Plant origin, processing, and domestic preparation may affect the postprandial glycemic response (PGR) of starchy foods. The objective of this study was to examine the possibility of integrating domestically cooked non-cereal starchy foods commonly consumed in Northeast Asia into glycemic management diet, and compare their glycemic characteristics with those of waxy and non-waxy whole grains and starchy beans. In a randomized crossover trial, ten healthy subjects consumed dried lily bulb (LB), lotus seed (LS), adlay (AD), waxy black rice (BR), millet (MI), and adzuki bean (AB), pre-soaked and each cooked for two time durations. Acute PGR tests and in vitro carbohydrate digestion were carried out for each test food. Both the LS and AB meals achieved low glycemic index (GI 21–51), while the other starchy foods failed to show significant difference with rice (GI 83–109). The hydrolysis indexes of LS and AB were 37.7%–61.1%, significantly lower than other test foods. The in vitro tests indicated that pre-soaking resulted in high rapidly digestible starch (RDS) and low resistant starch (RS). Careful choice of whole grain materials, minimized pre-soaking, and moderate cooking may be critical factors for successful postprandial glycemic management for diabetic and pre-diabetic.

AbstractThe addition of vegetable to carbohydrate-based meals was shown to contribute to glycaemic management. The aim of this study was to investigate the impact of homogenisation on vegetables added to rice meals in terms of acute glycaemic responses (GR). In a randomised crossover trial, sixteen healthy volunteers completed thirteen test sessions, which included two sessions for glucose control, two for rice and nine for different vegetable-rice mixed meals: cooked pak choi and cooked rice (CP+R); cooked cauliflower and cooked rice (CC+R); cooked eggplant and cooked rice (CE+R); and their homogenised counterparts, both raw or cooked. Postprandial GR tests, in vitro carbohydrate digestion and chemical analyses were carried out for each test meal. Compared with pure rice, CE+R, CP+R and CC+R meals achieved significantly lower glycaemic indexes (GI) of 67, 71 and 73, whereas their homogenised counterparts failed to show significant difference with rice. The hydrolysis indexes (HI) of CE+R, CP+R and CC+R were 69·6, 83·8 and 80·6 % of the HI of the rice control. CE had the greatest effect on lowering the GI, the incremental area under the blood glucose curve from 0 to 120 min, the peak glucose value, the maximum amplitude of glucose excursion in 0–120 min (MAGE0–120), the HI and rapid available starch. Both in vitro and in vivo tests demonstrated that incorporating non-homogenised cooked vegetables into a rice meal could slow the carbohydrate digestion and improve postprandial GR. Texture properties of vegetable may play an important role in underlying glycaemic control mechanisms.

Abstract Six patients received platelet concentrate transfusions from their HLA- identical siblings. Platelet concentrates were administered either fresh, or after being frozen in 10% dimethylsulfoxide, at a slow controlled rate (1 degree C/min) or rapidly (approximately 8 degrees C/min) in the vapor-phase of a liquid nitrogen refrigerator. The median freeze-thaw loss was 13.5%. The mean 1-hr and 20-hr corrected increments in platelet count were calculated for fresh platelet concentrates transfused before and after transfusion with controlled- rate frozen and vapor-phase frozen platelet concentrates. There was no significant difference among the first and second transfusion of fresh platelet concentrates, nor was the difference observed between fresh and controlled-rate frozen platelet concentrates significant. The difference between fresh and vapor-phase frozen platelet concentrates, and between controlled-rate frozen and vapor-phase frozen platelet concentrates were highly significant (p < 0.01). In vitro tests of aggregation using ristocetin and platelet ultrastructural studies paralleled the transfusion experience. Our results indicate that HLA- identical platelet concentrates can be successfully frozen and thawed for transfusion if a slow, controlled rate of freezing is employed. The use of HLA-identical frozen platelet concentrates may be important in emergency situations for the refractory patient and potentially for the establishment of a platelet concentrate bank.

Background: cattle tick Rhipicephalus (Boophilus) microplus poses serious problems for farmers in Brazil, especially because the parasite easily develops resistance to pesticide agents. For this reason, together with other factors including environmental, human and animal contamination and costs, alternative approaches have been sought for. Aims: this study sough to evaluate the efficiency of a tick biotherapic on tick-infested cows. Methods: 34 dairy Dutch cows were divided in 2 groups: one group received 100g/day of mineral salt supplement impregnated with tick biotherapic 12cH for 6 months, and then in alternate days with tick biotherapic 30cH to complete 28 months of treatment; the other group (control) received only the mineral salt supplement. After 28 months of treatment, engorged Rhipicephalus (boophilus) microplus females were collected in both groups, counted and weighed; in vitro tests were carried out to assess mass of ticks; egg mass; egg-hatching rate; and reproductive efficiency. Results: There was significant difference between both groups for all parameters evaluated; tick-mass (p = 0.0008); egg mass (p=0.0044); egg-hatching rate (p= 0.0017); and reproductive efficiency (p = 0.0044). Conclusion: treatment with tick biotherapic significantly decreased the mass of engorged females, deposition and hatching rate of eggs, resulting consequently in the decrease of the reproductive efficiency of ticks.

In this study, Licochalcone A (LicoA) was investigated in in vitro and in vivo assays. The survival of worms in culture, the pattern of oviposition, the count of intact tubers and the integrity of the coat were adopted in the in vitro tests. After the animals were perfused, the number of worms recovered, their location and the oogram study were the parameters analyzed to signal the existence of potential schistosomicidal activity in vivo. We observed reduction on the survival, integument integrity and reproduction of adult worms in vitro. Murine models did not show a significant difference in the parasitological parameters analyzed that indicate activity against the worms with an oral single dose of 25 mg/ kg of LicoA or two intraperitoneal injection of 50 mg/ kg LicoA. Nevertheless, it is too early to completely exclude the schistosomicide activity of LicoA, considering that the used dosage form could not provide a regular absorption of the drug.

Atherosclerosis is a chronic inflammatory disease, whose progression and stability are modulated, among other factors, by an innate and adaptive immune response. Prodiginines are bacterial secondary metabolites with antiproliferative and immunomodulatory activities; however, their effect on the progression or vulnerability of atheromatous plaque has not been evaluated. This study assessed the therapeutic potential of prodigiosin and undecylprodigiosin on inflammatory marker expression and atherosclerosis. An in vitro and in vivo study was carried out. Migration, low-density lipoprotein (LDL) uptake and angiogenesis assays were performed on cell types involved in the pathophysiology of atherosclerosis. In addition, male LDL receptor null (Ldlr-/-) C57BL/6J mice were treated with prodigiosin or undecylprodigiosin for 28 days. Morphometric analysis of atherosclerotic plaques, gene expression of atherogenic factors in the aortic sinus and serum cytokine quantification were performed. The treatments applied had slight effects on the in vitro tests performed, highlighting the inhibitory effect on the migration of SMCs (smooth muscle cells). On the other hand, although no significant difference in atherosclerotic plaque progression was observed, gene expression of IL-4 and chemokine (C-C motif) ligand 2 (Ccl2) was downregulated. In addition, 50 µg/Kg/day of both treatments was sufficient to inhibit circulating tumor necrosis factor alpha (TNF-α), interleukin-2 (IL-2) and interferon-gamma (IFN-γ) in serum. These results suggested that prodigiosin and undecylprodigiosin modulated inflammatory markers and could have an impact in reducing atherosclerotic plaque vulnerability.

Forty-three clinical isolates of Sporothrix schenckii derived from humans and animals were evaluated in vitro for their susceptibility to amphotericin B, itraconazole, and terbinafine. MICs were determined by the method of micro dilution in liquid media, using protocols M27-A2 for the yeast form and M38-A for the mycelial form, both standardized by the Clinical Laboratory Standards Institute. In general, higher MICs were found for the mycelial form (intervals of up to two dilutions). In the case of amphotericin B, a significant difference in activity was observed, with higher values (p < 0.05) found for the mycelial form. MICs for itraconazole and terbinafine were similar for both yeast and mycelial forms but slightly higher for mycelia. Although data presented here indicate different levels of susceptibility when both growth forms were compared, indicating an intrinsic difference between them, it is still difficult to draw a consensus as to which form correlates better with clinical findings. More studies are necessary to determine the criteria for in vitro tests that will lead to efficient therapeutic choices.Key words: Sporothrix schenckii, susceptibility testing, antifungal drug.

AbstractThe distal femur is the predominant site for benign bone tumours and a common site for fracture following tumour removal or cementation. However, the lack of conclusive assessment criterion for post-operative fracture risk and appropriate devices for cement augmentation are serious concerns. Hence, a validated biomechanical tool was developed to assess bone strength, depending on the size and location of artificially created tumorous defects in the distal femora. The mechanics of the bone–cement interface was investigated to determine the main causes of reconstruction failure. Based on quantitative-CT images, non-linear and heterogeneous finite element (FE) models of human cadaveric distal femora with simulated tumourous defects were created and validated using in vitro mechanical tests from 14 cadaveric samples. Statistical analyses demonstrated a strong linear relationship (R2 = 0.95, slope = 1.12) with no significant difference between bone strengths predicted by in silico analyses and in vitro tests (P = 0.174). FE analyses showed little reduction in bone strength until the defect was 35% or more of epiphyseal volume, and reduction in bone strength was less pronounced for laterally located defects than medial side defects. Moreover, the proximal end of the cortical window and the most interior wall of the bone–cement interface were the most vulnerable sites for reconstruction failure.

Abstract This study was carried out to determine the phenolic contents as well as to evaluate the antioxidant and antimicrobial activities of Genista microcephala. Total phenolic and flavonoid compounds were quantified. Antioxidant activity was evaluated by various in vitro tests, including Ferric Ion Reducing Activity (FRP), Cuprac Ion Reducing Antioxidant Capacity (CUPRAC), and total antioxidant capacities (TAC). The antimicrobial activity of ethyl acetate and butanolic extracts of G. microcephala in addition of fatty acids content of different parts of the studied plant was evaluated against human pathogenic bacteria, fish bacteria and two yeast. The used methods were agar diffusion method and the determination of minimal bactericidal and fungicidal concentrations (MBC and MFC). No significant difference was found between the two used solvents on total phenolic content (TPC). While, difference in total flavonoid content (TFC) were found to be significant. Ethylene acetate extract was the most actif regarding antioxidant assay. The tested activity seems to be due to the high content of this extract on isoflavonoids. The tested extracts demonstrated the lowest inhibition concentration for B.cereus RSKK 863.

Background and aims. The aim of this study was to compare the biocompatibility of calcium-enriched mixture cement (CEM), composite resin and nano-particled mineral trioxide aggregate (NP-MTA) using human gingival fibroblasts. Materials and methods. A comparative in vitro cell culture study was carried out using 60 single-rooted teeth which were assigned to the following four groups: 1) untreated healthy group (control); 2) restored with composite resin; 3) CEM cement; 4) NP-MTA. The MTT assay was used to measure the viability of fibroblasts attached to each specimen and scanning electron microscopy (SEM) was used for describing cell morphology. Results. After 24 hours of incubation, the survival rates for composite resin and NP-MTA were 74.1% and 76.9%, respectively, which were significantly lower than the value in the control group, while both were equally biocompatible. No statistically significant difference was found between the control group and CEM cement samples (94.3%). After 3 days of incubation, some increases in the viability of fibroblasts were detected in the composite resin and NP-MTA groups, with their survival rates being 89% and 93%, respectively. Conversely, in the CEM cement group, the survival rate decreased to 80.7%, which was significantly lower than that in the control group (P = 0.0001). Conclusion. The results of in vitro tests indicated that on days 1, 3 and 5 after incubation, composite resin, CEM cement and NP-MTA exhibited acceptable biocompatibility, provided they were allowed to set for 24 hours before exposure to the cells.
