The aim of this study is to design, develop and evaluate new biohybrid sponges based on polymers (collagen and polyvinyl alcohol) with and without indomethacin as anti-inflammatory drug model to be used for tissue regeneration in wound healing. Type I fibrillar collagen in the form of a gel and different concentrations of polyvinyl alcohol were mixed together to prepare composite gels. Both control samples, without indomethacin and with indomethacin, were obtained. All samples were crosslinked with glutaraldehyde. By freeze-drying of hydrogels, the spongious forms (matrices) were obtained. The matrices were characterized by FT-IR spectroscopy, scanning electron microscopy (SEM), water absorption, enzymatic degradation and in vitro indomethacin release. The pharmacological effect of the spongious biohybrid matrices was determined on an experimental model of burns induced to Wistar rats. The SEM images showed a porous structure with interconnected pores. Collagen sponges present a structure with pore sizes between 20 and 200 µm, which became more and more compact with polyvinyl alcohol addition. The FT-IR showed interactions between collagen and polyvinyl alcohol. The enzymatic degradation indicated that the most stable matrix is the one with the ratio 75:25 of collagen:polyvinyl alcohol (ACI75), the other ones being degradable in time. The kinetic data of indomethacin release from matrices were fitted with different kinetic models and highlighted a biphasic release of the drug. Such kinetic profiles are targeted in skin wound healing for which important aspects are impaired inflammation and local pain. The treatment with sponges associated with anti-inflammatory drug had beneficial effects on the healing process in experimentally induced burns compared to the corresponding matrices without indomethacin and the classical treated control group.

AbstractThe purpose of this study was to evaluate whether cryopreserved (frozen) adipose-derived regenerative cells (ADRCs) have a therapeutic effect on burn wound healing as well as freshly isolated (fresh) ADRCs.Full thickness burns were created on dorsum of nude mice and burn wound was excised. The wound was covered by artificial dermis with; (i) fresh ADRCs, (ii) frozen ADRCs, and (iii) PBS (control). The assessment for wound healing was performed by morphological, histopathological and immunohistochemical analyses.In vivo analyses exhibited the significant therapeutic effect of frozen ADRCs on burn wound healing up to the similar or higher level of fresh ADRCs. There were significant differences of wound closure, epithelized tissue thickness, and neovascularization between the treatment groups and control group. Although there was no significant difference of therapeutic efficacy between fresh ADRC group and frozen ADRC group, frozen ADRCs improved burn wound healing process in dermal regeneration with increased great type I collagen synthesis compared with fresh ADRCs.These findings indicate that frozen ADRCs allow us to apply not only quickly but also for multiple times, and the cryopreserved ADRCs could therefore be useful for the treatment of burn wounds in clinical settings.

Inflamasi merupakan respon alami tubuh terhadap adanya kerusakan jaringan. Salah satu medikamen untuk mengatasi inflamasi adalah antiinflamasi non steroid (AINS). Penggunaan AINS mempunyai beberapa efek samping dan dalam beberapa hal penggunaan tanaman obat dinilai lebih aman. Rosela merupakan salah satu tanaman obat yang mempunyai potensi sebagai antiinflamasi. Penelitian ini bertujuan untuk mengetahui efek pemberian sistemik ekstrak etanolik rosela terhadap ekspresi COX-2 dan jumlah neutrofil fase inflamasi pada proses penyembuhan luka. Bunga rosela didapatkan dari perkebunan di Dusun Bulusari Desa Pojok Kecamatan Tarokan Kabupaten Kediri Jawa Timur. Pembuatan ekstrak rosela dilakukan di LPPT unit I UGM Yogjakarta dengan cara perkolasi. Tikus putih galur Wistar sebanyak 36 ekor diberi perlukaan dengan punch biopsi ɵ 3 mm pada mukosa bukal. Subjek dibagi menjadi 3 kelompok, masing-masing kelompok 12 ekor tikus. Pembagian kelompok terdiri dari kontrol negatif (saline), kontrol positif (ibuprofen 20 mg/kg BB) dan perlakuan (ekstrak rosela 500 mg/kg BB). Pemberian minum sesuai kelompoknya sehari sekali selama 4 hari. Pada hari ke-1, ke-2, ke-3 dan ke-4 tikus dikorbankan lalu jaringan mukosa yang mengalami perlukaan dibuat preparat histologis. Pewarnaan Hematoksilin Eosin (HE) dilakukan untuk mengamati jumlah neutrofil. Ekspresi COX-2 diamati pada preparat dengan pewarnaan imunohistokimia menggunakan rabbit polyclonal antibody COX-2 (Lab Vision, USA). Jumlah neutrofil dan ekspresi COX-2 dihitung di bawah mikroskop cahaya lalu data dianalisi menggunakan ANAVA dan LSD. Hasil penelitian menunjukkan bahwa ekspresi COX-2 dan jumlah neutrofil lebih rendah pada kelompok perlakuan dibanding kontrol. Pengamatan klinis pada hari ke-4 juga tampak luka seluruh subjek telah menutup sempurna setelah pemberian minum rosela. Disimpulkan bahwa ekstrak etanolik rosela mempunyai kemampuan menghambat ekspresi COX-2 dan menurunkan jumlah neutrofil sehingga dapat digunakan sebagai bahan anti-inflamasi. Expression of COX-2 and The Number of Neutrophil in Inflammation stage of Wound Healing Process after Systemic Administration of Ethanolic Extract Rosela. Inflammation is an initial stage of body’s natural response to tissue damage.The use  empirically plants often used for traditional medicine because it is easily found in the community and fewer side effects. Flavanoid presence of roselle (Hibiscus sabdariffa) is thought to have anti inflammatory effects. This study aimed to know the effect of systemic administration of Roselle ethanolic extract toward COX-2 expression and neutrophils number in the inflammatory phase of wound healing processes. Roselle was obtained from plantations in Bulusari hamlet, Tarokan, Kediri, EastJava. Making roselle extract was performed in LPPT unit 1 UGM Yogyakarta by percolation ways. Wistar rats were given a total of 36 injuries with ɵ 3 mm punch biopsy of the buccal mucosa. Subjects were divided into three groups, each group of 12 rats. The division consists of the negative control group (saline), positive control (ibuprofen 20 mg/kg) and treatment (roselle extract 500 mg/kg). Giving drink suitable group once daily for four days. On day 1, the 2nd, 3rd and fourth rats were sacrificed, and mucosal tissue injury was made histological preparat. Hematoxylin eosin staining (HE) was performed to observe the number of neutrophils. COX-2 expression was found in preparations for immunohistochemical staining using rabbit polyclonal COX-2 antibody (Lab Vision, USA). The number of neutrophils and expression of COX-2 is calculated under a light microscope data were analyzed using Two-way ANOVA and LSD. The results showed that the expression of COX-2 and neutrophil number were least in the treatment group compared to the control. Clinical observation on day four also appears around the wound has completely closed the subject after administration of roselle drink. It was concluded that the ethanolic extract of roselle can inhibit COX-2 expression and decrease the number of neutrophils that can be used as an anti-inflammatory ingredient.

 Abstract: One phase of wound healing is the inflammatory phase. Haruan fish extract has shown to accelerate the healing process up because it has anti-inflammatory effects. Toman fish originates from the same genus as the Haruan fish, but its anti-inflammatory effect remains unknown. It was done to analyzed anti-inflammatory effects of Toman fish extract. This study was true experimental design with posttest-only control design. Twenty-seven male mice of Balb-C strain were divided into 3 groups. Incision wound of 1 cm was made along the back of the mice. Nine mice in each group were sacrificed on 3rd days, then histopathology examination was conducted with Haematoxylin eosin. There were significant differences between treatment group given Toman fish extract and positive control group given Haruan fish extract, and also compared to the negative control group in PMN cell examination. There were significant differences between treatment group given Toman fish extract and positive control group given Haruan fish extract compared to negative controls on the number of macrophage cells. Toman fish extract could lower the number of PMN cells and was able to increase the number of macrophages on the 3rd days. Background: Wound healing requires processes like cell migration, proliferation of extracellular matrix deposition, remodeling and angiogenesis. Also, anti-oxidative and anti-inflammatory reactions play key roles in the progression of wound healing. Previous studies showed that sesame oil has anti-inflammatory, antimicrobial and antioxidant activities. In addition, it is shown that sesame oil stimulates fibroblast production in vitro and in skin wounds. Therefore, in this study we investigated the effects of sesame ointment on the process of second-degree burn wound healing in rat models according to stereological parameters. Materials and Methods: In this experimental study, forty male Wistar rats (200±20 g) were randomly divided into four groups (n=10): ointment-base treated group (vehicle), silver sulfadiazine (SDD) treated group, sesame treated group (E1) and the control group which received no treatment. A 2×3 cm2 standard second-degree burn wound was induced on the posterior surface of animal’s neck under general anesthesia. After sacrifice, the animal’s skin sample was fixed in buffered formaldehyde for stereological evaluations. The data was analyzed by SPSS statistical software (version 14.0). P<0.05 was considered statistically significant. Results: According to our results, the mean of reduction in wound areas, volume density of collagen bundles and hair folicules, fibroblast populations, length density of vessels in E1 group was significantly higher than control group (P<0.05). The differences between E1 and SSD were not statistically noticeable regarding the stereological parameters. Conclusion: According to stereological analysis, administration of sesame ointment showed the ability to improve wound healing process and tissue regeneration in the treatment of skin damages.[GMJ.2016;5(2):56-62]

AbstractEndoscopic mucosal resection (EMR) and endoscopic submucosal dissection (ESD) are well-established therapeutics for gastrointestinal neoplasias, but complications after EMR/ESD, including bleeding and perforation, result in additional treatment morbidity and even threaten the lives of patients. Thus, designing biomaterials to treat gastric bleeding and wound healing after endoscopic treatment is highly desired and remains a challenge. Herein, a series of injectable pH-responsive self-healing adhesive hydrogels based on acryloyl-6-aminocaproic acid (AA) and AA-g-N-hydroxysuccinimide (AA-NHS) were developed, and their great potential as endoscopic sprayable bioadhesive materials to efficiently stop hemorrhage and promote the wound healing process was further demonstrated in a swine gastric hemorrhage/wound model. The hydrogels showed a suitable gelation time, an autonomous and efficient self-healing capacity, hemostatic properties, and good biocompatibility. With the introduction of AA-NHS as a micro-cross-linker, the hydrogels exhibited enhanced adhesive strength. A swine gastric hemorrhage in vivo model demonstrated that the hydrogels showed good hemostatic performance by stopping acute arterial bleeding and preventing delayed bleeding. A gastric wound model indicated that the hydrogels showed excellent treatment effects with significantly enhanced wound healing with type I collagen deposition, α-SMA expression, and blood vessel formation. These injectable self-healing adhesive hydrogels exhibited great potential to treat gastric wounds after endoscopic treatment.

Skin wound healing is a complex and dynamic process that aims to restore lesioned tissues. Collagen-based skin substitutes are a promising treatment to promote wound healing by mimicking the native skin structure. Recently, collagen from marine organisms has gained interest as a source for producing biomaterials for skin regenerative strategies. This preliminary study aimed to describe the application of a collagen-based skin-like scaffold (CBSS), manufactured with collagen extracted from sea urchin food waste, to treat experimental skin wounds in a large animal. The wound-healing process was assessed over different time points by the means of clinical, histopathological, and molecular analysis. The CBSS treatment improved wound re-epithelialization along with cell proliferation, gene expression of growth factors (VEGF-A), and development of skin adnexa throughout the healing process. Furthermore, it regulated the gene expression of collagen type I and III, thus enhancing the maturation of the granulation tissue into a mature dermis without any signs of scarring as observed in untreated wounds. The observed results (reduced inflammation, better re-epithelialization, proper development of mature dermis and skin adnexa) suggest that sea urchin-derived CBSS is a promising biomaterial for skin wound healing in a “blue biotechnologies” perspective for animals of Veterinary interest.

Abstract Introduction Burn is one of the most severe traumas that causes coagulative destruction of the skin. The use of various products that accelerate wound healing in patients with burn may affect the patient’s survival and reduce the complications that may be seen. In the present study we aimed effects of subcutaneous ozone injection on second degree burn wound. Methods A total of 72 Sprague-Dawley male rats included in the study were divided randomly into three groups (control group (CG), silver sulfadiazine group (SG), ozone group (OG)) and each group was divided randomly two subgroups (as sacrificed on d7 and on d14).A deep second degree scald burns were created on the lower back. In CG subcutaneous 0.9% serum saline was injected daily into the burn area. In SG, burns were dressed with silver sulfadiazine daily and in OG subcutaneous ozone was injected daily into the burn area. Tissue hydroxyproline level measurement and histopathological evaluation were done. Results When the groups were compared in terms of weight change, no significant difference was found on the 7th and 14th days. In the evaluation made in terms of tissue hydroxyproline, tissue hydroxyproline level in OG was found to be significantly higher on both the 7th and 14th days (p &lt; 0.001). In histopathological evaluations, it was determined that wound healing in OG was significantly higher than in the other groups. Conclusions According to the results, subcutaneous ozone therapy is more effective than silver sulphadiazine in the healing process of second-degree burn wounds and it can be safely used in the treatment of burn wounds.

<p>Two oil blends (sunflower/canola oils 85/15 (BL1) and canola/linseed oils 70/30 (BL2)), were prepared and enzymatically interesterified to be applied to surgically-induced wounds in rats. Following surgery, the animals were submitted to the Treatment with Physiological Saline (TPS) (control group), Blends (TBL), and Structured Lipids (TSL). The control group (TPS) received physiological saline solution for 15 days. In TBL, BL1 was administered during the inflammation phase (days 0-3) and BL2 in the tissue formation and remodeling phase (days 4-15). In TSL, Structured Lipid 1 (SL1) and Structured Lipid 2 (SL2) were used instead of BL1 and BL2, respectively. The aim of this study was to compare wound closure evolution among rats treated with the blends or structured lipids versus control rats treated with physiological saline. The wound healing process was evaluated by measuring the wound areas along the treatments and the concentrations of cytokines. An increase in the areas of wounds treated with the blends and structured lipids in the inflammatory phase was observed, followed by a steeper closure curve compared to wounds treated with physiological saline. The changes observed during the inflammatory phase suggest a potential therapeutic application in cutaneous wound healing which should be further investigated.</p>

ABSTRACT Background: Manipulating intestinal microbiota with probiotics might stimulate skin response. Understanding all stages of the healing process, as well as the gut-skin-healing response can improve the skin healing process. Aim: To evaluate the effect of perioperative oral administration of probiotics on the healing of skin wounds in rats. Methods: Seventy-two Wistar male adult rats were weighed and divided into two groups with 36 each, one control group (supplemented with oral maltodextrin 250 mg/day) and one probiotic group (supplemented with Lactobacillus paracasei LPC-37, Bifidobacterium lactis HN0019, Lactobacillus rhamnosus HN001, Lactobacillus acidophilus NCFM® at a dose of 250 mg/day), both given orally daily for 15 days. The two groups were subsequently divided into three subgroups according to the moment of euthanasia: in the 3rd, 7th and 10th postoperative days. Results: There were no significant changes in weight in both groups. Wound contraction was faster in probiotic group when compared to the controls, resulting in smaller wound area in the 7th postoperative day. As for histological aspects, the overall H&E score was lower in the probiotic group. The probiotic group showed increased fibrosis from 3rd to the 7th postoperative day. The type I collagen production was higher in the probiotic group at the 10th postoperative day, and the type III collagen increased in the 7th. Conclusion: The perioperative use of orally administrated probiotic was associated with a faster reduction of the wound area in rats probably by reducing the inflammatory phase, accelerating the fibrosis process and the deposition of collagen.

Abstract: Efectiveness Gel Extract Of White On The Process Of Healing Inflamation Phase Heating. The inflammatory phase is a favorable body response as a protection mechanism. In the process of wound healing becomes a very important phase. Management of inflammation that is often used Non-Steroid Anti-Inflammatory class of salicylates on the skin that have side effects. The content of allicin in garlic can be used for problems that begin with the inflammatory phase. The use of gel from garlic extract is also easier to use and easier to clean. The study aim to determine the effectiveness of garlic extract gel to process wound inflammatory phase healing. This research is an experimental research with pre and post test with control group method with 24 samples. Conducted injury to the back area of rat length of wound 1 cm, depth to dermis. Conducted wound care, given gel extract of garlic concentration of 20%, 40%, 80% of the control using 0.9% NaCl compress. Using Kruskal Wallis test and Anova oneway showed concentration of 20%, 40%, and 80% of sig <0,05 ie 0.00. It was concluded that 20%, 40%, 80% garlic extract gel was effective against inflammatory wound healing process. The use of garlic extract gel is more effective in the wound inflammatory wound healing process.Abstrak: Efektivitas Gel Ekstrak Bawang Putih  terhadap Proses Penyembuhan Luka Fase Inflamasi.  Fase inflamasi merupakan respon tubuh yang menguntungkan sebagai mekanisme perlindungan. Pada proses penyembuhan luka menjadi fase yang sangat penting. Penatalaksanaan inflamasi yang sering digunakan Anti-Inflamasi Non Steroid golongan salisilat pada kulit yang memiliki efek samping. Kandungan zat allicin pada bawang putih dapat dimanfaatkan untuk masalah yang diawali dengan fase inflamasi. Pemanfaatan gel dari ekstrak bawang putih pun dalam penggunaannya lebih mudah diabsorsi dan mudah dibersihkan. Penelitian ini bertujuan untuk mengetahui efektifitas gel ekstrak bawang putih terhadap proses penyembuhan luka fase inflamasi. Penelitian ini merupakan penelitian eksperiment dengan metode pre and post test with control grup dengan jumlah sampel sebanyak 24 ekor tikus. Dilakukan perlukaan pada daerah punggung tikus panjang luka 1 cm, kedalaman sampai dermis. Dilakukan perawatan luka, diberi gel ekstrak bawang putih konsentrasi berbeda yaitu 20%, 40%, 80%  kontrol menggunakan kompres NaCl 0,9%. Hasil uji Kruskal Wallis dan Anova oneway menunjukkan konsentrasi 20%, 40%, dan 80%  nilai sig <0,05 yaitu 0,00. Disimpulkan bahwa 20%, 40%, 80% gel ekstrak bawang putih efektif terhadap proses penyembuhan luka inflamasi. Penggunaan gel ekstrak bawang putih lebih efektif dalam proses penyembuhan luka inflamasi luka.   Disimpulkan bahwa 20%, 40%, 80% gel ekstrak bawang putih efektif terhadap proses penyembuhan luka inflamasi. Penggunaan gel ekstrak bawang putih lebih efektif dalam proses penyembuhan luka inflamasi luka.
