SummaryTo evaluate the basis for the hemorrhagic diathesis associated with oral anticoagulant therapy plus the hypolipidemic agent clofibrate, this interaction was studied in 8 normal subjects. Administration of clofibrate 2.0 g/day alone for 14 days had no effect on the platelet count, bleeding time, platelet aggregation, plasma adenosine diphosphatase, platelet release of adenosine diphosphate and adenosine triphosphate, platelet-collagen adhesion, one-stage prothrombin time, or vitamin K-dependent clotting factors (II, VII, IX and X) but significantly reduced platelet adhesiveness and epinephrine-induced platelet aggregation. After addition of large single doses of sodium warfarin, 1.5 mg/kg body weight, to the clofibrate regimen, all values remained within the normal range, including platelet adhesiveness, except epinephrine-induced platelet aggregation. The one-stage prothrombin activity and clotting factors II and X were significantly lower with warfarin plus clofibrate than with warfarin alone, but the plasma level of warfarin was unchanged. Co-administration of sodium warfarin and clofibrate for 21 days augmented the hypoprothrombinemia observed in long-term therapy with warfarin alone but caused no significant change in the plasma warfarin level. It is concluded that the hemorrhagic complications of therapy with warfarin plus clofibrate result primarily from the more rapid decline in the activities of clotting factors II and X and perhaps also from the reduced platelet aggregation.

SummaryCoumarin-induced skin necrosis is believed to be due to a transient hypercoagulable state resulting from a more rapid decline of the protein C activity relative to that of coagulation factors (F) II, IX and X during initiation of oral anticoagulant therapy. We studied hemostatic system activation during early oral anticoagulant treatment with a technique that investigates coagulation activation in the microcirculation.We determined in 10 healthy volunteers the concentrations of prothrombin fragment F1+2 (f1.2) and thrombin-antithrombin complex (TAT) in blood emerging from an injury of the microvasculature (bleeding time incision) before and after initiation of both high-inten- sity and low-intensity coumarin therapy. In addition, f1.2, TAT, activated F VII (F Vila) and the activities of FII, F VII, F X and protein C were measured in venous blood.A rapid decline of F VII and protein C was observed in venous blood with activities at 24 h of 7 ± 1% and 43 ± 2%, respectively, during the high-intensity regimen. A 20 to 30% reduction of f1.2 and TAT was seen in venous blood at 72 h with no major difference between the high- and the low-intensity regimen. F Vila levels were substantially affected by anticoagulation with a >90% reduction at 48 h during the high-intensity regimen. Following high-intensity coumarin, a >50% decrease in the fl.2 and TAT levels was found in shed blood at 48 h suggesting substantial inhibition of thrombin generation during early oral anticoagulation. An increase in the f1.2 and TAT levels was seen neither in shed blood nor in venous blood.Our data do not support the concept of a transient imbalance between generation and inhibition of thrombin as the underlying pathomechanism of coumarin-induced skin nekrosis.

During the first 20 years of a Pinus radiata tree rotation, tree growth and pasture yield were assessed under a range of tree spacings at Invermay and Akatore, two coastal sites in Eastern Otago. Pasture yield in association with trees thinned to 100 stems per hectare (sph) was comparable to that from open pasture up to a tree age of 12 years. By the 19th year, however, pasture production declined to 63% of open pasture yield at Invermay and to 42% at Akatore. At 200 and 400 sph at Akatore, pasture yield was similar to that from open pasture at tree age 12 years but declined to 27% and 0% of open pasture yield respectively by year 20. At both Invermay and Akatore, the ryegrass and clover content of open pasture was relatively constant throughout the term of the trial. However, both the ryegrass and clover content of pasture beneath trees began to decline by tree age 12 years with a very rapid decline at Akatore in the number of pasture species at 200 sph by the 19th year. No pasture remained at 400 sph, after 19 years. Livestock carrying capacity with sheep on tree treatments at Invermay decreased from 100% of open pasture at year 6 to 60% by year 10. At Akatore, livestock carrying capacity averaged over the 20-year life of the trial was 4.1 stock units per hectare with a maximum of 8.1 stock units at a tree age of 8 years. Tree growth at both sites was similar, averaging between 1 and 1.1 m/year in height over 20 years, with trees at Invermay at 100 sph averaging 9% greater height and diameter growth than at Akatore. Increasing tree stocking from 100 to 200 to 400 sph at Akatore, resulted in increased tree height, but decreased diameter at breast height. A comparison of the East Otago trees with those in a similar trial at Tikitere (Rotorua) 900 km further north indicated that the southern trees were about 6 years later in their growth pattern by tree age 20 years. On both sites, soil pH tended to be lower in the presence of trees and was significantly lower than in open pasture by year 20. The results and comparisons with the Tikitere data suggest that, in an integrated agroforestry regime, there will be livestock grazing under the trees further into the tree rotation in Otago than in North Island sites. However, slower tree growth would result in a longer rotation time to harvest. Current recommendations to farmers are to plant trees on the less productive areas of the farm and adopt a tree stocking rate which fully utilises the site. Южно-Уральский государственный аграрный университет Аннотация: В опыте были изучены титры антител против основных инфекционных заболеваний птицы (инфекционный бронхит кур ИБК, болезнь Гамборо, ньюкаслская болезнь) в крови у мясных кур кросса ISA Hubbard F-15, получавших пробиотические кормовые добавки на основе Saccharomyces cerevisiae или Васillus subtilis в количестве 0,50 кг/т комбикорма. Исследования показали, что цыплята суточного возраста уже имеют врожденный иммунитет к широко распространенным инфекционным заболеваниям птицы; титры антител к ИБК и болезни Гамборо до 59-суточного возраста имели тенденцию к снижению, затем повышались и достигали максимума к 151-суточному возрасту, затем плавно снижались. У птицы, получавшей добавки пробиотиков, титр антител к ИБК за продуктивный цикл был выше на 11,2-46,1%, к болезни Гамборо - на 7,2-26,8%, ньюкаслской болезни - на 14,4-27,9% по сравнению с контрольной группой. Установлено также, что периодическое применение пробиотиков в первые 45 сут. выращивания ремонтного молодняка, в периоды разноса кур, пика продуктивности и снижения яйцекладки повышает сохранность поголовья на 0,4-8,2%.
