This study is the first report about isolation, purification and optimization of lipase from Bacillus glycinifermentans. In this study, Bacillus glycinifermentansMK-840989 was isolated from a local petrol pump. The bacterium showed lipolytic zones of 0.19cm, 0.044cm, and 0.28cm on peptone yeast agar, olive oil hydrolysis agar and chromogenic plate agar, respectively. B. glycinifermentans also produced an extracellular lipase (55.1µmol/ml). This bacterium preferred acidic environment (pH 5) for growing optimally at 80˚C when the medium was supplemented with 1% olive oil. The olive oil induced its growth up to 9h. The protein content of the purified lipase was estimated about 75mg/ml as compared to its crude form, i.e. 350mg/ml. The purified lipase was found to be thermostable acidic in nature as its optimum activity was observed at 90˚C (0.08U/ml) and pH 5 (0.02U/ml). Other optimization factors included 1% olive oil (0.065U/ml), 0.1mM maltose (0.023U/ml), 0.1mM Ca (0.025U/ml), 1% yeast extract (16.8U/ml), 1% wheat waste (0.019U/ml), 1% commercial detergent (0.016U/ml) and 1% tween-20 (0.015 U/ml). The purified lipase showed a polypeptide of 26.7kDa on SDS-PAGE. These features such as thermostability, acidic nature, ability to show activity in wheat waste and tolerance to detergents render the lipase of B.glycinifermentans MK-840989 as an attractive choice for biotechnologists to employ it at industrial level. The purified lipase of B.glycinifermentans MK-840989 can be a potential candidate for detergent and oil-remediation industry. It can help to replace conventional synthetic detergent as it is cost-effective and eco-friendly.

An ongoing challenge in olive oil analytics is the development of a reliable procedure that can draw the consensus of all interested parties regarding the quantification of concentrations above the required minimum value of 5 mg of bioactive “olive oil polyphenols” per 20 g of the oil, to fulfill the health claim introduced by the European Commission (EC) Regulation 432/2012. An in-house validated ultra-high performance liquid chromatography (UHPLC) protocol fit for this purpose is proposed. It relies on quantification of the total hydroxytyrsol (Htyr) and tyrosol (Tyr) content in the virgin olive oil (VOO) polar fraction (PF) before and after acidic hydrolysis of their bound forms. PF extraction and hydrolysis conditions were as previously reported. The chromatographic run lasts ~1/3 of the time needed under high performance liquid chromatography (HPLC) conditions, this was also examined. Eluent consumption for the same piece of information was 6-fold less. Apart from being cost effective, a larger number of samples can be analyzed daily with less environmental impact. Two external curves, detection at 280 nm and correction factors for molecular weight difference are proposed. The method, which is fit for purpose, is selective, robust with satisfactory precision (percentage relative standard deviation (%RSD) values < 11%) and recoveries higher than 87.6% for the target analytes (Htyr, Tyr). Standard operational procedures are easy to apply in the olive oil sector.

The aim of the research was to verify the applicability of microsatellite (SSR) markers in High Resolution Melting (HRM) analysis for the identification of the olive cultivars used in the “Terra di Bari” PDO extra virgin olive oil. A panel of nine cultivars, widespread in Apulia region, was tested with seventeen SSR primer pairs and the PCR products were at first analysed with a Genetic Analyzer automatic sequencer. An identification key was obtained for the nine cultivars, which showed an unambiguous discrimination among the varieties constituting the “Terra di Bari” PDO extra virgin olive oil: Cima di Bitonto, Coratina, and Ogliarola. Subsequently, an SSR based method was set up with the DCA18 marker, coupled with HRM analysis for the distinction of the Terra di Bari olive oil from non-Terra di Bari olive oil using different mixtures. Thus, this analysis enabled the distinction and identification of the PDO mixtures. Hence, this assay provided a flexible, cost-effective, and closed-tube microsatellite genotyping method, well suited to varietal identification and authentication analysis in olive oil.

This study presents a cost-effective analysis by comparing the costs of measures (options) to improve the quality of bathing waters in the Gulf of Kalloni (island of Lesvos, Greece) in order to reduce the anthropogenic eutrophication in the coastal water of the Kalloni Gulf.The Gulf of Kalloni is a semi-enclosed gulf (115 km2) which receives municipal wastewater, agricultural activity drainage, and at times sewage from olive oil plants processing the local olives harvest. The area of study consists of the coastal waters and the river basin of which water run-off drains into the gulf. Four options are comparable in their environmental effectiveness to reduce eurtophication damages which are: municipal wastewater treatment plant, construction of dams, organic farming plus training and olive oil wastewater treatment plant.

The female reproductive tract may be the site of a wide variety of benign and malignant tumors, as well as non-neoplastic tumor-like conditions, most of which can be diagnosed by light microscopic examination including special stains and more recently immunoperoxidase techniques. Nevertheless there are situations where ultrastructural examination can contribute substantially to an accurate and specific diagnosis. It is my opinion that electron microscopy can be of greatest benefit and is most cost effective when applied in conjunction with other methodologies. Thus, I have developed an approach which has proved useful for me and may have benefit for others. In cases where it is deemed of potential value, glutaraldehyde-fixed material is obtained at the time of frozen section or otherwise at operation. Coordination with the gynecologic oncologist is required in the latter situation. This material is processed and blocked and is available if a future need arises.
