Elderberries, sea buckthorn, and sloe berries are fruits of wild-grown bushes, valued in folk medicine for their health-promoting properties but still rarely applied in food. The aim of the present study was to produce probiotic yoghurts with a 10% addition of sweetened purees prepared from elderberries (EPY), sea buckthorn (SBPY), and sloe berries (SPY) and to assess their chemical composition, acidity, content of polyphenols and anthocyanins, ferric reducing antioxidant power (FRAP) and antiradical power (ARP), level of starter microbiota, concentration of acetaldehyde and diacetyl, syneresis, instrumentally measured color and texture parameters, and sensory acceptance. The results were compared to those obtained for plain probiotic yoghurt (PPY) and the changes tracked during 1 month of cold storage at 2 week intervals. The addition of elderberry and sloe berries significantly increased the antioxidant capacity of probiotic yoghurts, probably due to a high content of polyphenols, especially anthocyanins. However, anthocyanins were more stable in the EPY when compared to the SPY. All yoghurt treatments were characterized by good sensory quality and viability of starter microorganisms, including probiotic strains during cold storage. Elderberries promoted the evolution of diacetyl in yoghurts during storage and, together with sloe berries, produced increased syneresis and the greatest changes in color profile compared to PPY.

Plectranthus neochilus é uma erva aromática conhecida popularmente como boldo-gambá, usada na medicina popular para tratar dispepsia e insuficiência hepática, usada também com efeito analgésico e anti-inflamatório. Tendo em vista o uso popular da planta, torna-se importante a investigação farmacológica dos compostos isolados de P.neochilus. A avaliação da Citotoxicidade em plantas é de extrema importância para posterior utilização em medicamentos. O objetivo deste trabalho é avaliar concentrações não citotóxicas de 1,6-di-O-acetil-9-deoxiforscolina um composto isolado análogo a forscolina oriundo do extrato hidroalcoólico das folhas e caules de P.neochilus e avaliar se este diterpeno possuicapacidade antioxidante. Para o teste de citotoxicidade foi utilizado o ensaio de viabilidade com sangue total, foi feita a coleta de sangue de 5 voluntários hígidos, e confeccionadas 6 culturas, sendo a primeira controle a segunda um controle de DMSO, que foi utilizado como solvente para o diterpeno e 4 culturas em concentrações diferentes do diterpeno, 40, 20, 10 e 5μg/ml, seguiu-se o protocolo de lise de hemácias com cloreto de amônio e marcou-se com azul de tripan, posteriormente, foi feita a leitura por citometria de fluxo. Também foi analisada a capacidadeantioxidante do diterpeno por meio da determinação do poder de redução do íon ferro, FRAP (do inglês Ferric Reducing Antioxidant Power) em que é analisada a produção do íon Fe2+ a partir da redução do íon Fe3+ presente no complexo 2,4,6-tripiridil-s-triazina (TPTZ). Quando o ocorre a redução, há alteração de cor na solução, a avaliação é realizada por espectrofotômetro em comprimento de onda de 593 nm. Os resultados demonstraram que nas concentrações testadas, 40, 20, 10 e 5 μg/ml o diterpeno extraído da planta P. neochilus não apresentou citotoxicidade seletiva a nenhuma população leucocitária avaliada (linfócitos, monócitos e neutrófilos) e ainda apresentou grande capacidadeantioxidante. Mais estudos devem ser realizados evidenciando os potenciais farmacológicos da planta P. neochilus, bem como de substâncias isoladas a partir de extratos da planta.Palavras-chave: Diterepeno. The evaluation of plant cytotoxicity is of extreme importance for subsequent use in medicinal products. The objective of this study is to evaluate non-cytotoxic concentrations of 1,6-di-O-acetyl-9-deoxyphosphine an isolated compound analogous to forskolin from the hydroalcoholic extract of P.neochilus leaves and stems and to evaluate whether this diterpene has antioxidant capacity. For the cytotoxicity test, the whole blood viability assay was performed, blood was collected from 5 healthy volunteers, and 6 cultures were made, the second control being a DMSO control, which is the diterpene solvent and 4 cultures at different concentrations of diterpene, 40, 20, 10 and 5 μg / ml, followed by the protocol of red blood cell lysis with ammonium chloride and labeled with tripan blue, and then read by flow cytometry. The antioxidant capacity of diterpene was also analyzed by means of the determination of the iron ion reductionpower (FRAP) in which Fe2 + ion production is analyzed from the Fe3 + ion present in complex 2, 4,6-tripyridyl-s-triazine (TPTZ). When the reduction occurs there is a color change in the solution, turning to an intense purple, the evaluation is performed by spectrophotometer at wavelength of 593 nm. The results demonstrated that at the tested concentrations, 40, 20, 10 and 5 μg / ml diterpene extracted from the plant P. neochilus did not present selective cytotoxicity to any leukocyte population evaluated (lymphocytes, monocytes and neutrophils) andstill presented a great antioxidant capacity. Further studies should be performed evidencing the pharmacological potentials of the plant P. neochilus, as well as of substances isolated from extracts of the plant.Keywords: Diterpene, cytotoxicity, antioxidant .

Abstract All parts of the sea buckthorn (Hippophae rhamnoides L.) plant are considered to contain large amounts of compounds that are believed to have beneficial health effects. Till now, different parts of sea buckthorn plant have been used for the treatment of diseases in traditional medicine in various countries. Nevertheless, sea buckthorn parts would be a good raw material not only for medicinal properties but also for food products with functional properties; therefore, the aim of the research was to determine the concentration of different phenolic compounds and antioxidant activity in various sea buckthorn parts. The study was conducted on parts of female and male sea buckthorn bushes. Phenolic compounds (total phenols, total flavonoids and condensed tannins) and antioxidative activity (ferric reducing antioxidant power (FRAP) free radical scavenging activity (using 2.2-diphenyl-1-picrylhydrazyl, DPPH)) in ethanolic extracts of leaves, shoots, flowers, and berries were determined using various spectrophotometric methods. The study showed that concentration of phenolic compounds differed among parts of sea buckthorn plant and among gender. Leaves of female plants proved to be the most valuable, as they contained the highest total phenol concentration (165.76 mg/g) and antioxidant activity (220.97 mg/g for FRAP and 43.76 mg/g for DPPH), while lowest values were found in young shoots of male plants (84.94 mg/g, 94.24 mg/g and 24.63 mg/g, respectively). The significant differences in chemical composition and biological activity of sea buckthorn leaves, shoots, berries, and buds indicate a need for detailed studies of their extracts, specific fractions and compounds during a whole vegetative season.

The present study is on Acacia seyal gum (ASG), which is an exudate from Talha tree. It provides a rich source of polyphenolics compounds that are used traditionally in folk medicine. The study aims to determine the antioxidant capacity (AC) and functional groups of ASG and Prebio-T-commercial (PTC) samples. The methanol crude extracts of both ASG and PTC have fractioned into chloroform (CHF), hexane (HF), acetone (AF) and methanol (MF) using solvent-solvent portion. Both ferric reducing antioxidant power (FRAP), and cupric reducing antioxidant capacity (CUPRAC) assays for each fraction examined. Crude methanol extracts (CME) and its active compositions also analysed carefully using Fourier Transform Infrared Spectroscopy (FTIR) technique. The findings presented a wide variety of functional groups provided by the FTIR spectra (eights bands approximately. Regarding cupric reducing antioxidant capacity (CUPRAC), the methanol crude extracts values are 888.6�4.57 mg TE/100g extract, for PTC as compared to 474.3� 2.23 mg TE/100g of extract for ASG. However, both methanol and acetone fractions revealed significantly (p = 0.05) high FRAP values ranged between 599.8�7.5 and 741.8�5.8 mg TE/100g fraction; for PTC and ASG, respectively. While CUPRAC showed insignificant (p = 0.05) same values 356.1�2.62 mg TE/100g of fraction; for MF of both PTC and ASG respectively. Therefore, in this study, methanolic fractions (MFs) are found to be more effective than acetone fractions (AFs), except for CHF and HF. Finally, the antioxidant activity of the active fraction has provided some evidence regarding its functional groups which may have used in traditional medicine.

The genus Minthostachys belonging to the Lamiaceae family, and is an important South American mint genus used commonly in folk medicine as an aroma in cooking. The phytochemical-rich samples of the aerial parts of Minthostachys diffusa Epling. were tested for pharmacological and health-promoting bioactivities using in vitro chemical and enzymatic assays. A range of radical scavenging activities of the samples against biological radicals such as nitric oxide and superoxide anion and against synthetic 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radicals, the ferric reducing antioxidant power and the lipid peroxidation inhibition were determined and ranked using the ‘relative antioxidant capacity index’ (RACI). The ethyl acetate fraction showed the highest RACI of +1.12. Analysis of the various fractions’ inhibitory ability against enzymes involved in diabetes (α-amylase and α-glucosidase), and against enzymes associated with Parkinson’s or Alzheimer’s diseases (acetylcholinesterase and butyrylcholinesterase) also suggested that the ethyl acetate fraction was the most active. Liquid chromatography–tandem mass spectrometry analysis of the ethyl acetate fraction showed more than 30 polyphenolic compounds, including triterpenes. The inhibitory cholinesterase effects of the triterpenes identified from M. diffusa were further analysed by in silico docking of these compounds into 3D-structures of acetylcholinesterase and butyrylcholinesterase. This is the first study on pharmacological activities and phytochemical profiling of the aerial parts of M. diffusa, showing that this plant, normally used as food in South America, is also rich in health-promoting phytochemicals.

The study investigated the impact of the fermentation process on the phenolic contents and antioxidant and anti-inflammatory activities in extracts of Miang, an ethnic fermented tea product of northern Thailand. The acetone (80%) extraction of Miang samples fermented by a non-filamentous fungi-based process (NFP) and filamentous fungi-based process (FFP) had elevated levels of total polyphenols, total tannins, and condensed tannins compared to young and mature tea leaves. The antioxidant studies also showed better the half-maximal inhibitory concentration (IC50) values for fermented leaves in both 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity assays as well as improved ferric reducing antioxidant power (FRAP) compared to young and mature tea leaves. Extracts of NFP and FFP samples at concentrations of 50 and 100 ppm showed better protective effects against hydrogen peroxide (H2O2)-induced intracellular reactive oxygen species (ROS) production in HT-29 colorectal cells without exerting cytotoxicity. Additionally, lipopolysaccharide (LPS)-induced production of nitric oxide (a proinflammatory mediator as well as a reactive nitrogen species) was also inhibited by these fermented Miang extracts with an IC50 values of 17.15 μg/mL (NFP), 20.17 μg/mL (FFP), 33.96 μg/mL (young tea leaves), and 31.33 μg/mL (mature tea leaves). Therefore, both NFP-Miang and FFP-Miang showed the potential to be targeted as natural bioactive functional ingredients with preventive properties against free radical and inflammatory-mediated diseases.

Tropical black bolete, Phlebopus portentosus, provides various nutritional benefits and natural antioxidants to humans. In this study, the chemical composition, phenolic compounds, and antioxidant and antimicrobial activities of fresh mushroom samples and samples stored for a period of one year using different preservation methods (drying, brining, and frozen) were investigated. The results indicated that the brining method significantly reduced the protein and fat contents of the mushrooms. The polyphenol and flavonoid contents of the frozen sample were not significantly different from that of the fresh sample. The results revealed that an inhibition value of 50% (IC50) for the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay of the extract of the dried and frozen samples was not statistically different from that of the fresh sample. The IC50 value of 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assay and ferric reducing antioxidant power (FRAP) value in the extract of the frozen sample were not found to be significantly different from those of the fresh sample. Furthermore, the lowest degree of antioxidant activity was found in the extract of the brined sample. Additionally, the antimicrobial activities of the extracts of the fresh and frozen samples were not significantly different and both extracts could have inhibited the growth of all tested Gram-positive bacteria and Pseudomonas aeruginosa.

The study investigated the phenols, sugar and the antioxidant capacities of date fruit extracts obtained by organic solvents and by hydrothermal treatment from six different Algerian cultivars at two ripening stages for the first time. The analyzed cultivars exhibited potent antioxidant properties (ferric reducing antioxidant power (FRAP), 1,1-Diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) scavenging capacities) and different phenols regardless of the solvents and the maturity stages. About 18 phenols were identified and quantified, mainly in the hydrothermal extracts. The earlier stages were characterized by high amounts of o-coumaric acid, cinnamic acid and luteolin, with a noticeable absence of quercetin. The tamr stage presented the highest sugar content (78.15–86.85 mg/100 mg dry weight (DW)) with an abundance of glucose. Galactose was present only in some cultivars from the kimri stage (tamjouhert). Uronic acids were mostly detected at the tamr stage (4.02–8.82 mg gallic acid equivalent/100 mg dried weight). The obtained results highlight the potential of using date fruit extracts as natural antioxidants, especially at industrial scales that tend use hydrothermal extraction.

AbstractThis study aimed to investigate the nutricosmetic effect of Asparagus officinalis extracts. The tip and spear of A. officinalis were successively extracted with 95% ethanol. The rutin, phenolic, and flavonoid contents of A. officinalis extracts were investigated. The antioxidant activities were determined by 2,2-azinobis (3-ethylbenzothiazoline-6-sulphonic acid) and a ferric reducing antioxidant power assay. Matrix metalloproteinase-1 (MMP-1), elastase, and hyaluronidase inhibition were determined by in vitro enzyme reaction assay. The cytotoxicity was analyzed on peripheral blood mononuclear cellss. Findings revealed that drying temperature and drying duration had significant effects on the chemical composition and biological activity of A. officinalis extract. A. officinalis tips dried at 50 °C for 24 h contained the (significantly) highest flavonoid and rutin content. The most potent extract was from A. officinalis spears since it possessed the (significantly) highest MMP-1, elastase, and hyaluronidase inhibition rates of 83.4 ± 1.5%, 70.4 ± 4.1%, and 75.2 ± 1.0%, respectively. Interestingly, at the same concentration, the A. officinalis spear extract was more potent in MMP-1 inhibition than oleanolic acid and epigallocatechin gallate, the well-known natural MMP-1 inhibitors. The results show that A. officinalis extract is an attractive source of natural anti-skin-wrinkle ingredients.

Abstract Background The main aim of this work was to synthesise a novel N-(substituted phenyl)-2-(3-(hydroxyimino) methyl)-1H-indol-1-yl) acetamide derivatives and evaluate their antioxidant activity. These compounds were prepared by a condensation reaction between 1H-indole carbaldehyde oxime and 2-chloro acetamide derivatives. The newly synthesised compound structures were characterised by FT-IR, 1H-NMR, mass spectroscopy and elemental analysis. Furthermore, the above-mentioned compounds were screened for antioxidant activity by using ferric reducing antioxidant power (FRAP) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) methods. Result The antioxidant activity result reveals that most of the compounds were exhibiting considerable activity in both methods and the values are very closer to the standards. Among the synthesised compounds, compound 3j, 3a and 3k were shown remarkable activity at low concentration. Conclusion Compounds 3j, 3a and 3k were shown highest activity among the prepared analogues due to the attachment of halogens connected at the appropriate place in the phenyl ring. Hence, these substituted phenyl rings considered as a perfect side chain for the indole nucleus for the development of the new antioxidant agents.
