This work investigated the preparation, characterization, antioxidant, and anti-inflammation capacities of Flammulina velutipes polyphenols (FVP) and fermented FVP (FFVP). The results revealed that the new syringic acid, accounting for 22.22%, was obtained after fermentation (FFVP). FFVP exhibits higher antioxidant and anti-inflammation activities than FVP, enhancing cell viability and phagocytosis, inhibiting the secretion of NO and ROS, and reducing the inflammatory response of RAW264.7 cells. This study revealed that FFVP provides a theoretical reference for in-depth study of its regulatory mechanisms and further development of functional antioxidants that are applicable in the food and health industry.

AbstractCirculating miR-150-5p has been identified as a prognostic marker in patients with critical illness and sepsis. Herein, we aimed to further explore the role and underlying mechanism of miR-150-5p in sepsis. Quantitative real-time-PCR assay was performed to detect the expression of miR-150-5p upon stimulation with lipopolysaccharide (LPS) in RAW264.7 cells. The levels of tumor necrosis factor-α, interleukin (IL)-6 and IL-1β were measured by ELISA assay. Cell apoptosis was determined using flow cytometry. Western blot was used to assess notch receptor 1 (Notch1) expression in LPS-induced RAW264.7 cells. Dual-luciferase reporter assay was employed to validate the target of miR-150-5p. Our data showed that miR-150-5p was downregulated and Notch1 was upregulated in LPS-stimulated RAW264.7 cells. miR-150-5p overexpression or Notch1 silencing alleviated LPS-induced inflammatory response and apoptosis in RAW264.7 cells. Moreover, Notch1 was a direct target of miR-150-5p. Notch1 abated miR-150-5p-mediated anti-inflammation and anti-apoptosis in LPS-induced RAW264.7 cells. miR-150-5p alleviated LPS-induced inflammatory response and apoptosis at least partly by targeting Notch1 in RAW264.7 cells, highlighting miR-150-5p as a target in the development of anti-inflammation and anti-apoptosis drugs for sepsis treatment.

AbstractMulberry leaves are used in traditional Chinese medicine and contain numerous active substances that are known to be beneficial for human health. The aim of this study was to investigate the phenolic compositions and antioxidant activities of the leaves from 23 mulberry cultivars. Qualitative LC-ESI-QTOF analysis revealed the presence of 11 phenolic compounds in the free phenolic extracts and 10 phenolic compounds in the bound fractions. Chlorogenic acid and caffeic acid were the major components in the free and bound fractions, respectively. The results revealed that the changguosang cultivar from Taiwan contained the greatest content of phenolic compounds as well as the highest antioxidant activity among the 23 cultivars examined, as determined using three separate antioxidant assays. The isoquercitrin, chlorogenic acid, and rutin contents of the free phenolic extracts displayed significant correlations with the antioxidant activities, while syringic acid and rutin were the main contributors to the antioxidant activities of the bound phenolic fractions. The obtained results demonstrate that mulberry leaves contain a variety of beneficial phenolic substances and may be suitable for further development as a herbal medicine.

The purpose of the article is to examine in detail the features of the historical fate of K. S. Stanislavsky's doctrine of the supertask, which is considered the cornerstone of his system, and to find out the reasons for the tendentiously limited interpretation of the term “supertask” in the research literature of the second half of the last century, which still remains unconsidered. The methodology of the research is the general logical method of cognition, which involves an analysis of the features of the historical era of the emergence of the doctrine of the super-task and specific measures for its dissemination among art workers. This method makes it possible to determine the causes of deformation of the defining term of the system of K. Stanislavsky. The method of comparison is also used as a cognitive operation, which makes it possible to clearly understand the primary content of the concept of "super task" and its differences from the interpretations of many other researchers of the theoretical heritage of K. S. Stanislavsky – those authors whose interpretation is based on the judgment about the similarity or difference of objects. Scientific novelty. An attempt is made to look at the fate of the Stanislavsky system as a whole and the doctrine of the super-task, in particular, from the standpoint of historical truth, in contrast to the standpoint of myth-making, which was characteristic of researchers of the Soviet era. The novelty also lies in the fact that for the first time in the theory of acting, an attempt was made to compare the true meaning put by the author of the system into the very term of the supertask itself, with the way it was interpreted by theoretical thought in the Soviet period in accordance with the ideological needs of the then society. Conclusions. As a result of a comparative analysis of the texts of K. Stanislavsky, dedicated to the disclosure of the content of the doctrine of the super-task with the formulations that tried to reveal its content in the period after the death of the author of the system, it becomes obvious that the true meaning of the doctrine has suffered a rather significant semantic deformation. This actualizes the need for a detailed and in-depth study of all the materials of his heritage, which are related to the disclosure of the content of the doctrine of the supertask, which will contribute to the further development of the system at the present stage.

Recent literature has highlighted the importance of chronic inflammation in psoriasis pathogenesis. Non-resolving inflammation can trigger progressive tissue damage and inflammatory mediator release which in turn perpetuate the inflammatory cycle. Under normal conditions, inflammatory responses are tightly controlled through several mechanisms that restore normal tissue function and structure. Defects in regulatory mechanisms of the inflammatory response can result in persistent unresolved inflammation and further increases of inflammation. Therefore, this review focuses on defects in regulatory mechanisms of inflammatory responses that lead to uncontrolled chronic inflammation in psoriasis. Databases such as Pubmed Embase, ISI, and Iranian databases including Iranmedex, and SID were researched to identify relevant literature. The results of this review indicate that dysregulation of the inflammatory response may be a likely cause of various immune-mediated inflammatory disorders such as psoriasis. Based on current findings, advances in understanding the cellular and molecular mechanisms involved in inflammation resolution are not only improving our knowledge of the pathogenesis of chronic inflammatory diseases but also supporting the development of new therapeutic strategies.

Purple sweet potato anthocyanins are kinds of natural anthocyanin red pigments extracted from the root or stem of purple sweet potato. They are stable and have the functions of anti-oxidation, anti-mutation, anti-tumor, liver protection, hypoglycemia, and anti-inflammation, which confer them a good application prospect. Nevertheless, there is not a comprehensive review of purple sweet potato anthocyanins so far. The extraction, structural characterization, stability, functional activity, application in the food, cosmetics, medicine, and other industries of anthocyanins from purple sweet potato, together with their biotransformation in vitro or by gut microorganism are reviewed in this paper, which provides a reference for further development and utilization of anthocyanins.

Apios americana Medikus was once widely accepted as staple food in India for a long time, and the tuber of which possesses high nutrients. During the past decades, most of the research has focused on the biological activity in the tubers of Apios americana Medikus whereas the leaves were ignored. In this study, the Apios americana Medikus leaf extract (ALE) was obtained and seven compounds were identified. LPS-induced RAW264.7 cells were used to study the anti-inflammation activity of ALE. As expected, ALE reduced the secretion of nitric oxide (NO) and inflammatory cytokines via inhibition of NF-κB and MAPK signaling together with activation of Nrf2-Keap1 and FOXO pathways, as well as alleviating the oxidative stress and mitochondrial dysfunction. In addition, ALE could activate HMGB1-Beclin1 and Sirt1-FoxO1 pathways and inhibit the Akt-mTOR signaling pathway to activate autophagy, protecting RAW264.7 cells from inflammation. In summary, our results suggested that ALE might help activate the anti-inflammation system, resulting in the prevention of LPS-induced damage in RAW264.7 cells.
