Workman and Langmore have recently proposed a procedure for isolating particular chromatin fragments. The method requires restriction endonuclease cutting of the chromatin and a probe, their digestion with two exonucleases which leave complimentary single strand termini and low temperature hybridization of these. We here report simple electron microscopic monitoring of the four reactions involved.Our test material was ϕX-174 RF DNA which is cut once by restriction endonuclease Xho I. The conversion of circles to linear molecules was followed in Kleinschmidt spreads. Plate I shows a circular and a linear DNA molecule. The rate of cutting is shown in Figure 1.After completion of the endonuclease cutting, one portion of the DNA was treated with exonuclease III, an enzyme known to digest the 3' terminals of double helical DNA. Aliquots when examined in the electron microscope reveal a decreasing length of double helix and increasing bushes at the ends.

The electropolymerization of 2,5-di(2-(thienyl)pyrrole) (SNS) on a Pt electrode from ethanolic solution with LiClO4 or LiCl as electrolyte has been studied by cyclic voltammetry (CV) and chronoamperometry (CA). In both media, a quasi-reversible process has been indicated by CV, reversing the scan at low oxidation potentials. Under these conditions, reducible positive charges formed in both oxidized polymers are compensated by the entrance of anions from solution. Elemental analysis reveals that polymers generated at a low oxidation potential by CA contain a 21.03% (w/w) of ClO4- or a 9.56% (w/w) of Cl-. The poly(SNS) doped with Cl- presents higher proportion of reducible positive charges, higher polymerization charge and lower productivity. A much higher electrical conductivity, however, has been found for the poly(SNS) doped with ClO4-. Both polymers are soluble in DMSO, acetone and methanol. The dimer, trimer, tetramer and pentamer have been detected as soluble and neutral linear oligomers by mass spectrometry-fast atom bombardment. The analysis of polymers by infrared spectroscopy confirms the predominant formation of linear molecules with α-α linkages between monomeric units. A condensation mechanism involving one-electron oxidation of all electrogenerated linear and neutral polymeric chains is proposed to explain the SNS electropolymerization.

The correlation function of the tensor polarization relevant for the depolarized Rayleigh line of a gas of rotating linear molecules is calculated for the pressure broadening regime. Point of depar­ture is the Waldmann-Snider equation for the distribution function of the gas. Due to the collisional coupling between the tensor polarization and other moments of the distribution function the cor­relation function turns out to be a sum of exponential functions. Consequently the depolarized Rayleigh line has a non-Lorentzian shape.

Modification of HAM/3 excitation energies for ΠΠ* transitions of linear molecules

The excitation energies calculated by the HAM/3 procedure for ΠΠ* transitions in linear molecules can be internally inconsistent by as much as ± 0.6 eV. In the recent study by Åsbrink etal., the problem was avoided by adopting Recknagel's expressions and requiring the proper average ΠΠ* excitation energy. In this paper, we trace the small inconsistency back to its origin in HAM/3 theory and derive the analytical expression for the energy correction as well as Recknagel's formulas. Numerical examples studied include all seven linear molecules investigated by Åsbrink etal. The explicit expression for the correction enables us to perform meaningful configuration-interaction calculations on the excited states, as illustrated by the carbon suboxide molecule.
