectively suppresses a switch to target error-based learning, which perhaps explains the unexpectedly poor performance by patients with cerebellar degeneration in visuomotor adaptation tasks.

ABSTRACTSaccadic eye-movements allow us to bring visual objects of interest to high-acuity central vision. Although saccades cause large displacements of retinal images, our percept of the visual world remains stable. Predictive remapping — the ability of cells in retinotopic brain areas to transiently exhibit spatio-temporal retinotopic shifts beyond the spatial extent of their classical receptive fields — has been proposed as a primary mechanism that mediates this seamless visual percept. Despite the well documented effects of predictive remapping, no study to date has been able to provide a mechanistic account of the neural computations and architecture that actively mediate this ubiquitous phenomenon. Borne out by the spatio-temporal dynamics of peri-saccadic sensitivity to probe stimuli in human subjects, we propose a novel neurobiologically inspired phenomenological model in which the underlying peri-saccadic attentional and oculomotor signals manifest as three temporally overlapping forces that act on retinotopic brain areas. These three forces – a compressive one toward the center of gaze, a convergent one toward the saccade target and a translational one parallel to the saccade trajectory – act in an inverse force field and specify the spatio-temporal window of predictive remapping of population receptive fields.

Rhythmic and discrete arm movements occur ubiquitously in everyday life, and there is a debate as to whether these two classes of movements arise from the same or different underlying neural mechanisms. Here we examine interference in a motor-learning paradigm to test whether rhythmic and discrete movements employ at least partially separate neural representations. Subjects were required to make circular movements of their right hand while they were exposed to a velocity-dependent force field that perturbed the circularity of the movement path. The direction of the force-field perturbation reversed at the end of each block of 20 revolutions. When subjects made only rhythmic or only discrete circular movements, interference was observed when switching between the two opposing force fields. However, when subjects alternated between blocks of rhythmic and discrete movements, such that each was uniquely associated with one of the perturbation directions, interference was significantly reduced. Only in this case did subjects learn to corepresent the two opposing perturbations, suggesting that different neural resources were employed for the two movement types. Our results provide further evidence that rhythmic and discrete movements employ at least partially separate control mechanisms in the motor system.

Previous studies have shown that cerebellar transcranial direct current stimulation (tDCS) leads to faster adaptation of arm reaching movements to visuomotor rotation and force field perturbations in healthy subjects. The first aim of the present study was to confirm a stimulation-dependent effect on motor adaptation. Second, we investigated whether tDCS effects differ depending on onset, that is, before or at the beginning of the adaptation phase. A total of 120 healthy and right-handed subjects (60 women, mean age 23.2 ± SD 2.7 yr, range 18–31 yr) were tested. Subjects moved a cursor with a manipulandum to one of eight targets presented on a vertically orientated screen. Three baseline blocks were followed by one adaptation block and three washout blocks. Sixty subjects did a force field adaptation task (FF), and 60 subjects did a visuomotor adaptation task (VM). Equal numbers of subjects received anodal, cathodal, or sham cerebellar tDCS beginning either in the third baseline block or at the start of the adaptation block. In FF and VM, tDCS and the onset of tDCS did not show a significant effect on motor adaptation (all P values >0.05). We were unable to support previous findings of modulatory cerebellar tDCS effects in reaching adaptation tasks in healthy subjects. Prior to possible application in patients with cerebellar disease, future experiments are needed to determine which tDCS and task parameters lead to robust tDCS effects. NEW & NOTEWORTHY Transcranial direct current stimulation (tDCS) is a promising tool to improve motor learning. We investigated whether cerebellar tDCS improves motor learning in force field and visuomotor tasks in healthy subjects and what influence the onset of stimulation has. We did not find stimulation effects of tDCS or an effect of onset of stimulation. A reevaluation of cerebellar tDCS in healthy subjects and at the end of the clinical potential in cerebellar patients is demanded.

Studies with patients and functional magnetic resonance imaging investigations have demonstrated that the cerebellum plays an essential role in adaptation to visuomotor rotation and force field perturbation. To identify cerebellar structures involved in the two tasks, we studied 19 patients with focal lesions after cerebellar infarction. Focal lesions were manually traced on magnetic resonance images and normalized using a new spatially unbiased template of the cerebellum. In addition, we reanalyzed data from 14 patients with cerebellar degeneration using voxel-based morphometry. We found that adjacent regions with only little overlap in the anterior arm area (lobules IV to VI) are important for adaptation in both tasks. Although adaptation to the force field task lay more anteriorly (lobules IV and V), lobule VI was more important for the visuomotor task. In addition, regions in the posterolateral cerebellum (Crus I and II) contributed to both tasks. No consistent involvement of the posterior arm region (lobule VIII) was found. Independence of the two kinds of adaptation is further supported by findings that performance in one task did not correlate to performance in the other task. Our results show that the anterior arm area of the cerebellum is functionally divided into a more posterior part of lobule VI, extending into lobule V, related to visuomotor adaption, and a more anterior part including lobules IV and V, related to force field adaption. The posterolateral cerebellum may process common aspects of both tasks.

Humans are able to adapt their motor commands to make accurate movements in novel sensorimotor environments, such as when wielding tools that alter limb dynamics. However, it is unclear to what extent sensorimotor representations, obtained through experience with one limb, are available to the opposite, untrained limb and in which form they are available. Here, we compared crosslimb transfer of force-field compensation after participants adapted to a velocity-dependent curl field, oriented either in the sagittal or the transverse plane. Due to the mirror symmetry of the limbs, the force field had identical effects for both limbs in joint and extrinsic coordinates in the sagittal plane but conflicting joint-based effects in the transverse plane. The degree of force-field compensation exhibited by the opposite arm in probe trials immediately after initial learning was significantly greater after sagittal (26 ± 5%) than transverse plane adaptation (9 ± 4%; P < 0.001), irrespective of whether participants learned initially with the left or the right arm or via abrupt or gradual exposure to the force field. Thus transfer was impaired when the orientation of imposed dynamics conflicted in intrinsic coordinates for the two limbs. The data reveal that neural representations of novel dynamics are only partially available to the opposite limb, since transfer is incomplete even when force-field perturbation is spatially compatible for the two limbs, according to both intrinsic and extrinsic coordinates.

The brain processes sensory and motor information in a wide range of coordinate systems, ranging from retinal coordinates in vision to body-centered coordinates in areas that control musculature. Here we focus on the coordinate system used in the motor cortex to guide actions and examine physiological and psychophysical evidence for an allocentric reference frame based on spatial coordinates. When the equations of motion governing reaching dynamics are expressed as spatial vectors, each term is a vector cross product between a limb-segment position and a velocity or acceleration. We extend this computational framework to motor adaptation, in which the cross-product terms form adaptive bases for canceling imposed perturbations. Coefficients of the velocity- and acceleration-dependent cross products are assumed to undergo plastic changes to compensate the force-field or visuomotor perturbations. Consistent with experimental findings, each of the cross products had a distinct reference frame, which predicted how an acquired remapping generalized to untrained location in the workspace. In response to force field or visual rotation, mainly the coefficients of the velocity- or acceleration-dependent cross products adapted, leading to transfer in an intrinsic or extrinsic reference frame, respectively. The model further predicted that remapping of visuomotor rotation should under- or overgeneralize in a distal or proximal workspace. The cross-product bases can explain the distinct patterns of generalization in visuomotor and force-field adaptation in a unified way, showing that kinematic and dynamic motor adaptation need not arise through separate neural substrates.
