Fruits contain a number of useful substances including antioxidants. Their bio-accessibility after passing through the digestive tract is of primary importance when considering their benefits. In this respect, we investigated the effect of in vitro digestion on the phytochemicals of eight fruit juices. Freshly prepared juices from pomegranate, orange and grapefruit were used as well as commercially available juices from cherry, black grapes and aloe vera, blackberry and chokeberry, and two types of chokeberry and raspberries. Spectrophotometric and HPLC methods were used in order to analyse the sugar content, the total phenolic (TPC) and flavonoid contents (TFC), anthocyanins, phenolic acids and antioxidant activity. Principle component analysis was used to explain the differentiation among the types of fruit juice. Sugar recovery variation was between 4–41%. The bio-accessibility of TPC ranged from 13.52–26.49% and of flavonoids between 24.25–67.00%. The pomegranate juice and the juice of black grapes and aloe vera kept 58.12 and 50.36% of their initial anthocyanins content, while for the other samples less than 1.10% was established. As a result, a maximum of 30% remaining antioxidant activity was measured for some of the samples, but for most this was less than 10%. In conclusion, fruit juices are a rich source of biologically active substances, but a more detailed analysis of food transformation during digestion is needed.

Nowadays, there is a growing interest for foods with a lower sugar content and rich in fiber and biologically active substances. The main purpose of this study was to prepare flakes from non-traditional pigmented cereals (Oryza sativa, Chenopodium quinoa, and Eragrostis tef) and to analyze their fibre, sugar, and in vitro digestibility values. Regarding phenolic antioxidants (soluble, soluble conjugated, and insoluble bound fractions), their content and antioxidant activity were measured using spectrophotometry and high performance liquid chromatography (HPLC) methods. Hydrothermally treated grains resulted in flakes with higher total dietary fibre contents (11.1–24.4%), quinoa and teff flakes were rich in maltose (up to 42.0 mg/g). Non-traditional flakes had lower in vitro digestibility, but conversely, they exhibited the highest phenolic contents corresponding with the highest antioxidant activity values (up to 2.33 mg Gallic acid equivalent/g of total phenolic content and 1.59 mg Trolox equivalent/g for 2,2-diphenyl-1-picrylhydrazyl (DPPH) in case of brown teff). Among free phenolics, the main contributors to an antioxidant activity were p-coumaric, o-coumaric, and gallic acids (r > 0.8186); among the soluble conjugated fractions, they were epigallocatechin, epicatechin, caffeic, and vanillic acids (r > 0.5935); while caffeic, protocatechuic, and ferulic acids (r > 0.5751) were the main contributors among the insoluble bound phenolics.

Belimbing wuluh merupakan tanaman jenis buah dan obat tradisional. Tanaman belimbing wuluh sudah sering dimanfaatkan masyarakat salah satunya untuk mengobati penyakit seperti batuk dan radang rektum. Tujuan penelitian ini adalah menguji senyawa fitokimia, menganalisis kandungan total fenolik dan flavonoid, serta aktivitas antioksidan dan antiinflamasi secara in vitro pada ekstrak etanol daun belimbing wuluh. Metode yang dilakukan pada penelitian ini adalah ekstraksi daun belimbing wuluh, skrining fitokimia, perhitungan total fenol dan flavonoid, uji aktivitas antioksidan, dan uji aktivitas antiinflamasi. Senyawa fitokimia yang terkandung pada ekstrak etanol daun belimbing wuluh adalah saponin, tanin, steroid, flavonoid, dan alkaloid. Kandungan total fenolik dan flavonoid ekstrak etanol daun belimbing wuluh secara berturut-turut sebesar 39,03 dan 97,28 µg QE/mg. Ekstrak etanol daun belimbing wuluh memiliki aktivitas antioksidan yang tergolong sangat kuat, sementara aktivitas antiinflamasinya terutama ditunjukkan pada konsentrasi ekstrak 200 µg/ ml, memiliki nilai persen inhibisi hemolisis yang paling tinggi. Kesimpulannya, ekstrak etanol daun belimbing wuluh dapat menjadi salah satu sumber antioksidan dan antiinflamasi alami.Ethanol Extracts of Averrhoa Bilimbi Leaf Demonstrated Antioxidative and Anti-inflammatory ActivityAbstractAverrhoa bilimbi, a fruit-bearing plant, has been traditionally used for medicinal purposes such as treatment of cough and rectal inflammation. In this current work, ethanol extract of Averrhoa bilimbi leaf was analyzed, with regard to phytochemical composition, i.e. total phenolic, and flavonoid, antioxidant activity, and in vitro antiinflammation activity. Extraction of carambola leaves, phytochemical screening, total phenolic and flavonoid contens, assay of antioxidant activity, and assay of anti-inflammatory activity were done in this research. As a result, the leaf extract positively contained some phytochemical compounds, i.e. saponin, tannin, steroid, flavonoid, and alkaloid. Furthermore, total phenolic and flavonoid of the leaf extract was found at 39.03 and 97.28 µg QE/mg extract, respectively. Additionally, antioxidant activity of the leaf extract was classified as very strong, while its anti-inflammatory feature at extract concentration of 200 µg/ml exhibited the highest inhibition of hemolysis. In summary, the ability of Averrhoa bilimbi leaf ethanol extracts to act as antioxidative and anti-inflammatory agents was determined and this may open the use for natural antioxidant and anti-inflammatory agents.

Objective: The present investigation focuses on the use of Cardiospermum halicacabum L. in their phytochemical and biological activities. Methods: In this study, in vivo stem and in vitro callus ethanolic extracts of C. halicacabum were tested for their phytochemical attributes by qualitative method, Fourier transform infrared (FTIR), antioxidant, antibacterial, and bioactive compound properties. The bactericidal activity of the in vivo stem and in vitro callus extract has been evaluated in both Gram+ve and Gram-ve microorganisms using the disk diffusion method. Results: The highest frequency (78%) of well developed, dark green organogenic callus was induced from stem explant on Murashige and Skoog (MS) medium supplemented with 0.7 mg/l 2,4-Dichlorophenoxyacetic acid (2, 4-D) and 0.5 mg/l benzyl adenine (BA). The results of FTIR spectra confirmed the presence of functional groups in wild stem and in vitro callus extract of C. halicacabum with various peaks. The total phenolic content in ethanolic extract of in vivo plant and in vitro callus was 80.46 mg gallic acid equivalent (GAE)/g dry weight and 76.4 mg GAE/g dry weight, respectively. The highest percentage of tannins was measured at 78.03 in wild stem ethanol extracts followed by 75.22 in callus extract. The antioxidant activity of 2,2-diphenyl-2- picrylhydrazyl (DPPH) ethanol extract was found to be 206.54 μg/ml. IC50 values of the stem extracts of C. halicacabum are 306 μg/ml and 286 μg/ml in callus extract, respectively. Antibacterial activity of the ethanol extract was higher for Staphylococcus aureus (S. aureus) with a 17 mm zone of inhibition. Conclusion: The present investigation recommended that the callus ethanolic extract function as a good source of biologically active compounds and natural antioxidants.

The aim of this study was to investigate the effect of raspberry juice addition on the content of phenolic compounds and the antioxidant activity of wafers. The research was carried out on non-supplemented wafers (control) and wafers in which water was replaced with raspberry juice in the amount of 10%, 20%, 50%, 75% and 100%. The potential bioavailability of the phenolic compounds after in vitro digestion was also determined. As shown by the consumer assessment, wafers in which the water was replaced with 100% raspberry juice turned out to be the best variant of the enriched wafers. The content of total phenolic compounds, flavonoids, phenolic acids, and anthocyanins in the tested products increased with the increasing amount of raspberry juice added to the wafers. The fortification of the wafers with raspberry juice had a positive effect on the antioxidant activity, expressed as the ability to neutralize free radicals ABTS (2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)) and DPPH (di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium), the ability to chelate iron ions, and as the reduction power. The simulated digestion increased the content of phenolic compounds and increased the antioxidant activity of the wafers. The ability to inhibit lipoxygenase and cyclooxygenase 1 and 2 (i.e., enzymes involved in the induction of inflammation), varied and depended on both the amount of raspberry juice added and the type of extract.

Raphanus sativus (L.) var. caudatus Alef (Thai rat-tailed radish), Brassica juncea (L.) Czern. (leaf mustard) and Brassica juncea (L.) Coss. var. sareptana Sinskaja (mustard green) are cruciferous vegetable commonly consumed in Thailand and Asian countries. The vegetables were extracted with different solvents namely methanol, hexane and water prior to total phenolic content (TPC), phenethyl isothiocyanate (PEITC) content and antioxidant activity by ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay were determined. Effects of gastrointestinal digestion on stability of these characteristics were investigated. It was found that the order of extraction efficiency for high bioactive compounds and antioxidant activity was water> methanol> hexane. Among vegetables, mustard green showed the highest TPC, PEITC, FRAP and DPPH values being 19.78±0.01 g GAE, 9.65 ± 1.08 µmol, 8.18±0.01 µM FeSO4 and 7.75±0.31 µM TE per 100g, respectively. Decreases in DPPH (30.6-53.5%), FRAP (49.0-88.0%) and PEITC (27.2-56.7%) values were slightly higher than TPC (27.9-41.6%) after in vitro digestion. It can be said that Thai cruciferous vegetables contain substantial chemical property and this may promote their health protection but the stability through digestive system should be warranted.

In this study, the effect of the addition of freeze-dried raspberry pomace on the content of phenolic compounds and the antioxidant and anti-inflammatory activity of wafers was investigated. Particular attention was paid to the biological activity of the potentially bioavailable fraction of polyphenols extracted via gastro-intestinal digestion. In the basic recipe for the waffle dough, flour was replaced with freeze-dried raspberry pomace in the amount of 10%, 20%, 30%, 50%, and 75%. The content of total phenolic compounds, phenolic acids, flavonoids, and anthocyanins in ethanol and buffer extracts and after in vitro digestion increased with the increase in the addition of pomace. A similar relationship was noted for antioxidant properties: ability to neutralize ABTS—2,2′-Azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) and DPPH—1,1-diphenyl-2-picrylhydrazyl radicals, iron II chelating ability, and reduction power. The extracts obtained after the simulated digestion showed the highest activities, which confirms that the polyphenols are a potentially bioavailable fraction. Extracts from the fortified wafers effectively inhibited the activity of enzymes involved in the generation of free radicals and induction of inflammation, i.e., xanthine oxidase (XO), lipoxygenase (LOX), and cyclooxygenase 2 (COX-2). The lowest IC50 values were determined for extracts after in vitro digestion. The sensory evaluation of the prepared wafers showed that the wafers fortified with 20% pomace achieved optimal scores. Enrichment of confectionery products with waste products from the fruit and vegetable industry can be a good way to increase the proportion of biologically active polyphenols in the diet and brings benefits to the environment.

ABSTRACTCyanobacteria are natural enormous sources of various biologically active compounds with great contributions in different industries. This study aimed to introduce molecular and biochemical characterization for four novel cyanobacterial isolates obtained from Egyptian wastewater canals. Besides, In vitro biological activity of these isolates and their potential ability to take up nutrients and heavy metals from wastewater were examined. The obtained accession numbers were KY250420.1, KY321359.1, KY296359.1 and KU373076.1 for Nostoc calcicola, Leptolyngbya sp, Nostoc sp, and Nostoc sp, respectively. The isolate Leptolyngbya sp (KY321359.1) showed the lowest identity (90%) with other deposited sequences in database. While the isolate Nostoc sp (KU373076.1) showed the highest total phenolic content as well as the highest levels of caffeic, ferulic and gallic acids. Consequently, it appeared the highest antioxidant scavenging activity. All cyanobacterial isolates revealed potent ability to take up nutrients and heavy metals from wastewater. Generally, this study provides a taxonomic and molecular evidence for four novel cyanobacterial isolates with antioxidant activity and potent phycoremediation ability.

The aim of the present study was to investigate the reducing sugar, total phenolic content, and in vitro antioxidant activity of 70% ( v / v ) ethanolic extract of seven medicinal plants grown in Nepal. The reducing sugar content and total phenolic content were determined by 3,5-dinitrosalicylic acid (DNSA) and the Folin–Ciocalteu method, respectively. The in vitro antioxidant activity was evaluated using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. The reducing sugar content of the studied plant samples ranged from 6.89 ± 2.44 to 674.13 ± 2.43  mg GE/g dry extract weight and the total phenolic content ranged from 14.87 ± 0.41 to 281.71 ± 1.47  mg GAE/g dry extract weight. The reducing sugar and total phenolic content were found highest in Ficus glaberrima. Antioxidant activity was found highest in Melastoma malabathricum (IC50 value = 6.27  μg/mL), followed by F. glaberrima (IC50 value = 11.7  μg/mL). A positive and significant correlation was found between (i) total phenolic content and reducing sugar content and (ii) total phenolic content and antioxidant activity. The present study is the first study for the analysis of reducing sugar content of selected plants and for the scientific exploration of F. glaberrima. The present result suggests that the various parts of these studied plants could be assumed as a rich source of biologically active compounds and considered beneficial for the food and pharmaceutical industries.
