The purpose of this study was to better understand the chemical characteristics and chain conformation of okra polysaccharides extracted by ultrasonic-assisted extraction. A pectic-polysaccharide, named OPP-D, was obtained, which was mainly composed of rhamnose, galacturonic acid, and galactose with a molar ratio of 1.01:1.00:2.31. Combined with NMR analysis, -4)-α-d-GalAp-(1,2,4)-α-l-Rhap-(1- were identified as the backbone with galactan side chains substituted partly at O-4 of Rhap. Molecular weight and radius of gyration of OPP-D were determined as 2.19 × 105 Da and 27.0 nm, respectively. OPP-D was determined as an air-core sphere with branching chains in 0.9% NaCl solution by high-performance size-exclusion chromatography coupled with multi-angle laser light scattering and dynamic light scattering for the first time. Moreover, OPP-D exhibited typical shear-thinning behavior. In addition, OPP-D exhibited remarkable in vitro antioxidant activities and prebiotic activities, while the relatively high molecular weight, high degree of esterification, high content of uronic acids, and highly branched globular conformation of OPP-D might contribute to its in vitro anti-diabetic activities and binding capacities. Results can contribute to a better understanding of the structure–bioactivity relationship of OPPs, and OPP-D has great potential applications in the functional food and pharmaceutical industries.

This paper studies the optimization of ultrasonic-assisted extraction of polysaccharides from Oenothera biennis L. leaves using Box-Behnken design, and their in vitro antioxidant activity and structure. It is demonstrated that the optimum extraction conditions are: the ultrasonic power at 400 W, solvent-tosample ratio of 23:1 (mL/g), ultrasonication temperature at 53 °C, and time for 36 min, under which the yield of polysaccharides from Oenothera biennis L. leaves, namely 3.54%, remains close to the predicted value in the model. The assays of the scavenging effect on DPPH and hydroxyl free radicals, and of the reducing power indicate that polysaccharides from Oenothera biennis L. leaves possess strong antioxidant activity and certain relationship exists between their dose and effect in the concentration range of the experiment; Fourier IR spectroscopy reveals that characteristic absorption peak of polysaccharides can be seen from the sample; the DE-52 cellulose column chromatography, HPLC profiles, and GC-MS chromatogram identify the obtained OCP-2 as heteropolysaccharides, whose major components, including arabinose, mannose, galactose, and talose, have an average molecular weight of 5435 Da.

In the present study, the crude polysaccharides were extracted from the microalgae Gracilaria lemaneiformis with ultrasonic-assisted extraction method and purified by DEAE-52 cellulose chromatography and Sephadex G-100 size-exclusion chromatography in that order. Two main fractions, GPC-1and GPC-2, were obtained through the extraction and purification steps. Then partial characterizations, such as chemical composition, ultraviolet (UV) spectrum and infrared (IR) spectrum of the two fractions were analyzed in this study. The results indicated that the total sugar contents of GPC-1and GPC-2 were 89.32% and 88.79%, and the protein contents of these two fractions were only 0.35% and 0.76%, compared with that of crude polysaccharides of 75.49% and 1.92%, respectively. Moreover, both the UV spectrum and the IR spectrum demonstrated that the two fractions were typical purified polysaccharides. As a consequence, these investigated characterizations could be considered as a favorable evidence for the identification of polysaccharides from Gracilaria lemaneiformis.

The Box-Behnken design combined with response surface method was employed to optimize ultrasonic-assisted extraction of flavonoids fromTaraxacum officinale. The optimized results showed that the highest extraction yield with ultrasonic-assisted extraction could reach 2.62% using 39.6% (v/v) ethanol and 59.5 : 1 (mL/g) liquid-solid ratio for 43.8 min. The crude extract was then purified by HPD-100 macroporous adsorption resin, and the flavonoids content in the purified extract increased to 54.7%. The antioxidant activity of the purified flavonoids was evaluatedin vitroby scavenging capacity of ABTS or DPPH,β-carotene bleaching, and FTC test. The knowledge obtained from this study should be useful to further develop and apply this plant resource.
