A filtering analysis of hydraulic head data deduced from slug tests injected in a confined aquifer with different porous media is proposed. Experimental laboratory tests were conducted in a large-scale physical model developed at the University of Calabria. The hydraulic head data were deduced from the records of a pressure sensor arranged in the injection well and subjected to a processing operation to filter the high-frequency noise. The involved smoothing techniques are the Fourier transform and two types of wavelet transform. The performances of the filtered hydraulic heads were examined for different slug volumes and four model layouts in terms of optimal fitting of the Cooper’s analytical solution. The hydraulic head variations in the confined aquifer were analyzed using wavelet transform in order to discover their energy contributions and frequency oscillations. Finally, the raw and smoothed hydraulic heads were adopted to calculate the hydraulic conductivity of the aquifer.

The article is devoted to the consideration of ways to solve one of the actual problems in theory and methodology of training and upbringing — the problem of developing professional skills of future informatics teacher. As a way to adapt students to the profession, the possibility of their involvement in social designing was chosen. Participation in social projects contributes to the approbation and introduction of new forms and methods in teaching informatics. Expanding the experience of future teachers in carrying out large-scale events contributes to the formation of a socially adapted personality competitive in modern society. The potential of a social project in consolidating the knowledge and skills obtained during the theoretical training at the university is indicated. In the article, theoretical reasoning is accompanied by examples of real social projects and activities aimed at the formation of professional competencies of future informatics teachers.

This paper traces the partnership between the University of Toronto and the non-profit Local Food Plus (LFP) to bring local sustainable food to its St. George campus. At its launch, the partnership represented the largest purchase of local sustainable food at a Canadian university, as well as LFP’s first foray into supporting institutional procurement of local sustainable food. LFP was founded in 2005 with a vision to foster sustainable local food economies. To this end, LFP developed a certification system and a marketing program that matched certified farmers and processors to buyers. LFP emphasized large-scale purchases by public institutions. Using information from in-depth semi-structured key informant interviews, this paper argues that the LFP project was a disruptive innovation that posed a challenge to many dimensions of the established food system. The LFP case study reveals structural obstacles to operationalizing a local and sustainable food system. These include a lack of mid-sized infrastructure serving local farmers, the domination of a rebate system of purchasing controlled by an oligopolistic foodservice sector, and embedded government support of export agriculture. This case study is an example of praxis, as the author was the founder of LFP, as well as an academic researcher and analyst.

AbstractInter-area oscillations are serious problems to large-scale power systems. A decentralized H ∞ generator excitation controller of a power system is proposed to damp the inter-area oscillations and to enhance power system stability. The design procedure for a linear composite system is presented in terms of positive semi-definite solutions to modified algebraic inequalities. The resulting controller guarantees closed-loop stability, robustness and an H ∞-norm bound on disturbance attenuation even under uncertainties such as high frequency noise. The control is decentralized in the sense that the control of each generator depends on local information only. The effectiveness of the H ∞ controller is demonstrated through digital simulation studies on a two-machine power system.

It is difficult to reconstruct the unknown information only by some sparse known data in the reconstruction of porous media. Multiple-point geostatistics (MPS) has been proved to be a powerful tool to capture curvilinear structures or complex features in training images. One solution to capture large-scale structures while considering a data template with a reasonably small number of grid nodes is provided by the multiple-grid method. This method consists in scanning a training image using increasingly finer multiple-grid data templates instead of a big and dense data template. The experimental results demonstrate that multiple-grid data templates and MPS are practical in porous media reconstruction.

Abstract Full wave field modeling of wide-aperture data is performed with a pseudospectral implementation of the elastic wave equation. This approach naturally produces three-component stress and two-component particle displacement, velocity, and acceleration seismograms for compressional, shear, and Rayleigh waves. It also has distinct advantages in terms of computational requirements over finite-differencing when data from large-scale structures are to be modeled at high frequencies. The algorithm is applied to iterative two-dimensional modeling of seismograms from a survey performed in 1985 by The University of Texas at El Paso and The University of Texas at Dallas across the Anadarko basin and the Wichita Mountains in southwestern Oklahoma. The results provide an independent look at details of near-surface structure and reflector configurations. Near-surface (<3 km deep) structure and scattering effects account for a large percentage (>70 per cent) of the energy in the observed seismograms. The interpretation of the data is consistent with the results of previous studies of these data, but provides considerably more detail. Overall, the P-wave velocities in the Wichita Uplift are more typical of the middle crust than the upper crust (5.3 to 7.1 km/sec). At the surface, the uplift is either exposed as weathered outcrop (5.0 to 5.3 km/sec) or is overlain with sediments of up to 0.4 km in thickness, ranging in velocity from 2.7 to 3.4 km/sec, generally increasing with depth. The core of the uplift is relatively seismically transparent. A very clear, coherent reflection is observed from the Mountain View fault, which dips at ≈40° to the southwest, to at least 12 km depth. Velocities in the Anadarko Basin are typical of sedimentary basins; there is a general increase from ≈2.7 km/sec at the surface to ≈5.9 km/sec at ≈16 km depth, with discontinuous reflections at depths of ≈8, 10, 12, and 16 km.

Globalization, informatization, digitalization, led to large-scale changes that have problematized the modern process of upbringing. The modern practice of upbringing in Kazakhstan is aimed at solving the problem of forming an intellectual nation. The key figure in the upbringing process is the teacher. The modernization of public consciousness taking place in Kazakhstan, the renewal of both the content of education and the system of upbringing require understanding not only the content, but also the methodology of the professional training of teachers for the upbringing of children, for the organization of the upbringing system in educational organizations. We believe that the analysis of traditional and clarification of modern methodological foundations of professional training of future teachers of Kazakhstan for upbringing work will give us the opportunity to develop a strategy for training future teachers in the conditions of spiritual renewal of Kazakhstan's society. The article reveals the experience of Abai KazNPU. As a result of the conducted research, we came to the conclusion that the process of training a teacher in Kazakhstan, who has a high degree of ethnic, cultural, and religious diversity, requires strengthening the upbringing and socializing components of the educational process of the university. The strategy of professional training of a modern teacher should be a polyparadigmatic concept with the leading role of ideas of personality-oriented, competence paradigm.

AbstractThe homogeneity of the genetically modified single-cells is a necessity for many applications such as cell line development, gene therapy, and tissue engineering and in particular for regenerative medical applications. The lack of tools to effectively isolate and characterize CRISPR/Cas9 engineered cells is considered as a significant bottleneck in these applications. Especially the incompatibility of protein detection technologies to confirm protein expression changes without a preconditional large-scale clonal expansion, creates a gridlock in many applications. To ameliorate the characterization of engineered cells, we propose an improved workflow, including single-cell printing/isolation technology based on fluorescent properties with high yield, a genomic edit screen (surveyor assay), mRNA rtPCR assessing altered gene expression and a versatile protein detection tool called emulsion-coupling to deliver a high-content, unified single-cell workflow. The workflow was exemplified by engineering and functionally validating RANKL knockout immortalized mesenchymal stem cells showing altered bone formation capacity of these cells. The resulting workflow is economical, without the requirement of large-scale clonal expansions of the cells with overall cloning efficiency above 30% of CRISPR/Cas9 edited cells. Nevertheless, as the single-cell clones are comprehensively characterized at an early, highly parallel phase of the development of cells including DNA, RNA, and protein levels, the workflow delivers a higher number of successfully edited cells for further characterization, lowering the chance of late failures in the development process.Author summaryI completed my undergraduate degree in biochemistry at the University of Ulm and finished my master's degree in pharmaceutical biotechnology at the University of Ulm and University of applied science of Biberach with a focus on biotechnology, toxicology and molecular biology. For my master thesis, I went to the University of Freiburg to the department of microsystems engineering, where I developed a novel workflow for cell line development. I stayed at the institute for my doctorate, but changed my scientific focus to the development of the emulsion coupling technology, which is a powerful tool for the quantitative and highly parallel measurement of protein and protein interactions. I am generally interested in being involved in the development of innovative molecular biological methods that can be used to gain new insights about biological issues. I am particularly curious to unravel the complex and often poorly understood protein interaction pathways that are the cornerstone of understanding cellular functionality and are a fundamental necessity to describe life mechanistically.

Eight in-service teachers, one pre-service education student, three observers from other universities, and two instructors from the University of Nebraska-Lincoln engaged in an inquiry-based geology field course from June 13 to 28, 2015 through Wyoming, South Dakota, and Nebraska. This commnity of learners spent three days working in the Grand Teton National Park area. Geological features and history present in Grand Teton National Park are an important part of the course curriculum. Large-scale extensional features of the Teton Range and Jackson Hole, and the glacial geomorphology and related climate changes of this area are some of the unique features examined here.
